Streptococcus agalactiae (S. agalactiae) is one of the main agents that causes mastitis in dairy cows, mainly inducing the subclinical form, which is characterized by a high somatic cell count (SCC). The aim of this study was to correlate the increase in SCC caused by S. agalactiae in cows with subclinical mastitis to the presence of genes related to adhesion and invasion in bovine mammary epithelial cells (BMEC) and biofilm formation. Considering the 145 isolates tested, 57.2% presented the capsular type Ia and 42.8% presented type III. We identified the virulence genes among the isolates and determined nine genetic profiles. The most common profile was identified in 69 isolates (47.5%): Ia, fbsA+, fbsB-, pI1-, pI2a-, pI2b+, and hylb+. All isolates produced biofilm, with 58.6% classified as strong producers, 29% as moderate producers and 12.4% as weak producers. No statistical correlation was found between the presence of virulence genes and increased SCC or biofilm production. However, biological evidence was observed between increased SCC and biofilm production. One isolate from each profile was randomly subjected to adhesion and invasion assays, and all of them adhered to BEMC, but none were able to invade. Our results showed that different genetic profiles do not provide advantages for bacteria to invade BMEC in vitro. In addition, biofilm production appears to be related to high SCC.

Different methods to analyze Streptococcus agalactiae biofilm formation have been investigated, but standardized protocols have not been developed. We compared S. agalactiae biofilm production among different atmospheres and growth media. Biofilm formation was studied in 32 isolates from bovine mastitis cases grown in Tryptone Soy Broth (TSB), Todd Hewitt Broth (THB), Luria Bertani Broth (LB) and Brain Heart Infusion (BHI), under two atmospheres, aerobic and 5% CO2. Regardless of the culture medium, growth under 5% CO2 resulted in a greater proportion of biofilm formation (65.63%), as compared with aerobic conditions (39.84%). Regardless of the atmosphere, the chances of biofilm formation were greater for isolates grown in TSB, as compared with THB [Odds ratio (OR) = 3.02], BHI (OR = 4.57), or LB (OR = 10.20). Thus, we suggest the use of 5% CO2 atmosphere and TSB in biofilm formation assays by Group-B streptococci (GBS) isolated from intramammary infections.

A Artrite Encefalite Caprina se caracteriza por ser multissistêmica e infecciosa, causada por um lentivírus. O estudo teve como objetivo avaliar a transmissibilidade do Lentivírus Caprino, para fêmeas e sua prole, por meio de sêmen infectado experimentalmente. Para tanto, onze fêmeas livres de CAEV foram inseminadas artificialmente com sêmen de bode livre de CAEV ao qual foi adicionado CAEV-Cork para obter título infectante com carga viral em 105 TCID50/ml. (grupo experimental 1). Destas, seis obtiverem prenhez confirmada, e a sua prole (n=6) constituiu o grupo experimental 2. Duas cabras livres de CAEV foram inseminadas artificialmente com sêmen do mesmo bode, sem o inócuo viral, constituindo-se o grupo controle. O diagnóstico da infecção pelo Lentivírus Caprino, foi realizado por IDGA, cELISA e nested-PCR. As fêmeas foram monitoradas durante 210 dias pós inseminação artificial. Já as proles foram imediatamente separadas das mães após o nascimento, e monitoradas nos momentos hora zero, aos quinze dias de idade e mensalmente, até doze meses de idade. Em relação às cabras, 56,96%(9/158) apresentaram positividade para cELISA, 24,05% (38/158) foram positivas a IDGA e nenhuma para nested-PCR. Em relação aos cabritos, 11,28% (15/133) amostras positivas para nested-PCR, 5,26% (7/133) amostras positivas para IDGA e nenhum para cELISA. As proles do grupo controle apresentaram resultados negativos para as três técnicas. A positividade encontrada em nested-PCR pode indicar grande importância para identificação de animais infectados, porém soronegativos, em situações de soroconversão tardia. De acordo com os resultados, concluiu-se que há a transmissão do Lentivírus caprino para a prole e para as mães pelo sêmen infectado.(AU)


Caprine Arthritis Encephalitis is a multisystemic infectious disease, caused by a lentivirus. The objective of this study was to evaluate the transmissibility of caprine lentivirus to goats and their offspring, through experimentally infected semen. Therefore, eleven free-CAEV goats were artificially inseminated using semen from a free-CAEV buck experimentally infected with CAEV-Cork strain (experimental group one). Pregnancy was confirmed in only six goats and their offspring (n=6) constituted the experimental group two. Two free-CAEV females were artificially inseminated with semen from the same seronegative buck, without viral inoculum to constitute the control group. The diagnosis of caprine lentivirus infection was performed using AGID, cELISA and nested-PCR. All females were monitored for 210 days after artificial insemination. Kids were immediately separated from their mothers after birth, and monitored at zero time, 15 days old and monthly until 12 months old. Regarding goat samples, 56.96% (9/159) were positive in cELISA, 24.05% (38/158) were positive in IDGA and none was positive in nested-PCR. Regarding to the offspring samples, 11.28% (15/133) and 5.26% (7/133) were positive in nested-PCR and IDGA, respectively, while no sample was positive in cELISA. The control group showed no positives in the three techniques. The positivity observed to nested-PCR may show its importance to identify infected, but seronegative animals, in late seroconversion situations. According to results, the transmission of caprine lentivirus to offspring and their mothers through infected semen is possible.(AU)

A diarreia neonatal pode ser consequência de infecções bacterianas, endoparasitarárias e virais. Enquanto esses agentes virais são extensamente estudados nos rebanhos bovinos, faltam informações sobre a importância destes para os rebanhos bubalinos brasileiros. O objetivo deste trabalho foi avaliar a presença de rotavírus e coronavírus em amostras de fezes diarreicas de búfalos (Bubalus buballis) criados em oito propriedades localizadas em Pariquera-açu, Registro, Pirassununga, Dourado, São João da Boa Vista e Congonhas, Estado de São Paulo. Foram coletadas 40 amostras de fezes diarreicas de bezerros búfalos (Bubalus bubalis). A detecção de coronavírus foi realizada pela nested-PCR, enquanto que a detecção de rotavírus foi realizada pela Eletroforese em Gel de Poliacrilamida (PAGE) com coloração com nitrato de prata. Enquanto rotavírus não foi identificado, duas amostras (2/40, 5,0%) foram positivas para coronavírus. Embora no presente trabalho tenha havido baixa detecção de coronavírus e a ausência de rotavírus nos rebanhos estudados, a possível interferência desses vírus no desenvolvimento dos quadros diarreicos não deve ser descartada. Considerando o escasso material encontrado na literatura a respeito da diarreia em bezerros búfalos, principalmente aquele relativo ao coronavírus, nossos resultados são um incentivo para que novos estudos sejam realizados para impulsionar o desenvolvimento da bubalinocultura em nosso país.(AU)


Neonatal diarrhea can be a consequence of bacterial, endoparasite and viral infections. Although virus involved in diarrhea is frequently studied in cattle herds, there is lack of studies in Brazilian buffalo herds. The aim of this study was evaluate the presence of rotavirus and coronavirus in diarrhea samples of buffaloes (Bubalus buballis) raised on eight farms in Pariquera-açu, Registro, Pirassununga, Dourado, São João da Boa Vista e Congonhas, State of São Paulo, Brazil. We collected 40 diarrhea samples from water buffalo calves. While coronavirus was detected using nested polymerase chain reaction, rotavirus was detected using Polyacrylamide Gel Electrophoresis (PAGE) with silver stain. Rotavirus was not detected, while two samples (2/40, 5.0%) were positive for coronavirus. Although we did not detect rotavirus, a low percentage of coronavirus was observed; possible interference of these viruses in the development of diarrhea should not be discarded. Considering the lack of literature about diarrhea in water buffalo calves, particularly the one related with coronavirus, our results encourage new studies to enhance buffalo health in our country.(AU)

The objectives of this study were to estimate the prevalence and incidence of subclinical mastitis (SM) in a large population of Brazilian dairy herds and to describe how these indices changed over time. A data set comprising individual cow somatic cell counts (SCC) from 18,316 test days (TD) of 1,809 herds that participated in a Dairy Herd Improvement Association (DHIA) program between January 2011 and May 2015 was available for analysis. Only tests that had ≥10 lactating cows and that were performed at 30 ± 10-d intervals were used for analysis. The final data set included 8,285 TD from 517 herds located in 5 regions of the country. Prevalence (%) of SM was defined as the number of cows with SCC ≥200,000 cells/mL divided by the total number of tested cows on a given TD. The incidence of SM was defined as the number of cows whose SCC increased from <200,000 to ≥200,000 cells/mL over 2 consecutive TD divided by the sum of each cow's days at risk during this interval, expressed as new cases per cow month at risk. Prevalence and incidence of SM were compared among years, regions, herd size categories, and frequency of DHIA testing during the study period. The overall mean prevalence and incidence of SM including all tests performed during the study period was 46.4% and 0.17 new cases per cow month at risk, respectively. The prevalence of SM varied little from 2011 to 2015, and an increasing trend was observed over the years. Prevalence was lower in herds that performed ≥60 DHIA tests during the study period than in those that performed fewer tests and was not different among regions or herd size categories. Incidence of SM varied little over the years and was not different among the regions studied. Prevalence and incidence of SM in the 517 herds studied were high and did not improve over the years. These trends were observed across all herd size categories and regions studied. Producers who had more DHIA tests performed per herd during the study period had lower prevalence of SM. Results of this study highlight the need to establish large-scale milk quality programs in Brazil.

A utilização de técnicas ultrassonográficas na área de Medicina Veterinária está cada vez mais presente e a capacitação nesta área tornou-se indispensável para o clínico, auxiliando-o na obtenção de informações rápidas e valiosas das afecções patológicas que podem acometer os animais. O exame ultrassonográfico renal revelou-se de grande importância neste âmbito, com o objetivo de avaliar e mensurar os parâmetros morfométricos renais normais de fêmeas caprinas (Capra hircus). Por meio de técnicas ultrassonográficas procedeu-se o estudo de 30 fêmeas da raça Saanen, divididas em três grupos: fêmeas com idade inferior a 6 meses (3,0±1,0 meses), de 6-18 meses (9,0±4,3 meses) e com idade superior a 18 meses (46,3±17,4 meses). Realizaram-se imagens dos rins, em secções longitudinais, medidas de comprimento e largura e, em secções transversais, medidas de altura (ou espessura). Com estes valores calcularam-se volumes renais, corticais e medulares, além da relação cortico-medular. Com relação ao comprimento renal os grupos com idade inferior a 6 meses, de 6-18 meses e com idade superior a 18 meses obtiveram média e desvio padrão de 4,20±0,36cm, 5,56±0,40cm e 6,77±0,64cm, respectivamente. Tratando-se do volume renal, estes grupos apresentaram média e desvio padrão de 17,02±3,99cm³, 19,99±5,86cm³; e, 41,23±13,05cm³. Comparou-se a equivalência métrica das médias entre os dois rins de forma que os parâmetros volumétricos e lineares renais com diferença entre si são comprimento renal, volume renal e volume cortical para o grupo de fêmeas com idade de 6-18 meses, e comprimento renal e comprimento medular para o grupo com idade superior a 18 meses. Entre diferentes grupos observou-se que somente o comprimento medular esquerdo apresentou média equivalente em todos os grupos, ou seja, entre o grupo com idade inferior a 6 meses e o grupo com idade de 6-18 meses e, entre este último e o grupo com idade superior a 18 meses. Os resultados mostraram correlações diretas e positivas entre peso corporal e idade com os parâmetros lineares e volumétricos, a relação cortico-medular esquerda foi a única que apresentou correlação significativa com o peso (r= -0,365; P = 0,047). Para aqueles parâmetros que apresentaram correlação significativa foi realizada análise de regressão, obtendo-se a linha de melhor ajuste das variáveis.


The veterinary ultrasonographic techniques have increased in the veterinary doctor's daily work, therefore training and knowing how to use ultrasound equipment have become needful to the animals healthcare professionals. It's important remember that ultrasound equipment makes possible getting valuable pathological data from animals' health. In this background, the renal ultrasonographic test has gotten an important place and, nowadays, it is possible measure with accuracy the caprine renal morphometrics parameters (Capra hircus). Through ultrasonographic techniques, it has done a stocktaking with three groups of normal goats: older than 18 months (46.3±17.4 months), younger than 6 months (3.0±1.0 months) and age between 6 and 18 months (9.0 4.3 months), totalizing 30 samples of the female Saanen kind. In longitudinal section of the kidneys, measured the length and breadth and, in transverse section, measured the height (or thickness). The values obtained calculated the renal, cortical and medullary volumes, besides corticomedullary correlation. The renal length of the groups younger than 6 months, age between 6 and 18 months and older than 18 months had mean and standard deviation of 4.20±0.36cm; 5.56±0,40cm; and, 6.77±0.64cm, respectively. To the renal volume theses groups had mean and standard deviation of 17.02±3.99cm³; 19.99±5.86cm³; and, 41.23±13.05cm³. It was compared the metrical equivalence of the mean between the two kidneys, to the group with age between 6 and 18 months the parameters showed difference were renal length, renal volume and cortical volume, in other hand to the group older than 18 months it was renal length and medullar length. Comparing different groups was noted that only the left medullar length had equivalent mean in all the groups, it means, the group younger than 6 months, older than 18 months and from 6 to 18 months of age. The results showed direct and positive correlation between corporal weight and age with the linear and volumetric parameters The left corticomedullary correlation was the only of its kind to exhibit significant correlation with the weight (r= -0.365; P value 0.047). The parameters with significant correlation had the regression analysis done, obtained the better line of adjust of the variables.
