RESUMO

Amanita phalloides is one of the most toxic mushrooms worldwide, being responsible for the majority of human fatal cases of mushroom intoxications. α-Amanitin, the most deleterious toxin of A. phalloides, inhibits RNA polymerase II (RNAP II), causing hepatic and renal failure. Herein, we used cyclosporine A after it showed potential to displace RNAP II α-amanitin in silico. That potential was not confirmed either by the incorporation of ethynyl-UTP or by the monitoring of fluorescent RNAP II levels. Nevertheless, concomitant incubation of cyclosporine A with α-amanitin, for a short period, provided significant protection against its toxicity in differentiated HepaRG cells. In mice, the concomitant administration of α-amanitin [0.45 mg/kg intraperitoneal (i.p.)] with cyclosporine A (10 mg/kg i.p. plus 2 × 10 mg/kg cyclosporine A i.p. at 8 and 12 h post α-amanitin) resulted in the full survival of α-amanitin-intoxicated mice, up to 30 days after the toxin's administration. Since α-amanitin is a substrate of the organic-anion-transporting polypeptide 1B3 and cyclosporine A inhibits this transporter and is a potent anti-inflammatory agent, we hypothesize that these mechanisms are responsible for the protection observed. These results indicate a potential antidotal effect of cyclosporine A, and its safety profile advocates for its use at an early stage of α-amanitin intoxications.

During infections, the host redistributes iron in order to starve pathogens from this nutrient. Several proteins are involved in iron absorption, transport, and storage. Ferritin is the most important iron storage protein. It is composed of variable proportions of two peptides, the L- and H-ferritins (FTL and FTH). We previously showed that macrophages increase their expression of FTH1 when they are infected in vitro with Mycobacterium avium, without a significant increase in FTL. In this work, we investigated the role of macrophage FTH1 in M. avium infection in vivo. We found that mice deficient in FTH1 in myeloid cells are more resistant to M. avium infection, presenting lower bacterial loads and lower levels of proinflammatory cytokines than wild-type littermates, due to the lower levels of available iron in the tissues. Importantly, we also found that FTH1 produced by myeloid cells in response to infection may be found in circulation and that it plays a key role in iron redistribution. Specifically, in the absence of FTH1 in myeloid cells, increased expression of ferroportin is observed in liver granulomas and increased iron accumulation occurs in hepatocytes. These results highlight the importance of FTH1 expression in myeloid cells for iron redistribution during infection.

The transcriptome is the complete set of transcripts in a cell or tissue and includes ribosomal RNA (rRNA), messenger RNA (mRNA), transfer RNA (tRNA), and regulatory noncoding RNA. At steady-state, the transcriptome results from a compensatory variation of the transcription and decay rate to maintain the RNA concentration constant. RNA transcription constitutes the first stage in gene expression, and thus is a major and primary mode of gene expression control. Nevertheless, regulation of RNA decay is also a key factor in gene expression control, involving either selective RNA stabilization or enhanced degradation. Transcriptome analysis allows the identification of gene expression alterations, providing new insights regarding the pathways and mechanisms involved in physiological and pathological processes. Upon perturbation of cell homeostasis, rapid changes in gene expression are required to adapt to new conditions. Thus, to better understand the regulatory mechanisms associated with gene expression alterations, it is vital to acknowledge the relative contribution of RNA synthesis and decay to the transcriptome. To the toxicology field, the study of gene expression regulation mechanisms can help identify the early and mechanistic relevant cellular events associated with a particular response. This review aims to provide a critical comparison of the available methods used to analyze the contribution of RNA transcription and decay to gene expression dynamics. Notwithstanding, an integration of the data obtained is necessary to understand the entire repercussions of gene transcription changes at a system-level. Thus, a brief overview of the methods available for the integration and analysis of the data obtained from transcriptome analysis will also be provided.

RESUMO

Two new eriophyoid mite species are described from the cocoa tree, Theobroma cacao L. (Malvaceae) - the Diptilomiopidae Davisella trevisani n. sp. Rodrigues, Navia & Oliveira, from Brazil; and the Eriophyidae Tetra theobromae n. sp. Rodrigues, Navia & Oliveira, from Costa Rica. The new Davisella species presents an enlarged subcylindrical tubercle bearing pedipalp genual seta d, a novel structure for eriophyoid mites; its functional morphology was discussed. The new Tetra species is the third in this genus described from plants of the Malvaceae. A closer look at Shevtchenkella biseta (Nalepa), described from the Malvaceae Hibiscus rosa-sinensis L., showed its taxonomic assignment to the Phytoptidae, Sierraphytoptinae and raised a hypothesis on its synonymy.

The side chain effect on transport in ionic aqueous salt solutions was investigated for [Formula: see text]-amino acids glycine, alanine, [Formula: see text]-amino butyric acid, norvaline, and norleucine --that together define a chemical homologous series based on the length of the characteristic side chain which increases from zero to four carbons, respectively. Binary mutual diffusion coefficients at infinitesimal concentration in aqueous solutions of NaCl (0.15 mol kg -1) are measured by means of Taylor dispersion technique for this series and significant differences were found against previous published results for identical systems in pure water. In this way, NaCl effect on the transport of each amino acid is thus assessed and discussed in terms of salting-out effects. Also, solvated Stokes hydrodynamic radii were computed for the series showing comparable results in water and NaCl solution. The new information should prove useful in the design and characterization of transport-controlled systems in physiological and pharmacological studies.

RESUMO

A new eriophyid genus and species, tentatively placed in the Tegonotini, namely Eriomacrotergum flechtmanni n. gen. n. sp., is described from the Amazonian fruit tree cupuaçu Theobroma grandiflorum (Willd. Ex Spreng.) K. Schum. (Malvaceae), collected in northern Brazil. The new taxon presents a peculiar morphology in having a large opisthosomal plate and a very reduced prodorsal shield, with ocellar-like structures and scapular setae displaced laterally. Discussion on the most remarkable taxonomic traits is presented as well as similarities of the new genus with other Eriophyidae genera.
