Weaning is a critical stage in the swine production cycle, as young pigs need to adjust to sudden and dramatic changes in their diet and environment. Among the various organ systems affected, the gastrointestinal tract is one of the more severely impacted during this transition. Traditionally, challenges at weaning have been managed by prophylactic use of antibiotics, which not only provides protection against diarrhea and other gut dysfunction but also has growth-promoting effects. With banning or major restrictions on the use of antibiotics for this purpose, various alternative products have been developed as potential replacements, including direct-fed microbials (DFMs) such as probiotics and postbiotics. As their efficiency needs to be improved, a continued effort to gain a deeper understanding of their mechanism of action is necessary. In this context, this report presents a study on the impact of a Lactobacillus-based probiotic (LPr) and a Bifidobacterium-based postbiotic (BPo) when added to the diet during the nursery phase. For animal performance, an effect was observed in the early stages (Day 0 to Day 10), as pigs fed diets supplemented with either DFMs were found to have higher average daily feed intake (ADFI) compared to pigs fed the control diet (p < 0.05). Histological analysis of intestinal morphology on D10 revealed that the ileum of supplemented pigs had a higher villus height/crypt depth ratio (p < 0.05) compared to controls, indicating a benefit of the DFMs for gut health. In an effort to further explore potential mechanisms of action, the effects of the DFMs on gut microbial composition were investigated using fecal microbial communities as a non-invasive representative approach. At the bacterial family level, Lactobacillaceae were found in higher abundance in pigs fed either LPr (D10; p < 0.05) or BPo (D47; p < 0.05). At the Operational Taxonomic Unit (OTU) level, which can be used as a proxy to assess species composition, Ssd-00950 and Ssd-01187 were found in higher abundance in DFM-supplemented pigs on D47 (p < 0.05). Using nucleotide sequence identity, these OTUs were predicted to be putative strains of Congobacterium massiliense and Absicoccus porci, respectively. In contrast, OTU Ssd-00039, which was predicted to be a strain of Streptococcus alactolyticus, was in lower abundance in BPo-supplemented pigs on D47 (p < 0.05). Together, these results indicate that the DFMs tested in this study can impact various aspects of gut function.

Bioaerosols originating from animal feeding operations (AFOs) may carry pathogens, allergens, and other hazardous biocomponents, such as endotoxins, posing a potential risk to community health and the environment when dispersed downwind. This review summarizes and synthesizes existing literature data on bioaerosols downwind from three major types of AFOs (swine, poultry, and cattle), covering their composition, concentration, dispersion patterns, measurement methodologies, potential health effects, and mitigation strategies. While many of these bioaerosols are typically detected only near AFOs, evidence indicates that certain bioaerosols, particularly viruses, can travel up to tens of kilometers downwind and remain infectious. Despite the critical importance of these bioaerosols, a refined modeling framework to simulate their transport and fate in downwind air has not yet been developed, nor have source attribution methods been established to track their origins in complex agricultural environments where multiple bioaerosols could co-exist. Therefore, it is imperative to further research downwind bioaerosols from AFOs, including their assessment, modeling, source attribution, and mitigation, to address the public health and environmental challenges associated with animal agriculture.

The human enteric nervous system, ENS, is a large network of glial and neuronal cell types with remarkable neurotransmitter diversity. The ENS controls bowel motility, enzyme secretion, and nutrient absorption and interacts with the immune system and the gut microbiome. Consequently, developmental and acquired defects of the ENS are responsible for many human diseases and may contribute to symptoms of Parkinson's disease. Limitations in animal model systems and access to primary tissue pose significant experimental challenges in studies of the human ENS. Here, a detailed protocol is presented for effective in vitro derivation of the ENS lineages from human pluripotent stem cells, hPSC, using defined culture conditions. Our protocol begins with directed differentiation of hPSCs to enteric neural crest cells within 15 days and yields diverse subtypes of functional enteric neurons within 30 days. This platform provides a scalable resource for developmental studies, disease modeling, drug discovery, and regenerative applications.

PURPOSE: Physiological aging is associated with microvascular dysfunction, including in the penis, and this may contribute to age-related erectile dysfunction (ED). Low-intensity extracorporeal shockwave therapy (Li-ESWT) is a non-invasive intervention for ED, but its effect on penile microvascular function, remains unclear. Our objectives are to (i) evaluate the effect of Li-ESWT (specifically radial type ESWT [rESWT]) on penile microvascular perfusion (PMP) in aging rats, (ii) elucidate a possible mechanism, and (iii) evaluate its impact on angiogenic and smooth muscle biomarkers in cavernosal tissue. MATERIALS AND METHODS: Male rats (n=9; 15-18 months) were anesthetized and subjected to rESWT while monitoring PMP. The nitric oxide (NO) pathway involvement was assessed by measuring the effect of rESWT on PMP following an intracavernosal injection of N(G)-nitroarginine methyl ester (L-NAME) (NO synthase inhibitor). To elucidate the cellular mechanism, another group of rats received repeated rESWT (n=4) or no treatment (n=4) three times/week for two weeks. Rats were euthanized at the end of the study and penile tissues were analyzed for angiogenic markers (vascular endothelial growth factor-A [VEGF-A], endothelial nitric oxide synthase [eNOS]) and smooth muscle content (α-actin) using immunostaining, Western blot, and quantitative polymerase chain reaction (qPCR). RESULTS: rESWT resulted in more than a 2-fold increase in PMP (from 68.5 arbitrary units; 163.7 AU). L-NAME injection produced a <40%-50% decrease (185.3 to 101.0 AU) in rESWT-induced PMP response. Immunostaining revealed increased α-actin, eNOS, and VEGF-A in the cavernosum and these findings were confirmed by qPCR and Western blot results. CONCLUSIONS: rESWT improved PMP, which may be mediated via increased VEGF expression, which stimulates the NO/cyclic guanosine monophosphate pathway, resulting in sustained PMP. rESWT devices could offer a safe, non-invasive treatment for age-related ED.

The enteric nervous system (ENS) is a tantalizing frontier in neuroscience. With the recent emergence of single cell transcriptomic technologies, this rare and poorly understood tissue has begun to be better characterized in recent years. A precise functional mapping of enteric neuron diversity is critical for understanding ENS biology and enteric neuropathies. Nonetheless, this pursuit has faced considerable technical challenges. By leveraging different methods to compare available primary mouse and human ENS datasets, we underscore the urgent need for careful identity annotation, achieved through the harmonization and advancements of wet lab and computational techniques. We took different approaches including differential gene expression, module scoring, co-expression and correlation analysis, unbiased biological function hierarchical clustering, data integration and label transfer to compare and contrast functional annotations of several independently reported ENS datasets. These analyses highlight substantial discrepancies stemming from an overreliance on transcriptomics data without adequate validation in tissues. To achieve a comprehensive understanding of enteric neuron identity and their functional context, it is imperative to expand tissue sources and incorporate innovative technologies such as multiplexed imaging, electrophysiology, spatial transcriptomics, as well as comprehensive profiling of epigenome, proteome, and metabolome. Harnessing human pluripotent stem cell (hPSC) models provides unique opportunities for delineating lineage trees of the human ENS, and offers unparalleled advantages, including their scalability and compatibility with genetic manipulation and unbiased screens. We encourage a paradigm shift in our comprehension of cellular complexity and function in the ENS by calling for large-scale collaborative efforts and research investments.

BACKGROUND: The proper surgical modality for large non-obstructing proximal ureteral stones is disputed. We compare effectiveness and safety of flexible ureteroscopic lithotripsy (FURL) and tubeless percutaneous nephrolithotomy (TPNL) in treatment of upper ureteral stones larger than 1.5 cm. METHODS: We reviewed the medical records of patients who performed FURL or TPNL for upper ureteral stones between June 2016 and November 2018. Comparative analysis was conducted regarding demographic parameters, stone free rate, postoperative pain and complications. RESULTS: This study included 58 patients treated with FURL and 60 patients treated with TPNL owing to upper ureteral stones larger than 1.5 cm. Stone size was similar in the FURL and TPNL groups (17.6 ± 2.6 vs. 18.0 ± 2.1 mm, p = 0.194). The overall 3-month stone clearance rate was 95.8% for FURL versus 96.0% for TPNL (p = 0.575). There was no difference between the FURL and TPNL groups for hospital stay (p = 0.280) and postoperative complications. On the other hand, patients treated with FURL had longer operative time (p = 0.012) and less postoperative pain (p = 0.008). CONCLUSIONS: Both surgical techniques were considered feasible and effective surgical procedures in the treatment of large upper ureteral stones.

A total of 1,170 pigs (PIC 800 × PIC, initially 38.6 kg) were used in a 98-d grow-finish study to determine the performance response of pigs fed increasing levels standardized ileal digestible (SID) Trp:Lys ratio in diets containing 40% dried distillers grains with solubles (DDGS). Five dietary treatments were fed and consisted of a corn-soybean meal (SBM) diet or diets containing 40% DDGS with SID Trp:Lys ratios of 15%, 18%, 21%, or 24%. Each treatment was replicated 9 times and pens contained 26 pigs with the equal number of gilts and barrows. Data was analyzed as a randomized complete block design with previous nursery treatment as a random blocking factor. Pair-wise comparisons were used to evaluate the impact of diets on performance and carcass traits; specifically to compare the corn-SBM dietary treatment against other dietary treatments. Single degree of freedom orthogonal polynomials were used to evaluate the dose-response of increasing the SID Trp:Lys ratio in 40% DDGS diets. Increasing the SID Trp:Lys ratio in diets containing 40% DDGS increased (linear, P ≤ 0.02) average daily gain (ADG), average daily feed intake (ADFI), final body weight (BW), hot carcass weight (HCW), carcass gain, and daily carcass gain. Increasing the SID Trp:Lys ratio in 40% DDGS diets did not impact (P ≥ 0.65) gain to feed (G:F) for the cumulative period. Pigs fed the corn-SBM diet had greater (P ≤ 0.01) ADG and a heavier (P ≤ 0.01) final BW compared to pigs fed 40% DDGS diets, regardless of the SID Trp:Lys ratio. Diets that contained 40% DDGS with a SID Trp:Lys ratio of 24% had no difference (P|t| = 0.25) in ADFI compared to the corn-SBM dietary treatment. The cumulative G:F of pigs did not differ (P = 0.32) between pigs fed corn-SBM diets and diets containing 40% DDGS regardless of the SID Trp:Lys ratio. However, pigs receiving the corn-SBM diets had heavier HCW, greater carcass yields, total carcass gain, and daily carcass gain (P < 0.01) compared to pigs fed the 40% DDGS diets regardless of SID Trp:Lys ratio. In conclusion, increasing the SID Trp:Lys ratio in 40% DDGS diets improved ADG and ADFI for the overall grow-finish period. However, when compared to a corn-SBM diet, increasing the SID Trp:Lys ratio in 40% DDGS diets did not correct the growth performance or carcass characteristics of pigs.


Corn by-products from the ethanol industry, such as dried distillers grains with solubles (DDGS), can be an economical feed ingredient in pig rations. However, when pigs are fed high dietary levels of DDGS, their growth performance can decrease. This decrease may be a result of the protein composition of the diet and more specifically the dietary amino acid composition. In the present study, the amino acid, tryptophan, was incrementally increased in diets containing 40% DDGS. The increase in dietary tryptophan resulted in increased pig feed intake and growth rate of pigs. These results suggest that increasing the dietary tryptophan level can help mitigate a portion of the decreased growth performance seen by pigs consuming higher dietary levels of DDGS. However, feeding 40% DDGS still resulted in a lower cumulative growth rate compared to a standard corn soybean meal diet. This indicates that other nutritional strategies are required to restore growth the performance of pigs fed 40% DDGS relative to a standard corn soybean meal diet, but increasing dietary tryptophan can help to partially restore growth performance.

Considering that pigs are naturally weaned between 12 and 18 weeks of age, the common practice in the modern swine industry of weaning as early as between two and four weeks of age increases challenges during this transition period. Indeed, young pigs with an immature gut are suddenly separated from the sow, switched from milk to a diet consisting of only solid ingredients, and subjected to a new social hierarchy from mixing multiple litters. From the perspective of host gut development, weaning under these conditions causes a regression in histological structure as well as in digestive and barrier functions. While the gut is the main center of immunity in mature animals, the underdeveloped gut of early weaned pigs has yet to contribute to this function until seven weeks of age. The gut microbiota or microbiome, an essential contributor to the health and nutrition of their animal host, undergoes dramatic alterations during this transition, and this descriptive review aims to present a microbial ecology-based perspective on these events. Indeed, as gut microbial communities are dependent on cross-feeding relationships, the change in substrate availability triggers a cascade of succession events until a stable composition is reached. During this process, the gut microbiota is unstable and prone to dysbiosis, which can devolve into a diseased state. One potential strategy to accelerate maturation of the gut microbiome would be to identify microbial species that are critical to mature swine gut microbiomes, and develop strategies to facilitate their establishment in early post-weaning microbial communities.

A study was conducted to identify the effects of standardized ileal digestible (SID) branched-chain amino acids (BCAA):lysine (Lys) ratios on the growth performance, plasma nitrogen (N) profile, carcass traits, and economic analysis of growing-finishing pigs fed diets with high corn distillers dried grains with solubles (cDDGS) inclusions. A total of 1,140 pigs (initial body weight [BW] = 28.7 ± 2.0 kg) were housed in 45 pens of 25 or 26 pigs and fed one of five diets in a randomized complete block design. Experimental diets were fed in four phases based on BW. Dietary treatments were a corn-soybean meal (SBM) based diet (PC), a corn-SBM-cDDGS-based diet (NC) with SID BCAA:Lys ratio of PIC (2020) recommendation and NC diets with SID BCAA:Lys ratios targeted for the 73% SID Val:Lys, 60% SID Ile:Lys, and 144% SID Leu:Lys during the growing phases (25 to 80 kg, Grow), targeted for the 78% SID Val:Lys, 70% SID Ile:Lys, and 160% to 170% SID Leu:Lys during the finishing phases (80 to 120 kg, finish), and both during the growing and finishing phases (Grow-Finish). One pig from each pen was bled at the end of 7 and 13 wk. After the 11-wk-feeding trial, pigs were sent to a commercial abattoir to investigate carcass traits. Pigs fed the Finish diet had a greater overall average daily gain (P < 0.05) than pigs fed the other cDDGS diets. Dietary treatments did not affect the hot carcass weight. However, feeding the Finish diet increased (P < 0.05) the iodine value of pork belly samples and decreased (P < 0.05) carcass yield. The plasma urea nitrogen (PUN) concentration at the end of the growing phase and plasma concentrations of Leu and Val were greater (P < 0.05) in pigs fed the Finish diet compared to the other cDDGS diets. Feeding pigs the cDDGS diets with different BCAA:Lys ratios had no difference in income over feed cost and income over feed and facility costs compared to the corn-SBM diet. Therefore, feeding pigs cDDGS diets with SID BCAA:Lys ratios adjusted for the previously determined finishing phase (from 80 to 120 kg of BW) recommendations by SBM inclusion supported growth performance and economic benefits equal to the corn-SBM diet.

Disorders of gut-brain interaction (DGBIs), formerly known as functional gastrointestinal disorders, are extremely common and historically difficult to manage. This is largely because their cellular and molecular mechanisms have remained poorly understood and understudied. One approach to unravel the molecular underpinnings of complex disorders such as DGBIs is performing genome wide association studies (GWASs). However, due to the heterogenous and non-specific nature of GI symptoms, it has been difficult to accurately classify cases and controls. Thus, to perform reliable studies, we need to access large patient populations which has been difficult to date. Here, we leveraged the UK Biobank (UKBB) database, containing genetic and medical record data of over half a million individuals, to perform GWAS for five DGBI categories: functional chest pain, functional diarrhea, functional dyspepsia, functional dysphagia, and functional fecal incontinence. By applying strict inclusion and exclusion criteria, we resolved patient populations and identified genes significantly associated with each condition. Leveraging multiple human single-cell RNA-sequencing datasets, we found that the disease associated genes were highly expressed in enteric neurons, which innervate and control GI functions. Further expression and association testing-based analyses revealed specific enteric neuron subtypes consistently linked with each DGBI. Furthermore, protein-protein interaction analysis of each of the disease associated genes revealed protein networks specific to each DGBI, including hedgehog signaling for functional chest pain and neuronal function and neurotransmission for functional diarrhea and functional dyspepsia. Finally, through retrospective medical record analysis we found that drugs that inhibit these networks are associated with an increased disease risk, including serine/threonine kinase 32B drugs for functional chest pain, solute carrier organic anion transporter family member 4C1, mitogen-activated protein kinase 6, and dual serine/threonine and tyrosine protein kinase drugs for functional dyspepsia, and serotonin transporter drugs for functional diarrhea. This study presents a robust strategy for uncovering the tissues, cell types, and genes involved in DGBIs, presenting novel predictions of the mechanisms underlying these historically intractable and poorly understood diseases.

For commercial swine producers, the natural variation in body weight amongst pigs in a herd presents a challenge in meeting the standards of meat processors who incentivize target carcass weights by offering more favorable purchase prices. Body weight variation in a swine herd is evident as early as birth, and it is typically maintained throughout the entire production cycle. Amongst the various factors that can affect growth performance, the gut microbiome has emerged as an important factor that can affect efficiency, as it contributes to vital functions such as providing assimilable nutrients from feed ingredients that are inedible to the host, as well as resistance to infection by a pathogen. In this context, the objective of the study described in this report was to compare the fecal microbiomes of light and heavy barrows (castrated male finishing pigs) that were part of the same research herd that was raised under commercial conditions. Using high-throughput sequencing of amplicons generated from the V1-V3 regions of the 16S rRNA gene, two abundant candidate bacterial species identified as operational taxonomic units (OTUs), Ssd-1085 and Ssd-1144, were found to be in higher abundance in the light barrows group. Ssd-1085 was predicted to be a potential strain of Clostridium jeddahitimonense, a bacterial species capable of utilizing tagatose, a monosaccharide known to act as a prebiotic that can enhance the proliferation of beneficial microorganisms while inhibiting the growth of bacterial pathogens. OTU Ssd-1144 was identified as a candidate strain of C. beijerinckii, which would be expected to function as a starch utilizing symbiont in the swine gut. While it remains to be determined why putative strains of these beneficial bacterial species would be in higher abundance in lower weight pigs, their overall high levels in finishing pigs could be the result of including ingredients such as corn and soybean-based products in swine diets. Another contribution from this study was the determination that these two OTUs, along with five others that were also abundant in the fecal bacterial communities of the barrows that were analyzed, had been previously identified in weaned pigs, suggesting that these OTUs can become established as early as the nursery phase.

The neural crest (NC) is highly multipotent and generates diverse lineages in the developing embryo. However, spatiotemporally distinct NC populations display differences in fate potential, such as increased gliogenic and parasympathetic potential from later migrating, nerve-associated Schwann cell precursors (SCPs). Interestingly, while melanogenic potential is shared by both early migrating NC and SCPs, differences in melanocyte identity resulting from differentiation through these temporally distinct progenitors have not been determined. Here, we leverage a human pluripotent stem cell (hPSC) model of NC temporal patterning to comprehensively characterize human NC heterogeneity, fate bias, and lineage development. We captured the transition of NC differentiation between temporally and transcriptionally distinct melanogenic progenitors and identified modules of candidate transcription factor and signaling activity associated with this transition. For the first time, we established a protocol for the directed differentiation of melanocytes from hPSCs through a SCP intermediate, termed trajectory 2 (T2) melanocytes. Leveraging an existing protocol for differentiating early NC-derived melanocytes, termed trajectory 1 (T1), we performed the first comprehensive comparison of transcriptional and functional differences between these distinct melanocyte populations, revealing differences in pigmentation and unique expression of transcription factors, ligands, receptors and surface markers. We found a significant link between the T2 melanocyte transcriptional signature and decreased survival in melanoma patients in the cancer genome atlas (TCGA). We performed an in vivo CRISPRi screen of T1 and T2 melanocyte signature genes in a human melanoma cell line and discovered several T2-specific markers that promote lung metastasis in mice. We further demonstrated that one of these factors, SNRPB, regulates the splicing of transcripts involved in metastasis relevant functions such as migration, cell adhesion and proliferation. Overall, this study identifies distinct developmental trajectories as a source of diversity in melanocytes and implicates the unique molecular signature of SCP-derived melanocytes in metastatic melanoma.

PURPOSE: To evaluate the efficacy of percutaneous nephrolithotomy (PCNL) access tract sealing agent. Fibrin glue and Tachosil® were used for sealing the access tract and compared to the control. Post operative computed tomography (CT) scan was used to evaluate those efficacies. METHODS: A total of 108 patients were randomized to three groups: In group 1, the access tract was sutured, and compressive dressing was done. In group 2, the fibrin glue was injected into the access tract with a tip applicator at the end of operation. And group 3, Tachosil® was rolled on its longitudinal axis and plugged into the access tract. Non-contrast CT at POD 1 was taken and perirenal hematoma thickness was measured and graded. Hemoglobin, hematocrit, VAS score, stone-free status, and hospital stay were analyzed. RESULTS: Preoperative demographic differences were not significant in all three arms. Postoperative CT scans in all groups demonstrated mostly minimal grade access tract hematomas. Mean perirenal hematoma thickness showed no significant differences (2.66 ± 3.74, 2.73 ± 3.85, 2.54 ± 4.37 mm, p = 0.981), respectively. Postoperative hemoglobin drop (0.75 ± 0.58, 0.84 ± 0.47, 0.91 ± 0.60 g/dl, p = 0.74), stone-free rate (93.75, 87.87, 87.87%, p = 0.121), VAS (p = 0.499) and hospital stay (1.81 ± 0.84, 1.48 ± 0.71, 1.59 ± 0.75 day, p = 0.127) were not significantly different between the groups. CONCLUSION: Fibrin glue and Tachosil® in tubeless PCNL were not necessary for postoperative access tract control.

Scenedesmus dimorphus was cultivated in raw and pretreated swine wastewater (SW) with 6-L photobioreactors (PBRs) to investigate the effect of solid-liquid separation on algal growth. The same aerated PBRs containing no algae were used as control. Moderate COD and nitrogen removal from the SW was achieved with the algal PBRs. However, compared to the control reactors, they offered no consistent treatment boost. Improved algal growth occurred in the pretreated SW, as measured by maximum algal cell count (3202 ± 275 × 106 versus 2286 ± 589 × 106 cells L-1) and cell size. The enhanced algal growth in the pretreated SW resulted in relatively high nitrogen (5.7 %) and organic matter contents in the solids harvested at the end of cultivation experiments, with ∼25.6 % of nitrogen in the SW retained in the solids and ∼9.1 % absorbed by algae. The pretreatment also resulted in elevated phosphorus removal. This study is anticipated to foster the development of microalgae-based SW treatment processes.

High concentrations of total dissolved solids (TDS) in water have been reported to increase the incidence of diarrhea and reduce nursery pig growth performance. The objective of this study was to investigate the effects of drinking water with high concentrations of TDS from sulfate salts on nursery pigs. Weaned pigs sorted to equalize gender were placed in 44 pens with 26 pigs per pen. One of four water treatments was randomly assigned to each pen: (1) combination of CaSO4, MgSO4, and NaSO4; (2) CaSO4; (3) MgSO4; (4) NaSO4. Access to water and feed (nursery diets fed in four phases) was unrestricted throughout. The weights of pens were measured, feed remaining on weigh days was calculated, and pen water meters were read on d 0, 7, 21, 35, and 42. Water treatment did not affect (p > 0.07) average daily gain, average daily feed intake, or gut integrity of pigs. Water disappearance tended to be greater (p = 0.10) in pens receiving the CaSO4 water compared to the combination treatment from d 21 to 35. The TDS concentrations from sulfate salts used in this experiment did not impact the growth performance or feed or water disappearance of newly weaned nursery pigs.

A survey of 23 South Dakota pork producers in 2019 reported that 68% of the waterers in finishing barns had water flow rates above the recommended rate of 500-1,000 mL/min. The objective of the two studies was to determine the impact of water flow rate on finishing pig performance in the summer months. Study 1 used a total of 396 pigs in two groups in a 77-day trial (35.0 to 104.3 kg BW) with 6 pigs/pen and 1 cup waterer/pen. Study 2, conducted in a commercial style barn, used a total of 1,227 pigs in an 84-day trial (60.9 to 117.4 kg BW) with 26 pigs/pen and 2 cup waters/pen. Pens were assigned to one of three water flow rates (high, medium, low) based on the 3-hole settings of the water nipples (2.0, 1.0, and 0.8 mm; n = 22 and 16 pens/treatment for Study 1 and 2, respectively). Room temperature, outside temperature and relative humidity were recorded daily for both studies. In Study 1, water disappearance was recorded daily, and individual pen water flow rates were recorded every two weeks. At every diet phase change (26 ±â€…2.6 days), feed disappearance and individual pig body weights were recorded. Water flow rates averaged 1856 ±â€…188, 906 ±â€…214, 508 ±â€…100 mL/min for high, medium, and low flow settings, respectively. In Study 2, individual pen water flow rate, water disappearance, BW, and feed disappearance were recorded every two weeks. Water flow rates averaged 1115 ±â€…98, 906 ±â€…209, and 605 ±â€…203 mL/min for high, medium, and low flow settings, respectively. In both studies, there were no differences in final BW, cumulative ADG, or G:F. Due to the variability of water flow rate within a setting, data was further analyzed using regression with flow rate as the independent variable. Apart from average daily water disappearance (adj. R 2 = 0.87), there was a low relationship between pig performance and water flow rate (adj. R 2 < 0.09). The low R 2 values associated with pig performance and the high association with water disappearance suggests that water flow rate above current recommendations has little impact on finishing pig performance but does contribute to water wastage and its associated costs.

Microbial interactions within a natural or engineered consortium of microbes play an important role in the functions of the consortium. Better understanding these interactions is also important for engineering microbial consortia for specific applications. As such, tools that can enable investigating microbial interactions are highly valuable. One aspect of microbial interactions that impacts community formation is how the spatial organization of individual microbes impacts interactions leading to community formation. Here, we report the development of a tool that can manipulate the spatial organization of microorganisms to investigate the role of these interactions in community formation. Our developed microfluidic platform utilizes dielectrophoretic (DEP) force to perform on-demand spatial arrangement of microorganism-encapsulated agarose gel microparticles. To demonstrate this concept, three gel microparticle manipulators composed of three independently controllable DEP electrodes were utilized for the on-demand spatial arrangement of a specific combination of microparticles, each containing Escherichia coli cells expressing red fluorescence protein, green fluorescent protein, or blank content. The spatially arranged microparticles suspended in carrier oil were first trapped in a downstream particle trapping structure to form a defined microparticle array, followed by the application of an electric field to disrupt the carrier oil barrier so that all gel microparticles were within the same aqueous solution while the individual gel microparticles remain intact, thereby maintaining their spatial arrangements. We demonstrated that this method can be utilized to generate various arrays with differing number of "spacer microparticles", which were blank microparticles, between the two different E. coli-containing microparticles, enabling precise control over spatial distances between the two different cell populations. This method paves the way for more easily investigating bacterial interactions, especially those that depend on their spatial arrangement such as where cell-cell communication plays a major role.

In order to determine the standardized ileal digestible (SID) Ile:Lys requirement in finishing diets containing 20% DDGS, a 56-d study was conducted utilizing 2,268 pigs (DNA 600 x Topigs Norsvin 70, initially 82.3 kg). A total of six dietary treatments were fed, including a standard corn-soybean meal (SBM) diet and five diets containing 20% DDGS with SID Ile:Lys ratios of 55%, 60%, 65%, 70%, and 75%. Dietary treatments were assigned to pens, incompletely balancing for previous treatment, with each treatment being replicated 14 times. Pair-wise comparisons were used to evaluate the impact of dietary treatment on performance and carcass traits, whereas single degree of freedom orthogonal polynomials were used to evaluate dose response of SID Ile:Lys in 20% DDGS diets. Increasing the SID Ile:Lys ratio in diets containing 20% DDGS did not impact pig growth performance criteria in a quadratic or linear fashion (P ≥ 0.18). However, increasing the SID Ile:Lys ratio in 20% DDGS diets resulted in decreased back fat (BF; Quadratic, P = 0.01), increased loin depth (Quadratic, P = 0.03), and tended to increase percent lean (Quadratic, P = 0.07) with optimal carcass parameters occurring when 65% SID Ile:Lys was supplied in 20% DDGS diets. Pigs fed the corn-SBM diet had a similar final body weight (BW; P = 0.26) and cumulative average daily gain (ADG; P = 0.12) compared to pigs fed a 20% DDGS diet containing 70% SID Ile:Lys ratio and 3% greater cumulative average daily feed intake (ADFI) compared to pigs receiving diets with SID Ile:Lys ratios of 65% and 75% (P ≤ 0.01). In conclusion, these results suggest that when feeding 20% DDGS in late finishing swine diets, a SID Ile:Lys ratio of 70% should be utilized when attempting to achieve similar overall growth performance relative to a corn-SBM diet.


The use of corn by-products, such as dried distillers grains with solubles (DDGS), from the ethanol industry in swine rations is a common practice when economical. However, the inclusion of these by-products can potentially impact the nutrient requirements of pigs. It is important to understand how one should adjust diets to meet these changes in nutrient requirements and to minimize the reduction in growth performance. Previous research has indicated that the excess of a specific amino acid, leucine, can negatively impact the performance of pigs through the increased breakdown of other amino acids, more specifically valine and isoleucine. This increased breakdown of valine and isoleucine can result in inadequate amounts being available for protein deposition and, therefore, reduce growth performance. In the present study, late finishing pigs fed a diet containing 20% DDGS required a ratio of 70% between the amino acids isoleucine and lysine, on a digestible basis, to perform similarly to pigs receiving a standard corn soybean meal diet. Further research is necessary to more accurately define the optimal dietary isoleucine level for finishing pigs when fed 20% DDGS.

Droplet microfluidic systems have been widely deployed to interrogate biological and chemical systems. The major limitations of these systems are the relatively high error rates from critical droplet manipulation functions. To address these limitations, we describe the development of FIDELITY (Flotation and Interdigitated electrode forces on Droplets to Enable Lasting system IntegriTY), a highly sensitive and accurate size-based droplet bandpass filter that leverages the natural buoyancy of aqueous droplets and highly localized dielectrophoretic force generated by interdigitated electrode arrays. Droplet manipulation accuracies greater than 99% were achieved at a throughput of up to 100 droplets/s and separation of droplets that differed in diameter by only 6 µm was demonstrated. Last, the utility of FIDELITY was demonstrated in a droplet size quality control application and also in a droplet-based in vitro transcription/translation workflow. We anticipate FIDELITY to be integrated into a broad range of droplet microfluidic configurations to achieve exceptional operational accuracy.

A total of 2,430 pigs (DNA 600 × Topigs Norsvin 70, initially 39.4 kg) were used in a 28-d trial to determine the standardized ileal digestible (SID) Val:Lys requirement for pigs fed with diets containing 30% DDGS. Treatments included five diets containing 30% DDGS with SID Val:Lys ratios of 60%, 65%, 70%, 75%, and 80%, plus a corn soybean meal (SBM) diet, for a total of six dietary treatments. Diets were formulated to be isocaloric through the inclusion of fat and to contain equal amounts of SID Lys within phase. Pens were assigned to dietary treatment in a randomized complete block design with initial body weight (BW) as the random blocking factor. Each dietary treatment was replicated 15 times and pens contained 27 pigs, balanced for sex. Increasing the SID Val:Lys ratio in diets containing 30% DDGS increased (Quadratic; P ≤ 0.007) 14-d BW, final BW, average daily gain (ADG), average daily feed intake (ADFI), and gain:feed (G:F) for all periods. Providing an SID Val:Lys ratio of 75% resulted in the heaviest 14-d BW and final BW as well as greater ADG and ADFI of pigs fed with 30% DDGS for all periods. The G:F response was maximized when an SID Val:Lys ratio of 70% was provided from day 0 to 14 while an SID Val:Lys ratio of 75% maximized G:F from day 14 to 28 and for the cumulative period. The SID Val:Lys requirement was estimated at 66.6% (95% CI [65.9, 67.4]), 65.7 (95% CI: [64.8, 66.5]), and 68.4% (95% CI [66.0, 70.8]) for ADG, ADFI, and G:F, respectively, using the straight broken line (SBL) method and 69.9% (95% CI [68.2, 71.5]), 67.6 (95% CI [65.4, 69.8]), and 72.8% (95% CI [69.8, 75.8]) for the quadratic broken line (QBL) method. Pigs fed the corn-SBM diet had heavier 14-d BW, final BW, and greater ADG, ADFI, G:F (P ≤ 0.032) compared to pigs fed diets containing 30% DDGS, except for cumulative ADFI compared to pigs receiving 75% SID Val:Lys (P = 0.167). In conclusion, these results suggest that when feeding 30% DDGS during the growing period, an SID Val:Lys ratio of 68% would yield more than 99% and 97% of the maximum ADG and G:F response for the 39 to 68 kg pigs. However, growth performance of pigs fed diets containing 30% DDGS did not equate to pigs consuming the corn-SBM diet regardless of the SID Val:Lys ratio.


Understanding the nutrient requirements of pigs across various conditions is important to determine the most economical method of feeding pigs. When feeding pigs corn byproducts from the ethanol industry, such as dried distiller grains with solubles (DDGS), the growth performance of pigs decreases. This is thought to be due to the composition of the dietary protein, more specifically the amino acid concentration of the diet. In the present study, one specific amino acid, valine, was incrementally increased in diets containing 30% DDGS. The growth performance of pigs increased when digestible valine increase until a ratio of 68% was reached between digestible valine and the amino acid lysine. These results indicate that the digestible valine requirement for pigs between 39 and 68 kg of body weight is 68% of the lysine requirement. However, when growth performance of pigs fed with 30% DDGS was compared to a standard corn soybean meal diet, pigs fed with the standard corn soybean meal diet had greater performance, regardless of digestible valine level. More research is required to understand why the addition of 30% DDGS in pig diets negatively impacts the growth performance relative to a standard corn soybean meal diet.
