RESUMO

We report a full experimental comparison study on the injection of a Ti:Sa multi-terawatt amplifier chain with a standard 15 fs Ti:Sa oscillator and 35 fs frequency-doubled fiber oscillator. The study highlights that the Ti:Sa oscillator, with high performance in terms of pulse duration and spectral width, can be replaced by the frequency-doubled fiber oscillator to seed Ti:Sa amplifier chains almost without any compromise on the output pulse duration and picosecond contrast. Finally, we demonstrate for the first time to our knowledge a 30 TW and 33 fs Ti:Sa amplifier injected by a fiber oscillator.

Independent control over phase separation and photo-cross-linking allows the structure and porosity of hydrogels to be patterned in a single photolithographic step. This observation is based upon a temperature-triggered spinodal decomposition of a ternary mixture of water, salt, and polymer into a salt-rich aqueous phase and a polymer-rich phase. Importantly, subsequent exposure to light arrests the phase separation, allowing the porosity state to be frozen in a cross-linked hydrogel network. Tuning the delay between the application of heat and illumination allows the pore size to be tuned between 400 nm and 4 µm. By utilizing gray-scale photomasks, a single process can be used to define regions of pure hydrogel, porous hydrogel with a programmed average pore size, and blank substrate with no hydrogel. In addition to representing a combination of top-down and bottom-up processes that enables the realization of complex samples, the simplicity of this process and the versatility of the resultant patterns could provide a useful capability for the definition of hydrogel samples for the development of advanced biomaterials.

The impact of age and physical health on processing speed was investigated in 42 non-demented HIV+ individuals ranging in age from 30 to 75. We used the Medical Outcomes Study-HIV Healthy Survey (MOS-HIV) to measure self-reported physical health, neuropsychological tests to measure psychomotor and cognitive processing speed (Delis-Kaplan Executive Function System Trail Making Test, Grooved Pegboard Test, letter and category fluency), and a test of the foreperiod effect to measure reaction time under increasing attentional load. Results indicated that aging and worse physical health each independently contributed to slowing on different processing speed measures, while the interaction between aging and physical health did not contribute to processing speed. These findings highlight the importance of considering physical health separately from age when measuring cognitive function in HIV+ adults.

We implement a novel strategy for harnessing the power of high-copy transposons for functional analysis of the maize genome, and report behavioral features of the Mutator system in a uniform inbred background. The unique UniformMu population and database facilitate high-throughput molecular analysis of Mu-tagged mutants and gene knockouts. Key features of the population include: (i) high mutation frequencies (7% independent seed mutations) and moderation of copy number (approximately 57 total Mu elements; 1-2 MuDR copies per plant) were maintained by continuous back-crossing into a phenotypically uniform inbred background; (ii) a bz1-mum9 marker enabled selection of stable lines (loss of MuDR), inhibiting further transpositions in lines selected for molecular analysis; (iii) build-up of mutation load was prevented by screening Mu-active parents to exclude plants carrying pre-existing seed mutations. To create a database of genomic sequences flanking Mu insertions, selected mutant lines were analyzed by sequencing of MuTAIL PCR clone libraries. These sequences were annotated and clustered to facilitate bioinformatic subtraction of ancestral elements and identification of insertions unique to mutant lines. New insertions targeted low-copy, gene-rich sequences, and in silico mapping revealed a random distribution of insertions over the genome. Our results indicate that Mu populations differ markedly in the occurrence of Mu insertion hotspots and the frequency of suppressible mutations. We suggest that controlled MuDR copy number in UniformMu lines is a key determinant of these differences. The public database (http://uniformmu.org; http://endosperm.info) includes pedigree and phenotypic data for over 2000 independent seed mutants selected from a population of 31 548 F2 lines and integrated with analyses of 34 255 MuTAIL sequences.
