RESUMO

Curcumin has been used in traditional medicine forages. The present study aimed to develop a curcumin-based hydrogel system and assess its antimicrobial potential and wound healing (WH) activity on an invitro and in silico basis. A topical hydrogel was prepared using chitosan, PVA, and Curcumin in varied ratios, and hydrogels were evaluated for physicochemical properties. The hydrogel showed antimicrobial activity against both gram-positive and gram-negative microorganisms. In silico studies showed good binding energy scores and significant interaction of curcumin components with key residues of inflammatory proteins that help in WH activity. Dissolution studies showed sustained release of curcumin. Overall, the results indicated wound healing potential of chitosan-PVA-curcumin hydrogel films. Further in vivo experiments are needed to evaluate the clinical efficacy of such films for wound healing.

Activity coefficient values offer insight into the intermolecular interactions between the solute and the solvent and the deviation from the ideal behavior. CO2 capture from different industrial processes is a globally pertinent issue and the search for suitable chemicals is required. To address the issue, knowledge of activity coefficient values is crucial for CO2 separation-based process. In this regard, a correlation is developed that predicts the coefficient of CO2 activity in ionic liquids by multi-nonlinear regression analysis. The correlation is developed between the pressure range of 1-50 bar and the temperature range of 298.15-33.15 K for mole fractions of 0.3, 0.5, and 0.7. Outliers' analysis is performed using the boxplot method to determine the suitability of ranges of the selected input parameters. The preceding literature does not predict the activity coefficient in relatively lower to higher temperature and pressure ranges for CO2 solubility in ionic liquids. Initially, the activity coefficient values from COSMO-RS were obtained and compared with the correlation results. The COSMO-RS and the correlation predicted results were subsequently validated with the experimental data. The average absolute error (AAE%) of the predicted correlation values is 19.53% while the root mean square error (RMSE) value is 0.465. The correlation can be used in the future to predict the CO2 activity coefficient values in ionic liquids to facilitate qualitative analyses of their CO2 capture efficiency.

Lepidoptera is the second most diverse insect order outnumbered only by the Coeleptera. Acetylcholinesterase (AChE) is the major target site for insecticides. Extensive use of insecticides, to inhibit the function of this enzyme, have resulted in the development of insecticide resistance. Complete knowledge of the target proteins is very important to know the cause of resistance. Computational annotation of insect acetylcholinesterase can be helpful for the characterization of this important protein. Acetylcholinesterase of fourteen lepidopteran insect pest species was annotated by using different bioinformatics tools. AChE in all the species was hydrophilic and thermostable. All the species showed lower values for instability index except L. orbonalis, S. exigua and T. absoluta. Highest percentage of Arg, Asp, Asn, Gln and Cys were recorded in P. rapae. High percentage of Cys and Gln might be reason for insecticide resistance development in P. rapae. Phylogenetic analysis revealed the AChE in T. absoluta, L. orbonalis and S. exigua are closely related and emerged from same primary branch. Three functional motifs were predicted in eleven species while only two were found in L. orbonalis, S. exigua and T. absoluta. AChE in eleven species followed secretory pathway and have signal peptides. No signal peptides were predicted for S. exigua, L. orbonalis and T. absoluta and follow non secretory pathway. Arginine methylation and cysteine palmotylation was found in all species except S. exigua, L. orbonalis and T. absoluta. Glycosylphosphatidylinositol (GPI) anchor was predicted in only nine species.

OBJECTIVE: This study aimed to investigate the knowledge, usage and attitude of physiotherapists towards non-steroidal anti-inflammatory drugs (NSAIDs) in Karachi, Pakistan. METHODS: A cross-sectional study was conducted among clinical physiotherapists working in Karachi who were enrolled through convenient sampling. A questionnaire was distributed to the participants, which consisted of questions regarding physiotherapists' current knowledge, current practice and their attitude towards NSAIDs prescription. RESULTS: Responses of 310 physiotherapists out of the 350 questionnaires distributed were received (response rate = 88%). Results show that most physiotherapists (60.3%) recommended NSAIDs to their patients despite having a variable understanding of the contraindications, adverse reactions and drug interactions of NSAIDs. Most physiotherapists demonstrated poor knowledge of the legislation on NSAIDs prescription as most of them believed that they were allowed to advise (71.9%), prescribe (66.1%) and recommend (72.2%) NSAIDs to their patients. CONCLUSION: This study implicates that physiotherapists should be trained through proper educational sessions such as certification courses on pharmacotherapy (specifically pain medications) in order to become certified to prescribe NSAIDs and other analgesics.
