The rapid growth of industrialization and urbanization in China has led to an increase in soil heavy metal pollution, which poses a serious threat to ecosystem safety and human health. The advancement of spectral technology offers a way to rapidly and non-destructively monitor soil heavy metal content. In order to explore the potential of rice leaf spectra to indirectly estimate soil heavy metal content. We collected farmland soil samples and measured rice leaf spectra in Xushe Town, Yixing City, Jiangsu Province, China. In the laboratory, the heavy metals Cd and As were determined. In order to establish an estimation model between the pre-processed spectra and the soil heavy metals Cd and As content, a genetic algorithm (GA) was used to optimise the partial least squares regression (PLSR). The model's accuracy was evaluated and the best estimation model was obtained. The results showed that spectral pre-processing techniques can extract hidden information from the spectra. The first-order derivative of absorbance was more effective in extracting spectral sensitive information from rice leaf spectra. The GA-PLSR model selects only about 10% of the bands and has better accuracy in spectral modeling than the PLSR model. The spectral reflectance of rice leaves has the capacity to estimate Cd content in the soil (relative percent difference [RPD] = 2.09) and a good capacity to estimate As content in the soil (RPD = 2.97). Therefore, the content of the heavy metals Cd and As in the soil can be estimated indirectly from the spectral data of rice leaves. This study provides a reference for future remote sensing monitoring of soil heavy metal pollution in farmland that is quantitative, dynamic, and non-destructive over a large area.

Pharmacological inhibition of IDO1 exhibits great promise as a strategy in cancer therapy. However, the failure of phase III clinical trials has raised the pressing need to understand the underlying reasons for this outcome. To gain comprehensive insights into the reasons behind the clinical failure of IDO1 inhibitors, it is essential to investigate the entire tumor microenvironment rather than focusing solely on individual cells or relying on knockout techniques. In this study, we conducted single-cell RNA sequencing to determine the overall response to apo-IDO1 inhibitor administration. Interestingly, although apo-IDO1 inhibitors were found to significantly activate intratumoral immune cells (mouse colon cancer cell CT26 transplanted in BALB/C mice), such as T cells, macrophages, and NK cells, they also stimulated the infiltration of M2 macrophages. Moreover, these inhibitors prompted monocytes and macrophages to secrete elevated levels of IL-6, which in turn activated the JAK2/STAT3 signaling pathway in tumor cells. Consequently, this activation enables tumor cells to survive even in the face of heightened immune activity. These findings underscore the unforeseen adverse effects of apo-IDO1 inhibitors on tumor cells and highlight the potential of combining IL-6/JAK2/STAT3 inhibitors with apo-IDO1 inhibitors to improve their clinical efficacy.

The developmental trajectory of audiovisual speech perception in Mandarin-speaking children remains understudied. This cross-sectional study in Mandarin-speaking 3- to 4-year-old, 5- to 6-year-old, 7- to 8-year-old children, and adults from Xiamen, China (n = 87, 44 males) investigated this issue using the McGurk paradigm with three levels of auditory noise. For the identification of congruent stimuli, 3- to 4-year-olds underperformed older groups whose performances were comparable. For the perception of the incongruent stimuli, a developmental shift was observed as 3- to 4-year-olds made significantly more audio-dominant but fewer audiovisual-integrated responses to incongruent stimuli than older groups. With increasing auditory noise, the difference between children and adults widened in identifying congruent stimuli but narrowed in perceiving incongruent ones. The findings regarding noise effects agree with the statistically optimal hypothesis.

The present study aimed to examine whether Mandarin-speaking children on the autism spectrum showed differences in comprehending spatial demonstratives ("this" and "that", and "here" and "there"), as compared to typically developing (TD) children. Another aim of this study was to investigate the roles of theory of mind (ToM) and executive functions (EF) in the comprehension of spatial demonstratives. Twenty-seven autistic children (mean age 6.86) and 27 receptive-vocabulary-matched TD children (mean age 5.82) were recruited. Demonstrative comprehension was assessed based on participants' ability to place objects in certain locations according to experimenters' instructions which involved these demonstratives in three different conditions (same-, opposite-, and spectator-perspective conditions). Four false-belief tasks were administered to measure ToM, and the word-span task and the dimensional change card sort task were used to measure two subcomponents of EF - working memory and mental flexibility - respectively. Children on the autism spectrum were found to score below TD children in the comprehension of spatial demonstratives. In addition, the results showed that ToM and working memory were conducive to the correct interpretation of spatial demonstratives. The two cognitive abilities mutually influenced their respective roles in spatial demonstrative comprehension in the three different conditions. The findings suggest that the comprehension of spatial demonstratives comprehension is an area of need in Mandarin-speaking children on the autism spectrum, and it might be linked to their differences in cognitive abilities.

Proprotein convertase subtilisin/kexin type 9 (PCSK9) is an enzyme that promotes the degradation of low-density lipoprotein receptors. It is involved in hyperlipidemia as well as other diseases, such as cancer and skin inflammation. However, the detailed mechanism for PCSK9 on ultraviolet B (UVB)-induced skin lesions was not clear. Thus, the role and possible action mechanism of PCSK9 in UVB-induced skin damage in mice were studied here using siRNA and a small molecule inhibitor (SBC110736) against PCSK9. Immunohistochemical staining revealed a significant increase in PCSK9 expression after UVB exposure, indicating the possible role of PCSK9 in UVB damage. Skin damage, increase in epidermal thickness, and keratinocyte hyperproliferation were significantly alleviated after treatment with SBC110736 or siRNA duplexes, compared with that in the UVB model group. Notably, UVB exposure triggered DNA damage in keratinocytes, whereas substantial interferon regulatory factor 3 (IRF3) activation was observed in macrophages. Pharmacologic inhibition of STING or cGAS knockout significantly reduced UVB-induced damage. In the co-culture system, supernatant from UVB-treated keratinocyte induced IRF3 activation in macrophages. This activation was inhibited with SBC110736 and by PCSK9 knockdown. Collectively, our findings reveal that PCSK9 plays a critical role in the crosstalk between damaged keratinocytes and STING activation in macrophages. The interruption of this crosstalk by PCSK9 inhibition may be a potential therapeutic strategy for UVB-induced skin damage.

LAY ABSTRACT: Some theories suggested that autistic people have better pitch perception skills than non-autistic people. However, in a context where pitch patterns are used to differentiate word meanings (i.e. lexical tones), autistic people may encounter difficulties, especially those with less language experience. We tested this by asking language-delayed autistic children to identify and discriminate two Mandarin lexical tones (/yi/ with Tone 1, meaning 'clothes'; /yi/ with Tone 2, meaning 'aunt'; /yi/: the standard romanization of Mandarin Chinese). On average, these autistic children were 7.35 years old, but their developmental age in language ability was 4.20, lagging behind 7-year-old non-autistic children in terms of language ability. Autistic children's performance in identifying and discriminating lexical tones was compared with two groups of non-autistic children: one group was matched with the autistic group on age, and the other was matched based on language ability. Autistic children performed differently from the non-autistic children matched on age, while autistic and non-autistic children matched on language ability exhibited seemingly similar performance. However, both the non-autistic groups have developed the perceptual ability to process lexical tones as different categories, but this ability was still developing in autistic children. Finally, we found autistic children who performed worse in identifying lexical tones had poorer language ability. The results suggest that language disability might have adverse influence on the development of skills of speech sound processing.

Chemical recycling of polymers is critical for improving the circular economy of plastics and environmental sustainability. Traditional thermoset polymers have generally been considered permanently crosslinked materials that are difficult or impossible to recycle. Herein, we demonstrate that by activating 'dormant' covalent bonds, traditional polycyanurate thermosets can be recycled into the original monomers, which can be circularly reused for their original purpose. Through retrosynthetic analysis, we redirected the synthetic route from forming conventional C-N bonds via irreversible cyanate trimerization to forming the C-O bonds through reversible nucleophilic aromatic substitution of alkoxy-substituted triazine derivatives by alcohol nucleophiles. The new reversible synthetic route enabled the synthesis of previously inaccessible alkyl-polycyanurate thermosets, which exhibit excellent film properties with high chemical resistance, closed-loop recyclability and reprocessing capability. These results show that 'apparently dormant' dynamic linkages can be activated and utilized to construct fully recyclable thermoset polymers with a broader monomer scope and increased sustainability.

While it has been proposed, following relevance theory, that similes can be understood at a purely literal level on a par with literal statements, it remains unclear whether children with high-functioning autism spectrum disorders (HFASD) perform similarly to typically developing (TD) children in comprehending literal statements and similes. The present study investigated comprehension of literal statements and similes in Cantonese-speaking children with HFASD and TD children matched on both chronological age and verbal mental age. An utterance-picture matching task was devised to assess their comprehension of literal statements and similes in Cantonese. Overall results showed that Cantonese-speaking children with HFASD performed worse than TD children in comprehending literal statements and similes, and both groups showed more difficulty in comprehending similes than literal statements. After the effects of chronological age and verbal mental age were controlled for, no group difference was found between children with HFASD and TD children in comprehending literal statements, whereas the group difference in simile comprehension still existed, suggesting that children with HFASD showed deficits in comprehending similes relative to TD children. These findings challenge the proposal that similes can be understood at a purely literal level on a par with literal statements. Future studies should investigate the role of different aspects of language ability and different levels of theory-of-mind skills in comprehension of similes and metaphors in children with HFASD.

Built on a previous finding that children with autism spectrum disorders (ASD) had difficulty comprehending presuppositions, a domain of knowledge which is crucial for successful communication, the present study investigated the comprehension of seven types of presupposition trigger - namely definite descriptions, factive predicates, change-of-state verbs, implicative verbs, iteratives, temporal clauses, and counterfactual conditionals - in Cantonese-speaking children with and without ASD. Twenty-seven children with ASD (mean age 9.07) were compared with 23 typically developing (TD) children matched on chronological age and 21 TD children matched on language ability (LA). Knowledge of presupposition triggers was evaluated on the basis of children's ability to judge whether a given utterance was a correct presupposition of a preceding utterance. Children with ASD were found to perform significantly worse than TD children matched on chronological age in comprehending the seven types of presupposition trigger, but they performed similarly to TD children matched on language ability. After the effects of chronological age, language ability, and non-verbal intelligence were controlled for, children with ASD were still found to show a deficit in comprehending the presupposition triggered by a temporal clause, relative to the two groups of TD children. Future studies should investigate the factors contributing to this specific deficit in children with ASD, such as executive functioning and theory of mind.
