RESUMO

In order to prove that porous piles are effective in remediating muddy tidal flat sediments and increasing the biomass, field experiments were carried out at the tidal flat of a brackish river located in Hiroshima City, Japan. Porous piles with a diameter of 16cm and height of 50cm were installed in the muddy sediment that covers the sand layer of the tidal flat. After installation, concentrations of dissolved oxygen in interstitial water in and around the porous piles increased to a maximum concentration of 6mg/l due to enhancement of the groundwater flow. The increase of dissolved oxygen in the interstitial water produced a decrease in the concentration of ammonia and an increase in the individual number of benthos at the porous pile site. From these results, we concluded that the porous pile is an effective technology for remediation of muddy tidal flats.

Edaravone is a neuroprotective agent with a potent free radical scavenging and antioxidant actions. In the present study we investigated the influence of edaravone on cisplatin ototoxicity in auditory cells. Cell viability was determined using a 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide cell proliferation assay. Oxidative stress and apoptosis were assessed by reactive oxygen species (ROS) measurement, Hoechst 33258 staining, caspase-3 activity assay, and immunoblotting of PARP. Pretreatment with 100 µM of edaravone prior to application of 15 µM of cisplatin increased cell viability after 48 h of incubation in HEI-OC1 cells (from 51.9% to 64. 6% viability) and also, attenuated the cisplatin-induced increase in reactive oxygen species (ROS) (from 2.3 fold to 1.9 fold). Edaravone also decreased the activation of caspase-3 and reduced levels of cleaved poly-ADP-ribose polymerase (PARP). We propose that edaravone protects against cisplatin-induced ototoxicity by preventing apoptosis, and limiting ROS production in HEI-OC1 cells.

BACKGROUND AND OBJECTIVES: The purpose of this study is to identify the effectiveness of intratympanic (IT) steroids when used as an initial therapy in sudden sensorineural hearing loss (SSNHL), and when it is used as a salvage therapy in a refractory SSNHL. SUBJECTS AND METHODS: This retrospective study included 122 patients with SSNHL. First, patients were divided into three groups according to the initial treatment; IT steroid, systemic steroid and combined (systemic and IT steroid) groups. Second, patients were divided into two groups according to the salvage treatment in a refractory SSNHL; IT steroid and the control (non IT group). Fifty eight patients who did not respond to initial therapy were included in the second analysis. Hearing was assessed immediately before the treatment and 2 weeks, 4 weeks and 3 months after the treatments. Hearing recovery was defined as an improvement of >15 dB and the final hearing of 25-45 dB in the audiogram. RESULTS: When we analyzed the hearing recovery in initial treatment, the comparison of audiogram among three groups did not result in significantly different outcomes. There were no differences in the recovery rate in 2 and 4 weeks throughout all the frequencies. Also, the analysis of the salvage treatment demonstrated that neither IT steroid group nor control group was significantly effective in treating the refractory SSNHL. CONCLUSIONS: The results suggest that initial treatment of SSNHL with IT steroid alone is as effective as systemic steroid alone or a combination therapy. Also, salvage IT steroids for refractory SSNHL did not have any additional beneficial effects.

Alkalin-reduced water (ARW) is known to exert several anti-cancer effects, as well as to scavenge reactive oxygen species (ROS) and reduce blood-glucose levels. This study was performed in order to determine the effects of ARW on the control of spontaneous diabetes in Otsuka Long-Evans Tokushima Fatty (OLETF) rats. We assigned 16 male OLETF rats (4 wk) to two groups: an experimental group, which was given ARW, and a control group, which received laboratory tap water. From week 6 to 32, the body weight, lipid composition, and glucose levels in the blood of the rats were measured. The glucose levels of both groups tended to increase. However, the ARW group's glucose levels were significantly lower than those of the control group after 12 weeks (p<0.05). The total cholesterol and triglyceride levels in the ARW group were found to be significantly lower than those of the control group during the experimental period. These results suggest that ARW spurred the growth of OLETF rats during the growth stage, and that long-term ingestion of ARW resulted in a reduction in the levels of glucose, triglycerides, and total cholesterol in the blood.
