Solanum nigrum plant has rich therapeutic potential and different developed countries utilize this plant as a chief element for oriental medicinal practice including cancer therapy. The current study determines the isolation and purification of S. nigrum bioactive constituents through column chromatography from ethyl acetate extract of the plant followed by the Gas Chromatography-Mass Spectroscopy (GC-MS) to analyze the isolated compounds Different gradient elutions followed by thin layer chromatography of collected fractions were done and structural analysis of the isolated compounds was performed following GCMS analysis, More specifically the compounds were identified as 1, 2 benzene dicarboxylic acid, diisooctyl ester (95%) and as Bis (2-Ethylhexyl) phthalate (84%) along with 3 hydroxy 4 carboxy 2 methyl 6 pyridine (58%) with reference to chemical abstract service which may be responsible for its pharmacological properties. According to the best information available, no documented information exists regarding GC-MS based identification of the isolated chemical compounds from the S. nigrum. Present findings will help in exploring the therapeutic potential of 1, 2 benzene dicarboxylic acid, diisooctyl ester (95%), Bis (2-Ethylhexyl) phthalate (84%) and 3 hydroxy 4 carboxy 2 methyl 6 pyridine (58%) will be helpful in the development of new composites in pharmaceutical fields.

The renin-angiotensin system (RAS) is involved in body fluid regulation, but one of its enzymes, angiotensin-converting enzyme (ACE), indirectly causes hypertension by constricting blood vessels. Autoimmune illness is linked to the increased risk of hypertension and cardiovascular disease. In this study, ACE-inhibiting peptides were studied from Artemisia annua proteins. In silico hydrolysis of proteins was performed by BIOPEP-UWM using proteolytic enzymes from plant, microbial, and digestive sources. The physicochemical properties of 1160 peptides were determined using the peptide package of R studio. Di- and tripeptides were mostly released with a molecular weight of 170 to 350 Da. PeptideRanker was used to select 16 peptides from a pool of 1160 peptides based on their likelihood of being bioactive. Molecular docking was performed by DS 2020 and AutoDock Vina, which revealed that the stability of the ligand-receptor complex is due to hydrogen bonding and electrostatic and hydrophobic interactions. Their binding energies ranged from -31.81 to -20.09 kJ/mol. For drug-likeness evaluation, an online tool SwissADME was used that follows the ADME rule (absorption, distribution, metabolism, and excretion) to check the pharmacokinetics and drug-likeness of the compound. In the future, the released peptides can be used to make functional nutraceutical foods against hypertension.

Eclipta alba (L.) is a valuable medicinal plant. However, its medicinal efficacy can be affected by geminivirus infection. Therefore, identification of healthy specimen is essential before to use as medicine. The present study provided the taxonomic characterization of geminivirus infected and healthy E. alba plant by studying apparent morphology and microscopic features through light and scanning electron microscopy. Before taxonomic characterization infected and healthy specimens were separated through molecular detection of geminivirus. Results of morphological studies reported that geminivirus infected E. alba plant showed systematic symptoms of infection like stunted growth, distortion and chlorosis of leaves, decrease in size of root, shoot and fruit, and so forth in comparison to healthy specimen. Anatomical findings reported that in both plants anomocytic and anisocytic types of stomata with multicellular warty trichomes were present. However, variations were observed in quantitative measures such as size of trichomes, epidermal, subsidiary and guard cells. Palynological observations identifies that both plants possessed tricolporate type of pollen but variation was mainly observed in size and shape of pollen, thickness of exine and intine, P/E ratio, pore size, interspecific difference, size of colpi, and pollen ornamentation. Overall this study concluded that both healthy and infected E. alba do not reported much variations in qualitative taxonomic features, but can be differentiated in terms of quantitative taxonomic evidences. Future studies are recommended for pharmacological analysis of both healthy and virus infected plants. RESEARCH HIGHLIGHTS: Eclipta alba has incredible therapeutic worth, but due to geminivirus infection the plant is affecting badly. Hence, the present studies give a comprehensive taxonomic report on the geminivirus infected and healthy plant species.

Molecules isolated and identified from plant origin are used to manufacture most chemotherapeutic drugs for cancer treatment. We assumed that these plant extracts contain prolific bioactive compounds with potent antiproliferative activities and could be effective against different human cancer cells. Ethanolic extracts were prepared from Chelidonium majus, Myrica cerifera, Fumaria indica, Nigella sativa, and Silybum marianum, and the antiproliferative assay was performed in HepG2 and HeLa human cancer cell lines. All plants extract exhibited antiproliferative potential against studied cancer cell lines in the dose and time-dependent manner. Chelidonium majus and Silybum marianum have shown promising results against HepG2 and HeLa cells, respectively, followed by Myrica cerifera, Fumaria indica, and Nigella sativa. Results indicated that utilization of whole plant extract as anticancer compounds could be of great value in generating novel chemotherapeutic drugs.

The anatomical variations of two plants from the Nyctaginaceae family, Bougainvillea spectabilis and Bougainvillea glabra, were studied using light and scanning electron microscopy methods in this work. Bougainvillea is a dicotyledonous with defensive traits that can withstand extreme (hot and dry) settings; according to the findings, crystal inclusions in cells, woody spines, and an abnormal development pattern are all features that help them survive against predators and are unique to this species. The Bougainvillea plant's leaves are arranged in simple pattern, alternate to each other along stem having an undulate leaves edge and an oval form. The xylem and phloem, palisade, parenchyma midrib, spongy mesophyll, raphide crystal bundles, and trichomes were all visible when bracts and leaves were transversally sectioned and dyed with toluidine blue O (TBO). The presence of crystals was confirmed by a detailed examination of the transverse leaves by using bright-field and cross-polarizing microscopy. Dissecting microscopic examination showed that all the leaves revealed leaves venation pattern that had midvein, lateral veins areoles, and trichomes. Although trichomes have been identified on both sides, a closer look at a cleaned leaf dyed with TBO showed multicellular abundant trichomes on adaxial surface. Stomata complexes were typically found on the abaxial surface of the leaf according to epidermal peels. Present studies also showed that on adaxial side, stomata were lesser in number or were absent and also showed that the morphologies of the pavement cells on the adaxial and abaxial sides of the leaf differed.

Present research work was carried out to clarify the variations among species of genus Paspalum morphologically and palynologically as this genus is taxonomically difficult due to having multiple similar morphologically overlapping characters which make it difficult to identify. Henceforth present research work was carried out to delimit taxa within the same genus by morphological and palynological tools through light microscopy (LM) and scanning electron microscopy (SEM). Both these tools are considered as the most useful taxonomic characters for taxonomically problematic genera. The results showed a lot of variations among morphological characters. In Paspalum dilatatum, the upper glume was ovate whereas in the other two species, the upper glume was elliptic. The upper glume apex found in P. dilatatum and Paspalum scrobiculatum was obtuse whereas in Paspalum distichum, upper glume apex was acute. Glume nerves showed variation in all three species. Paspalum distichum was 3 nerved, P. scrobiculatum was 5-7 nerved, and P. dilatatum was 5-9 nerved. All three species showed variation in lemma nerves. Paspalum scrobiculatum had 3 nerved lemma whereas in P. distichum 3-5 nerved and P. dilatatum 5-9 nerved lemma were present. In polar and equatorial view, pollen grains ranged from 25 (20-30) to 37.5 (30-45) µm. Paspalum distichum appeared to be the smallest in size whereas P. dilatatum was the largest. Exine thickness ranged from 0.75 (0.5-1) to 1.35 (1.2-1.5) µm. The higher value of pollen fertility was found in P. scrobiculatum as 87.69% and the lowest value was in P. distichum as 78.08%. Morphological keys were also given for correct identification.

Paspalum is one of the 10 largest genera in Poaceae and is considered as a complex and taxonomically difficult genus due to its similar morphological characters within the species which makes it difficult to delimit taxa within the same genus. The current research work establishes the distinction among species of this genus and resolves taxonomic confusions among this genus. And this distinction is done by micro morphological foliar anatomical characterization by light and scanning electron microscopy. Both the abaxial and adaxial epidermal cuticles of three different species of genus Paspalum were studied and their phytoliths were observed through elemental dispersive spectrophotometric analysis by SEM first time in Pakistan. Results showed great variation in studied anatomical features. P. dilatatum Poir. was comprised of macro-hairs in the intercostal regions only whereas no macro-hairs were found in P. distichum Linn. and P. scrobiculatum Linn. Beak-shaped prickles were found in the intercostal regions on adaxial surface only in P. distichum Linn. and P. scrobiculatum Linn. whereas in P. dilatatum Poir., knife-shaped prickles were present in intercostal regions only on both abaxial and adaxial surfaces of leaf epidermis. No papillae were seen in P. dilatatum Poir whereas in P. distichum Linn. and P. scrobiculatum Linn. papillae were found abundantly. In genus paspalum, P. dilatatum Poir. phytoliths showed lowest mass percentage of silicon as 15.13% whereas P. distichum Linn. phytoliths showed highest mass percentage of silicon as 22.00%. Based on micro morphological foliar characteristics a taxonomic key of studied species is also provided to delimit the taxa.

Grass taxonomy is quite problematic and insignificant taxonomic work has been done on Echinochloa with special reference to Pakistan. Therefore, the present study was carried out to provide taxonomic keys for the identification of this genus through microscopic epidermal leaf anatomical parameters. Light microscopy and scanning electron microscopy were done to delimit the different species. Special structures like hook cells, short and long cells, stomata, macro- and micro-hairs, prickles and silica bodies showed a lot of variation among each species. For instance, E. frumentacea can be distinguished from E. walteri by the presence of macro hairs between the veins. Furthermore, E. crus-galli can be distinguished from E. frumentacea in terms of macro hair size, that is, 45-60 µm and 20-60 µm, respectively. Similarly, E. colona showed marked variations in terms of prickles abundance from E. frumentacea. Overall results of this research reports valuable qualitative and quantitative diagnostic futures for the genus Echinochloa.

Abstract A wide variety of cellular mechanisms such as cell division and metabolic processes are maintained by protein-protein interactions (PPIs). The identification of PPI through laboratory techniques is costly, time-consuming, difficult, and challenging. However, computational techniques were generated for PPIs prediction. In alfalfa (Madicago sativa), PPI was predicted among 12 MsMAPKs and 4 MsPP2Cs using a docking approach. For homology modelling, the Swiss model was employed while PROCHECK, ERRAT, and Verify3D were used to validate 3D models. The Ramachandran plots were obtained from PROCHECK which showed value more than 90% (nPP2C1, PP2C1, PP2C, and MSK-3 revealed 92.9%, 94.2%, 92.4%, and 91.1% respectively) for high-quality structures. The HawkDock server and the BIPSPI server were used to analyse protein docking and predict interaction sites, respectively. Our findings demonstrated that MsPP2C docking sites play an important role in the identification and docking of MsMAPKs. The binding free energy ranged from -0.16Kcal/mol to -49.15Kcal/mol for all MsMAPKs and MsPP2Cs, indicating that they interact. Docking site analysis showed that there were 48 pairs of PPIs which indicated that MsPP2Cs can perform a vital role in other signaling pathways. This study found that all MsPP2Cs have docking sites for MsMAPKs, indicating that this method can accurately determine protein-protein interactions.

Fertile plain of Central Punjab Pakistan is rich with fodder grasses and from centuries the local inhabitants of this area have been using their regional grasses for ruminant feeding. However, they always faced difficulties in identification because of their overlapping vernacular names, more or less identical leaf shapes, indefinite variations in stem branching pattern, and reduced floral parts. Hence, the current study has provided a detailed and comprehensive micro-morphological analysis of 53 ethnobotanical fodder grass taxa. A variety of quantitative and qualitative leaf epidermal micromorphological traits was studied and results reported epidermal characters like stomatal index, silica bodies, prickles, microhairs, hook cells as most diagnostic in delimitation of species, and genera. As stomatal index was 79% in Poa annua while in its closely related species Poa infirmai was 85%. Similarly, Cenchrus ciliaris can be differentiated from Cenchrus pennisetiformis on the basis of silica body shape as butterfly shaped in former and dumbbell shaped in later one. Moreover, prickles were present in Chrysopogon aucheri while absent in Chrysopogon zizanioides. Hence, overall this study declared that diversity and variations in foliar micromorphological characters are valuable and supportive in the identification of grasses at the specific and generic level.

Plants transformed with single Bt gene are liable to develop insect resistance and this has already been reported in a number of studies carried out around the world where Bt cotton was cultivated on commercial scale. Later, it was envisaged to transform plants with more than one Bt genes in order to combat with resistant larvae. This approach seems valid as various Bt genes possess different binding domains which could delay the likely hazards of insect resistance against a particular Bt toxin. But it is difficult under field conditions to develop homozygous plants expressing all Bt genes equally after many generations without undergoing recombination effects. A number of researches claiming to transform plants from three to seven transgenes in a single plant were reported during the last decade but none has yet applied for patent of homozygous transgenic lines. A better strategy might be to use hybrid-Bt gene(s) modified for improved lectin-binding domains to boost Bt receptor sites in insect midgut. These recombinant-Bt gene(s) would express different lectin domains in a single polypeptide and it is relatively easy to develop homozygous transgenic lines under field conditions. Enhanced chloroplast-localized expression of hybrid-Bt gene would leave no room for insects to develop resistance. We devised and successfully applied this strategy in cotton (Gossypium hirsutum) and data up to T3 generation showed that our transgenic cotton plants were displaying enhanced chloroplast-targeted Cry1Ac-RB expression. Laboratory and field bioassays gave promising results against American bollworm (Heliothis armigera), pink bollworm (Pictinophora scutigera) and fall armyworm (Spodoptera frugiperda) that otherwise, were reported to have evolved resistance against Cry1Ac toxin. Elevated levels of hybrid-Bt toxin were confirmed by ELISA of chloroplast-enriched protein samples extracted from leaves of transgenic cotton lines. While, localization of recombinant Cry1Ac-RB protein in chloroplast was established through confocal laser scanning microscopy.
