RESUMO

Proteinase-inhibitor balance in the lacrimal fluid in eye burns was studied in rabbits with modified and intact immune status. Stimulation of the immune system was induced by complete Freund's adjuvant. Clinical picture of the disease was studied in parallel. Immunization accelerated the local adaptive and defense reaction of the proteolytic enzymes and their inhibitors, which was paralleled by a more benign course of burn disease of the eyes, in comparison with that in non-immunized animals.

RESUMO

The influence of multibilayer phosphatidylcholine liposomes on the properties of rat hepatocyte mitochondria membranes damages caused by hepatotropic toxin--CCl4 was investigated. Alterations of the membrane structure were estimated by the decrease in phospholipid/protein ratio /by 33%/ and via changes of quantitative and qualitative composition of phospholipid components. The possibility of reparation of the hepatocytes mitochondria damaged membrane by egg yolk phosphatidylcholine liposomes is shown. Phosphatidylcholine reparative effect on the damaged membranes is revealed in quantitative phospholipid fractions redistribution.

RESUMO

Rat hepatocyte plasmatic membrane damages caused by the administration of tetrachloromethane of heliotrine was investigated. Phospholipid content in plasmatic membranes decreased. Heliotrine caused complete and CCL4 twofold inhibition of Na, K-ATPase. Intraperitoneal administration of egg yolk phosphatidylcholine liposomes had a reparative effect on the damaged membranes consisting in the restoration of phospholipid content and enzyme activities.

RESUMO

The effect of phospholipid multilayer liposomes on the properties of endoplasmic reticular membranes has been studied in hepatic cells damaged with CCl4. It has been shown that the repair effect of phosphatidylcholine liposomes was manifested in vivo by normalization of phospholipid membrane composition, reduction in the degree of cytochrome P-450 inactivation, partial normalization of its hydroxylase activity and recovery of glucose-6-phosphatase activity. The degree of phosphatidylcholine unsaturation, and the introduction of antioxidants--SH-compounds, sphingomyelin, phosphatidyl inositol--into liposomes did not influence the efficacy of liposomes.

RESUMO

Hepatocyte membranes destruction in experimental toxic hepatitis caused by heliotrine administration was accompanied by a 10-fold increase in blood serum activity of aldolase fructose-I-monophosphate, a decrease in cytochrome P-450 content, an increase in the rate of cytochrome P-450 inactivation, as well as a decrease in microsomal glucose-6-phosphatase activity. Administration of phosphatidylcholine liposomes decreased the activity of aldolase twofold, which indirectly shows partial reconstitution of liver cell membranes. Phosphatidylcholine protective action is also manifested in an increase in the activity of glucose-6-phosphatase, a microsomal marker enzyme, up to its control level and in a 20% reduced rate of cytochrome P-450 inactivation. It has been shown that destroyed liver cell membranes may be repaired by the introduction of phosphatidylcholine in the form of multilayer liposomes.

RESUMO

Level of fructose-1-monophosphate aldolase was decreased in blood serum after administration of phosphatidyl choline-containing liposomes into male rats with experimental toxic hepatitis caused by treatment with hepatotropic poison CCl4 at a dose of 0.25 ml/100 g of body mass. The rate of the aldolase level normalization depends on composition of liposomes as well as on the pathway of their administration.

RESUMO

A method for biomembrane reconstitution from microsomal proteins and lipids solubilized by sodium cholate consisting in a removal of the detergent by its dialysis followed by treatment with 10% albumin has been developed. A comparison of the original and reconstituted membranes showed that the phospholipid, protein and enzymatic composition of the latter is similar or only slightly different from that of the original ghosts. The reconstituted membranes contained 1.5 times more cytochrome b5 and an equal amount of cytochrome P-450. No more than 20% of cytochrome P-450 was represented by the inactive form. The inactivation rate of the reduced hemoprotein in the reconstituted membranes was lower than in the ghosts. Both in the reconstituted and original membranes the similarity of solubilization patterns of microsomal electron carries upon trypsin treatment was indicative of identical topography of these proteins. The most effective was the reconstitution of NADH and cumole hydroperoxide-dependent N-demethylase, whereas the p-hydroxylase and O-dealkylase activities of the reactivated P-450 were not retained. Hence, no complete reconstitution of the properties of the microsomal membrane and its redox chain was observed in spite of the effective removal of the detergent, similar localization of microsomal electron carriers in the reconstituted membranes and ghosts and the reconversion of cytochrome P-420 into cytochrome P-450.
