We have studied the reduction reactions of two cytosolic human peroxiredoxins (Prx) in their disulfide form by three thioredoxins (Trx; two human and one bacterial), with the aim of better understanding the rate and mechanism of those reactions, and their relevance in the context of the catalytic cycle of Prx. We have developed a new methodology based on stopped-flow and intrinsic fluorescence to study the bimolecular reactions, and found rate constants in the range of 105 -106 m-1 s-1 in all cases, showing that there is no marked kinetic preference for the expected Trx partner. By combining experimental findings and molecular dynamics studies, we found that the reactivity of the nucleophilic cysteine (CN ) in the Trx is greatly affected by the formation of the Prx-Trx complex. The protein-protein interaction forces the CN thiolate into an unfavorable hydrophobic microenvironment that reduces its hydration and results in a remarkable acceleration of the thiol-disulfide exchange reactions by more than three orders of magnitude and also produces a measurable shift in the pKa of the CN . This mechanism of activation of the thiol disulfide exchange may help understand the reduction of Prx by alternative reductants involved in redox signaling.

The characterization of suitable microsites for tree seedling establishment and growth is one of the most important tasks to achieve the restoration of native forest using natural processes in disturbed sites. For that, we assessed the natural Quercus petraea forest expansion in a 20-year-old reclaimed open-cast mine under sub-Mediterranean climate in northern Spain, monitoring seedling survival, growth, and recruitment during 5 years in three contrasting environments (undisturbed forest, mine edge, and mine center). Seedling density and proportion of dead branches decreased greatly from undisturbed forest towards the center of the mine. There was a positive effect of shrubs on Q. petraea seedling establishment in both mine environments, which increase as the environment undergoes more stress (from the mine edge to the center of the mine), and it was produced by different shrub structural features in each mine environment. Seedling survival reduction through time in three environments did not lead to a density reduction because there was a yearly recruitment of new seedlings. Seedling survival, annual growth, and height through time were greater in mine sites than in the undisturbed forest. The successful colonization patterns and positive neighbor effect of shrubs on natural seedlings establishment found in this study during the first years support the use of shrubs as ecosystem engineers to increase heterogeneity in micro-environmental conditions on reclaimed mine sites, which improves late-successional Quercus species establishment.

OBJECTIVES: The aim of our study was to evaluate the safety and midterm clinical results of dexamethasone-eluting stent (DexES) implantation in ST-segment elevation acute myocardial infarction (STEMI). BACKGROUND: Inflammation plays a pivotal role in both inestabilization of coronary atherosclerotic plaques and development of restenosis after stent placement. Antiinflammatory agents may attenuate those mechanisms and improve clinical outcomes. There is little information about clinical results of DexES and no data are available about their utilization during percutaneous coronary intervention (PCI) in STEMI. METHODS: Consecutive patients with STEMI that underwent primary or rescue PCI in our institution were treated with DexES. Clinical follow-up with routine realization of noninvasive test for detection of myocardial ischemia and coronariography if necessary, were performed. The objective of the study was to evaluate the rate of MACE (death, reinfarction, or target lesion revascularization) during midterm follow-up. RESULTS: The procedure was successful in 96.7% of cases. There were no in-hospital deaths or reinfarctions. One acute stent thrombosis occurred and no subacute thrombosis were observed. During a mean follow-up period of 384 days, cardiac-related death was 1.1%, there were no reinfarctions or late stent thrombosis and target lesion revascularization rate was 4.2%. CONCLUSION: We conclude that utilization of DexES for PCI in STEMI is safe and provides good midterm clinical outcomes.
