RESUMO

This experiment was conducted to evaluate the effects of feeding a Bacillus-based direct-fed microbial (DFM) on performance, nutrient digestibility, rumen fermentation, and metabolic response of lactating dairy cows. Sixty-eight lactating (50 ± 6 d in milk) Holstein-Friesian (n = 20) and Swedish Red (n = 48) cows were enrolled to a 15-wk experiment. Cows were blocked by breed, lactation number, and days in milk and, within blocks, assigned to 1 of the 2 treatments: (1) basal partial mixed ration (PMR) without DFM (n = 34; CON) or (2) basal PMR with the addition of 3 g/head per day of a DFM containing Bacillus licheniformis and Bacillus subtilis (n = 34; DFM). The DFM was mixed in a protein pellet, whereas the CON group was fed the same pellet without DFM (1 kg/cow per day). The PMR contained 53% clover grass silage and 47% compound feed plus 3 kg of a concentrate (dry matter basis) offered during milking. Milk yield and production efficiency were recorded daily, whereas milk samples were collected for 24 h every second week of the study for milk composition. During the experimental period, fecal, rumen fluid, and blood samples were collected from each cow for apparent nutrient digestibility, rumen fermentation, and metabolic responses, respectively. All data were analyzed using the MIXED procedure of SAS. No treatment effects were observed on cows final body weight and daily dry matter intake. However, cows fed DFM had a greater milk yield, milk production efficiency, lactose and total solids yield, and also tended to have a greater energy-corrected milk production efficiency and milk protein yield. No significant differences were observed on nutrient digestibility and total volatile fatty acids, but molar proportion of acetate was greater for cows fed DFM. In contrast, molar proportion of propionate was greater and butyrate tended to be greater for CON. Cows fed DFM had greater mean plasma insulin-like growth factor-I (IGF-I), but no differences were observed for plasma glucose and insulin. In summary, supplementing a Bacillus-based DFM benefited productive responses of lactating dairy cows, while also modulating rumen fermentation and serum IGF-I.

Volatile fatty acids (VFAs) supplementation in ruminants' diet as a source of energy and chemical precursors and their effect on animal's physiology and well-being has long been of scientific interest. Production of VFAs through anaerobic digestion of agro-industrial residues not only creates value but also presents an alternative sustainable approach for ruminant feed supplementation. Therefore, this study aimed to investigate the bioconversion of agro-industrial residues produced in large quantities such as apple pomace (AP), thin stillage (Ts), and potato protein liquor (PPL) to VFAs, fully complying to regulations set for ruminant feed supplement production. In this regard, batch acidogenic fermentation assays (pH 6-10) and semi-continuous immersed membrane bioreactor (iMBR) were applied. In batch assays, at pH 10 the co-digestion of Ts and PPL produced the highest VFAs concentration (14.2 g/L), indicating a yield of 0.85 g CODVFAs/g volatile solids (VS)added. The optimum batch condition was then applied in the iMBR for in situ fermentation and recovery of VFAs at different organic loading rates (OLR). With increasing the OLR to 3.7 gVS/L.day, the highest VFAs concentration of 28.6 g/L (1,2 g CODVFAs /gVSadded) was achieved. Successful long-term (114 days) membrane filtration was conducted in a media with a maximum of 40 g/L of total solids (TS), facing irreversible membrane fouling in the final stages. Acidogenic fermentation using an iMBR has the potential to play an important role in the future of feed additive provision through the biorefining of agro-industrial wastes via the carboxylate platform, given the role of VFAs production from organic residues.

A control strategy against Fasciola hepatica infection based on selective treatment of non-lactating animals was evaluated in four Swedish dairy herds. The study was conducted over the course of two consecutive seasons in moderately to highly F. hepatica infected herds with robotic milking, where heifers and dry cows received an oral drench with albendazole (10 mg/kg) during three visits in January, February and March in both 2017 and 2018. This resulted in an anthelmintic coverage between 38 % and 58 % of the animals. Furthermore, on each visit, the infection status of all dewormed animals along with 15 randomly selected milking cows were monitored by detection of F. hepatica coproantigens. Individual milk samples were also collected quarterly from the whole herds for measurements of individual antibody levels against the parasite using milk ELISA. In addition, individual data on milk yield and quality were collected on a monthly basis between 2016 and 2018. To further study the impact of the infection on milk production, truly F. hepatica positive and negative cows in the first lactation were identified based on the results from coproantigen and milk ELISA assays. Total F. hepatica coproantigen prevalence in the herds varied between 28 % and 85 % in the first year, and between 27 % and 68 % in the second year of the study. We found that two years of treatments resulted in a significant decrease of coproantigen-positivity especially on the two most heavily infected farms. These results were confirmed by a similar drop in within-herd prevalences obtained by milk ELISA results. The infection had a significant negative impact on milk yields in untreated F. hepatica positive cows. No consistent long-term effect was observed at the herd level probably due to the influx of animals infected before puberty and/or adult animals that were re-infected at dry-off. This is the first study of the effects of F. hepatica infection on milk yield and quality in dairy herds in Sweden.
