RESUMO

Objective@#To investigate the awareness of Internet based sex education and associated among middle school students, and to provide scientific basis to improve sex education using online resources.@*Methods@#By using stratified cluster random sampling method, 449 students from 8 middle schools in Zhejiang Province were investigated with a self designed questionnaire by Chi square test, Logistic regression, and Importance Performance Analysis (IPA).@*Results@#The awareness of Internet based sex education was 66.59%(299), among which 17.15%(77) chose "WeChat", 7.35%(33) chose "sex education website", and 42.09%(189) chose"video platform" as the primary resources of online sex education, respectively. In the sexual health literacy of middle school students, the correct rate in sexual safety and sexual assault was higher than 40%, with STD transmission and prevention higher than 70%. The correct rate was more than 80% when it came to sexual physiology and sexual development. Logistic regression analysis showed that higher score of sexual health literacy was associated with higher awareness of Internet based sex education ( OR =1.06, 95% CI =1.01-1.11, P <0.05). According to the IPA, the teaching effect and content in area A (the dominant area) had higher scores of importance and satisfaction.The funniness of content in area B (retention area) had a lower importance score and a higher satisfaction score. The practicality and interactivity in area C (opportunity area) had low scores of importance and satisfaction. The curriculum model in area D (improvement area) scored higher in importance and lower in satisfaction.@*Conclusion@#The awareness of Internet based sex education needs to be improved among middle school students. Efforts should be made to improve sexual health literacy. Online platforms providing sex education should focus on improving the relevant course model, strengthening the practicability and interactivity of the course, and continue to maintain the advantage of teaching effect, teaching content and interesting aspects of the course.

RESUMO

The SARS-CoV-2 Omicron with increased fitness is spreading rapidly worldwide. Analysis of cryo-EM structures of the Spike (S) from Omicron reveals amino acid substitutions forging new interactions that stably maintain an "active" conformation for receptor recognition. The relatively more compact domain organization confers improved stability and enhances attachment but compromises the efficiency of viral fusion step. Alterations in local conformation, charge and hydrophobic microenvironments underpin the modulation of the epitopes such that they are not recognized by most NTD- and RBD-antibodies, facilitating viral immune escape. Apart from already existing mutations, we have identified three new immune escape sites: 1) Q493R, 2) G446S and 3) S371L/S373P/S375F that confers greater resistance to five of the six classes of RBD-antibodies. Structure of the Omicron S bound with human ACE2, together with analysis of sequence conservation in ACE2 binding region of 25 sarbecovirus members as well as heatmaps of the immunogenic sites and their corresponding mutational frequencies sheds light on conserved and structurally restrained regions that can be used for the development of broad-spectrum vaccines and therapeutics.

Emergence of variants of concern (VOC) with altered antigenic structures and waning humoral immunity to SARS-CoV-2 are harbingers of a long pandemic. Administration of a third dose of an inactivated virus vaccine can boost the immune response. Here, we have dissected the immunogenic profiles of antibodies from 3-dose vaccinees, 2-dose vaccinees and convalescents. Better neutralization breadth to VOCs, expeditious recall and long-lasting humoral response bolster 3-dose vaccinees in warding off COVID-19. Analysis of 171 complex structures of SARS-CoV-2 neutralizing antibodies identified structure-activity correlates, revealing ultrapotent, VOCs-resistant and broad-spectrum antigenic patches. Construction of immunogenic and mutational heat maps revealed a direct relationship between "hot" immunogenic sites and areas with high mutation frequencies. Ongoing antibody somatic mutation, memory B cell clonal turnover and antibody composition changes in B cell repertoire driven by prolonged and repeated antigen stimulation confer development of monoclonal antibodies with enhanced neutralizing potency and breadth. Our findings rationalize the use of 3-dose immunization regimens for inactivated vaccines. One sentence summaryA third booster dose of inactivated vaccine produces a highly sifted humoral immune response via a sustained evolution of antibodies capable of effectively neutralizing SARS-CoV-2 variants of concern.

Objective:To comprehensively and systematically measure and analyze deaf children's gray matter cortex and white matter fibers by surface-based morphometry (SBM) and tract-based spatial statistics(TBSS).Methods:Twenty-seven deaf children aged 9-13 years old and twenty-seven age and sex matched normal controls were selected. T1 structural images and diffusion tensor imaging data were collected and analyzed by SBM and TBSS to calculate the cortical thickness, back index and anisotropic index (fractional anisotropy, FA). The SPSS 20.0 software and FSL software were used for data analysis.Results:Compared with the control group, the thickness of the cortex in the left cerebral hemisphere central posterior gyrus, the superior lobule, the central lateral lobule, and the anterior lobe were significantly reduced(cluster size 4 150, P<0.05), and in the right cerebral hemisphere squats and the middle sacral region reduced(cluster size 2 592, P<0.05). The local regression index of the left anterior wedge region was significantly increased(cluster size 3 225, P<0.05). The DTI results showed a decrease in FA values in the areas of radiation crown, cortical bundle, cingulate gyrus, corpus callosum, thalamus radiation, and sub-occipital bundle( P<0.05, TFCE corrected). Conclusion:There are different degrees of damage in the cerebral cortex and white matter microstructure of deaf children, and the brain structure remodeling and compensatory reconstruction appeared in the anterior wedge, which provide strong evidence for in-depth study of relationship between the loss of auditory function and changes in the brain structure.

Objective:To explore the relationship between executive dysfunction and brain functional connectivity in deaf children and its mechanisms.Methods:Twenty-eight deaf children were recruited from special education schools and twenty-seven children with normal hearing were treated as controls matched with deaf children in age, sex and education. All human subjects were scheduled to complete brain magnetic resonance scanning, based on the voxel-based morphometry (VBM) technique for detecting differences in gray matter volume (GMV) and a set of executive function (EF) tasks for detecting differences in EF behavior performance between deaf and normal hearing children. All human subjects were also performed resting-state fMRI, which were analyzed by independent component analysis (ICA) and the executive control network was selected and compared between the deaf and normal hearing children.Results:Compared with the children with normal hearing, deaf children had smaller volume of gray matter in the left angular gyrus (MIN coordinates: -48, -72, 45; 700 voxels), right superior parietal gyrus (MIN coordinates: 20, -62, 57; 594 voxels) and left middle occipital gyrus(MIN coordinates: -32, -87, 42; 782 voxels). Deaf children had weaker functional connectivity (FC) in the left middle frontal gyrus, orbital part/left inferior frontal gyrus (MIN coordinates: -24, 21, 3; 54 voxels) with normal hearing children. In the behavioral test, deaf children had defects in the switching function compared with normal children, which was statistically significant ( t(42)=2.39, P=0.02). Conclusion:The EF of deaf children has some defects, and its neural mechanism may be related with the reduced functional connectivity and gray matter volume in the brain regions related to executive function.

Objectives To investigate the relationship between the degeneration of lumbar multifidus mus-cles and facet joint osteoarthritis(FJOA)by MRI. Methods This retrospective study included 248 patients with low back and leg pain,aged from 55 to 70.MRI was used to measure TCSA,FCSA,fCSA and FI% of the L4~5and L5~S1segments on the both sides of their multifidus muscle in the cross section in T2weighted image.All the cases were divided into three grades:grade 0~1,grade 2 and grade 3 according to their FJOA grades. The relationship between the FJOA grades at the same level and TCSA,FCSA and FI% of both sides was analyzed retrospectively. Results There were significant differences of the FI% of the multifidus muscle between both sides of the two seg-ments(P < 0.05);The TCSA,FCSA and fCSA of the left and right multifidus muscle all showed an increasing trend from L4~5to L5~S1segment and the difference was statistically significant(P<0.05);the FI% of two sides of multifidus muscle was negatively correlated with the right FJOA grade at L5~S1segment(P<0.05).There was a re-lationship between females and right FJOA grade at L5~S1segment(all P < 0.05). Whereas,no significant differ-ence was found in other parameters(all P>0.05).Conclusions There is no relationship between the cross-sec-tional area of the multifidus muscle and the FJOA grade. The higher FI% degree of multifidus muscle in the L5~S1 segment is accompanied with a lower grade of FJOA.The degeneration of multifidus muscle occurs earlier than the degeneration of articular process joints,so the earlier exercise of lumbar muscle is recommended.

RESUMO

Objective To investigate the therapeutic mechanism of bazedoxifene, the third-generation selective ER modulator (SERM), on endometriosis lesions in a rat model. Methods Endometriosis was induced by transplanting pieces of endometrium from other syngeneic rats that were as donors onto the subcutaneous of other unmated female rats. The rats with successful ectopic implants were divided into two groups:control group (n=10) and bazedoxifene group (n=10). The macroscopic morphology, volume, histopathology of ectopic implant and rats uterine wet weight were determined before and after the treatment. Expression of proliferation cell nuclear antigen (PCNA), ER and PR in the eutopic endometrium and endometriosis lesions detected by immunohistochemistry in the two groups. Results (1) The gross morphology and histological changes of endometriosis lesions in rats after treatment: compared with the control group, it was obviously depauperated and had more less glands and blood vessels in the stroma. (2) The change of rats′weight, the volume of endometriosis lesion before and after treatment and rats uterine wet weigh after treatment respectively in the control group and the bazedoxifene group:rats′ weight were respectively before treatment: (201±17) g, (202±18) g, that were respectively after treatment: (266±16) g, (261±16) g, which showed no significant difference between two groups before and after treatment (P>0.05). The volume of ectopic implant before treatment were respectively (85±17) mm3, (85±12) mm3, and showed no significant difference between two groups;that were respectively (48±11) mm3, (24±9) mm3 afte rtreatment, which was significantly decreased compared with the control group (P0.05). Conclusion Bazedoxifene could obviously reduce the size of endometriosis lesions, the mechanism may be related with suppressing estrogen-induced proliferation, the expression of ER and direct ER antagonism by this SERM.
