RESUMO

In Brazil, acanthocephalans parasitise anurans in several biomes. In the present study, we performed an analysis of acanthocephalan infections across 175 anuran individuals from the Cerrado biome, belonging to ten species: Boana raniceps, Pithecopus hypochondrialis, Scinax fuscomarginatus, Scinax x-signatus, Leptodactylus pustulatus, Leptodactylus macrosternum, Leptodactylus vastus, Physalaemus cuvieri, Adenomera hylaedactyla, and Elachistocleis piauiensis. We also verified the specificity of the parasites using the STD* index. Additionally, we conducted a survey of acanthocephalan infection in anurans in South America. The studied assemblage in the Brazilian Cerrado presented 57 parasitised hosts of 175 specimens (overall prevalence: 32.6%). In total, 437 acanthocephalans cystacanths were recorded, among which 286 presented the same morphotype but could not be identified, 148 belonged to the genus Centrorhynchus, and three belonged to Oncicola. Unidentified acanthocephalans had a higher prevalence in L. vastus (53.85%) and the highest intensity was in L. pustulatus (17±16). The highest prevalence of Centrorhynchus sp. was in the species S. fuscomarginatus (28.57%), while the highest intensity was observed in L. vastus (111). The taxon Oncicola sp. it had a prevalence of 3.23% and an intensity of 3 only in S. x-signatus. The highest specificity was recorded for Oncicola sp. (STD*= 1), whereas the lowest was found in Centrorhynchus sp. (STD*= 2.21). Finally, according to the survey for South America, we found ten records of acanthocephalan taxa parasitizing 58 species of anurans distributed in seven countries (Brazil with the most records).

Abstract In this study was evaluated the influence of glutamine supplementation on the endogenous content of amino acids, proteins, total phenolics, flavonoids and proanthocyanidins in Bacupari callus. The explants were inoculated in MS medium, MS with half concentration of the nitrogen salts (MS½) and nitrogen-free MS, supplemented with glutamine (5, 10, 30 and 60mM) named as Gln5, Gln10, Gln30 and Gln60. Amino acids and proteins were analyzed after 20, 80 and 140 days and the secondary metabolites on the 140th day. There was no difference in the amino acids on the 20th day. On the 80th day the treatments MS and MS½ presented the lowest levels. On the 140th day MS and MS½ presented the lowest amino acid concentration and Gln10 the highest. Concerning proteins, there was difference only on the 140th day, being the highest concentrations observed in Gln5, and the lowest in MS½ treatment. Total phenolics content was higher in the treatment Gln60 and lowest in MS. Treatments Gln5, Gln10, Gln30 and MS½ were statistically equal. For flavonoids, the highest values occurred in the treatments Gln30, Gln60 and MS½ and the lowest in Gln5, Gln10 and MS. Similarly, for the proanthocyanidins the highest concentrations were observed in treatment Gln60 and the lowest in Gln5 and MS. In conclusion, the treatment with 60mM of glutamine favors the protein accumulation and production of secondary metabolites in Bacupari callus.


Resumo Nesse estudo foi avaliado o efeito da suplementação com glutamina no conteúdo endógeno de aminoácidos, proteínas, fenólicos totais, flavonoides e proantocianidinas em calos de Bacupari. Os explantes foram inoculados em meio MS, meio MS com metade da concentração de dos sais de nitrogênio (MS½) e meio MS sem nitrogênio suplementado com glutamina (5, 10, 30 e 60mM) denominados como Gln5, Gln10, Gln30 e Gln60. Os aminoácidos e as proteínas foram analisados após 20, 80 e 140 dias e os metabólitos secundários no 140° dia. Não houve diferença nos aminoácidos no 20° dia. No 80° dia os tratamentos MS e MS½ apresentaram os menores níveis. No 140° dia, MS e MS½ apresentaram as menores concentrações de aminoácidos e o Gln10 as maiores. A respeito das proteínas, houve diferença apenas no 140° dia, sendo as maiores concentrações observadas nos tratamentos Gln, e as menores no MS½. O conteúdo de fenólicos totais foi maior no tratamento Gln60 e menor no MS. Os tratamentos Gln5, Gln10, Gln30 e MS½ foram estatisticamente iguais. Para os flavonóides, os maiores valores ocorreram nos tratamentos Gln30, Gln60 e MS½ e os menores no Gln5, Gln10 e MS. Da mesma forma, para as proantocianidinas, as maiores concentrações foram observadas no tratamento Gln60 os menores no Gln5 e MS. Em conclusão, o tratamento com 60 mM de glutamina favorece o acúmulo de proteínas e a produção de metabólitos secundários em calos de Bacupari.

Abstract Some macrophytes species show a high growth potential, colonizing large areas on aquatic environments. Cattail (Typha angustifolia L.) uncontrolled growth causes several problems to human activities and local biodiversity, but this also may lead to competition and further problems for this species itself. Thus, the objective of this study was to investigate anatomical modifications on T. angustifolia plants from different population densities, once it can help to understand its biology. Roots and leaves were collected from natural populations growing under high and low densities. These plant materials were fixed and submitted to usual plant microtechnique procedures. Slides were observed and photographed under light microscopy and images were analyzed in the UTHSCSA-Imagetool software. The experimental design was completely randomized with two treatments and ten replicates, data were submitted to one-way ANOVA and Scott-Knott test at p<0.05. Leaves from low density populations showed higher stomatal density and index. These modifications on stomatal characteristics were more evident on the leaf abaxial surface. Plants from low density populations showed thicker mesophyll and higher proportion of aerenchymal area. Roots from low density populations showed a higher proportion of the vascular cylinder. Whereas, plants from higher density populations showed greater thickness of the endodermis, exodermis, phloem and root cortex. Higher density populations showed a higher proportion of aerenchymal gaps in the root cortex. Therefore, cattail plants from populations growing under high density population show anatomical traits typical of plants under stress, which promotes the development of less functional anatomical modifications to aquatic environments.


Resumo Algumas espécies de macrófitas podem apresentar um elevado potencial de crescimento, colonizando extensas áreas de ambientes aquáticos. O crescimento descontrolado da taboa (Typha angustifolia L.) causa problemas para a biodiversidade local e para atividades antrópicas, isso também pode levar à competição e problemas para a própria espécie. Dessa forma, o objetivo desse trabalho foi verificar se ocorrem modificações anatômicas em plantas de Typha angustifolia L. de diferentes densidades populacionais, uma vez que essas podem auxiliar na compreensão da biologia dessa espécie. Raízes e folhas de T. angustifolia foram coletadas de populações naturais com alto e baixo adensamento populacional. Esse material vegetal foi fixado e submetido a procedimentos usuais de microtécnica vegetal. As lâminas foram observadas e fotografadas em microscopia de luz e as imagens analisadas no software UTHSCSA-Imagetool. O delineamento experimental foi inteiramente casualizado com dois tratamentos e dez repetições, os dados foram submetidos à análise de variância e ao teste de Scott-Knott para p<0,05. As folhas das populações com baixa densidade demonstraram maior densidade e índice estomáticos. Essas características foram mais evidentes na superfície abaxial em comparação com a superfície adaxial. As plantas de populações com baixo adensamento demonstraram maior espessura do mesofilo e área de aerênquima foliar. As raízes das populações com baixo adensamento demonstraram maior proporção do cilindro vascular. Por outro lado, as plantas de populações com alto adensamento demonstraram maior espessura da endoderme, exoderme, floema e do córtex nas raízes. Populações de alto adensamento demonstraram maior proporção de aerênquima no córtex radicular. Portanto, plantas de taboa sob alto adensamento populacional estão sob estresse o qual promove o desenvolvimento de modificações anatômicas menos funcionais para os ambientes aquáticos.

Androgens may directly modulate early ovarian follicular development in preantral stages and androgen excess before puberty may disrupt this physiological process. Therefore, the aim of this study was to investigate the dynamics of follicular morphology and circulating androgen and estradiol levels in prepubertal Wistar rats acutely exposed to androgens. Prepubertal female Wistar rats were distributed into three groups: control, equine chorionic gonadotropin (eCG) intervention and eCG plus dehydroepiandrosterone (DHEA) intervention (eCG+DHEA). Serum DHEA, testosterone and estradiol levels were determined, and ovarian morphology and morphometry were assessed. The eCG+DHEA group presented increased serum estradiol and testosterone levels as compared with the control group (P<0.01), and higher serum DHEA concentration v. the eCG-only and control groups (P<0.01). In addition, the eCG+DHEA group had a higher number of, and larger-sized, primary and secondary follicles as compared with the control group (P<0.05). The eCG group presented intermediate values for number and size of primary and secondary follicles, without significant differences as compared with the other two groups. The number of antral follicles was higher in the eCG+DHEA and eCG groups v. controls (P<0.05). The number of primordial, atretic and cystic follicles were similar in all groups. In conclusion, the present experimental model using an acute eCG+DHEA intervention was useful to investigate events involved in initial follicular development under hyperandrogenic conditions, and could provide a reliable tool to study defective follicular development with possible deleterious reproductive consequences later in life.

Este trabalho teve por objetivo identificar as plantas medicinais de uso popular e o perfil socioeconômico de seus usuários em área urbana de Ouro Preto, MG. O levantamento utilizou entrevistas semiestruturadas e amostragem aleatória, perfazendo 10% das residências. O grau de conhecimento sobre plantas medicinais foi medido pelo número de espécies citadas. Analisou-se a relação entre o saber popular sobre as plantas medicinais e características socioeconômicas dos entrevistados (renda, escolaridade, sexo, idade e forma de aquisição do conhecimento). Foram questionadas 6.713 pessoas, onde mais de 90% usam plantas medicinais para se tratarem. Identificou-se 342 espécies, reunidas em 94 famílias. Para cada espécie foram referidos os nomes populares, hábito, procedência, uso medicinal, parte usada, e forma de preparo. As principais moléstias tratadas com plantas foram: diarreia, insônia, gripe, hidropisia, distúrbios hepáticos, renais e do trato respiratório. Há um grande número de espécies nativas utilizadas. Dentre as exóticas, a maioria é de origem europeia. O uso místico de espécies, embora presente na cultura popular do município, foi pouco citado. Algumas espécies identificadas figuram na lista das espécies ameaçadas de extinção. O grau de conhecimento sobre plantas medicinais pela população de Ouro Preto independe, tanto do nível econômico, como da escolaridade ou do sexo. A idade e a forma de aquisição do conhecimento influenciam no saber popular das ervas medicinais. As pessoas com maior saber popular sobre as plantas adquiriram esses conhecimentos principalmente pelo costume familiar, por livros, ou por outras pessoas. Pessoas mais jovens conhecem menos espécies medicinais que as mais idosas, sugerindo risco de perda desse conhecimento tradicional. A grande riqueza de plantas citadas neste trabalho denota a importância de estudos etnobotânicos no resgate do conhecimento tradicional em áreas urbanas, tanto pelo seu valor histórico-cultural, como pela importância científica.


This work aimed to identify the medicinal plants of popular use and the socioeconomic profile of the users in the urban area of Ouro Preto, state of Minas Gerais, Brazil. The survey was carried out with random sampling and semi-structured interviews, amounting to 10% of households. The degree of knowledge about medicinal plants was measured by the number of species mentioned. The relationship between the popular knowledge about medicinal plants and the socioeconomic characteristics of the users (income, education, gender, age and type of knowledge acquisition) was also studied. In this survey, 6,713 inhabitants were questioned, of whom more than 90% use medicinal plants. A total of 342 species grouped in 94 families were identified. The popular names, growth habit, habitat, medicinal uses, part used and method of preparation are listed for each identified species. The main ailments treated with plants were diarrhea, insomnia, flu, dropsy and liver, renal and respiratory tract disorders. There is a large number of native species. Among the exotic ones, most are of European origin. The spiritual use of species, although present in the popular culture of the city, was negligible. Some species figure on the Brazilian red lists. It was noted that knowledge of medicinal plants by the population of Ouro Preto, in species richness, is not related to income, class, educational level and gender. The age and manner of acquisition of empirical knowledge about medicinal plants are associated with the number of species listed. Persons with greater popular knowledge about medicinal plants acquired information through family tradition, books and from other persons. Younger persons know fewer medicinal plants than the older ones, what suggests a loss of this traditional knowledge. The richness of the plants mentioned in this paper demonstrates the importance of ethnobotanical studies in the rescue of traditional knowledge in urban areas, for its scientific, historical and cultural values.

BACKGROUND: Haplotypes of adiponectin gene single nucleotide polymorphisms (SNP) might be related to metabolic disorders. AIM: To assess whether the prevalence of SNP 45T/G and 276G/T of the adiponectin gene and their haplotypes differ between polycystic ovary syndrome (PCOS) and non-hirsute cycling controls and to investigate the relationship between these haplotypes and risk factors for cardiovascular disease. SUBJECTS AND METHODS: In this case-control study, 80 women with PCOS and 1500 non-hirsute controls with regular cycles underwent clinical and laboratory measurements. Genotype distribution was analyzed by conventional PCR-restriction fragment length polymorphism. RESULTS: Compared to controls, PCOS women had greater body mass index (BMI) (31.0±7.9 kg/m² vs 23.4±4.6 kg/m²; p<0.001), waist circumference (92.2±18.8 cm vs 74.5±10.2 cm; p<0.001), and systolic and diastolic blood pressure (124.6±19.9 vs 111.5±13.0 mmHg and 79.2±12.5 vs 71.8±10.6 mmHg; p<0.025), as well as a worse lipid profile (p<0.007), even after adjustment for age and BMI. Genotype distribution was similar in PCOS and controls (45T/G: p=0.399; 276G/T: p=0.135). Six haplotypes were inferred and their frequencies differed significantly between the groups (p=0.001). The TGTG haplotype was more frequent in PCOS than controls (41.3 vs 18.9%). In PCOS, the GG genotype for SNP 276 (p=0.031) and the TGTG haplotype (p=0.023) were associated with higher systolic blood pressure vs other genotypes and haplotypes. Body composition, glucose, insulin, and lipid profile were similar across genotypes and haplotypes in both groups. CONCLUSIONS: Haplotype TGTG from adiponectin gene variants 45T/G and 276G/T is related to susceptibility to PCOS, and might be associated with increased blood pressure in PCOS.

RESULTS: Genotype GG of the ApaI single nucleotide polymorphism (SNP) was more frequent in PP (30.6%) than in controls (16.2%) [odds ratio (OR): 2.269; confidence interval 95% (95%CI): 1.015-5.076; p=0.042]. This genotype was also associated with lower estradiol [35.30 (14.80-50.48) pg/ml vs 12.22 (6.49-23.69) pg/ml; p=0.025] and total testosterone levels (0.52 (0.39-0.84) ng/ml vs 0.20 (0.11-0.47) ng/ml; p=0.005) as compared with the TT + TG genotypes in girls with PP. The distribution of TaqI, BsmI, and Fokl SNP was similar in PP and controls, and no association was found between these polymorphisms and sex steroid levels. CONCLUSIONS: The ApaI SNP of the VDR gene was associated with PP in the studied population and may modulate ovarian steroid secretion in these girls.

CONCLUSIONS: The ApaI SNP of the VDR gene was associated with PP in the studied population and may modulate ovarian steroid secretion in these girls.

BACKGROUND: The 2NN Study was a randomised comparison of the non-nucleoside reverse-transcriptase inhibitors (NNRTI) nevirapine and efavirenz. METHODS: In this multicentre, open-label, randomised trial, 1216 antiretroviral-therapy-naive patients were assigned nevirapine 400 mg once daily, nevirapine 200 mg twice daily, efavirenz 600 mg once daily, or nevirapine (400 mg) and efavirenz (800 mg) once daily, plus stavudine and lamivudine, for 48 weeks. The primary endpoint was the proportion of patients with treatment failure (less than 1 log(10) decline in plasma HIV-1 RNA in the first 12 weeks or two consecutive measurements of more than 50 copies per mL from week 24 onwards, disease progression [new Centers for Disease Control and Prevention grade C event or death], or change of allocated treatment). Analyses were by intention to treat. FINDINGS: Treatment failure occurred in 96 (43.6%) of 220 patients assigned nevirapine once daily, 169 (43.7%) of 387 assigned nevirapine twice daily, 151 (37.8%) of 400 assigned efavirenz, and 111 (53.1%) of 209 assigned nevirapine plus efavirenz. The difference between nevirapine twice daily and efavirenz was 5.9% (95% CI -0.9 to 12.8). There were no significant differences among the study groups in the proportions with plasma HIV-1 RNA concentrations below 50 copies per mL at week 48 (p=0.193) or the increases in CD4-positive cells (p=0.800). Nevirapine plus efavirenz was associated with the highest frequency of clinical adverse events, and nevirapine once daily with significantly more hepatobiliary laboratory toxicities than efavirenz. Of 25 observed deaths, two were attributed to nevirapine. INTERPRETATION: Antiretroviral therapy with nevirapine or efavirenz showed similar efficacy, so triple-drug regimens with either NNRTI are valid for first-line treatment. There are, however, differences in safety profiles. Combination of nevirapine and efavirenz did not improve efficacy but caused more adverse events.

RESUMO

This research has a quality input that aims to reveal the concepts of popular class families caregivers of frail elderly people at their home, concerning the aspects that involve the meaning of care taking to the frail elderly in the family context. The methodology used is a case study of the ethnological kind. The data were collected by means of participant observation and interview with four families. Through the data analysis four themes related to the families conceptions were identified: the meaning of health-sicken, the meaning of family, the meaning of old age, the meaning of taking care of a frail elderly family member.

RESUMO

This is a descriptive study done in a qualitative perspective. It aims to describe the conceptions of health care units' managers about the work in public health. The subjects perceptions are that this work goes from interdisciplinary thought with a preventive focus to the curative practice of ambulatory care. On what concerns Nursing, nurses' management functions and auxiliaries assistance activities are pointed out. They believe that strategies to approximate to the SUS principles are necessary in the context of Public Health.

We determined the genotypes of the alcohol dehydrogenase (ALDH) and aldehyde dehydrogenase (ALDH2) loci of different ethnic groups living in Brazil, using saliva DNA amplified by PCR and allele-specific oligonucleotides. Self-reports of flushing reaction after drinking were also studied. The allelic frequencies of ADH2 and ALDH2 were found to be lower than those reported other authors, which might be a result of the admixture origin of the Brazilian population. Variability in facial flushing reaction suggests that other factors play a role in the expression of alcohol-induced flushing.

RESUMO

It approaches the influences of family relationship in the construction of the gender identity process, through the Theory of Objectal Relationship referential, proposed by Chorodow (1979). This theoretical support, emphasizes that the gender identity is basically influenced by the primary bonds established between child and adults, through the continuous personal relationship developing mother and father role. It points out that the gender construction for the boy and for the girl happens in a different way. It concludes pointing out some lines to be adopted by the nurse in the practice with families, as well as, some limitation of the Theory of Objectal Relationship.

The use of microcomputers in medical education to assess the training process in Primary Health Care is analyzed. The experimental study consisted of 8 modules related to immunization campaigns, nursing care of healthy children, sanitation, nursing care of women, epidemiology and statistics, and nursing care of those diseases presenting the highest prevalence. The program is based on the basic software "Evaluation of Training", and designed in basic language. Each module consisted of a set of questions and each set had 3 different forms of feedback: the given form, the controlled form, and the form for searching. The independent variable of this investigation was the evaluation of training by microcomputers, and the dependent variable was the cognitive domain and development. The sample comprises 16 individuals studying on the eighth semester of a nursing course, and each group is made up by 8 students (a control group and an experimental group). Data obtained from this investigation were assessed by the test of nonparametric indicators. Results evidenced that the evaluation of training by a microcomputer provided a significantly higher performance in comparison with noncomputer assisted evaluation. Students were satisfied regarding the interaction with the microcomputer and with the performance of the evaluation. Further investigations in this field are recommended.

Self reports of flushing reaction after drinking, cutaneous sensitivity to alcohol (patch test), and genotypic determination of ADH2, ADH3, and ALDH2 were studied in 53 Brazilian volunteers of different ethnic groups. Genotypes were determined using single-strand conformation polymorphism in discontinuous buffer electrophoresis. Analysis of the results indicated several cases of a reported flushing reaction among ALDH2 1/1 individuals, while all but 2 cases of ALDH2 heterozygotes reported a flushing reaction. The latter subjects also had a negative result in the patch test. These preliminary results indicate that variability in the facial flushing reaction to alcohol seems to be a phenomenon resulting not only from the presence of a deficient ALDH2*2 allele, but also from other polymorphisms of alcohol-metabolizing enzymes.
