RESUMO

Xylosandrus crassiusculus is an invasive ambrosia beetle comprising two differentiated genetic lineages, named cluster 1 and cluster 2. These lineages invaded different parts of the world at different periods of time. We tested whether they exhibited different climatic niches using Schoener's D and Hellinger's I indices and modeled their current potential geographical ranges using the Maxent algorithm. The resulting models were projected according to future and recent past climate datasets for Europe and the Mediterranean region. The future projections were performed for the periods 2041-2070 and 2071-2100 using 3 SSPs and 5 GCMs. The genetic lineages exhibited different climate niches. Parts of Europe, the Americas, Sub-Saharan Africa, Asia, and Oceania were evaluated as suitable for cluster 1. Parts of Europe, South America, Central and South Africa, Asia, and Oceania were considered as suitable for cluster 2. Models projection under future climate scenarios indicated a decrease in climate suitability in Southern Europe and an increase in North Eastern Europe in 2071-2100. Most of Southern and Western Europe was evaluated as already suitable for both clusters in the early twentieth century. Our results show that large climatically suitable regions still remain uncolonized and that climate change will affect the geographical distribution of climatically suitable areas. Climate conditions in Europe were favorable in the twentieth century, suggesting that the recent colonization of Europe is rather due to an increase in propagule pressure via international trade than to recent environmental changes.

Xylosandrus crassiusculus, a fungus-farming wood borer native to Southeastern Asia, is the most rapidly spreading invasive ambrosia species worldwide. Previous studies focusing on its genetic structure suggested the existence of cryptic genetic variation in this species. Yet, these studies used different genetic markers, focused on different geographical areas and did not include Europe. Our first goal was to determine the worldwide genetic structure of this species based on both mitochondrial and genomic markers. Our second goal was to study X. crassiusculus' invasion history on a global level and identify the origins of the invasion in Europe. We used a COI and RAD sequencing design to characterize 188 and 206 specimens worldwide, building the most comprehensive genetic data set for any ambrosia beetle to date. The results were largely consistent between markers. Two differentiated genetic clusters were invasive, albeit in different regions of the world. The markers were inconsistent only for a few specimens found exclusively in Japan. Mainland USA could have acted as a source for further expansion to Canada and Argentina through stepping stone expansion and bridgehead events. We showed that Europe was only colonized by Cluster II through a complex invasion history including several arrivals from multiple origins in the native area, and possibly including bridgehead from the United States. Our results also suggested that Spain was colonized directly from Italy through intracontinental dispersion. It is unclear whether the mutually exclusive allopatric distribution of the two clusters is due to neutral effects or due to different ecological requirements.

Xylosandrus compactus and X. crassiusculus are two polyphagous ambrosia beetles originating from Asia and invasive in circumtropical regions worldwide. Both species were recently reported in Italy and further invaded several other European countries in the following years. We used the MaxEnt algorithm to estimate the suitable areas worldwide for both species under the current climate. We also made future projections for years 2050 and 2070 using 11 different General Circulation Models, for 4 Representative Concentration Pathways (2.6, 4.5, 6.0 and 8.5). Our analyses showed that X. compactus has not been reported in all potentially suitable areas yet. Its current distribution in Europe is localised, whereas our results predicted that most of the periphery of the Mediterranean Sea and most of the Atlantic coast of France could be suitable. Outside Europe, our results also predicted Central America, all islands in Southeast Asia and some Oceanian coasts as suitable. Even though our results when modelling its potential distribution under future climates were more variable, the models predicted an increase in suitability poleward and more uncertainty in the circumtropical regions. For X. crassiusculus, the same method only yielded poor results, and the models thus could not be used for predictions. We discuss here these results and propose advice about risk prevention and invasion management of both species.

The Asian and Citrus longhorned beetles, Anoplophora glabripennis (ALB) and A. chinensis (CLB) respectively, are two closely related invasive species with overlapping native ranges. Although both species have rather similar biological characteristics, they differ in their invasion patterns. ALB shows numerous, but local, outbreaks in urban areas of North-East America, Western and Central Europe, whereas CLB has colonized a large part of Northern Italy. Temperature is pivotal in setting distribution limits of ectotherms. Low temperature may be limiting for larvae since they are the main overwintering stage for both species. To investigate whether differential cold tolerance may contribute to setting the respective limits of the range invaded by each species, we monitored larval metabolic rate before and after exposure to a one-week ecologically relevant moderate cold stress (-2/+2°C, 14/10h). We tested two distinctive fluctuating regimes before the cold exposure to check whether larval acclimation significantly altered their cold tolerance. Survival was high in all conditions for both species. Visual examination showed temporary locomotor inactivity during the stress but respiration rates were not altered after the stress suggesting that larvae could rapidly resume their initial metabolic activity. The respiration rate was globally higher in ALB than in CLB. Together, these results tend to indicate that both species have similar tolerance to the moderate cold stress tested, but also that ALB may be better at maintaining metabolic activity at cold than CLB. These observed differences could affect phenology in both species and in turn their establishment potential.

Thaumetopoea pityocampa causes allergies and skin and ocular lesions. No commercial tools are currently available for the clinical diagnosis of this allergy. We aimed to develop an in vitro method for the diagnosis of this allergy to avoid patients undergoing in vivo tests with insect extracts. Recombinant Tha p 2 was produced and used in an ELISA validated with 15 allergic patients. Subsequently, 42 subjects recruited from a random sampling cross-sectional study were analysed. The ELISA sensitivity and specificity were 93.3% and 100%, respectively, for the allergic patients and 71.4% and 95.3%, respectively, for the epidemiological study. The positive ELISA results correlated with the skin prick test areas with the whole body and the setae extracts. Professional exposure and short latency of symptoms onset were risk factors for a positive result in the ELISA. In conclusion, our ELISA is very useful for T. pityocampa allergy diagnosis and for epidemiologic testing.

The mango blossom gall midge, Procontarinia mangiferae, is a multivoltine species that induces galls in inflorescences and leaves of the mango tree, Mangifera indica. In subtropical Reunion Island, populations of P. mangiferae are observed all-year round, but the pattern and the role of dormancy in their life cycle have never been documented. We performed field and laboratory experiments using more than 15,000 larvae. We demonstrated that a larval diapause may affect a part of the midge population, regardless of the season. The total duration of the diapause varied from 6 weeks to more than 1 year. One year of field monitoring showed that the highest incidence of diapause was observed in larvae collected during the summer from mango leaves, where it affected approximately one-third of the individuals. This facultative diapause allows the permanent presence of P. mangiferae in the orchards. By recording diapause duration during 22 weeks under controlled conditions, we showed that high temperatures (26 °C) increased diapause duration and extended the range of the dates of diapause emergence, whereas cool temperatures (20 °C) shortened diapause duration and shortened the range of the dates of emergence from diapause. A temperature decrease from 26 to 20 °C triggered the emergence of diapausing individuals. These mechanisms ensure the synchronization of the emergence of diapausing individuals with the appearance of mango inflorescences, which is also induced by cool winter temperatures.

Among eukaryotes, sexual reproduction is by far the most predominant mode of reproduction. However, some systems maintaining sexuality appear particularly labile and raise intriguing questions on the evolutionary routes to asexuality. Thelytokous parthenogenesis is a form of spontaneous loss of sexuality leading to strong distortion of sex ratio towards females and resulting from mutation, hybridization or infection by bacterial endosymbionts. We investigated whether ecological specialization is a likely mechanism of spread of thelytoky within insect communities. Focusing on the highly specialized genus Megastigmus (Hymenoptera: Torymidae), we first performed a large literature survey to examine the distribution of thelytoky in these wasps across their respective obligate host plant families. Second, we tested for thelytoky caused by endosymbionts by screening in 15 arrhenotokous and 10 thelytokous species for Wolbachia, Cardinium, Arsenophonus and Rickettsia endosymbionts and by performing antibiotic treatments. Finally, we performed phylogenetic reconstructions using multilocus sequence typing (MLST) to examine the evolution of endosymbiont-mediated thelytoky in Megastigmus and its possible connections to host plant specialization. We demonstrate that thelytoky evolved from ancestral arrhenotoky through the horizontal transmission and the fixation of the parthenogenesis-inducing Wolbachia. We find that ecological specialization in Wolbachia's hosts was probably a critical driving force for Wolbachia infection and spread of thelytoky, but also a constraint. Our work further reinforces the hypothesis that community structure of insects is a major driver of the epidemiology of endosymbionts and that competitive interactions among closely related species may facilitate their horizontal transmission.

Orthopaedic implants experience large cyclic loads, and pre-clinical analysis is conducted to ensure they can withstand millions of loading cycles. Acetabular cup developments aim to reduce wall thickness to conserve bone, and this produces high pre-stress in modular implants. As part of an implant development process, we propose a technique for preclinical fatigue strength assessment of modular implants which accounts for this mean stress, stress concentrating features and material processing. A modular cup's stress distributions were predicted computationally, under assembly and in vivo loads, and its cyclic residual stress and stress amplitude were calculated. For verification against damage initiation in low-cycle-fatigue (LCF), the peak stress was compared to the material's yield strength. For verification against failure in high-cycle-fatigue (HCF) each element's reserve factor was calculated using the conservative Soderberg infinite life criterion. Results demonstrated the importance of accounting for mean stress. The cup was predicted to experience high cyclic mean stress with low magnitude stress amplitude: a low cyclic load ratio (Rl=0.1) produced a high cyclic stress ratio (Rs=0.80). Furthermore the locations of highest cyclic mean stress and stress amplitude did not coincide. The minimum predicted reserve factor Nf was 1.96 (HCF) and 2.08 (LCF). If mean stress were neglected or if the stress ratio were assumed to equal the load ratio, the reserve factor would be considerably lower, potentially leading to over-engineering, reducing bone conservation. Fatigue strength evaluation is only one step in a broader development process, which should involve a series of verifications with the full range of normal and traumatic physiological loading scenarios, with representative boundary conditions and a representative environment. This study presents and justifies a fatigue analysis methodology which could be applied in early stage development to a variety of modular and pre-stressed prosthesis concepts, and is particularly relevant as implant development aims to maximise modularity and bone conservation.

Most invasive species established in Europe originate from either Asia or North America, but little is currently known about the potential of the Anatolian Peninsula (Asia Minor) and/or the Near East to constitute invasion sources. Mediterranean forests are generally fragile ecosystems that can be threatened by invasive organisms coming from different regions of the Mediterranean Basin, but for which historical data are difficult to gather and the phylogeographic patterns are still poorly understood for most terrestrial organisms. In this study, we characterized the genetic structure of Megastigmus schimitscheki, an invasive seed-feeding insect species originating from the Near East, and elucidated its invasion route in South-eastern France in the mid 1990s. To disentangle the evolutionary history of this introduction, we gathered samples from the main native regions (Taurus Mountains in Turkey, Lebanon and Cyprus) and from the invaded region that we genotyped using five microsatellite markers and for which we sequenced the mitochondrial Cytochrome Oxidase I gene. We applied a set of population genetic statistics and methods, including approximate Bayesian computation. We proposed a detailed phylogeographic pattern for the Near East populations, and we unambiguously showed that the French invasive populations originated from Cyprus, although the available historical data strongly suggested that Turkey could be the most plausible source area. Interestingly, we could show that the introduced populations were founded from an extremely restricted number of individuals that realized a host switch from Cedrus brevifolia to C. atlantica. Evolutionary hypotheses are discussed to account for this unlikely scenario.

We analysed 54 alumina ceramic-on-ceramic bearings from total hip replacements retrieved at one centre after a mean duration of 3.5 years (0.2 to 10.6) in situ. These implants were obtained from 54 patients (16 men and 38 women) with a mean age of 67 years (33 to 88) who underwent revision for a variety of reasons. Posterior edge loading was found in the majority of these retrievals (32 out of 54). Anterosuperior edge loading occurred less often but produced a higher rate of wear. Stripe wear on the femoral heads had a median volumetric wear rate of 0.2 mm(3)/year (0 to 7.2). The wear volume on the femoral heads corresponded to the width of edge wear on the matching liner. Anteversion of the acetabular component was found to be a more important determinant than inclination for wear in ceramic bearings. Posterior edge loading may be considered to be a normal occurrence in ceramic-on-ceramic bearings, with minimal clinical consequences. Edge loading should be defined as either anterosuperior or posterior, as each edge loading mechanism may result in different clinical implications.

Two-stage revision surgery for infected total knee replacement offers the highest rate of success for the elimination of infection. The use of articulating antibiotic-laden cement spacers during the first stage to eradicate infection also allows protection of the soft tissues against excessive scarring and stiffness. We have investigated the effect of cyclical loading of cement spacers on the elution of antibiotics. Femoral and tibial spacers containing vancomycin at a constant concentration and tobramycin of varying concentrations were studied in vitro. The specimens were immersed and loaded cyclically to 250 N, with a flexion excursion of 45°, for 35 000 cycles. The buffered solution was sampled at set intervals and the antibiotic concentration was established so that the elution could be calculated. Unloaded samples were used as a control group for statistical comparison. The elution of tobramycin increased proportionately with its concentration in cement and was significantly higher at all sampling times from five minutes to 1680 minutes in loaded components compared with the control group (p = 0.021 and p = 0.003, respectively). A similar trend was observed with elution of vancomycin, but this failed to reach statistical significance at five, 1320 and 1560 minutes (p = 0.0508, p = 0.067 and p = 0.347, respectively). However, cyclically loaded and control components showed an increased elution of vancomycin with increasing tobramycin concentration in the specimens, despite all components having the same vancomycin concentration. The concentration of tobramycin influences both tobramycin and vancomycin elution from bone cement. Cyclical loading of the cement spacers enhanced the elution of vancomycin and tobramycin.

Biological invasions usually start with a small number of founder individuals. These founders are likely to represent a small fraction of the total genetic diversity found in the source population. Our study set out to trace genetically the geographical origin of the horse-chestnut leafminer, Cameraria ohridella, an invasive microlepidopteran whose area of origin is still unkown. Since its discovery in Macedonia 25 years ago, this insect has experienced an explosive westward range expansion, progressively colonizing all of Central and Western Europe. We used cytochrome oxidase I sequences (DNA barcode fragment) and a set of six polymorphic microsatellites to assess the genetic variability of C. ohridella populations, and to test the hypothesis that C. ohridella derives from the southern Balkans (Albania, Macedonia and Greece). Analysis of mtDNA of 486 individuals from 88 localities allowed us to identify 25 geographically structured haplotypes. In addition, 480 individuals from 16 populations from Europe and the southern Balkans were genotyped for 6 polymorphic microsatellite loci. High haplotype diversity and low measures of nucleotide diversities including a significantly negative Tajima's D indicate that C. ohridella has experienced rapid population expansion during its dispersal across Europe. Both mtDNA and microsatellites show a reduction in genetic diversity of C. ohridella populations sampled from artificial habitats (e.g. planted trees in public parks, gardens, along roads in urban or sub-urban areas) across Europe compared with C. ohridella sampled in natural stands of horse-chestnuts in the southern Balkans. These findings suggest that European populations of C. ohridella may indeed derive from the southern Balkans.

Information relevant to invasion processes and invasive alien insect species management in Central Europe was extracted from two databases: a compilation of two inventories of alien insects in Austria and Switzerland, and a list of interceptions of non-indigenous plant pests in Europe gathered by the European and Mediterranean Plant Protection Organisation (EPPO) for the period 1995-2004. For one-third of the insects established in Switzerland and Austria, the region of origin is unclear. Others come mainly from North America, Asia and the Mediterranean region. Among the intercepted insects, 40% were associated with commodities from Asia, 32% from Europe and only 2% from North America. Sternorrhyncha, Coleoptera and Psocoptera were particularly well represented in the alien fauna compared to the native fauna. In the interception database, Sternorrhyncha were also well represented but Diptera accounted for the highest number of records. Sap feeders and detritivores were the dominant feeding niches in the alien insect fauna. In contrast, external defoliators, stem borers, gall makers, root feeders, predators and parasitoids were underrepresented. Nearly 40% of the alien insects in Switzerland and Austria live only indoors. Another 15% live outdoors but exclusively or predominantly on exotic plants. Less than 20% are found mainly in 'natural' environments. The majority of introductions of alien insects in Europe are associated with the international trade in ornamental plants. An economic impact was found for 40% of the alien insects in Switzerland and Austria, whereas none is known to have an ecological impact. The implications of these observations for further studies and the management of alien species in Europe are discussed.

Phylogeographic structure of the eastern pine processionary moth Thaumetopoea wilkinsoni was explored in this study by means of nested clade phylogeographic analyses of COI and COII sequences of mitochondrial DNA and Bayesian estimates of divergence times. Intraspecific relationships were inferred and hypotheses tested to understand historical spread patterns and spatial distribution of genetic variation. Analyses revealed that all T. wilkinsoni sequences were structured in three clades, which were associated with two major biogeographic events, the colonization of the island of Cyprus and the separation of southwestern and southeastern Anatolia during the Pleistocene. Genetic variation in populations of T. wilkinsoni was also investigated using amplified fragment length polymorphisms and four microsatellite loci. Contrasting nuclear with mitochondrial data revealed recurrent gene flow between Cyprus and the mainland, related to the long-distance male dispersal. In addition, a reduction in genetic variability was observed at both mitochondrial and nuclear markers at the expanding boundary of the range, consistent with a recent origin of these populations, founded by few individuals expanding from nearby localities. In contrast, several populations fixed for one single mitochondrial haplotype showed no reduction in nuclear variability, a pattern that can be explained by recurrent male gene flow or selective sweeps at the mitochondrial level. The use of both mitochondrial and nuclear markers was essential in understanding the spread patterns and the population genetic structure of T. wilkinsoni, and is recommended to study colonizing species characterized by sex-biased dispersal.

A parallel study of the genetic structure of two oligophagous species associated with the same hosts was conducted to determine the main factors shaping the distribution of genetic diversity. The bark beetle Tomicus piniperda and the pine processionary moth (PPM) Thaumetopoea pityocampa are both associated with the genus Pinus and belong to different guilds (xylophagous vs defoliating species). The PPM is an ectophagous species that feeds on the needles of living trees, whereas T. piniperda is endophagous and bores galleries in the inner bark of weakened trees. Both species were sampled in the main regions of France, and their genetic structure was assessed after genotyping with five microsatellite markers. Populations of the PPM were significantly structured. A pattern of isolation by distance was found when distances were calculated as bypassing the Massif Central, whereas no such pattern could be found with raw geographic distances. On the contrary, most populations of T. piniperda were not differentiated. No effect of host species could be detected in either of the two species. We conclude that the two taxa have contrasting effective dispersal rates per generation, and we hypothesize that this reflects the different selection pressures acting on individual fitness via different strategies of host use.

AIM: To measure ketonemia in a control population of pregnant women and in a population of women with gestational diabetes (GDM). To define a normal ketonemia threshold for the controls and to determine whether or not this value could play a role in the clinical management of women with GDM. METHOD: Fifty-six women with a normal OGTT and 49 women with GDM were included and monitored from the 25th to the 37th week of pregnancy. Control subjects agreed to perform glycaemia and ketonemia self-monitoring 3 times a day. In addition, women with GDM were asked to measure their postprandial glycaemia. Glycaemia and ketonemia measurements were performed using Optium meters. Subjects kept a 24-hour food record twice a week. RESULTS: The mean ketonemia was lower in the control group than in the GDM group (0.01+/-0.10 vs. 0.04+/-0.009 mmol/l; P<0.001). Ketonemia values measured before the midday meal and prior to the evening meal were lower for control subjects than for GDM patients (P=0.002 and P=0.005). Fasting ketonemia was unrelated to ketonuria in the GDM group, whereas there was a correlation in the control group (P=0.006). At least one chronic increase in ketonemia levels was observed in 47% of the women with GDM, compared with only 12% of controls. The lowest levels of evening glycaemia correlated with the highest levels of ketonemia; women with GDM reported lower food and carbohydrate intakes than controls (P<0.001). CONCLUSION: This work has enabled the establishment of ketonemia reference standards in non-diabetic pregnant women. If ketonemia does indeed indicate overly restrictive dietary behavior, this parameter could be employed for monitoring adherence to the nutritional recommendations for GDM.
