RESUMO

Multiple comorbidities related to arsenic exposure through drinking water continue to be public problems worldwide, principally in chronically exposed populations, such as those in the Comarca Lagunera (CL), Mexico. In addition, this relationship could be exacerbated by an early life exposure through the placenta and later through breast milk. This study conducted a comparative analysis of arsenic levels in multiple biological samples from pregnant women and their neonates in the CL and the comparison region, Saltillo. Total arsenic levels in placenta, breast milk, blood, and urine were measured in pregnant women and their neonates from rural areas of seven municipalities of the CL using atomic absorption spectrophotometry with hydride generation methodology. The average concentrations of tAs in drinking water were 47.7 µg/L and 0.05 µg/L in the exposed and non-exposed areas, respectively. Mean levels of tAs were 7.80 µg/kg, 77.04 µg/g-Cr, and 4.30 µg/L in placenta, blood, urine, and breast milk, respectively, in mothers, and 107.92 µg/g-Cr in neonates in the exposed group, which were significantly higher than those in the non-exposed area. High levels of urinary arsenic in neonates were maintained 4 days after birth, demonstrating an early arsenic exposure route through the placenta and breast milk. In addition, our study suggested that breastfeeding may reduce arsenic exposure in infants in arsenic-contaminated areas. Further studies are necessary to follow up on comorbidities later in life in neonates and to provide interventions in this region.

BACKGROUND: The COVID-19 pandemic had infected more than 3.5M people around the world and more than 250K people died in 187 countries by May 2020. The causal agent of this disease is a coronavirus whose onset of symptoms to death range from 6 to 41 days with a median of 14 days. This period is dependent on several factors such as the presence of comorbidities, age and the efficiency of the innate or adaptive immune responses. METHODS: The effector mechanisms of both types of immune responses depend on the pathogen involved. In the case of a viral infection, the innate immune response may approach the harmful virus through pattern recognition receptors inducing an antiviral state. RESULTS: On the other hand, the adaptive immune response activates antibody production to neutralize or eliminate the virus. Phenolics are plant secondary metabolites with many biological activities for plants and humans against infection. Chemical modification of proteins may enhance their biological properties; thus, a protein of medical interest, for instance, a viral protein can be used as a scaffold to build a biopharmaceutical conjugated or complexated with phenolics exhibiting structural complexity or biological activities to achieve effective phenolic-protein-based therapeutics like vaccine adjuvant complexes, immunogen conjugates, and antiviral conjugates. CONCLUSION: Pharmaceutical biotechnology applies the principles of biotechnology to develop biopharmaceuticals for protein-based therapeutics; such as adjuvants, recombinant proteins, monoclonal antibodies, and antivirals. As neither a vaccine nor a treatment for COVID-19 is currently available, this manuscript focuses on insights from pharmaceutical biotechnology into phenolic biopharmaceuticals against COVID-19.

BACKGROUND: The rate of lead poisoning has decreased in recent years due to increased health control in industries that use this metal. However, it is still a public health problem worldwide. The use of various plants with chelating properties has been a topic of research today. In traditional medicine, it is said that Coriandrum sativum has chelating properties, but there is no scientific evidence to support this fact. The purpose of this research is to evaluate the chelating effect of methanol extract of coriander and its fractions on Wistar rats intoxicated with lead. MATERIALS AND METHODS: In this research, male Wistar rats were poisoned with 50 mg/kg of lead acetate and treated with 50 mg/kg of methanol extract and its fractions. The extract and its fractions were administered to four treatment groups. Positive and negative controls were established. Hemoglobin, hematocrit and lead concentrations were analyzed; liver was evaluated histologically in control and treatment groups. RESULTS: The methanol extract of coriander presented a LD50 >1000 mg/dL. The group administered with the methanol extract showed significant difference in the levels of hemoglobin and hematocrit compared to the negative control group. Lead concentration in treatment groups showed a decrease compared to the positive control. Histological evaluation of tissue showed less damage in groups administered with methanolic extract and its fractions compared to the positive control which presented structural alterations. CONCLUSION: Coriander extracts protect liver and lower lead concentration in rats intoxicated with lead in contrast to the positive control group.

BACKGROUND: Since the time when drugs began to be used, it became evident that they could produce a therapeutic effect, but also a clinical condition of toxicity or no effect at all on humans, despite using the same doses in different patients. Such untoward effects were termed "drug idiosyncrasy" and also "idiosyncratic drug effects", but the factors producing such diverse responses were never taken into account. MATERIALS AND METHODS: Ruta chalepensis L. (fringed rue) is an herbaceous plant of the Rutaceae family used in traditional medicine due to its properties, such as its analgesic and antipyretic effects. This study used 25 male rats divided into five groups. Plant extract was administered to Groups 1 and 2 at doses of 100 and 30 mg/kg/day, respectively, for three days; Group 3 was administered 100 mg/kg/day of dexamethasone (DEX), as well as 100 mg/kg/day of Ruta chalepensis extract; Group 4 was administered 100 mg/kg/day of DEX and treated as positive control; Group 5 was treated as negative control and was administered a physiological solution. Twenty-four hours after the the last dose, the animals were sacrificed and their livers were extracted. RESULTS: The aqueous extract of Ruta chalepensis, intraperitoneally administered, was able to induce cytochrome 3A1 in doses of 30 mg/kg/day, and a greater inducing effect occurs when the plant is co-administered in doses of 100 mg/kg/day with dexamethasone. CONCLUSION: This study suggests that aqueous extract of Ruta chalepensis can induce cytochrome 3a1. This study helps provide a better understanding of CYP3a regulation. Future in vitro work is needed to determine the compounds that produce the cytochrome modulation.

OBJECTIVE: To assess whether in Mexican population the frequencies of ATM polymorphisms IVS24-9delT, IVS38-8-T>C, and 5557G>A in breast cancer (BC) cases and healthy controls were different from those found in other countries. MATERIALS AND METHODS: Frequencies of polymorphisms conferring BC risk IVS24-9delT, IVS38-8T>C, and 5557G>A were analyzed by PCR-RFLP in 94 patients with familial and/or early onset BC, and 97 healthy controls randomly selected. Allele frequencies analysis was done using χ(2) and Hardy-Weinberg test. RESULTS: Frequencies of heterozygous were: for 5557G>A, 13% cases, 0%controls (p=0.0009); for IVS24-9delT, 21% cases, 8% controls (p=0.0122); for IVS38-8T>C, only one case. 5557G>A and IVS24-9delT were more frequent in cases than in controls. The allelic frequencies found in 5557G>A are similar to those described by González-Hormazábal in Chile. CONCLUSION: The similarity of results in this polymorphism between Chilean and Mexican populations may be due to both being crossbred with an Amerindian-Spanish component, while differences may be due to fact that Chilean population has a greater European component than Mexican's.

Evaluar si en la población mexicana las frecuencias de los polimorfismos IVS-9delT, IVS38-8T>C y 5557G>A en casos de cáncer de mama y en controles sanos son diferentes de las encontradas en otros países. Material y métodos. Los polimorfismos IVS24-9delT, IVS38-8T>C y 5557G>A fueron analizados mediante PCR-RFLPs en 94 pacientes con CM de tipo familiar o de inicio temprano y 97 testigos seleccionadas de forma aleatoria. El análisis de la frecuencia alélica se hizo mediante χ² y equilibrio de Hardy-Weinberg. Resultados. Las frecuencias de heterocigotos fueron 5557G>A, 13% de casos, 0% de testigos (p=0.0009); IVS24-9delT, 21% de casos, 8% de testigos (p=0.0l22); IVS38-8T>C, sólo un caso. 5557G>A y IVS24-9delT fueron más frecuentes en casos que en testigos. Las frecuencias alélicas encontradas en 5557G>A son similares a las descritas por González-Hormazábal en Chile. Conclusión. La similitud de resultados en este polimorfismo entre la población chilena y mexicana puede ser debida a que ambas son mestizas con un componente amerindio-español. Las diferencias encontradas podrían explicarse porque la población chilena tiene mayor componente europeo que la mexicana.

Desde tiempos antiguos la medicina tradicional ha usado extensamente las especies del género Lippia como analgésicos, antiinflamatorios, antipiréticos, antifúngicos, etc. Numerosos estudios describen diversos compuestos presentes en extractos acuosos, metanólicos, o aceites esenciales de estas plantas, siendo los flavonoides los compuestos más abundantes. Sin embargo, la composición y cantidad de los metabolitos secundarios dependen de la zona geográfica, factores climáticos, altitud, época de cosecha y estado de crecimiento de estas plantas. El objetivo de este estudio fue evaluar la actividad antioxidante del extracto acuoso de orégano (Lippia graveolens HBK) del Norte de México y su efecto tóxico in vitro e in vivo. La capacidad antioxidante del extracto acuoso se midió por el método de DPPH en seis diluciones del extracto (5-160 mg/mL) y se utilizó Trolox como referencia; para el efecto tóxico in vitro se usó el ensayo de citotoxicidad con larvas de Artemia salina. Para el efecto in vivo se emplearon 24 ratones árabes machos divididos en 6 grupos de animales (n=4): 4 grupos experimentales con 10, 100, 1000 y 5000 mg del extracto/ kg de peso administrados vía oral respectivamente, además de un grupo control positivo (5 mg de colchicina/kg de peso vía i.p) y un grupo control negativo (solo agua destilada). Después del tratamiento los ratones se sacrificaron y se colectaron muestras de hígado y riñón que se sometieron a estudios histológicos e histoquímicos, además se realizó un análisis cuantitativo. La actividad antioxidante del extracto acuoso de orégano se presentó a 160 mg/mL. La CL50 fue mayor a 1,000 ug/mL por lo que el extracto se considera no tóxico. En el análisis morfológico in vivo con distintas dosis del extracto acuoso de orégano no se observó un efecto tóxico. Since ancient times, traditional medicine has widely used species of the genus Lippia as analgesics, anti-inflammatory, antipyretic, antifungal, etc. Numerous studies describe several compounds present in aqueous extracts, methanol, or essential oils of these plants, being flavonoids the most abundant compounds. However, the composition and quantity of secondary metabolites depend on the geographical area, climatic factors, altitude, time of harvest and growth status of these plants. The objective of this study was to evaluate the antioxidant activity of aqueous extract of oregano (Lippia graveolens HBK) from the North of Mexico and its toxic effect in vitro and in vivo. The antioxidant activity of the aqueous extract was measured by DPPH method in six dilutions of the extract (5-160 mg / mL), Trolox was used as a reference. For the in vitro toxic effect, cytotoxicity assay with larvae of Artemia salina was used. For the in vivo effect, 24 males mice were used and divided into 6 groups (n = 4): 4 experimental groups with 10, 100, 1000 and 5000 mg extract / kg body weight administered orally respectively, also we used a group positive control (5 mg of colchicine / kg body weight administered via ip) and a negative control group (distilled water only). After treatment all mice were sacrificed, and samples from liver and kidney were collected and analyzed by histological and histochemical studies. Also a quantitative analysis was done. The antioxidant activity of aqueous extract of oregano was presented at 160 mg/mL. The LC50 was greater than 1.000 mg/mL, so the extract is considered nontoxic. In the morphological analysis in vivo with different doses of aqueous extract of oregano, no toxic effect was observed. The results validate the use of oregano in traditional medicine...

RESUMO

El propósito de este estudio fue establecer un modelo de agregación plaquetaria que permitiera evaluar "in vivo" (en conejos Nueva Zelanda) parámetros hemodinámicos y microscópicos. Se indujo la agregación plaquetaria con la administración de colágena I.V. 75 µg/kg/min, la cual produjo disminución de la presión arterial sistólica de x- = 69 ax- = 55 mm Hg y diastólica de x- = 43 ax- = 27 mmHg, con aumento de la ventricular de x- = 25 ax- = 41 mm Hg. Se administraron aspirina, dipiridamol o sulfinpirazona 10 mg/kg, media hora antes de la administración de colágena y prostaciclina 100 mg/kg/min desde 3 minutos antes, hasta 10 minutos después de la colágena y aspirina, colágena y dipiridamol, disminuyó la presión arterial tanto sistólica como diastólica, sin modificación de la ventricular. No se observaron cambios hemodinámicos con la administación conjunta de colágena sulfinpirazona o colágena prostaciclina. Se observaron por histología trombosis vasculares pulmonares múltiples con la administración de colágena y, en menor intensidad, cuando se administró ésta conjuntamente con un fármaco antiagregante. Este modelo no permitió valorar hemodinámica e histológicamente susbtancias pro y antiagregantes plaquetarias.
