RESUMO

Shanxi is one of the main producing areas of Forsythia suspensa in China. In order to explore the safety of the soil in the areas where Forsythia suspensa grows,70 surface (0-25 cm) soil samples were collected from the main growing areas of F. suspensa in the eastsouth of Shanxi Province in July 2017. The concentration and composition characteristics of 16 polycyclic aromatic hydrocarbons (PAHs) in the sample soils were analyzed using chemical extraction and gas chromatography-mass spectrometry (GC-MS). The diagnostic ratio method was used to determine the source of PAHs in the areas. The potential ecological risk was assessed by using the method of calculating the equivalent carcinogenic concentration of benzo[a]pyrene. The results showed that the average concentration of total PAHs (Σ16PAHs) in all of the soil samples was 1.85 µg·g-1, which was dominated by three ring number PAHs, accounting for 76.7% of the total PAHs. The detection rates of phenanthrene (Phe) and anthracene (Ant) were both 100% of all the sample sites. The soil PAHs in the wild F. suspensa growing areas mainly originated from coal, biomass burning, and motor vehicle exhaust emissions, which resulted from air transport and sedimentation pathways. In all of the sample sites, the concentration of Σ16PAHs the limit standard level (0.2 µg·g-1) of Maliszewska-Kordybach for agricultural soil pollution and exceeded the soil heavy pollution level limit value (1.0 µg·g-1) in 41.4% of the sample sites. The concentration of BaP was above the risk control standard for soil contamination of agricultural land (0.55 µg·g-1) in 10% of all the soil samples. A total of 11.4% of the sample soil ΣBaPeq16PAHs and ΣBaPeq8BPAHs exceeded the agricultural soil screening value (0.55 µg·g-1). These results indicate that the contamination of PAHs was at a detectable level in the soil of wild F. suspensa growing in Shanxi, and thus their potential ecological risks should not be ignored. It is necessary to enhance the research regarding these areas to ensure the safe production of medicinal plants.

BACKGROUND: Climate heterogeneity not only indirectly shapes the genetic structures of plant populations, but also drives adaptive divergence by impacting demographic dynamics. The variable localized climates and topographic complexity of the Taihang Mountains make them a major natural boundary in Northern China that influences the divergence of organisms distributed across this region. Opisthopappus is an endemic genus of the Taihang Mountains that includes only two spatially partitioned species Opisthopappus longilobus and Opisthopappus taihangensis. For this study, the mechanisms behind the genetic variations in Opisthopappus populations were investigated. RESULTS: Using SNP and InDel data coupled with geographic and climatic information, significant genetic differentiation was found to exist either between Opisthopappus populations or two species. All studied populations were divided into two genetic groups with the differentiation of haplotypes between the groups. At approximately 17.44 Ma of the early Miocene, O. taihangensis differentiated from O. longilobus under differing precipitation regimes due to the intensification of the Asian monsoon. Subsequently, intraspecific divergence might be induced by the dramatic climatic transformation from the mid- to late Miocene. During the Pleistocene period, the rapid uplift of the Taihang Mountains coupled with violent climatic oscillations would further promote the diversity of the two species. Following the development of the Taihang Mountains, its complex topography created geographical and ecological heterogeneity, which could lead to spatiotemporal isolation between the Opisthopappus populations. Thus the adaptive divergence might occur within these intraspecific populations in the localized heterogeneous environment of the Taihang Mountains. CONCLUSIONS: The localized environmental events through the integration of small-scale spatial effects impacted the demographic history and differentiation mechanism of Opisthopappus species in the Taihang Mountains. The results provide useful information for us to understand the ecology and evolution of organisms in the mountainous environment from population and species perspective.

Genetic diversity and differentiation are revealed particularly through spatio-temporal environmental heterogeneity. Acer ginnala, as a deciduous shrub/small tree, is a foundation species in many terrestrial ecosystems of Northern China. Owing to its increased use as an economic resource, this species has been in the vulnerability. Therefore, the elucidations of the genetic differentiation and influence of environmental factors on A. ginnala are very critical for its management and future utilization strategies. In this study, high genetic diversity and differentiation occurred in A. ginnala, which might be resulted from its pollination mechanism and species characteristics. Compared with the species level, relatively low genetic diversity was detected at the population level that might be the cause for its vulnerability. There was no significant relationship between genetic and geographical distances, while a significant correlation existed between genetic and environmental distances. Among nineteen climate variables, Annual Mean Temperature (bio1), Mean Diurnal Range (bio2), Isothermality (bio3), Temperature Seasonality (bio4), Precipitation of Wettest Month (bio13), Precipitation Seasonality (bio15), and Precipitation of Warmest Quarter (bio18) could explain the substantial levels of genetic variation (> 40%) in this species. The A. ginnala populations were isolated into multi-subpopulations by the heterogeneous climate conditions, which subsequently promoted the genetic divergence. Climatic heterogeneity played an important role in the pattern of genetic differentiation and population distribution of A. ginnala across a relatively wide range in Northern China. These would provide some clues for the conservation and management of this vulnerable species.

Opisthopappus Shih is an endemic and endangered genus restricted to the Taihang Mountains that has important ornamental and economic value. According to the Flora Reipublicae Popularis Sinicae (FRPS, Chinese version), this genus contains two species (Opisthopappus longilobus and Opisthopappus taihangensis), whereas in the Flora of China (English version) only one species O. taihangensis is present. The interspecific phylogenetic relationship remains unclear and undefined, which might primarily be due to the lack of specific molecular markers for phylogenetic analysis. For this study, 2644 expressed sequence tag-simple sequence repeats (EST-SSRs) from 33,974 unigenes using a de novo transcript assembly of Opisthopappus were identified with a distribution frequency of 7.78% total unigenes. Thereinto, mononucleotides (1200, 45.39%) were the dominant repeat motif, followed by trinucleotides (992, 37.52%), and dinucleotides (410, 15.51%). The most dominant trinucleotide repeat motif was ACC/GGT (207, 20.87%). Based on the identified EST-SSRs, 245 among 1444 designed EST-SSR primers were selected for the development of potential molecular markers. Among these markers, 63 pairs of primers (25.71%) generated clear and reproducible bands with expected sizes. Eventually, 11 primer pairs successfully amplified all individuals from the studied populations. Through the EST-SSR markers, a high level of genetic diversity was detected between Opisthopappus populations. A significant genetic differentiation between the O. longilobus and O. taihangensis populations was found. All studied populations were divided into two clusters by UPGMA, NJ, STRUCTURE, and PCoA. These results fully supported the view of the FRPS, namely, that O. longilobus and O. taihangensis should be regarded as two distinct species. Our study demonstrated that transcriptome sequences, as a valuable tool for the quick and cost-effective development of molecular markers, was helpful toward obtaining comprehensive EST-SSR markers that could contribute to an in-depth assessment of the genetic and phylogenetic relationships between Opisthopappus species.

In order to evaluate the potential agricultural risks of soil contaminated by polycyclic aromatic hydrocarbons (PAHs), Lactuca sativa L. was used as a model leaf vegetable plant to investigate the enrichment characteristics of PAHs in the different tissues of Lactuca sativa L, such as its underground parts (GS) and aboveground parts (YS), which were studied through an experiment involving potted cultivation in PAHs contaminated soil that was taken from the agricultural soil around a coking enterprise area. The concentrations of the different PAHs in the soil and plant tissues were analyzed using ultrasonic oscillation extraction and high-performance liquid chromatography-mass spectrometry (GC-MS) analysis methods. The results show that the enrichment of the total PAHs (Σ16PAHs) in the YS is higher than that in the GS. The components enriched in the YS mainly consist of 3-5 ring PAHs, and those in the GS consist of 4-6ring PAHs. The coefficients of the different PAHs enriched in the YS were higher than those of the GS. The enrichment coefficient of anthracene (Ant) was the largest and that of fluoranthene (Fla) was the smallest in the YS, while the enrichment coefficient of benzene[a]pyre (Bap) was the largest and that of Fla was the smallest in the GS. The transfer coefficients of the different PAHs from the GS to the YS were greater than the rate from the initial soil (SS) to the GS; the value is less than 1 from the SS to GS. The correlations and goodness of fit were analyzed for the concentrations of PAHs in the SS, GS and YS. The Σ16PAHs in the SS showed positive correlations with the Σ16PAHs in the YS and in GS. The goodness of fit of the correlation of the PAHs in YS to those in the SS was the largest (R2=1.0), while the goodness-of-fit for the correlation between the GS and SS was lower (R2=0.71). The benzo[a]pyrene equivalent concentration values (Σ16PAHsBapeq) of YS and GS were 11.8 and 12.7 times the maximum value of Bap level allowed in food. The study indicates that contaminated farmland soil would present a high health risk when it was used to grow the plant.

Lishan Mountain is belonged to the Zhongtiao Ranges in Southeast Shanxi, and situated between 35 degrees 16'30" - 35 degrees 27'20" N and 111 degrees 51'-- 112 degrees 5'35" E. The climate is cool and arid in winter, but warm and rainy in summer. This paper studied the species diversity and community structure of forest communities there, based on field investigation data and by using diversity, richness and evenness indices. The results showed that the species diversity and evenness of the communities were in the order of Acer davidii + Acer mono > Acer davidii + Carpinus turczaninouii > Quercus liaolugensis > Quercus varibilis > Pinus armandii + Betula albo-sinensis > Pinus tabulaeformis + Quercus glandulifera > Betula platyphylla > Platycladus orientalis + Quercus variabilis > Betula albo-sinensis > Pinus armandii > Platycladus orientalis > Pinus tabulaeformis > Populus davidiana + Betula albo-sinensis. The diversity and richness of shrub layer were higher than those of herb layer and arbor layer, but the evenness of arbor, shrub and herb layers had no significant difference. There were positive correlations (P < 0.05) between Shannon-Wiener index, Hill index, and species richness from 1 000 m to 1 920 m above sea level, but negative correlations (P < 0.05) from 1920 m to the top of the Mountain. The species diversity showed a "mid-altitude bulge" pattern with the change of altitude, and the reason was that the combination of temperature and moisture conditions from 1000 m to 1920 m above sea level was improved, and human disturbance was decreased gradually.
