RESUMO

Using structural similarity approach we identified dillapiole, a phenylpropanoid, the main component of Piper aduncum L. and Anethum graveolens L. essential oils as potential PPARγ agonist. Molecular docking revealed that dillapiole binds to the active site of PPARγ, similar to pioglitazone binding. In silico ADME studies showed that dillapiole has high water solubility and GI absorption. Dillapiole was also observed to be partial agonist of PPARγ receptors with EC50 of 43.95 µM. In BHK-21 cells cultured under hyperglycaemic conditions, dillapiole administration reduced oxidative stress and prevented decrease in histone H3 acetylation (k9/14) levels. In HFD + STZ induced diabetic mice, dillapiole treatment for 7 days was able to improve renal functions and decrease plasma glucose level to 138.39 ± 12.36 mg/dl along with decreasing total cholesterol (29%), triglycerides (48.8%), LDL (24.7%), and VLDL (65%) levels in serum. These results show that dillapiole is a potential PPARγ-agonist and thus needs to explore further.

RESUMO

Even though climate change involves much more than warming, it is the name given to a set of physical phenomena. It is a long-term change in weather patterns that characterises different regions of the world. The warming effect in the earth's atmosphere has dramatically increased through the influence of some heat-taping gases emitted by various human activities, especially fossil fuel burning. The more the input of such gases, the more will be the warming effect in the coming times. Global climate change is already visible in various parts of the larger ecosystems like forests, fisheries, biodiversity, and agriculture; however, it is now also influencing the supply of freshwater, human health, and well-being. This paper reviews climate change drivers, its global scenario, major global events, and assessing climate change impacts. The most daunting problem of economic and ecological risks, along with the threats to humanity, is also discussed. The paper further reviews the species' vulnerability to climate change and the heat waves and human migration vis-à-vis climate change. Climate change politics and coverage of climate change episodes in mass media is the special focus of this review that concludes with a few mitigation measures.

RESUMO

Intense research interests have been observed in establishing PPAR gamma as a therapeutic target for diabetes. However, PPARγ is also emerging as an important therapeutic target for varied disease states other than type 2 diabetes like neurodegenerative disorders, cancer, spinal cord injury, asthma, and cardiovascular problems. Furthermore, glitazones, the synthetic thiazolidinediones, also known as insulin sensitizers, are the largely studied PPARγ agonists and the only ones approved for the treatment of type 2 diabetes. However, they are loaded with side effects like fluid retention, obesity, hepatic failure, bone fractures, and cardiac failure; which restrict their clinical application. Medicinal plants used traditionally are the sources of bioactive compounds to be used for the development of successful drugs and many structurally diverse natural molecules are already established as PPARγ agonists. These natural partial agonists when compared to full agonist synthetic thiazolidinediones led to weaker PPARγ activation with lesser side effects but are not thoroughly investigated. Their thorough characterization and elucidation of mechanistic activity might prove beneficial for counteracting diseases by modulating PPARγ activity through dietary changes. We aim to review the therapeutic significance of PPARγ for ailments other than diabetes and highlight natural molecules with potential PPARγ agonistic activity.

RESUMO

In recognition of the importance of bacteria as ecological indicators of the aquatic systems a comprehensive and systematic analysis was carried out on Manasbal Lake, the deepest spring fed valley lake of Kashmir. The main objective envisaged was to analyze bacterial community composition (BCC) and for this purpose systematic and regular sampling of waters from ten different sampling stations, predetermined in the Lake according to differences in degree of human interference and also as zones of special ecological interests were selected. The isolated species were identified according to Bergey's Manual specification by examining their micro and macro morphological characteristics and biochemical characteristics on different culture media. Further confirmation was done by sequencing the 16s rRNA gene by using universal bacterial primers 27F and 1429R. From all the sampling stations the class Bacilli showed a maximum relative abundance with a contribution of 16 bacterial species. The whole process resulted in the identification of Bacillus aerius, Bacillus altitudinis, Bacillus anthracis, Bacillus cereus, Bacillus ginsengisoli, Bacillus pumilus, Bacillus safensis, Bacillus stratosphericus, Bacillus subtilis, Bacillus tequilensis, Bacillus thermocopriae, Bacillus thuringiensis, Brevibacillus agri strain, Lysinibacillus boronitolerans, Lysinibacillus pakistanensis and Lysinibacillus sphaericus.

RESUMO

The primary aspiration in the microbial observatory is to advance the understanding of freshwater bacterioplankton, whose diversity and population dynamics are currently the least understood off all freshwater planktonic organisms. Through identification and characterization of bacterial populations in a suite of fresh water bodies, we are able to gain significant new insight into the ecological niches of bacteria in diverse freshwater ecosystems. Given the facts that lakes and other inland freshwaters play a more critical role in the global carbon budget and that lakes have been described as early indicators of both regional and global environmental change, the role of microbes in these processes is of renewed interest. In this review, general overview will be given highlighting the characteristic features of bacterial species thriving in different water bodies. In the following sections, different cultural approaches vis a vis the controlling factors of bacterial diversity have been elaborated. In the concluding sections, the prospects of aquatic microbial diversity are well mentioned.

RESUMO

Endophytic fungi are those that live internally in apparently healthy and asymptomatic hosts. Endophytic fungi appear to be ubiquitous; indeed, no study has yet shown the existence of a plant species without endophytes. High species diversity is another characteristic of endophytic mycobiota which is depicted by the fact that it is quite common for endophyte surveys to find assemblages consisting of more than 30 fungal species per host plant species. Medicinal plants had been used to isolate and characterize directly the bioactive metabolites. However, the discovery of fungal endophytes inside these plants with capacity to produce the same compounds shifted the focus of new drug sources from plants to fungi. Bioactive natural products from endophytic fungi, isolated from different plant species, are attracting considerable attention from natural product chemists and biologists alike which is clearly depicted by the steady increase of publications devoted to this topic during the recent years. This review will highlight the chemical potential of endophytic fungi with focus on the detection of pharmaceutically valuable plant constituents as products of fungal biosynthesis. In addition, it will cover newly discovered endophytic fungi and also new bioactive metabolites reported in recent years from fungal endophytes. It summarizes the up-to-date and comprehensive information on bioactive compounds from endophytic fungi by having done a thorough survey of literature.
