RESUMO

Many species of freshwater prawns, large and small, inhabit Southeast Asian countries. In Thailand alone, there have been recent reports of new species in the genus Macrobrachium. Morphological differentiation and, recently, DNA sequence differences have been used to distinguish new species from valid ones. Macrobrachium chainatense (Decapoda: Palaemonidae), a small freshwater prawn found in Thailand, is now established as a new species based on both morphological and genetic criteria. Among Thailand Macrobrachium prawns, M. chainatense has smallest body size, largest egg size, and 3-5 teeth on the cutting edges. The most closely related Macrobrachium species to M. chainatense in terms of COI and 18S DNA sequences is M. niphanae. Their COI sequences differ by 7.3% while their 18S sequences are basically identical in all but one site. Observations of some aspects of this prawn in captivity are also utilized to establish its unique species status. This prawn may be useful as a source of cooked food for human consumption.

RESUMO

A small freshwater prawn in Thailand has been found to be a new species and is named Macrobrachium suphanense (Decapoda: Palaemonidae). Fully grown male M. suphanense appeared very different from the most closely related male of M. sintangense: size smaller, second pereiopod smaller and less robust, rostrum form different and, in females, fewer eggs. Less closely related, fully grown M. dolatum has sharper distal cutting edge on fixed finger and M. hungi has longer rostrum than M. suphanense. DNA analyses put M. suphanense, M. sintangense and M. nipponense in the same clade with M. nipponense sister to the other two. Two more clades consist of M. dienbienphuense and M. niphanae on the one hand and M. lanchesteri and M. rosenbergii on the other. The relationship among the three clades is not clearly resolved.

Air-breathing fish species of the genus Trichopsis have been reported in Cambodia, Lao PDR, Indonesia, Malaysia, Singapore, Thailand and Vietnam. It is only in Thailand that all three recognized species (Trichopsis vittata, Trichopsis schalleri and Trichopsis pumila), as judged by distinct external features, are found. Cambodia and Lao PDR harbor two species each. The present work involves first-time DNA sequencing and analysis based on mitochondrial (COI) and nuclear (RAG1) DNA of numerous specimens of these species and specimens of a controversial Phetchaburi (Thailand) fish population with a mixed outward appearance. In addition to confirming the morphologically clear-cut taxonomic division of the three fish species, our DNA results show that whereas the T. pumila populations form one single species, there are cryptic species in the T. vittata and T. schalleri populations and possibly a new one in the latter. Members of the putative Phetchaburi fish population have been proven to be hybrids between T. pumila and T. vittata. In addition, a new the phylogenetic tree indicating ancestral relationships is also presented. This study should generate further research to find new/cryptic species of the genus Trichopsis in all countries harboring the fish.

In Thailand, there are currently five recognized species members of the bubble-nesting Betta genus, namely Betta splendens, B. smaragdina, B. imbellis, B. mahachaiensis and B. siamorientalis. In 2010, we indicated the possibility, based on COI barcoding evidence, that there might be two additional species, albeit cryptic, related to the type-locality B. smaragdina in some provinces in the northeast of Thailand. In the present study, after a more extensive survey of the northeast, and phylogenetic analyses based on COI and ITS1 sequences, the B. smaragdina group may be composed of at least 3 cryptic species members. The phylogenetic positions of these B. smaragdina group members in the bubble-nesting bettas' tree together with those of their congeners have been consolidated by better DNA sequence quality and phylogenetic analyses. With a better supported tree, the species statuses of B. siamorientalis and the Cambodian B. smaragdina-like fish, B. stiktos, are also confirmed.

Fighting fish species in the genus Betta are found in several Southeast Asian countries. Depending on the mode of paternal care for fertilized eggs and hatchlings, various species of the betta fish are classified as mouth brooders or nest builders whose members in turn have been grouped according to their similarities mainly in morphology. The mouth brooders as well as some nest builders involved in the present study include fishes discovered and identified subsequent to previous reports on species groupings and their positions on phylogenetic trees based on DNA sequences that differ from those used by us in this study. From the mitochondrial COI gene and nuclear ITS1 gene sequences and more accurate analyses we conclude that the following members of the mouth-brooding pairs, named differently previously, are virtually identical, viz the Betta prima-Betta pallida pair and Betta ferox-Betta apollon pair. The Betta simplex, hitherto believed to be one species, could possibly be genetically split into 2 distinct species. In addition, several other established type-locality fishes could harbor cryptic species as judged by genetic differences. Assignments of fish species to groups reported earlier may have to be altered somewhat by the present genetic findings. We propose here a new Betta fish phylogenetic tree which, albeit being similar to the previous ones, is clearly different from them. Our gene-based evidence also leads to assignments of some fishes to new species groups and alters the positions of some species on the new phylogenetic tree, thus implying different ancestral relationships.

RESUMO

This research aimed to develop an STS-based learning unit on biological control to enhance scientific inquiry capability of secondary school students through community science projects. It was framed by the science-technology-society (STS) teaching approach, and focused on collaboration between individual students, peers, teachers, agriculturists, and local experts. Students were assessed for their scientific inquiry ability at the beginning and the end of the learning process using five instruments: experimental skill test, students' laboratory reports, students' science projects, semi-structured interview and classroom observation. The results showed that the STS-based learning unit on biological control helped students gain significant improvement in scientific inquiry. They were able to apply and integrate the scientific knowledge learned in both classroom and field studies to help solve agricultural problems in their own communities. Moreover, this learning unit encouraged students' skills in solving problems in other situations.

First clinical exposures are stressful situations for nursing students, especially, when practicing on the labour ward. The purpose of this study was to develop intrapartum nursing care web-based learning to facilitate students' acquisition of conceptual knowledge and performance skills. This web-based learning unit integrated the 5E-model and information technology with the lecture content. Eighty four nursing students were recruited in the study. The control group received traditional teaching, while the experimental group was supplemented with the web-based learning unit on intrapartum nursing care. The results showed that the students in the experimental group had significant higher scores in conceptual knowledge and performance skill. The students also had significant lower scores in ignorance - related stress when compared to those of the control group. The students supplemented with the web-based course showed a strong positive attitude toward the new learning method.

Textbook illustrations of 3D biopolymers on printed paper, regardless of how detailed and colorful, suffer from its two-dimensionality. For beginners, computer screen display of skeletal models of biopolymers and their animation usually does not provide the at-a-glance 3D perception and details, which can be done by good hand-held models. Here, we report a study on how our students learned more from using our ordered DNA and protein models assembled from colored computer-printouts on transparency film sheets that have useful structural details. Our models (reported in BAMBED 2009), having certain distinguished features, helped our students to grasp various aspects of these biopolymers that they usually find difficult. Quantitative and qualitative learning data from this study are reported.

RESUMO

This study aimed to develop a web-based learning media on the process and mechanism of labour for the third-year university nursing and midwifery students. This media was developed based on integrating principles of the mechanism of labour with the 5Es inquiry cycle and interactive features of information technology. In this study, the web-based learning unit was used to supplement the conventional lecture as in the traditional teaching. Students' achievements were assessed by using the pre- and post-test on factual knowledge and semi-structured interviews on attitude to the unit. Supplementation with this learning unit made learning significantly more effective than the traditional lecture by itself. The students also showed positive attitude toward the learning unit.

From our teaching of the contractile unit of the striated muscle, we have found limitations in using textbook illustrations of sarcomere structure and its related dynamic molecular physiological details. A hand-held model of a striated muscle sarcomere made from common items has thus been made by us to enhance students' understanding of the sliding filament mechanism as well as their appreciation of the spatial arrangements of the thick and thin filaments. The model proves to be quite efficacious in dispelling some alternative conceptions held by students exposed previously only to two-dimensional textbook illustrations and computer graphic displays. More importantly, after being taught by this hand-held device, electronmicrographic features of the A and I bands, H zone, and Z disk can be easily correlated by the students to the positions of the thick and thin elements relatively sliding past one another. The transverse expansion of the sarcomere and the constancy of its volume upon contraction are also demonstrable by the model.

RESUMO

Due to constraints of increasing number of nursing students and a disproportionate shortfall of faculty members in nursing schools, it was necessary to supplement traditional lecture or lecture-demonstration with on-line multimedia materials and/or multimedia compact discs for enhancing learning. The authors have developed a computer-assisted learning (CAL) multimedia on vital signs with animation and audio features for teaching in the classroom based on the 5Es inquiry cycle. When the CAL was tried on second-year undergraduates in two comparable schools, all groups of students gained significantly higher performance skills regardless of whether the groups were subjected to CAL/lecture or CAL/lecture/demonstration. However, they did not gain in factual knowledge.

Haloperoxidase enzymes are of interest for basic and applied bioscientists because of their increasing importance in pharmaceutical industry and environmental cleanups. In a guided inquiry-based laboratory experiment for life-science, agricultural science, and health science undergraduates, the bromoperoxidase from a red seaweed was used to brominate bromophenol red, a novel starting substrate. Substrate and enzyme concentration dependence of this enzyme-catalyzed reaction as followed colorimetrically yielded initial rates of reaction that the students compared with those of their peers. The students worked as collaborative groups partially designing their own experiments and carrying them out. In performing the laboratory experiment, they were minimally guided by the instructor and teaching assistants. To engage the students before the laboratory, a short activity involving the enzyme-induced color change was carried out. At the end of the laboratory session, student groups discussed their results in front of the class and reached their own conclusions. Most students had better understanding of important concepts in enzyme kinetics and showed good attitude toward the overall student-centered laboratory exercise.

Instructions are given for building physical scale models of ordered structures of B-form DNA, protein α-helix, and parallel and antiparallel protein ß-pleated sheets made from colored computer printouts designed for transparency film sheets. Cut-outs from these sheets are easily assembled. Conventional color coding for atoms are used for both types of biopolymers. Arrows facilitate following chain direction for the polypeptides. For DNA, the 5' to 3' direction is guided by a 5' phosphate group and a free hydroxyl group. Important chiral centers, for example, α-carbon, deoxyribose C1', are easily made. The main advantages of this version of DNA are the proportional major and minor grooves as in the actual molecule. More importantly, because of transparency of the film one can see successive base-pair stacking very clearly and also the sense of relative base-pair rotation. Because of the introduction of two central metal wire axes, the model of B-form DNA can be twisted to give a rather good representation of A-form and even a semblance of a left-handed helix. The models of secondary structure of protein allow a better insight into the axial alignment of side chains, the formation of hydrogen bond, the handedness of the α-helix, and the backbone connection between the ß-strands. Students taught by these models understand 3D features of the biopolymers better than from textbook illustrations, computer graphic representations, and even common paper and plastic versions.
