RESUMO

The article deals with analysis of morphologic characteristics of microscopic fungi of genus of Coccididoides spp. under cultivation on culture of mouse splenocytes culture. During two days, the strains of C. imitis and C. posadasii converse from filamentous to spherulic form. This process makes it possible to apply this test to identify agents of coccidioidomycosis.

RESUMO

Molecular diagnostic kits for detection and identification of agents of melioidosis and glanders on environmental objects and in clinical material are described. It was demonstrated that PCR with use of specific primers on the basis of different genetic targets could be useful for determination of generic, inter- and intraspecies belonging of pathogenic Burkholderia as well as for epidemiologic inspection of territories where melioidosis is enzootic.

RESUMO

The cellular fatty acid compositions of studied two strains of Burkholderia mallei and a strain of Burkholderia pseudomallei are represented by saturated and monounsaturated straight chain fatty acids with 14-18 carbon atoms, cyclopropane fatty acids C(17 inverted delta) and C(19 inverted delta), and hydroxy acids 3-OH-C14:0, 2-OH-C(16:0), and 3-OH-C(16:0). The strain variation of cyclopropane and unsaturated fatty acid levels was observed. The cellular fatty acid spectra of studied bacteria did not depend essentially on growth medium. The levels of cyclopropane fatty acids increased and those of unsaturated ones decreased with culture age, a tendency to increasing the levels of hydroxy fatty acids was observed too.

RESUMO

In this review modern methods for the identification and differential diagnostics of the causative agents of glanders and melioidosis, recently included into the genus Burkholderia, are presented. The known phenotypic signs and genetic markers permitting the identification of two pathogenic microorganisms on the definite taxonomic level are described.

RESUMO

The dynamics of the antigen 8 synthesis in Burkholderia pseudomallei and B. mallei under conditions of their submerged was studied. Differences in the intensity of this antigen synthesis in two pathogenic Burkholderia species were established and the producer strains, most effective with respect to this sign, were selected.

RESUMO

The studied strains of Yersinia pseudotuberculosis (5 st.), Y. enterocolitica (2 st.), Y. intermedia (1 st.), Y. frederiksenii (1 st.), Y. kristensenii (1 st.) and Y. ruckeri (1 st.) have similar fatty acid profiles of cells mainly represented by straight chain and cyclopropanic fatty acids with chain of 12 to 19 carbon atoms. Prevalence of C16:1, C16:0, C17v, in the spectrum and C18:1 under growth in meat-peptone broth are observed. Strains of Y. pseudotuberculosis as well as the already studied vaccine strain Y. pestis differ from yersiniae of other species by the lower level of dodecanoic acid. Bacteria of species Francisella tularensis (6 st.) are characterized by a wide range of straight-chain saturated and monounsaturated fatty acids with 10-26 carbon atoms. Yersiniae and francisellae preserve the proper fatty acid profiles under cultivation on various nutrient media and on the same medium (agarized medium with aminopeptide). Results obtained confirm close relationship of yersiniae of the studied species as well as phylogenetic remoteness and taxonomic isolation of yersiniae and francisellae.

RESUMO

A transporting medium was designed for the transportation and preservation for 5 to 7 days of material infected with agents of malleus and melioidosis. Foreign contaminating microorganisms gradually die in this medium, whereas solitary cells of malleus and melioidosis agents (100 bacterial cells per 0.1 ml) survive.

RESUMO

The fatty acid composition of Y. pestis strains, the causative agent of plaque, has been studied. Y. pestis cells have been found to contain great amounts of palmitoleic acid, methylenehexadecanoic acid, oleic acid with elaidic acid, palmitic acid and pentadecanoic acid. Lauric acid, myristic acid, 3-oxymyristic acid and methyleneoctadecanoic acid have been detected in moderate amounts. Vaccine strains, virulent museum and newly isolated strains, while differing in their antigenic structure, have proved to be uniform in their fatty acid composition. In Y. pestis cels, grown on a solid medium for a longer period and at higher temperature, an increased proportion of saturated acids is observed.

RESUMO

Fatty acid composition of lipopolysaccharides (LPS) and cytoplasmic membranes (CPM) of Yersinia pestis [correction of the plaque microbe] has been studied. Concentration of certain acids in the LPS content proved sharply different and thus the strains were separated into groups. High content of 3-oxymiristinic acid was a distinction of vaccine strains. Virulent strains of one of the groups regularly differed from the rest of virulent strains by high content of laurinic acid. Laurinic and two nonidentified acids, which were not found in the virulent cultures, were present in CMP lipids of vaccine strains.

RESUMO

The interaction of the aqueous saline extracts of 55 plant species with the antigens of the causative agents of plague, glanders and melioidosis in the reaction of immunodiffusion (RID) in gel has been studied. The aqueous saline extracts obtained from the seeds of 12 plant species have been revealed to possess precipitating capacity. At the same time these extract have been found to ensure, as a rule, RID with several antigens under test.
