Stochastic wave-function unravelling of the generalized Lindblad equation.

We investigate generalized non-Markovian stochastic Schrödinger equations (SSEs), driven by a multidimensional counting process and multidimensional Brownian motion introduced by A. Barchielli and C. Pellegrini [J. Math. Phys. 51, 112104 (2010)JMAPAQ0022-248810.1063/1.3514539]. We show that these SSEs can be translated in a nonlinear form, which can be efficiently simulated. The simulation is illustrated by the model of a two-level system in a structured bath, and the results of the simulations are compared with the exact solution of the generalized master equation.

Dynamics of behavioral disorders in transgenic mice with modeled Alzheimer's disease.

Age-related development of behavioral disorders in transgenic mice with modeled Alzheimer's disease carrying V6S3-Tg(APP695)85Dbo Tg(PSENI)85Dbo) genotype was assessed at the age of 7.5, 10 and 20 months in the following tests: open-field, plus maze, T-maze, conditioned passive avoidance response, rotarod, conflict situation with water deprivation, behavioral despair, and arecoline tremor. The main behavioral disorder in transgenic mice at all observation terms was memory impairment in conditioning with positive (but not negative) reinforcement. At the age of 7.5 and 10 months, transgenic mice also showed signs of nonspecific excitement and anxiety, depression-like state, and symptoms of cholinergic deficit. Our results suggest that appropriate age for behavioral tests in studies of effects of potential anti-Alzheimer drugs in transgenic V6S3-Tg(APP695)85Dbo Tg(PSENI)85Dbo) mice is 7.5-10 months.

Possible involvement of neurokinin receptors in CNS and neuromuscular synapse in the realization of the effects of CAPAH and substance P.

Nootropic non-anticholinesterase organophosphorus preparation CAPAH and substance P produced similar and dose-dependent effects on the amplitude and temporal parameters of miniature endplate potentials in mammalian neuromuscular synapse. Neurokinin receptor antagonist Win-51.708 abolished the effects of these agents. In behavioral experiments substance P moderated the mnemotropic and antidepressant effects of CAPAH. It was assumed that neurokinin receptors are the targets of CAPAH and substance P in CNS and neuromuscular synapse.

[Neuroprotective effect of kapakh, representative of new nootropic class non-anticholinesterase organophosphoric compounds]. / Neroprotektivnoe deistvie kapakh, predstavitelia novogo klassa nootropov-neantikholinésteraznykh fosforoorganicheskikh soedinenii.

In vivo and in vitro experiments revealed the neuroprotective activity of CAPAH, a representative of the new nootropic class phosphorylated carboxylic acid hydrazides. The mechanisms of this action, which were due to the membranous stabilizing activity of the drug and its affinity for glycine strychnine-sensitive sites of MMDA receptors (Ki = 8.8.10(-5) M). Six analogues of CAPAH have been also found to have affinity for these types of receptors; based on the examination of structure-affinity relationship, putative pharmacophores (only radicals of the benzene ring of a molecule in the para position exerted effects on affinity changes) were revealed. The findings give a deep insight into the mechanism of nootropic activity and open vistas for synthesizing phosphorylated carboxylic acid hydrazides as potential neuroprotective agents interacting with glycine strychnine-sensitive sites of MMDA receptor.

Some physicochemical properties of B.pertussis antigenic complexes isolated from synthetic and semisynthetic culture media have been studied. The chemical composition of these complexes has been determined. Proteins, polypeptide subunits and lipopolysaccharide forming these complexes have been characterized. The presence of two main protective substances of B.pertussis has been revealed: fimbrial hemagglutinin and B.pertussis toxin. The influence of culture medium on the level of the synthesis of B.pertussis adenylate cyclase has been studied.

A comprehensive intraspecies typing of the cultures of MRSA collected during inspection of drug resistance in causative agents of intrahospital infections was performed. The following parameters were investigated: antibiotic resistance, toxin production, sensitivity to the phages of the International Set and the phages of an experimental collection providing the isolation of strains with definite specificity of the restriction-modification system. Different clones of methicillin resistant S. aureus were found to be circulating on the territory of the CIS.

[The safety and protective properties of a new cell-free pertussis vaccine in animal trials]. / Issledovanie bezvrednosti i protektivnykh svoistv novoi beskletochnoi kokliushnoi vaktsiny v opytakh na zhivotnykh.

The preclinical trial of a new cell-free pertussis vaccine has been carried out with a view to its comparison with whole-cell pertussis vaccine in respect of the toxic and protective properties of the preparations under experimental conditions. The whole-cell pertussis vaccine is characterized by pronounced toxic action causing leukocytosis and histamine-sensitization, produces a negative effect on the detoxifying function of the liver and possesses sensitizing activity. The cell-free pertussis vaccine has pronounced protective activity. In animal experiments the supposed immunization dose of this vaccine has proved to be nontoxic.

The composition of antigenic complexes isolated from the supernatant fluid of B. pertussis culture has been studied by means of immunoblotting techniques. In preparations obtained from B. pertussis strains 305 and 475 fragments of the molecule of fimbrial hemagglutinin, three subunits of B. pertussis toxin and agglutinogens 2 and 3 have been detected with the use of antisera to B. pertussis protective substances.

[Physicochemical and biological characteristics of preparations of an antigenic complex isolated from the cultivation medium of Bordetella pertussis]. / Fiziko-khimicheskaia i biologicheskaia kharakteristika preparatov antigennogo kompleksa, vydelennogo iz sredy kul'tivirovaniia kokliushnykh mikrobov.

The physicochemical and biological properties of antigenic complexes isolated from the supernatant fluid of the culture medium of B. pertussis, strains 305 and 475, were studied. The preparations obtained from both strains contained proteins, carbohydrates, nucleic acids and lipids. Electrophoresis in polyacrylamide gel revealed the presence of filamentous hemagglutinin, 4 subunits of B. pertussis toxin and agglutinogens in the antigenic complexes of both strains. The preparations of both strains possessed similar toxic properties and, after their detoxification, produced a pronounced protective effect.

A new restriction endonuclease Pmi I was detected in Proteus mirabilis 1667. The enzyme hydrolyzes DNA of the phage lambda into 10 electrophoretically separating fragments with molecular weights of 1.3-7.9 mD. With the use of two-stage chromatography on blue sepharose and phosphocellulose it is possible to obtain restriction endonuclease Pmi I free of the admixtures of ballast proteins, nonspecific nucleases and phosphatases.

[Production of immobilized SalI and PvuII restrictase preparations]. / Poluchenie preparatov immobilizovannykh restriktaz SalI i PvuII.

Conditions for the immobilization of specific endonucleases Sal I and Pvu II on BrCN-activated Sepharose 4B have been selected. Some physico-chemical properties of the preparations of immobilized restrictases Sal I and Pvu II have been characterized. The specific and general activity values of the preparations thus obtained have been established. The immobilized enzymes have been used for the multiple restriction of the DNA of phage lambda and the DNA of Neisseria meningitidis.

The present article deals with the study of the multicyclic process of the cultivation of B. amyloliquefaciens in a liquid medium prepared from natural raw materials and with obtaining restrictase Bam H1 from the biomass with different microbiological characteristics. The selected medium has been found to be suitable for the realization of the multicyclic cultivation process on an industrial scale. The tendency towards a drop in the level of nonspecific nucleases simultaneously with a decrease in the specific growth rate has been revealed. A simplified scheme for the purification of the restricting endonuclease is proposed.

Hybrid plasmids containing B. pertussis DNA insertions have been constructed with the use of the vector plasmid pBR 322 and the Pst I fragments of B. pertussis DNA. Some properties of the hybrid plasmids are characterized. The possibility of the expression of B. pertussis genes in the protein-synthetizing cell-free system obtained from E. coli has been demonstrated.

A number of physico-chemical properties of Bordetella pertussis DNA has been studied. The values of its floating density and melting point have been established, which has allowed one to calculate the GC composition of B. pertussis DNA. The average molecular weights of the fragments of B. pertussis DNA, resulting from its hydrolysis with specific endonucleases EcoR, BamH 1, Sal 1, Pst 1, have been determined, and thus the basis has been provided for establishing the number of clones necessary for obtaining the complete lset of B. pertussis genes.

New restriction endonuclease has been isolated from Bordetella pertussis vaccine strain 305 and purified in 1 stage on Sepharose covalently bound with blue dextran. The isolated restrictase has been found capable of breaking down lambda-phag DNA into 7 fragments. According to its specificity, Bpe I is the isoschizomer of Hind III obtained from Haemophilus influenzae strain Rd.

[Structural characteristics (reassociation and melting kinetics) of kinetoplast DNA from Crithidia oncopelti]. / Kharakteristika struktury kinetoplastnoi DNK Crithidia oncopelti po kinetike reassotsiatsii i plavleniiu.

A highly purified associate of kinetoplast DNA is isolated from C. oncopelti, and its physico-chemical properties are studied. Both native associate and its ultrasonic fragments are found to have a complex character of melting. 5-6 melting zones (3 of them being the main) are found on the melting curve. Analysis of reassociation kinetics of sonicated associate of kinetoplast DNA has revealed the presence of at least two components: fast reassociating component (65-70% of complex DNA), which reassociation kinetics is equivalent to the unique sequence with molecular weight of 2.3. - 10(6) daltons, and slow reassotiating component (15% of complex DNA), having reassociation kinetics equivalent to unique sequence of 26 - 10(6) daltons. The data obtained suggest that complex associate of kinetoplast DNA is heterogenous for its nucleotide sequence and base composition.
