We studied the possibility of introduction of heterogeneous genetic information in adult animals through intraperitoneal injection of plasmid DNA precipitated by calcium phosphate. Accumulation of heterogeneous DNA in the liver was shown by Southern hybridization and polymerase chain reaction. Unlike the published data, we obtained a more prolonged (up to 11 days) preservation of heterogeneous DNA in the animals.

Attempts to transfer gene p53 via pAT153 vector to the mouse genome are described. Transgenic mice were obtained that carried sequences homologous to the pAT153 only. No transmission of foreign nucleotide sequences to progeny was observed.

The literature is reviewed concerning the factors of the foreign DNA integration into the genome and the regulation of tissue and stage specific expression of the introduced genes in transgenic animals. Examples are given of utilizing transgenic mice for testing various hypotheses in immunology, oncology, virology, etc.
