Thechemical composition, antioxidant and antimicrobial activities of the essential oil from aerial parts (leaves and flowers) of Chuquiraga arcuataHarling grown in the Ecuadorian Andes were studied. One hundred and twenty-six compounds were identified in the essential oil. Monoterpene hydrocarbons (45.8%) and oxygenated monoterpenes (44.1%) had the major percentages. The most abundant compounds were camphor (21.6%), myrcene (19.5%), and 1,8-cineole (13.4%). Antioxidant activity was examined using DPPH, ABTS,and FRAP assays. The essential oil had a moderate scavenging effect and reduction of ferric ion capacity through FRAP assay. Antimicrobial activity of the essential oil was observed against four pathogenic bacteria and a fungus. The essential oil exhibited activity against all microorganism strains under test, particularly against Candida albicansand Staphylococcus aureuswith MICs of 2.43-12.10 µg/mL.


Se estudió la composición química, actividades antioxidantes y antimicrobianas del aceite esencial procedente de las partes aérea (hojas y flores) de Chuquiraga arcuataHarling cultivadas en los Andes ecuatorianos. Se identificaron 126 compuestos en el aceite esencial. Los hidrocarburos monoterpénicos (45,8%) y los monoterpenos oxigenados (44,1%) tuvieron el mayor porcentaje. Los compuestos más abundantes fueron alcanfor (21,6%), mirceno (19,5%) y 1,8-cineol (13,4%). La actividadantioxidante se examinó mediante ensayos DPPH, ABTS y FRAP. El aceite esencial tuvo un efecto eliminador moderado y una reducción de la capacidad de iones férricos mediante el ensayo FRAP. Se observó actividad antimicrobiana del aceite esencial contra cuatro bacterias y un hongo patógenos. El aceite esencial mostró actividad contra todas las cepas de microorganismos bajo prueba, particularmente contra Candida albicansy Staphylococcus aureuscon CMI de 2,43-12,10 µg/mL.

Exposure to acute and chronic stress has significant effects on the basic mechanisms of associative learning and memory. Stress can both impair and enhance associative learning depending on type, intensity, and persistence of the stressor, the subject's sex, the context that the stress and behavior is experienced in, and the type of associative learning taking place. In some cases, stress can cause or exacerbate the maladaptive behavior that underlies numerous psychiatric conditions including anxiety disorders, obsessive-compulsive disorder, post-traumatic stress disorder, substance use disorder, and others. Therefore, it is critical to understand how the varied effects of stress, which may normally facilitate adaptive behavior, can also become maladaptive and even harmful. In this review, we highlight several findings of associative learning and decision-making processes that are affected by stress in both human and non-human subjects and how they are related to one another. An emerging theme from this work is that stress biases behavior towards less flexible strategies that may reflect a cautious insensitivity to changing contingencies. We consider how this inflexibility has been observed in different associative learning procedures and suggest that a goal for the field should be to clarify how factors such as sex and previous experience influence this inflexibility.

Lithium-sulfur (Li-S) batteries have attracted considerable interest as next-generation high-density energy storage devices. However, their practical applications are limited by rapid capacity fading when cycling cells with high mass loading levels. This could be largely attributed to the inferior electron/ion conduction and the severe shuttling effect of soluble polysulfide species. To address these issues, composites of sulfur/ferroelectric nanoparticles/ho ley graphene (S/FNPs/hG) cathodes were fabricated for high-mass-loading S cathodes. The solvent-free and binder-free procedure is enabled using holey graphene as a unique dry-pressable electrode for Li-S batteries. The unique structure of the holey graphene framework ensures fast electron and ion transport within the electrode and affords enough space to mitigate the electrode's volume expansion. Moreover, ferroelectric polarization due to FNPs within S/hG composites induces an internal electric field, which effectively reduces the undesired shuttling effect. With these advantages, the S/FNPs/hG composite cathodes exhibit sustainable and ultrahigh specific capacity up to 1409 mAh/gs for the S/BTO/hG cathode. A capacity retention value of 90% was obtained for the S/BNTFN/hG battery up to cycle 18. The high mass loading of sulfur ranging from 5.72 to 7.01 mgs/cm2 allows maximum high areal capacity up to ∼10 mAh/cm2 for the S/BTO/hG battery and superior rate capability at 0.2 and 0.5 mA/cm2. These results suggest sustainable and high-yielding Li-S batteries can be obtained for potential commercial applications.

OBJECTIVE: The mortality rate of schizophrenia patients is higher than that of the general population; cardiovascular disease (CVD) is their leading cause of death. This issue must be studied since people with schizophrenia are disproportionately burdened with CVD. Therefore, our goal was to identify the prevalence of CVD and other comorbidities, stratified by age and gender, in patients with schizophrenia living in Puerto Rico. METHODS: A retrospective, case-control, descriptive study was conducted. Subjects in this study were admitted to Dr. Federico Trilla's hospital from 2004 through 2014 for both psychiatric- and non psychiatric conditions. The sample populations were stratified by the confounding variables of tobacco use and alcohol abuse, and the resulting stratification was analyzed with the Cochran-Mantel-Haenszel method. RESULTS: A higher frequency of CVDs was noted in the patients with schizophrenia compared to those in the control group. Although hypertension was the most frequent pathology encountered in both groups, ischemic heart disease was approximately four times more frequent in the patients with schizophrenia. CVD represented 58.4% and 52.7% in the schizophrenia and non-schizophrenia groups, respectively, although a statistically significant difference was not observed. The prevalence of malignancies in patients without schizophrenia was higher than in patients with schizophrenia. Moreover, the prevalence of asthma was 10.9% in the control group compared to 5.3% in the schizophrenia group. CONCLUSION: These findings should motivate a systematic approach to prioritizing the aggressive management, early diagnosis, and prevention of comorbid risk factors in patients with schizophrenia.

Discrimination against lesbian, gay, bisexual, transgender, and queer (LGBTQ+) persons in health care creates barriers to serious illness care, including patients avoiding or delaying necessary care, providers disrespecting wishes of surrogates, and adverse outcomes for patients and families. A cross-sectional mixed-methods study using an online survey was used to determine the extent to which LGBTQ+ patients and spouses, partners, and widows experienced disrespectful or inadequate care due to sexual orientation or gender identity. A total of 290 LGBTQ+ patients and partners reported high levels of disrespectful and inadequate care, including 35.2% stating their provider was insensitive to them because of their identity; 30% reporting their provider was unaware of LGBTQ+ health needs; 23.1% feeling judged; 20.7% experiencing rudeness; 20.3% stating providers did not use their correct pronouns; and 19.7% reporting their treatment decisions were disregarded. Black and Hispanic patients were 2-4 times more likely than non-Hispanic White patients to report discrimination. This study demonstrated high levels of disrespectful and inadequate care towards patients and partners due to being LGBTQ+, which was especially problematic for Black and Hispanic patients and those living in politically conservative regions. Recommendations include federal and state civil rights laws to prohibit LGBTQ+ discrimination and institutional practices to address discrimination, including cultural sensitivity training for staff.

OBJECTIVES: Pulmonary disease is a significant cause of morbidity and mortality in adults and children, but most of the world lacks diagnostic imaging for its assessment. Lung ultrasound is a portable, low-cost, and highly accurate imaging modality for assessment of pulmonary pathology including pneumonia, but its deployment is limited secondary to a lack of trained sonographers. In this study, we piloted a low-cost lung teleultrasound system in rural Peru during the COVID-19 pandemic using lung ultrasound volume sweep imaging (VSI) that can be operated by an individual without prior ultrasound training circumventing many obstacles to ultrasound deployment. DESIGN: Pilot study. SETTING: Study activities took place in five health centres in rural Peru. PARTICIPANTS: There were 213 participants presenting to rural health clinics. INTERVENTIONS: Individuals without prior ultrasound experience in rural Peru underwent brief training on how to use the teleultrasound system and perform lung ultrasound VSI. Subsequently, patients attending clinic were scanned by these previously ultrasound-naïve operators with the teleultrasound system. PRIMARY AND SECONDARY OUTCOME MEASURES: Radiologists examined the ultrasound imaging to assess its diagnostic value and identify any pathology. A random subset of 20% of the scans were analysed for inter-reader reliability. RESULTS: Lung VSI teleultrasound examinations underwent detailed analysis by two cardiothoracic attending radiologists. Of the examinations, 202 were rated of diagnostic image quality (94.8%, 95% CI 90.9% to 97.4%). There was 91% agreement between radiologists on lung ultrasound interpretation among a 20% sample of all examinations (κ=0.76, 95% CI 0.53 to 0.98). Radiologists were able to identify sequelae of COVID-19 with the predominant finding being B-lines. CONCLUSION: Lung VSI teleultrasound performed by individuals without prior training allowed diagnostic imaging of the lungs and identification of sequelae of COVID-19 infection. Deployment of lung VSI teleultrasound holds potential as a low-cost means to improve access to imaging around the world.

ConspectusHoley graphene (hG) is a structural derivative of graphene with arrays of through-thickness holes of a few to tens of nanometers in diameter, randomly distributed across the nanosheet surfaces. In most bulk preparation methods, the holes on hG sheets are preferentially generated from the pre-existing defects on graphene. Therefore, contrary to intuitive belief, hG is not necessarily more defective than the intact graphene. Instead, it retains essential parent properties, including high electrical conductivity, high surface area, mechanical robustness, and chemical inertness. Furthermore, the added holey structural motif imparts unique properties that are not present in unmodified graphene, making hG advantageous in numerous applications such as sensing, membranes, reinforcements, and electrochemical energy storage. In particular, the presence of holes enhances the mass transport through the nanosheet plane and thus significantly reduces tortuosity. This difference is a key advantage for using hG in energy storage applications where the transport of ions through the thickness becomes more hindered as the electrode thickness increases to meet practical energy density requirements.An unexpected discovery is that the holes of the hG sheets enable the dry hG powder to be directly compressed into robust monoliths. hG not only can be pressed into monoliths by itself but also can host other electrochemically active materials as a compressible matrix. This important yet unique property, which is not available for other carbon materials including intact graphene, significantly broadens the application horizon in energy storage applications. With the dry compressibility, electrodes with ultrahigh mass loading and thus ultrahigh areal capacity may be conveniently fabricated without toxic solvents or parasitic binders, which are required in conventional slurry-based approaches for electrode fabrication. The dry-press electrode preparation process can be completed within minutes regardless of mass loading. In comparison, high-mass-loading electrodes for advanced battery chemistries using conventional fabrication methods often need stringent and time-consuming process control. hG can also be combined with electrochemically active battery materials while maintaining dry compressibility. This has allowed the unprecedented, convenient manipulation of a wide variety of thick electrode compositions and architectures, which provides not only outstanding performance but also new physical insights for various battery chemistries.In this Account, we first present some basic observations on the dry compressibility of hG as well as the mechanistic investigations from atomistic modeling rationalizing this unique property. We then showcase the applications of neat and composite dry-pressed hG electrodes for various energy storage platforms including supercapacitors, lithium (Li) ion batteries, Li-O2 batteries, and Li-S/Se batteries. The preparation and performance of thick electrodes with practical mass loadings and unique electrode architecture manipulation, both enabled by the dry compressibility of hG, are highlighted and discussed.

Sickle cell disease (SCD) is the most common hereditary blood disorder in the United States. SCD is frequently associated with osteonecrosis, osteoporosis, osteopenia, and other bone-related complications such as vaso-occlusive pain, ischemic damage, osteomyelitis, and bone marrow hyperplasia known as sickle bone disease (SBD). Previous SBD models have failed to distinguish the age- and sex-specific characteristics of bone morphometry. In this study, we use the Townes mouse model of SCD to assess the pathophysiological complications of SBD in both SCD and sickle cell trait. Changes in bone microarchitecture and bone development were assessed by using high-resolution quantitative micro-computed tomography and the three-dimensional reconstruction of femurs from male and female mice. Our results indicate that SCD causes bone loss and sex-dependent anatomical changes in bone. SCD female mice in particular are prone to trabecular bone loss, whereas cortical bone degradation occurs in both sexes. We also describe the impact of genetic knockdown of cathepsin K- and E-64-mediated cathepsin inhibition on SBD.

Two-point microrheology (TPM) is used to infer material properties of complex fluids from the correlated motion of hydrodynamically interacting probes embedded in the medium. The mechanistic connection between probe motion and material properties is propagation of disturbance flows, encoded in current TPM theory for unconfined materials. However, confined media e.g. biological cells and particle-laden droplets, require theory that encodes confinement into the flow propagator (Green's function). To test this idea, we use Confined Stokesian Dynamics simulations to explicitly represent many-body hydrodynamic couplings between colloids and with the enclosing cavity at arbitrary concentration and cavity size. We find that previous TPM theory breaks down in confinement, and we identify and replace the underlying key elements. We put forth a Confined Generalized Stokes-Einstein Relation and report the viscoelastic spectrum. We find that confinement alters particle dynamics and increases viscosity, owing to hydrodynamic and entropic coupling with the cavity. The new theory produces a master curve for all cavity sizes and concentrations and reveals that for colloids larger than 0.005 times the enclosure size, the new model is required.

Resumen: Introducción: el manejo de antibióticos en las unidades de cuidados intensivos (UCI) es un tema prioritario. Conocer la epidemiología bacteriana y su sensibilidad es fundamental para aumentar la sobrevida de nuestros pacientes. Material y métodos: se realizó un estudio tipo cohorte retrospectiva en la Unidad de Cuidados Intensivos del Hospital Ángeles del Carmen durante el periodo de 2018 a 2020 en pacientes con infección documentada y con cultivo positivo. Se obtuvo el patrón de sensibilidad antimicrobiana y se analizó de acuerdo al origen Gram, tipo de infección, reactantes de fase aguda y mortalidad. Se realizó comparación de medias y proporciones con χ2, t de Student y ANOVA. Se obtuvieron razones de desventajas (OR) para identificar variables asociadas a resolución. Se consideró un valor de p < 0.05 para significancia estadística. Resultados: se analizaron 308 cultivos bacterianos obtenidos de 188 pacientes, principalmente de origen respiratorio, urinario y torrente sanguíneo (76.7%), de origen nosocomial (65.3%), con predominio de gram-negativos (65%) multidrogorresistentes. La procedencia comunitaria se asoció más a infección que la nosocomial (85 versus 61.7%, OR 3.5, IC 95% 1.93-6.45, p < 0.001). El porcentaje de infección fue mayor en gram-negativos (71.8 versus 66%, OR 1.10, IC 95% de 0.91-1.32, p = 0.297). Las infecciones por gram-positivos tuvieron menor porcentaje de mortalidad que aquéllas por gram-negativos (17.9 versus 30.7%, OR 0.49, IC 95% de 0.27-0.88, p = 0.016) así como las infecciones comunitarias en comparación con nosocomiales (17.8 versus 30.8%, OR 0.48, IC 95% de 0.27-0.86, p = 0.013). Conclusión: las bacterias predominantes en nuestra unidad de cuidados críticos son bacilos gram-negativos multidrogorresistentes, provenientes de infecciones respiratorias, urinarias y de torrente sanguíneo. Las infecciones por gram-positivos y adquiridas en la comunidad se asociaron a menor riesgo de mortalidad.


Abstract: Introduction: local identification of antimicrobial susceptibility and resistance patterns must be a priority in intensive care units. Material and methods: a cohort study was conducted in the intensive care unit from 2018 to 2020, identifying patients with an infectious diagnosis and a positive culture, with prospective clinical and laboratory follow-up. Antimicrobial resistance patterns were analyzed according to source, gram, type of infection, acute phase reactants and outcome, comparing means and proportions with χ2, Student t and ANOVA. OR were obtained to identify resolution-associated variables. A p < 0.05 value was considered as statistically significant. Results: 308 cultures were analyzed, obtained from 188 patients. Primary souces were respiratory, urinary and bloodstream (76.7%), 65.3% were from in-hospital infections, and 65% were caused by gram-negative multi-drug resistant bacteria. Community cultures were more associated with infection compares with in-hospital cultures (85 vs 61.7%, OR 3.5, 95% CI 1.93-6.45, p < 0.001). Gram-negative bacteria had a greater association with infection compared with gram-positive (71.8 vs 66%, OR 1.10, 95% CI 0.91-1.32, p = 0.297), but infections caused by gram-positive bacteria had a greater association with resolution (82.1 vs 68.8%, OR 2.07, 95% CI 1.16-3.70, p = 0.019), as well as community infections (82.2 vs 68.7%, OR 2.11, 95% CI 1.18-3.77, p = 0.016). Conclusion: multi-drug resistant gram-negative bacteria were the principal isolates found in respiratory, urine and bloodstream infections in our intensive care unit. Community infections and gram-positive isolates were associated with greater resolution rates.

Conhecer a epidemiologia bacteriana e sua suscetibilidade é essencial para aumentar a sobrevida de nossos pacientes. A comparação de médias e proporções foi feita com χ2, teste t de Student e ANOVA. Razões de desvantagem (OR) foram obtidas para identificar variáveis ​​associadas à resolução. Um valor de p < 0.05 foi considerado para significância estatística. Resultados: foram analisadas 308 culturas bacterianas obtidas de 188 pacientes, principalmente de origem respiratória, urinária e sanguínea (76.7%), de origem nosocomial (65.3%), com predominância de gram-negativos (65%) multirresistentes. A origem comunitária foi mais associada à infecção do que a nosocomial (85 vs 61.7%, OR 3.5, IC 95% 1.93-6.45, p < 0.001). Clear cell renal cell carcinoma is the most common and deadly type of cancer affecting the kidney, and is characterized histologically by large intracellular lipid deposits. These deposits are thought to result from lipid metabolic reprogramming occurring in tumor cells, but the exact mechanisms and implications of these metabolic alterations are incompletely understood. Obesity is an independent risk factor for clear cell renal cell carcinoma, and is also associated with lipid accumulation in noncancerous epithelial cells of the proximal tubule, where clear cell renal cell carcinoma originates. This article explores the potential link between obesity-associated renal lipid metabolic disturbances and lipid metabolic reprogramming in clear cell renal cell carcinoma, and discusses potential implications for future research.

We develop a scaling theory that predicts the dynamics of symmetric and asymmetric unentangled liquid coacervates formed by solutions of oppositely-charged polyelectrolytes. Symmetric coacervates made from oppositely-charged polyelectrolytes consist of polycations and polyanions with equal and opposite charge densities along their backbones. These symmetric coacervates can be described as mixtures of polyelectrolytes in the quasi-neutral regime with a single correlation length. Asymmetric coacervates are made from polycations and polyanions with unequal charge densities. The difference in charge densities results in a double semidilute structure of asymmetric coacervates with two correlation lengths, one for the high-charge-density and the other for the low-charge-density polyelectrolytes. We predict that the double-semidilute structure in asymmetric coacervates results in a dynamic coupling which increases the friction of the high-charge-density polyelectrolyte. This dynamic coupling increases the contribution to the zero-shear viscosity of the high-charge-density polyelectrolyte. The diffusion coefficient of the high-charge-density polyelectrolyte is predicted to depend on the concentration and degree of polymerization of the low-charge-density polyelectrolyte in the coacervate if the size of the low-charge-density polymer is smaller than the correlation length of the high-charge-density polymer. We also predict a non-monotonic salt concentration dependence of the zero-shear viscosity of asymmetric coacervates.

Community-based conservation and resource management (CBCRM) programs often incorporate the dual goals of poverty alleviation and conservation. However, robust assessments of CBCRM program outcomes are relatively scarce. This study uses a multidisciplinary, systems approach to assess the ecological and social dimensions of success of an internationally acclaimed CBCRM program. This program, located in one of the largest protected areas in the Peruvian Amazon (Pacaya-Samiria National Reserve), strives for the sustainable harvest and trade of a turtle species (Podocnemis unifilis). We used mixed methods analysis, including interviews and population viability modeling, to understand three elements: how local perceptions of changes in the managed population compare to changes inferred by ecological analyses, the indicators stakeholders use to measure success, and the barriers to long-term program success and social-ecological system sustainability. We find that stakeholders perceive a growth trend in the managed turtle population, but this perception may diverge from our ecological understanding of the system under current management. Population viability analyses with a 1:1 sex ratio suggested population size will decline under two of three management scenarios (different degrees of harvest). Yet this and similar studies are plagued by a lack of species- and site-specific population parameters that could improve understanding of the system. Significant vulnerabilities exist for system sustainability, notably the recent decrease in foreign demand for the traded resource. Identifying a sustainable species-specific harvest rate, developing locally-grounded ecological and social indicators, and focusing on data-driven adaptive management will facilitate the identification of key leverage points for future management interventions.

ABSTRACT: Beef slaughter establishments employ many interventions to help minimize the occurrence of pathogens in their products. This study explored the effectiveness of various common interventions on microbial load using the results of the Beef-Veal Carcass Baseline Survey conducted in 2014 to 2015. The Food Safety and Inspection Service analyzed swab samples taken from 1,135 carcasses at 139 establishments. These included paired samples from post-hide removal (before evisceration) and prechill (after evisceration). Samples were tested for pathogens (Salmonella and Shiga toxin-producing Escherichia coli) and indicators (E. coli, Enterobacteriaceae, coliforms, and aerobic count [AC]). The sample size for pathogen-positive samples was small, impeding the establishment of a direct correlation between interventions and pathogens. However, we observed associations between pathogen-positive rate and log AC, indicating similar intervention effectiveness of pathogens and indicators in this study. Generally, the use of interventions reduced indicator concentrations. Each intervention produced a range of effectiveness, suggesting that how interventions are applied may be as important as which interventions are applied. The range of effectiveness for single interventions was a 0.4- to 1.9-log AC reduction; for multihurdle interventions, it ranged from 1.6- to 2.9-log AC reduction. The results of this study may be used by slaughter establishments to help identify effective intervention options for pathogen reduction.

To define morphological changes in carotid and cerebral arteries in sickle cell transgenic mice (SS) as they age, a combination of ultrasound and microcomputed tomography of plastinated arteries was used to quantify arterial dimensions and changes in mice 4, 12, and 24 weeks of age. 12-week SS mice had significantly larger common carotid artery diameters than AS mice, which continued through to the extracranial and intracranial portions of the internal carotid artery (ICA). There were also side specific differences in diameters between the left and right vessels. Significant ICA tapering along its length occurred by 12- and 24-weeks in SS mice, decreasing by as much as 70%. Significant narrowing along the length was also measured in SS anterior cerebral arteries at 12- and 24-weeks, but not AS. Collectively, these findings indicate that sickle cell anemia induces arterial remodeling in 12- and 24-weeks old mice. Catalog of measurements are also provided for the common carotid, internal carotid, anterior cerebral, and middle cerebral arteries for AS and SS genotypes, as a reference for other investigators using mathematical and computational models of age-dependent arterial complications caused by sickle cell anemia.

Purpureocillium lilacinum (P. lilacinum) is an emergent pathogenic mold that presents more commonly as an ocular infection, cutaneous and/or subcutaneous infections in patients that are usually immunocompromised. A pulmonary presentation is rare, the clinical presentation is fever and cough with radiographic presentation as pleural effusion, single-lung consolidation, and cavitary pulmonary disease. We present a case of a patient with hematologic malignancy with febrile neutropenia; after receiving chemotherapy, the patient developed a pulmonary infection with multiple bilateral consolidations shown in the thoracic computed tomography scan. Fever persisted in spite of the use of wide-spectrum antibiotics and amphotericin. Bronchoalveolar lavage was performed and the samples were cultured, isolating in the Sabouraud Dextrous Agar a filamentous fungi growth with purple colonies that were identified morphologically as P. lilacinum and later it was confirmed by molecular methods. Once the infectious agent was identified, we continued amphotericin and oral voriconazole was added to the treatment with complete resolution of the infection. The report aims to create awareness of this emerging infectious disease, as there is little information concerning the treatment and the prognosis of patients infected by P. lilacinum with a pulmonary presentation.

Aversive or rewarding experiences are remembered better than those of lesser survival significance. These emotional memories, whether negative or positive, leave traces in the brain which can later be retrieved and strongly influence how we perceive, how we form associations with environmental stimuli and, ultimately, guide our decision-making. In this review aticle, we outline what constitutes an emotional memory by focusing on threat- and reward-related memories and describe how they are formed in the brain during learning and reformed during retrieval. Finally, we discuss how the field is moving from understanding emotional memory brain circuits separately, towards studying how these two opposing brain systems interact to guide choices during conflict. Here, we outline two novel tasks in rodents that model opposing binary choices (approach or avoid) guided by competing emotional memories. The prefrontal cortex (PFC) is a major integration hub of emotional information which is also known to be critical for decision-making. Consequently, brain circuits that involve this brain region may be key for understanding how the retrieval of emotional memories flexibly orchestrates adaptive choice behavior. Because several mental disorders (e.g., drug addiction and depression) are characterized by deficits in decision-making in the face of conflicting emotional memories (maladaptively giving more weight to one memory over the other), the development of choice-based animal models for emotional regulation could give rise to new approaches for the treatment of these disorders in humans.

OBJECTIVE: Sickle cell anemia (SCA) causes chronic inflammation and multiorgan damage. Less understood are the arterial complications, most evident by increased strokes among children. Proteolytic mechanisms, biomechanical consequences, and pharmaceutical inhibitory strategies were studied in a mouse model to provide a platform for mechanistic and intervention studies of large artery damage due to sickle cell disease. Approach and Results: Townes humanized transgenic mouse model of SCA was used to test the hypothesis that elastic lamina and structural damage in carotid arteries increased with age and was accelerated in mice homozygous for SCA (sickle cell anemia homozygous genotype [SS]) due to inflammatory signaling pathways activating proteolytic enzymes. Elastic lamina fragmentation observed by 1 month in SS mice compared with heterozygous littermate controls (sickle cell trait heterozygous genotype [AS]). Positive immunostaining for cathepsin K, a powerful collagenase and elastase, confirmed accelerated proteolytic activity in SS carotids. Larger cross-sectional areas were quantified by magnetic resonance angiography and increased arterial compliance in SS carotids were also measured. Inhibiting JNK (c-jun N-terminal kinase) signaling with SP600125 significantly reduced cathepsin K expression, elastin fragmentation, and carotid artery perimeters in SS mice. By 5 months of age, continued medial thinning and collagen degradation was mitigated by treatment of SS mice with JNK inhibitor. CONCLUSIONS: Arterial remodeling due to SCA is mediated by JNK signaling, cathepsin proteolytic upregulation, and degradation of elastin and collagen. Demonstration in Townes mice establishes their utility for mechanistic studies of arterial vasculopathy, related complications, and therapeutic interventions for large artery damage due to SCA.

The Food Safety and Inspection Service (FSIS) conducts microbiological baseline studies to determine national prevalence of select foodborne pathogens in federally inspected meat and poultry products and to obtain data for risk assessments. The FSIS conducted a baseline study from 1 June 2017 through 31 May 2018 to characterize and determine the prevalence of Salmonella and assess the occurrence of Shiga toxin-producing Escherichia coli (STEC) in a variety of raw pork products. In total, 4,014 samples from slaughter and processing establishments were analyzed for Salmonella; a subset of these samples (1,395) from slaughter establishments were also analyzed for STEC. Analyses determined that the national prevalence of Salmonella in raw pork products was highest in comminuted products (28.9%), followed by intact cuts (5.3%) and nonintact cuts (3.9%). Less than 1% of samples analyzed were positive for the top seven STEC. Our findings indicate there is a need for additional pathogen reduction strategies for raw pork products.
