To achieve the health benefit from the natural of Basella. Albafruit. This study intended to figure out the bioactive compounds in the two varieties of B. alba (native and hybrid) fruit extract and measurement its biological activities like antioxidant, anti-inflammatory, cytotoxic activities and a molecular docking were performed to observed the pharmaceutical impact on the anti-inflammatory Cyclooxygenase-2 (COX-2) enzyme. The cold extractions along with GC-MS were used for the extraction of and analysis of phytoconstituents from B. alba fruit. The hemolytic inhibitory and BSA (Bovine serum albumin)-denaturation assay, DPPH(2,2-diphenyl-1-picrylhydrazyl) and H2O2-free radical scavenging analysis, and brine shrimp lethalness test were performed to measure the biological activities of the extracted The biological activities assay results showed that the ethanol extract of native malabar spinach exhibited dose-dependent antioxidant activity. The IC50 value 21.55 ± 1.51 µg/mL was for DPPH scavenging assay and 23.36 ± 0.36 µg/mL was for H2O2 scavenging analysis. In anti-inflammatory activity assessment study, the IC50 values of the ethanol extracts were 20.52 ± 0.91 µg/mL for BSA inhibition and 20.43 ± 1.30 µg/mL for RBC hemolytic inhibitory study. In this study, cytotoxicity test results reveal that aqueous extract exhibited no cytotoxicity as compared to ethanol and ethyl acetate extract (LD50 = 875.27 µg/mL). Conversely, the current study insist the in silico analysis, to find out the anti-inflammatory activity of the investigated two fruit varieties due to pharmacokinetics analysis, toxicity properties analysis, ADMETand molecular docking. The result of this study signified that both (native and hybrid) malabar spinach fruit varieties contain phytoconstituents with potent antioxidant, anti-inflammatory, and cytotoxic action.Moreover, the in vitro and in silico results suggest that the native and hybrid fruit varieties of the extracts could be a superior striver for future appraisal as a prospective therapeutically active ingredient.

In this study, the levels of indicator PCBs congeners and PAHs compounds were reported in cow milk from selected areas in Dhaka, Bangladesh, and the potential human health risks were assessed. A total of 100 milk samples were collected and analyzed using gas chromatography-tandem mass spectrometry (GC-MS/MS). Method validation was performed using recovery performance, linearity, limit of detection (LOD), and limit of quantification (LOQ) assays. PCBs congeners, including PCB No. 52 (2,2´,5,5´-tetrachlorobiphenyl), PCB No. 101 (2,2´,4,5,5´-pentachlorobiphenyl), PCB No. 153 (2,2,4,4,5,5-hexachlorobiphenyl), and PCB No 209 (Decachloro-1,1'-biphenyl perchlorobiphenyl) were detected, whereas PCB No 28 (2,2 ´,4-trichlorobiphenyl), PCB No. 138 (2,2´,3,4,4´,5´-hexachlorobiphenyl), and PCB No 180 (2,2´,3,4,4´,5,5´-heptachlorobiphenyl) were not detected in the analyzed milk samples. Among the 16 PAHs compounds, benzo (a) anthracene and chrysene were detected in milk samples. The Σ hazard risk index (HI) of all detected PCBs congeners was below the limit set by the European Food Safety Authority, which indicates limited health risks to animals and humans in the study area. However, the presence of PCBs and PAHs in milk samples from industrial areas may negatively affect human health, and further detailed studies are required to ensure public health safety.

In the present study, the presence of organophosphorus (OPs) and carbamates (CBs) residues in the pond water and cultured Pangas catfish (Pangasius pangasius) samples collected from Comilla and Mymensingh areas were detected and assessed for their potential health risks. A total of 100 samples from each category were analysed among which 17% of the pond water samples and 9% of the fish samples were detected contaminated with OP and CB residues. The pond water and fish samples were extracted by liquid-liquid extraction (LLE), quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction, and ultrasonic extraction, respectively, and analysed through gas chromatography tandem mass spectrometry (GC-MS/MS). Among the detected OPs, Dursban (chlorpyrifos) and dichlorvos were detected, while among CB pesticides, carbofuran and sevin (Carbaryl) were detected in fish muscle samples. The detected OP and CB residual levels were below than the maximum residue limits (MRLs). The risk assessment study indicated no potential health risks. However, the level of compliance should be maintained through proper monitoring and controlling the overuse of pesticides in agricultural fields for public health safety.

The present study aimed to determine acrylamide traces in180 heat-treated carbohydrate-rich foods through gas chromatography coupled to mass spectrometry (GC-MS/MS) in Bangladesh. Detected acrylamide contents were 730 ± 293 µg/kg, 244 ± 83 µg µg/kg, 598 ± 222 µg/kg, 340 ± 189 µg/kg, 548 ± 278 µg/kg, 217 ± 77 µg/kg, 558 ± 297 µg/kg, 391 ± 263 µg/kg and 679 ± 285 µg/kg in potato chips, chanachur (a locally processed food), potato crisps, biscuits, cake, bread, crackers, breakfast cereals and French fries respectively. The use of different ingredients during the manufacturing process might affect on acrylamide formation as different ingredients contained variable amounts of free asparagines and reducing sugars to form acrylamide. Among the analyzed samples, 20% of potato chips, 5% of chanachur, 15% of potato crisps, 15% of biscuit, 10% of cake, 15% of bread, 20% of crackers, 10% of breakfast cereal and 20% of French fries samples, representing only a few samples in each category, were found to have acrylamide contents above benchmark levels set by the European Commission [EC]. This study provided an estimation of the presence of acrylamide traces in heat treated carbohydrate rich foods consumed by local population.

Blumea lacera (Burm. f.) DC. is attracting scientific interest due to the diverse biological activities of its various parts and its use in folk medicine. The present study was undertaken to investigate the tissue-specific differential expression pattern of its total bioactive compounds. The study was further extended to whole plant phenolics profiling, in vitro enzyme inhibition activities, followed by in silico enzyme inhibition analysis to assess its potential as herbal medicine. The amount of total phenolics in different tissues was followed in decreasing order as old leaf, flower bud, root, young leaf, flower, old stem, and young stem, while that for the flavonoids was old leaf, root, young leaf, flower bud, flower, young stem, and old stem. This study identified rosmarinic acid, quercetin, and kaempferol in this plant for the first time. The solvent extracts demonstrated strong inhibition of lipase and tyrosinase activity, along with varying degrees of inhibition of acetylcholinesterase and butyrylcholinesterase activity. Among the detected compounds, ten displayed strong in silico binding affinities with the tested enzymes. The findings provide a new insight into further investigation of the medicinal potential of this species against obesity, neurological disorders, and aberrant skin color.

Human exposure to nonessential trace elements occurs from food crops that are contaminated by the soil. The present study aimed to determine the level of heavy metals in soil and rice samples using an atomic absorption spectrophotometer from three major industrial areas in Bangladesh: Savar, Gazipur, and Ashulia. Heavy metals were detected in the order Fe > Zn > Ni > Cr > Pb > Co > Cu > Cd > As and Zn > Cu > Cr > Co > Fe > Cd > Pb > Ni > As in the soil and rice samples, respectively. From this analysis, it was observed that the detected concentrations of Zn, Cd, Cr, and Co were higher than the WHO/FAO recommended maximum tolerance values. The transfer factor of the detected heavy metals from soil to rice was detected in the following order: Zn > Cu > Cr > Co > Cd > Pb > Fe > As > Ni. The accumulation of heavy metals in rice is a major public health concern. Therefore, we recommend strict regulations for the safety of food crops grown in the study areas.

Bacteriocins have become biological weapons against harmful food pathogens and have attracted interest as tools for biopreservation. The aim of this study was to isolate, identify and characterize lactic acid bacterial (LAB) strains from cheese samples, partially purify potential bacteriocins and characterize their antimicrobial activity against pathogens. Bacteriocin-producing organisms were screened by Agar spot assay test. Initially, 25 LAB isolates were isolated from the cheese samples and identified as Lactobacillius spp., among them five strains were able to produce bacteriocin whose antimicrobial activates were analysed by agar-well-diffusion assay test against pathogenic organisms. Bacillus subtilis, Bacillus cereus, Staphylococcus aureus, Streptococcus thermophillus and Listeria monocytogens were inhibited, while Enterococcus faecalis, Salmonella typhi, Escherichia coli and Pseudomonas aeruginosa were resistant to the antimicrobial substances from LAB isolates. For optimal production of bacteriocin, LAB broth cultures were harvested at exponential phase. The molecular weights of the bacteriocins are between 7.0-15.0 kDa. The bacteriocins were characterized on the basis of their sensitivity to heat, pH, enzymes, NaCl and treatments with organic solvents. These results revealed that the bacteriocins from Lactobacillius spp. isolated from the cheese might have potential antimicrobial properties and give new insight in the development of bio-preservative agents to prevent and control pathogenic bacterial infection.

Blumea laciniata (Roxb.) DC. is a folk medicinal annual herb of the Asteraceae family that grows in South and Southeast Asia. In order to evaluate its phytopharmaceutical potential against diabetic, obesity, and Alzheimer's, a comprehensive phytochemical profile, in vitro and in silico enzyme inhibitory activity against α-amylase, α-glucosidase, lipase, cholinesterases, and tyrosinase along with in vitro antioxidant activity were performed. Additionally, in vivo antidiabetic activity and acute toxicity were also evaluated. The total phenolic content in various organs follows the following order: old leaf > flower bud > young leaf > flower > young stem > old stem > root, while total flavonoids followed the order: flower bud > old leaf > young leaf > flower > young stem > old stem > root. The identified phenolic compounds are 3,4-dihydroxybenzoic acid, caffeic acid, vanillic acid, p-coumaric acid, syringic acid, rosmarinic acid, trans-cinnamic acid, catechin, catechol, (-) epicatechin, rutin, quercetin, myricetin, and kaempferol, which are also expressed differently in various organs. Solvent extracts demonstrated strong antioxidant activity as well as varying levels of inhibition against the enzymes tested, with strong inhibitory activity against α-amylase, α-glucosidase, and lipase. Thirteen phenolic compounds displayed strong binding affinity in silico against studied enzymes, thus documented as bioactive. Furthermore, solvent extracts significantly suppressed blood glucose levels in mice with induced diabetes and extracts were not acutely toxic. The results suggest that Blumea laciniata (Roxb.) DC. could be a potential candidate for developing new phytopharmaceuticals and bioactive ingredients.

The hybrid variety of Cucurbita maxima is commercially available and frequently used as food in Bangladesh compared to the native variety. Although the seeds of Cucurbita maxima have nutritional and therapeutic values, people in Bangladesh typically throw them away as waste. If we can explore their physicochemical and biochemical properties, this will add new knowledge to nutrition and food science. Thus, in this study, we compare the physicochemical and biochemical properties of native and hybrid varieties of pumpkin seed and seed oil. Solvent extraction methods were used to obtain oil. The physicochemical properties of the extracted seed oils were examined by titration methods and proximate compositions of pumpkin seeds were determined using the standard method. Tocopherols were analyzed by HPLC and fatty acids were identified by GC/MS as N-acylpyrolidines. The specific gravity, refractive index, viscosity, acid value, saponification value, iodine value and ester values were determined and among them iodine values and acid values were significant (p < 0.05) at native but saponification and ester values were very highly significantly (p < 0.001) at hybrid. The comparative lipid content in the seed of native variety was higher than hybrid (p < 0.01) and protein contents were higher but not significant. However, crude fiber content was higher in the hybrid (p < 0.01). There was no significant differences of moisture, ash, sugar and starch contents in between the two varieties. The total tocopherol, contents were very high (p < 0.001) in native. In hybrid, saturated fatty acids -capric acid, myristic acids were more (p < 0.001) but in native unsaturated fatty acid linoleic and linolenic acids were predominant (p < 0.05). Owing to the considerable differences in the physicochemical composition, the fatty acids and other properties, this study may suggest that the both varieties of pumpkin seed oil may be an alternative good source of edible oil.

The objective of this study was to elucidate if Bisphenol A (BPA) administration modulates T helper (Th) cell component of immune responses in a mouse challenged with ovalbumin (OVA), a major food antigen. BALB/c mice, (6 weeks old, female) were orally given either OVA (OVA-fed) or water (Water-fed), immunized intraperitoneally with OVA and injected with either BPA in corn oil or the vehicle alone. After subsequent 2nd immunization, serum titers of total IgE, OVA-specific IgE, IgG, IgG1 IgG2a and ability of their splenocytes for production of interferon (IFN) -gamma, interleukin (IL) -4 and IL-12 were examined by ELISA. Lymphocyte proliferation assay against concanavalin A (Con A) or OVA was also performed for 3H-Thymidine incorporation. In Water-fed groups, treatment with BPA resulted in lower titers of total IgE (P<0.01) and higher levels IgG2a (P<0.05) followed by a higher IFN-gamma (P<0.05) and IL-12 (P<0.05) with an intact IL-4. When OVA-fed groups were examined, the compound did not change production of total and OVA-specific IgE and -IgG2a but resulted in lower production of IFN-gamma (P<0.05). Also, BPA resulted in impaired lymphocyte proliferation to Con A in Water-fed groups (P<0.05) but not in tolerated animals. The findings indicate that BPA results in augmentation of Th1 immune responses but no significant effect on an established tolerance to OVA.

Oral tolerance is a potential strategy for preventing or minimizing aberrant immune responses. Although, oral tolerance has been extensively studied, to date the effects of dietary protein on the induction of oral tolerance are poorly understood. We have previously shown that restricted dietary protein induces oral tolerance to ovalbumin. This study was designed to investigate whether or not such tolerance occurs with beta-lactoglobulin (BLG) instead of ovalbumin (OVA) and if the tolerance resulting from this feeding regimen involves Th1-mediated immune response. Female BALB/c mice fed either 20% or 5% dietary protein were given 5 mg BLG or water orally for four consecutive days and then immunized intraperitoneally (ip) twice with BLG at 3-wk intervals. Oral tolerance induction was compared in BLG-fed and water-fed mice by measuring total IgE, BLG-specific antibodies, footpad reactions, splenocyte proliferation, and cytokine production. When mice were given BLG orally before ip immunization, the Th1-mediated immune responses (production of IL-2, IFN-gamma, and IgG2a) were significantly reduced, whereas the Th2-mediated immune responses (production of IL-4 and IgG1) were unchanged. The Th1-mediated immune responses were markedly down-regulated in mice fed 5% protein as compared to those in mice fed 20% protein. Moreover, the production of total IgE, BLG-specific IgE, splenocyte proliferation, and footpad reactions were more reduced in mice fed 5% protein than those in mice fed 20% protein. The present study provides evidence that dietary protein plays an important role in the induction of oral tolerance against BLG as the result of, clear down-regulation of Th1 helper activity accompanied by a reduction in IgE.

The suitable development of oral tolerance against ingested dietary foods is of critical importance to escaping food allergy. Using mice as an animal model for oral tolerance against ovalbumin (OVA) as a dietary antigen, we investigated the effects of dietary protein on their immunological tolerance. Female BALB/c mice fed either a 20% or 5% protein diet were orally administered 5 mg of OVA for four consecutive days, then immunized intraperitoneally with 100 microg of OVA. The immunized group of mice were fed and treated in the same manner, except that they received orally distilled water for four consecutive days before receiving intraperitoneal immunization with the antigen. Immunization alone with OVA elevated the total IgE and induced the production of OVA-specific antibodies IgE, IgG, IgG1, and IgG2a in the sera of both the 20% and 5% protein diet groups. The oral administration of OVA to mice before intraperitoneal immunization significantly reduced the total IgE and OVA-specific antibodies in mice fed 5% protein diet, but it had hardly any effect on those in mice fed a 20% protein diet. When spleen cells from these groups of mice were cultured with OVA as a mitogen, they responded substantially to OVA in the immunized groups fed 20% and 5% protein diets and in the presensitized group fed 20% protein, but those from the presensitized group fed a 5% protein diet did not respond. Furthermore, when IL-4 was assayed in the spleen cell cultures of the 20% and 5% groups, mice in the presensitized group fed a 5% protein diet produced a significantly less amount of IL-4 than those fed a 20% protein diet. Moreover, irrelevant to the protein amount in the diet, the production of IFN-gamma from spleen cell cultures dramatically decreased in the group without presensitization and profoundly increased in the presensitized group of mice fed a 5% protein diet. These findings suggest that a low-protein diet leads to an induction of oral tolerance against dietary antigens; this appears to involve a clear down-regulation of Th2 cytokine, IL-4.
