RESUMO

This study aimed to evaluate the antibacterial, leishmanicidal, and cytotoxic potential of metabolites produced by bacteria isolated from rhizosphere soil samples. The bacterium was identified by genome sequencing as Streptomyces kronopolitis. A preliminary screening was carried out for the antimicrobial activity of S. kronopolitis, demonstrating activity against Staphylococcus aureus ATCC 6538, Corynebacterium diphtheriae ATCC 27010, C. diphtheriae ATCC 27012, and Mycobacterium abscessus, with inhibition halos of sizes 25, 36, 29, and 33 mm, respectively. To obtain secondary metabolites, the bacteria were subjected to submerged fermentation, and the metabolites were extracted using the liquid-liquid method with ethyl acetate. There was a similar MIC for M. abscessus and the two strains of C. diphtherium, reaching a concentration of 12.5 µg/mL, while that of S. aureus was 0.048 µg/mL. Assays for leishmanicidal activity and cytotoxicity against HEp-2 cells and red blood cells were performed. The metabolite showed an IC50 of 9.0 ± 0.9 µg/mL and CC50 of 221.2 ± 7.0 µg/mL. This metabolite does not have hemolytic activity and is more selective for parasites than for mammalian cells, with a selectivity index of 24.6. Thus, the studied metabolite may be a strong candidate for the development of less toxic drugs to treat diseases caused by pathogens.

ABSTRACT: This study focused on analyzing the inclusive advances for the population of Special Education in ordinary classrooms in Basic Education, considering teachers' training need in this specific area. It consists of a quantitative and qualitative approach study, using as empirical sources microdata from Prova Brasil, 2021 edition, more specifically the questionnaires applied to teachers of the 5th grade of Elementary School, which are available on the webpage of the National Institute of Educational Studies and Research Anísio Teixeira (INEP). The extraction of this information and statistical analysis were carried out with the aid of the Statistical Package for Social Science (SPSS). The result of the investigation reveals that the percentage of enrollments for students with disabilities, autism spectrum disorder (ASD) and giftedness in ordinary classrooms, in Brazil, between 2013 and 2019, rose 7.4%. It also points out that 86.5% of the teachers in the investigated sample declared that they had moderate to high training needs. It was concluded that the population of Special Education still needs to face some issues in favor of a fairer and more inclusive school to everyone, which implies fighting so that the principles of equality and equity materialize both in the real field and in the ideal field.

RESUMO

The relationship between helminth infection and allergic diseases has long intrigued the scientific community. This interaction was previously studied in a horse family with high incidence of severe equine asthma and in non-related severely asthmatic horses from equine hospital referrals in Switzerland. Our aim was to determine if this interaction would also be observed in a group of non-related client-owned severely asthmatic horses living in a Mediterranean climate and recruited through a first-opinion veterinarian group. Fecal samples from severe equine asthma-affected and healthy horses living in the same farms and subjected to identical environmental and deworming management were evaluated qualitatively and quantitatively. Strongyle-type eggs and Cyathostomum sensu latum larvae were the most abundant parasites in the studied population of horses; no significant differences between the groups were observed regarding the types of egg and infective larvae. However, we observed significant differences in the number of eggs and infective larvae per gram of feces shed, as this number was significantly lower in the SEA group than in the healthy horses. This may indicate that severely asthmatic horses have an intrinsic resistance to gastrointestinal helminths. Further studies in a larger population of horses are required to ascertain the immunological mechanisms responsible for these findings.

RESUMO

From previous studies, the International Society for Equitation Science has advised that further research be conducted on the physiological/psychological effects of less-exacerbated poll flexion angles. We aimed to evaluate the effects of two riding poll flexion positions with a difference of only 15° on the respiratory systems and behaviour of horses through an evaluation of dynamic airway collapse via over-ground endoscopy, the pharyngeal diameter, pleural pressure, arterial oxygenation and lactate, HR/RR, and the occurrence of conflict behaviours. Twenty high-level dressage and twenty show-jumping horses underwent a 40 min ridden test at a ground angle of 85°; 3 weeks later, they underwent a ridden test at a 100° ground angle (the angle between the ground and the line from the forehead to the muzzle) and in a cross-over design. Using a mixed model for repeated measures, Wilcoxon/Friedman tests were carried out according to the experimental design and/or error normality. For both groups, at 100°, conflict behaviours and upper airway tract abnormalities were significantly more frequent, and the pleural pressure was higher, and the pharyngeal diameter was lower. At 85°, relaxation behaviours were significantly more frequent. Lactate was significantly higher at 100° only in the dressage horses. Compared to the first test at 85°, the HR/RR were significantly lower at the beginning of the second test (at 100°) but higher at the end. The significant differences identified in these dressage and show-jumping horses support the idea that an increase of just 15° in riding poll flexion can have negative effects on the respiratory system and behaviour of a horse and therefore on its welfare.

RESUMO

BACKGROUND: Studies in human medicine have concluded that acetazolamide reduces pain associated with carbon dioxide insufflation during laparoscopic surgery. However, there are no published reports regarding the use of acetazolamide for this purpose in companion animals, despite the increasing popularity of laparoscopic techniques in veterinary medicine due to their advantages over open surgeries. OBJECTIVES: Thirty mixed-breed female dogs were included in the study and randomly assigned to one of three groups: OVE (median celiotomy ovariectomy; n = 10), OVEL (laparoscopic ovariectomy, n = 10) and OVELA (laparoscopic ovariectomy with acetazolamide preoperative administration; n = 10). Experienced surgeons performed all procedures, and the anaesthetic and analgesic protocols were identical for all animals. Acetazolamide was administered orally (at a dose of 25 mg/kg) 2 h prior to induction in the OVELA group. Postoperative pain was evaluated using serum cortisol, salivary cortisol, and the University of Melbourne Pain Scale (UMPS) Score. RESULTS: Any statistical differences were observed in the UMPS scores when the OVELA group was compared to the OVEL group at 1 h after surgery (p = 0.515), 12 h (p = 0.375) and 24 h (p = 0.242). Animals undergoing open surgery (OVE group) had significantly higher pain scores at all times after surgery when compared with OVEL and OVELA groups. A high positive correlation (r = 0.792; p = 0.01) was found between serum and saliva cortisol concentrations. Mean saliva cortisol concentration was not significantly lower for the OVELA group compared to the other groups. CONCLUSIONS: This study found evidence that preoperative administration of acetazolamide may be beneficial in managing postoperative pain in dogs after laparoscopic surgeries. However, further research with a larger sample size is needed to confirm this and to determine if acetazolamide should be included in a multimodal postoperative analgesia protocol for laparoscopic ovariectomy in dogs.

RESUMO

The objective of this study is to analyze the validity evidence of the Academic Procrastination Scale (APS) in a sample consisting of 1979 students from public and private universities, from different courses, 67% of the students being women, aged 18 to 68 years old (M = 22.84). The principal component analysis revealed two components, also validated by confirmatory analysis, which showed a good overall fit of a two-dimensional model: Daily Study Procrastination (a = 0.75) and Exam Study Procrastination (a = 0.75). Internal consistency was estimated through Cronbach's alpha, obtaining 0.83 for the total scale. The study concludes that APS has satisfactory psychometric properties and suggests further validation studies and also analysis of academic procrastination in higher education students. (AU)

BACKGROUND: Neurofibromatosis type 1 (NF1) is a chronic and progressive autosomal dominant genetic and sporadic disease characterized by cutaneous and neurological abnormalities. Plexiform neurofibroma (PN), a significant cause of clinical complications in NF-1, is a benign tumor of the peripheral nerve sheath that involves multiple nerve fascicles. Although there is an important number of patients who are affected by NF1 in Brazil, there is little data on the behavior of the disease in the national literature as well as in other low- and middle-income countries. METHODS: We performed a retrospective analysis of 491 patients with NF1 followed at two reference centers in Brazil. RESULTS: Approximately 38% of patients had PNs, resulting in reduced life quality. The median patient age with PNs was 30 years (range: 6 to 83 years). Head and neck, and extremity were the main affected locations with 35.8 and 30.6%, respectively. PNs were classified as asymptomatic in 25.1% of patients, while 52.5% presented symptomatic and inoperable tumors. The most common manifestations related to PNs were disfigurement and orthopedic involvement. Twenty patients developed neoplasms and ten (50%) presented with malignant peripheral nerve sheath tumors (MPNST). The prevalence of MPNST in our study was 2.9%. CONCLUSIONS: Patients with NF1 experience clinically significant morbidity, especially when it is associated with PN. Though there are many patients affected by NF1 in Brazil and other low- and middle-income countries, there is little data available in the corresponding literature. Our results are comparable to the previous results reported from higher-income countries and international registries.

RESUMO

This paper aimed to study the feasibility of a new laparoscopic castration technique in male dogs, evaluate the pain associated with it, and compare it with the classical orchiectomy. Surgical times, pain scores, blood and salivary cortisol, and CRP were recorded and compared between the two groups. The use of high-frequency bipolar forceps allowed quick and uneventful laparoscopic procedures. The laparoscopic group had significantly lower pain scores, cortisol, and PCR values than the orchiectomy group. No complications were seen in any group. Our results suggest that this laparoscopic castration is a safe and beneficial surgical alternative to traditional orchiectomy in dogs.

RESUMO

Severe equine asthma (SEA) syndrome is a chronic recurrent respiratory disease, common among adult horses. The disease occurs in genetically susceptible individuals after their exposure to organic dust. Thus, environmental management has proved essential in controlling airway challenge and disease exacerbation. This is a demanding process that can only be achieved through the horse owners' cooperation. One year after initial diagnosis of SEA in a group of 39 horses, owner compliance to an environmental management protocol was evaluated. The overall compliance to the protocol was poor and the horses' clinical health and need for pharmacological management was related to the successful implementation of the environmental recommendations provided on disease diagnosis.

RESUMO

Objectives We aimed to evaluate children with type 1 diabetes (T1D) with early age at onset (EAO) for clinical, immune and metabolic features in order to identify age-related disease phenotypes. Methods Comparative study of two groups of T1D children: EAO (≤5 years) and later age at onset (LAO; >5 years), regarding the presence of other autoimmune (AI) diseases, diabetes ketoacidosis and immunologic profile at onset and metabolic data 1 year after diagnosis. Statistical analysis was performed with significance set for p < 0.05. Results The study included 137 children (EAO = 52, mean age 3.6 ± 1.5 [mean ± standard deviation (SD)] and LAO = 85, mean age 10.4 ± 2.9). EAO was more associated with concomitant AI diseases (p = 0.032). Despite no differences in disease onset, EAO presented with lower C-peptide levels (p = 0.01) and higher absolute lymphocyte number (p < 0.0001), with an inverse correlation between these two variables (p = 0.028). Additionally, the EAO group had a higher frequency of serum detection of three antibodies (Abs) (p = 0.0008), specifically insulin Abs (p = 0.0001). One year after diagnosis, EAO had higher total daily insulin (TDI) dose (p = 0.008), despite similar hemoglobin A1c (HbA1c). Conclusions Our data show an association of EAO T1D with more AI diseases, higher number of Abs, lower initial insulin reservoir and higher insulin requirements 1 year after diagnosis. In this group, immune imbalance seems more evident and disease progression faster, probably reflecting distinct "immune environment" with different ages at disease onset. Further studies in the field of immunogenetics and immune tolerance are required, to improve patient stratification and find novel targets for therapeutic intervention.

RESUMO

Staging methods are useful tools for monitoring disease and response to treatment, and because Severe Equine Asthma Syndrome (SEAS) has a high prevalence in the equine population, a clinical staging method can provide important information to optimize equine care. Our team has previously developed and published a clinical staging method for SEAS and in the present study we further evaluated information provided by lung function tests, in order to determine their contribution to disease staging. Using discriminant analysis we set out to produce a new staging method with applicability in the field. Differences between group means (P < .05) were observed for clinical score, bronchoalveolar lavage fluid neutrophil percentage, pleural pressure (ΔPpl), PaO2 and histamine concentration and the linear functions obtained explained 99.3% of the data variability, with 94.7% of cases grouped correctly and a cross-validation of 86.8%. Thus this staging model showed very good results and the discriminant linear functions may be used to identify and stage SEAS. This method can be used in the field and also in diagnostic and research centres.

RESUMO

An elective laparoscopic ovariectomy on a healthy dog revealed a cystic structure in the left ovary. The surgical procedure was successful. Histopathological examination showed the presence of a teratoma adjacent to the ovary. To the best of the authors' knowledge, this is the first reported case of an ovarian teratoma removed by laparoscopic ovariectomy in a dog by using a multiport laparoscopic ovariectomy technique.

RESUMO

Premenopausal breast cancer (BC) is a core tumor of Li-Fraumeni (LFS) and Li-Fraumeni-like (LFL) Syndromes, predisposition disorders caused by germline mutations in TP53 gene. In the Southern and Southeastern regions of Brazil, a specific TP53 germline mutation, c.1010G>A (p.Arg337His), was identified at a population frequency of 0.3%, the highest value ever described for a TP53 germline variation. In Brazilian BC patients, carrier frequency can vary from 0.5% to 8.7%. The current study assessed carrier frequency by genotyping TP53 c.1010G>A in 2 BC groups: 1) 315 patients unselected for age of diagnosis and family history (FH) and 2) 239 patients diagnosed before 46 years and without Chompret criteria for LFS or LFL. One carrier was identified in group 1 (0.3%; CI 95% 0.1-1.76%) and six carriers in group 2 (2.5%; CI 95% 0.93-5.39%). The frequencies differed significantly between groups (p = 0.04). The mutation carrier frequency observed in group 2 could justify mutation testing in BC patients diagnosed before 46 years and without Chompret criteria for LFS or LFL. Further studies in larger samples of BC patients of different ages and regions of the country are necessary to provide more definitive TP53 p.Arg337His carrier frequencies in different scenarios.

jective: To develop an economic evaluation of lipegfilgrastin, long acting G-CSF, compared to other available G-CSF to reduce the duration of severe neutropenia (SN) and the incidence of febrile neutropenia (FN) in adult patients treated with cytotoxic chemotherapy for cancer in the Brazilian private Health System. Methods: Systematic literature review of scientific evidence evaluating the efficacy and safety of lipegfilgrastin and cost-minimization analysis comparing to pegfilgrastin or filgrastin in the Brazilian private Health System. The analysis includes time of treatment with filgrastin from clinical trials (scenario 1) or real world data (scenario 2 and 3) and considers direct medical costs. Results: Six studies were included in the analysis: three randomized controlled trials and three systematic reviews. Lipegfilgrastin resulted in a statistically significant reduction of median SN duration compared to placebo and non-inferior compared to pegfilgrastin. A study that performed an indirect comparison of lipegfilgrastin and filgrastin did not find any difference statistically significant reduction of SN duration and FN incidence. Lipefilgrastin resulted in a cost-difference of -R$ 3.673,50/patient compared to pegfilgrastin in all scenarios and -R$ 57.403,40; -R$ 19.183,67 and -R$ 42,50/patient compared to filgrastin in scenario 1, 2 and 3 respectively. Conclusions: Lipegfilgrastim showed a favorable cost-minimization profile compared to pegfilgrastin or filgrastin and is an important alternative treatment in reducing the duration of neutropenia and the incidence of febrile neutropenia during the course of chemotherapy, and may result in resource savings for the Brazilian Private Health System.

RESUMO

Canine dirofilariosis is a life-threatening parasitic disease that is increasingly reported worldwide. Once diagnosed the main treatment goals are to improve the animal's clinical condition and to eliminate all life stages of the parasite with minimal posttreatment side effects. This can be achieved through mechanical, surgical, or chemotherapeutical approaches. Currently, manual extraction is the preferred method to remove adult heartworms due to its diminished invasiveness, reduced damage to the vascular endothelium, and shortened anaesthesia duration. However, it remains an expensive technique that can be highly traumatic. To address this issue, a nontraumatic homemade catheter-guided snare was developed for heartworm removal by adapting and folding a 0.014-inch coronary wire (BMW, Abbott Vascular). Transvenous heartworm extraction was performed on a dog severely infected with adult heartworms by inserting the modified snare into a 6-F Judkins right coronary guiding catheter BMW (Cordis) and advancing it into the right ventricle under fluoroscopic guidance. Fifteen adult specimens of Dirofilaria immitis were successfully extracted from the pulmonary artery and right ventricle without complications. To assure the death of both larvae and adults, postoperative treatment was successfully managed using ivermectin, doxycycline, and melarsomine, with no recurrence after surgery.

Incorporation of native areas to production systems induced changes in ecosystems, especially in tropical regions, due to inappropriate land use, resulting in the rapid loss of organic carbon. This study aimed to evaluate impact of soil management and land use on biomass and microbial activity, as well as soil organic carbon and its fractions in an Oxisol and Quartzipsamment in Cerrado. Management and land use systems in Quartzipsamment were native vegetation, crop-livestock integration, pasture, no-till 1 (soybean/Brachiaria) and two no-till 2 (corn/Brachiaria). In Oxisol the systems were native vegetation, crop-livestock integration, conventional tillage, no-till 3 (soybean /radish/corn/radish) and no-till 4 (soybean/ corn/sorghum/soybean). Both soils evaluated, integrated crop-livestock improved the microbial biomass levels and soil carbon pools. In Quartzipsamment, management and land use systems not increased microbial biomass carbon, urease activity, hydrolysis of fluorescein diacetate and the carbon stock of the light fraction. In Oxisol, conventional tillage system decreased soil organic carbon and total carbon in the free light fraction soil. Furthermore, although it increased the organic carbon soil losses, the soil conservation systems enhanced the light fraction carbon in relation to native vegetation.

This is a cross-sectional study conducted to investigate quality of life, job satisfaction, presence of mental disorders, and level of physical activities among primary care workers in Montes Claros, Minas Gerais, Brazil. Validated instruments were used (WHOQOLBref, Effort-Reward Imbalance, General Health Questionnaire, and the International Physical Activity Questionnaire). Descriptive and bivariate analyses were performed. Occupational categories were aggregated into three groups in bivariate analysis (General Services/Community Health Workers; Technicians/assistants; nurses/dentists/doctors). 752 workers (95% of the total) were evaluated ? 470 Community Health Workers; 69 Technicians/Nursing Assistants; 51 Technicians/Dental Assistants; 52 Dentists; 71 Nurses; and 39 Doctors. The mean age was 32 years (± 8.3) - 80% were women. About 20% had job dissatisfaction. The Social Relations and Environmental dimensions had respectively the least and greatest impact on quality of life. 67% showed an imbalance between effort and reward at work. About 16% had mental disorders and 22% were sedentary / insufficiently active. Job satisfaction, effort-reward imbalance, mental health, and physical activity level showed statistically significant differences (p<0.05) among the three occupational groups. The results call attention to psychosocial risks posed for these workers, demonstrating the need for encouraging healthy lifestyles.

RESUMO

BACKGROUND/AIMS: In a previously described model of isolated rabbit eye, we detected myogenic intrinsic vascular tone of unknown origin in the ophthalmic artery. In order to better understand the origin of these low frequency oscillations, we analyzed their spectral characteristics using fast Fourier. METHODS: Hybrid New Zealand rabbits of either sex (n = 24) were used; they were divided into 2 groups according to age. The spectral characteristics of the myogenic behaviour of the rabbit external ophthalmic artery were analyzed using the fast Fourier algorithm. RESULTS: The frequency of the oscillations of the myogenic activity seen in the rabbit external ophthalmic artery varied between 0.033 and 0.066 Hz (mean 0.045 ± 0.012 Hz), all in the region of very low frequency (VLF) oscillations (VLF <0.07 Hz for the rabbit). The frequency of spontaneous oscillations was higher in younger animals. CONCLUSION: Fast Fourier analysis proved to be an adequate mathematical tool to analyze the myogenic tone oscillations, which were all in the range of VLF in the model we used. These results indicate that myogenic vascular function of ocular blood flow is composed of VLF oscillations, and they provide a new explanation for the origin of VLF in arterial spectra. They also suggest that the ocular local myogenic vascular function observed is more efficient in younger animals.

RESUMO

Enterococci are ubiquitous microorganisms found as part of the normal intestinal microbiota of many animals such as the free-ranging Eurasian otter (Lutra lutra Linnaeus, 1758). In this work, twenty-nine enterococci isolated from fecal samples of Eurasian otters free-living in reservoirs and associated river stretches in South Portugal were identified and typed by conventional/molecular methods and screened for virulence factors and antibiotic resistance. Identification allocated the isolates to the species Enterococcus faecalis (19), E. faecium (9) and E. durans (1) and PCR-fingerprinting revealed their high genomic diversity. Regarding virulence factors, three isolates produced cytolysin and six were gelatinase-positive. Genes ace and acm were detected in five enterococci each, ebpABC in seventeen, gelE in fourteen and cylA in three. All isolates showed resistance patterns and antibiotic resistance genes tet(M) and pbp5 were detected in seventeen isolates each, whereas vanB and vanD were identified in thirteen and five, respectively, being most van-harboring isolates members of E. faecium. The aac(6')-Ie-aph (2″) gene, encoding for gentamicin resistance, was observed in all gentamicin-resistant enterococci. Since all isolates harbor virulence and/or antibiotic resistance traits, the role of free-living Eurasian otters in the dissemination of virulent/resistant enterococci among other animals sharing the same ecological niche cannot be disregarded, as well as the health risk they may represent for humans directly interacting with them or their habitat.
