RESUMO

The genus Burkholderia (ß-Proteobacteria) currently comprises more than 60 species, including parasites, symbionts and free-living organisms. Several new species of Burkholderia have recently been described showing a great diversity of phenotypes. We examined the diversity of Burkholderia spp in environmental samples collected from Caatinga and Atlantic rainforest biomes of Bahia, Brazil. Legume nodules were collected from five locations, and 16S rDNA and recA genes of the isolated microorganisms were analyzed. Thirty-three contigs of 16S rRNA genes and four contigs of the recA gene related to the genus Burkholderia were obtained. The genetic dissimilarity of the strains ranged from 0 to 2.5% based on 16S rDNA analysis, indicating two main branches: one distinct branch of the dendrogram for the B. cepacia complex and another branch that rendered three major groups, partially reflecting host plants and locations. A dendrogram designed with sequences of this research and those designed with sequences of Burkholderia-type strains and the first hit BLAST had similar topologies. A dendrogram similar to that constructed by analysis of 16S rDNA was obtained using sequences of the fragment of the recA gene. The 16S rDNA sequences enabled sufficient identification of relevant similarities and groupings amongst isolates and the sequences that we obtained. Only 6 of the 33 isolates analyzed via 16S rDNA sequencing showed high similarity with the B. cepacia complex. Thus, over 3/4 of the isolates have potential for biotechnological applications.

Devido à importância das moscas-das-frutas como pragas da fruticultura, este trabalho teve como objetivos caracterizar as populações das moscas-das-frutas em pomares comerciais de goiaba na região do Cariri, Estado do Ceará, por meio de índices faunísticos e estudar a flutuação populacional dessas moscas ao longo do ano. Para isso, foram instaladas armadilhas McPhail nos Municípios de Barbalha, Crato e Juazeiro do Norte. As armadilhas foram examinadas semanalmente e as moscas capturadas foram triadas, conservadas em álcool a 70% e identificadas. Constatou-se que Anastrepha zenildae (Wiedemann), A. sororcula Zucchi, A. fraterculus (Wiedemann), A. obliqua (Macquart) e Ceratitis capitata (Wiedemann) foram as moscas-das-frutas associadas à goiaba. Dessas, A. zenildae. A. sororcula e A. obliqua foram dominantes e constantes na região estudada, enquanto que A. fraterculus e C. capitata foram não-dominantes, mas também constantes. A. zenildae ocorreu em quase todos os meses do ano, exceto em outubro e dezembro de 2008, nos três pomares de goiaba do Cariri e com picos populacionais nos meses mais chuvosos e com temperaturas amenas.


Due to the importance of fruit-flies as pests in fruit growing, the present study was aimed to characterize the populations of fruit-flies in commercial guava orchards in the Cariri region (state of Ceará), Brazil, by means of faunistic indexes and to study their population fluctuation during the year. For this purpose, McPhail traps were installed in the counties Barbalha, Crato and Juazeiro do Norte. The traps were examined weekly and the captured flies were separated, preserved in 70% alcohol and identified. Anastrepha zenildae (Wiedemann), A. sororcula Zucchi, A. fraterculus (Wiedemann), A. obliqua (Macquart) and Ceratitis capitata (Wiedemann) were associated with guava fruits. Of them, A. zenildae. A. sororcula and A. obliqua were dominant and constant in the studied region, while A. fraterculus and C. capitata were non-dominant but constant. A. zenildae occurred in almost all months of the year, except October and December of 2008, with population peaks in the rainy season with its milder temperatures.

New designs for Magnetic Resonance Imaging contrast agents are presented. Essentially, they all are host-guest inclusion complexes between y-cyclodextrins and polyazamacrocycles of gadolinium (III) ion. Substitutions have been made to the host to optimise the host-guest association. Molecular mechanics calculations have been performed, using the UFF force field for metals, to decide on the suitability of the substitutions, and to evaluate the host-guest energies of association. Interesting general conclusions have been obtained, concerning the improvement of Magnetic Resonance Imaging contrast agents; namely, a set of rational methodologies have been deduced to improve the association between the gadolinium (III) chelates and the cyclodextrins, and their efficiency is demonstrated with a large set of substituted complexes, opening new doors to increase the diagnostic capabilities of Magnetic Resonance Imaging.

We investigated the role of retinotectal projections on the distribution of nitric oxide synthase (NOS) in the retinoceptive layers of the rat superior colliculus (SC) using histochemical methods. Rats enucleated at birth showed no alteration in the temporal pattern of NOS expression. There was, however, a dramatic change in the NADPH-diaphorase staining pattern of NOS-positive cells. NOS was absent from the distal portions of the dendritic trees of the deafferented SC. Nevertheless, staining the dendritic trees of these cells with Lucifer yellow showed that they were not morphologically different from those of the ipsilateral SC of monoenucleated animals. The same results were obtained when enucleation was performed in adult rats. We conclude that NOS intracellular distribution in the SC cells can be regulated by retinotectal projections in both developing and adult animals.

Nitric oxide (NO) is synthesized in cells of both the central and peripheral nervous system and has been implicated in several forms of synaptic plasticity. The enzyme that produces NO, nitric oxide synthase (NOS), can be visualized in the brain by the reduced nicotinamide adenine dinucleotide phosphate diaphorase histochemistry technique (NADPH-d). We have used NADPH-d activity to detect the presence of NOS-positive cells in the developing rat superior colliculus. Our results showed that NOS is present in cells and neuropil in the developing and adult rat superior colliculus. The first NOS-positive cells appeared at postnatal day 7 and were weakly stained. The number and intensity of the NOS-positive cells increased progressively during the following days reaching a maximum at postnatal day 15. By the end of the third postnatal week, both the number and intensity of stained cells showed an adult-like pattern. The NOS-positive cells showed a Golgi-like morphology and we have found that all cell types present in the superior colliculus express the enzyme. The expression of NOS by tectal cells parallels the functional development of the retino-collicular and cortico-tectal projections and suggest that nitric oxide synthase-positive cells might be involved in this process. In this review we highlighted some of the recent descriptions of the expression of NOS in the mammalian visual system with emphasis in the superior colliculus and correlate these findings with several developmental events taking place in this structure.
