RESUMO

Background: Biostimulation is a management practice that improves the reproductive parameters, potentiates the desire, and improves the reproductive efficiency during the short breeding season in camels. Aim: This study aims to investigate the concurrent changes in hormonal profiles in response to management and behavioral situations on camel farms in the Eastern region of Saudi Arabia. Methods: A total of 10 male and 50 female camels were used in this study. The hormonal profile of male camels was evaluated weekly starting from December to August. Results: The results show that both serum testosterone and cortisol levels increased (p < 0.01) from December to March compared to April to August. A strong negative correlation was observed between testosterone levels and temperature (r = -0.81, p < 0.05), and a similarly robust negative correlation was found between cortisol levels and temperature (r = -0.83, p < 0.05). The dominant rutting males showed higher levels of testosterone than the submissive males. Serum testosterone levels increased (p < 0.01) in males out of rutting after hearing the sounds of other couples before and during mating. Conclusion: Hearing sounds emitted during mating increases the testosterone output, and improves the libido of male camels during the nonrutting period. It is important to keep two individually housed males for mating purposes. One male joins the herd alone in winter (winter rutter), and the other joins in spring (spring rutter), this could keep the libido extended rutting and improve the reproductive performance in camel herds.

RESUMO

Cancer is a life-threatening illness all over the world, and developing anticancer treatments with high efficacy and low side effects remains a challenge. The quinoline ring structure has long been recognized as a flexible nucleus in the design and synthesis of physiologically active chemicals. In this study, five new 2-morpholino-4-anilinoquinoline compounds were synthesized and their biological anticancer potential against the HepG2 cell line was assessed. The compounds produced demonstrated varying responses against HepG2 cells, with compounds 3c, 3d, and 3e exhibiting the highest activity, with IC50 values of 11.42, 8.50, and 12.76 µM, respectively. It is a critical requirement that anticancer medications are able to selectively decrease cancer growth while not causing damage to normal cells. Compound 3e exhibited increased activity while maintaining adequate selectivity. It was also the most effective chemical against cell migration and adhesion, which could play an important role in drug resistance and cell metastasis. In total, the findings revealed good possibilities for anticancer therapy, suggesting a target for future development of anticancer medication.

RESUMO

Heterocyclic amines and acetamide derivatives are known for their chemotherapeutic potential. Hence, in the present study, morpholine was taken as a principal product and novel morpholine derivatives were designed, formulated, characterized, and screened for the mechanism of inhibition of carbonic anhydrase and their anticancer potential. In addition, in vitro inhibition of hypoxia-inducible factor-1 (HIF-1) protein was also investigated. Results revealed that compounds 1c, 1d, and 1h possessed significant inhibitory activities against carbonic anhydrase with IC50 of 8.80, 11.13, and 8.12 µM, respectively. Interestingly, the carbonic anhydrase inhibitory activity of compound 1h was comparable with that of standard acetazolamide (IC50 7.51 µM). The compounds 1h and 1i significantly inhibited the proliferation of ovarian cancer cell line ID8 with IC50 of 9.40, and 11.2 µM, respectively while the standard cisplatin exhibited an IC50 8.50 µM. In addition, compounds 1c, 1b, 1h and 1i also exhibited significant inhibitory effects on HIF-1α. In conclusion, we report first time the biological potential of morpholine based compounds against ovarian cancer and HIF-1α that may serve as lead molecules for drug discovery.

RESUMO

Recently, the secondary use of by-products of the processing industry resulting from the production of poultry meat, which can be included in animal diets, has become a popular topic in the feed industry. For studying the effects of partial replacement of rapeseed cake (RC) with the by-product source of animal protein concentrate Agro-Matic (PCAM) on growth dynamics, nutrient absorption and nitrogen balance, as well as blood biochemical parameters during the growing period, a total of 48 Russian Ayrshire dairy heifers were selected for this experiment and they were divided into three experimental groups (16 in each group, including the control group). The heifers of the control group were fed the basal diet which contained rapeseed cake (30%), while the second (RC + PCAM) and third groups (PCAM + RC) were fed the basal diet after replacing a part of the rapeseed cake with 2.25% and 4.5% of protein concentrate Agro-Matic respectively. The results showed that the weight of heifers treated with PCAM at 3 months of age exceeded the control by 2.3 kg (p > 0.05) in group 2 by 4.4 kg (p < 0.05). Similar results were obtained at the age of 6 months of raising. Feeding 4.50% protein concentrate Agro-Matic has a positive effect on the digestibility of nutrients; in particular, there was a significant increase in the digestibility of crude protein in the PCAM + RC group (77.23 vs. 73.42%) compared with the control group. Moreover, a similar trend was found in the digestibility of nitrogen in the diet. At the age of 3 months, heifers showed a significant decrease in the concentration of ketone bodies in the second group (1.82 vs. 2.20 mmol/L) relative to the control group. Urea was significantly lower in the RC + PCAM group (5.05 vs. 6.62 mmol/L) relative to the PCAM + RC group, while acid capacity (alkaline reserve) was higher by 2.41% (p < 0.05) relative to the control. In the 10th month of age, a positive effect on the blood of heifers was observed, as in the second and the third experimental groups, ß-globulin and phosphorus increased (p < 0.05), while in the second group aspartate aminotransferase decreased (p < 0.05). Consequently, replacing the rapeseed cake with the protein concentrate Agro-Matic revealed an improvement in the dynamics of growth, nutrient digestibility and nitrogen balance, and it has an effect on improving some biochemical parameters of the blood.

RESUMO

Background and objective Diabetes is a major risk factor for the development of cardiovascular diseases. To tackle this issue, guidelines have emphasized the use of cardioprotective antihyperglycemic agents [glucagon-like peptide-1 receptor agonists (GLP1-RAs) and sodium-glucose co-transporter-2 (SGLT2) inhibitors] among type 2 diabetic patients at high risk of/with established atherosclerotic cardiovascular disease (ASCVD) or heart failure to curb morbidity and mortality in such individuals. The aim of this study was to assess physicians' adherence to the American Diabetes Association's (ADA) recommendations on the use of cardioprotective antihyperglycemic agents among such patients.  Methods This study involved a retrospective records review of physicians' treatment plans for all type 2 diabetic patients aged 18 years and older and attending outpatient clinics from the year 2019 to 2022 at King Abdulaziz University Hospital, Jeddah, Saudi Arabia, who (a) were at high risk of/with established ASCVD as defined by the American Diabetes Association (ADA) or (b) had heart failure with reduced ejection fraction (HFrEF) and required cardioprotective antihyperglycemic agents. Results We reviewed physicians' treatment plans for 202 type 2 diabetic patients in this study. All patients were either at high risk of or had established ASCVD. In addition, 36 (17.8%) of these patients had HFrEF. Although all patients were candidates for cardioprotective antihyperglycemic agents, only 56.9% of them received treatment as recommended by the ADA guidelines. Conclusion Despite being suitable candidates for cardioprotective antihyperglycemics, a significant number of patients were not treated based on the ADA guidelines, and this demonstrates a lack of adherence to the guidelines by physicians.

RESUMO

2-Mercaptobenzothiazole and its derivatives are widely known for their diverse biological activities, particularly antimicrobial and anticancer potential. In the present study, a series of new hybrid compounds consisting of 2-mercaptobenzothiazole and different aryl amines 2(a-j) were synthesized and characterized by Fourier transform infrared (FTIR), 1H NMR, and 13C NMR spectral data. The synthesized compounds were screened for in vitro antibacterial activities through agar well diffusion assay. Among the series, 2b, 2c, and 2i exhibited significant antibacterial activity comparable to the standard drug levofloxacin. Based on their antibacterial potential, these compounds were further tested for their antibiofilm activity. All of the three compounds showed promising antibiofilm potential, even better than the standard drug cefadroxil at 100 µg/100 µL concentration. Molecular docking studies were performed to explore the antibacterial mechanism of these compounds. Strikingly, the molecule 2i shared the same hydrophobic pockets as those of levofloxacin in case of bacterial kinases and DNA gyrases. In addition, 2i exhibited satisfactory antibiofilm activity in comparison to the standard. Our study therefore suggested that the synthetic compound 2i possesses remarkable antibacterial activity and may serve as a lead molecule for the discovery of potent antibacterial agents.

RESUMO

In this review we discuss several recent concepts regarding retinoblastoma control and its impact. In a cohort of 482 patients with solitary unilateral retinoblastoma revealed germline mutation in 16% and the likelihood of germline retinoblastoma was greater for younger children (≤1 year versus (vs.) >1 year at presentation) with odds ratio (OR) 2.96 (p = 0.001), and greatest for the youngest infants (≤3 months vs. >3-12 months) (OR 5.52) (p = 0.002). Retinocytoma/retinoma, a benign variant of retinoblastoma, was studied in 78 tumours and demonstrated transformation into retinoblastoma in 9.2% by 5 years and 15.3% by 10 years and 20 years. An international global study on retinoblastoma over 1.5 years revealed 4351 new patients and 85% from low- and middle-income countries, notably with older age at detection and greater risk for metastasis. Management of retinoblastoma in 964 eyes using intravenous chemotherapy showed 20-year globe salvage at 96% in group A, 90% in group B, 90% in group C, 68% in group D, and 32% in group E eyes. The 5-year globe salvage with intra-arterial chemotherapy for 160 eyes (655 infusions) with retinoblastoma showed success in 100% for group B, 80% for group C, 78% for group D, and 55% for group E. The psychological impact of retinoblastoma on the parents revealed depression (73%), anxiety (64%), and/or stress (100%), and on the patient revealed deficits in quality of life issues. Retinoblastoma is a challenging disease and chemotherapy provides reliable tumour control and globe salvage. Continuing efforts to improve quality of life issues is important.

RESUMO

BACKGROUNDConstant mutation in the SARS-COV2 virus genetic material is resulting in the appearance of new variants frequently hence the overall virulence, treatment resistance, replication modalities, transmissions rates and COVID-19 signs & symptoms are all changing regularly. METHODOLOGYFrom 1 January 2021 to 30 August 2022, the clinical lab at Fatima Jinnah General & Chest Hospital Quetta, Balochistan, determined a total of 3375 individuals to be COVID-19 positive because RT-PCR detected ORF, N, and E genes or their various Bi & Tri combinations in their samples. A questionnaire-based interview was conducted with each participant during sample collection. Body temperature more than 370c was recorded as Fever/Chill. Age, Comorbidities, A-symptomatic individuals & Vaccination status were all neglected during this study. Frequency tables were generated using MS-excel 2016, while Odds ratios were calculated using Chi-square test of association whereby 2x2 contingency tables between Mono, Bi & Tri combinations for ORF, N & E genes were cross associated with various generalized nonspecific COVID -19 signs and symptoms using Epi-info software. Absence of Genetic sequencing was the major limitation. RESULTSThe study showed that individually the presence of ORF gene was found to be strongly associated " Shortness of Breath/Difficulty in Breathing", Diarrhea, Head ache & Vomitting. While the presence of N- gene was found to be strongly associated with Loss of smell & taste, Head ache,Presistant Chest Pain & Bluish lips/Face. Where as the presence of E-gene was found to be strongly associated with Cough, Shortness of breath/ Difficulty in breathing, Sore throat, Diarrhea, Head ache & Laziness. In addition, the study also found that different Bi & Tri combinations of ORF, N & E genes in a COVID-19 positive patient expressed generalized non-specific COVID-19 signs & symptoms differently. DISCUSSION & CONCLUSIONThe presence of various SARS-COV2 genetic markers significantly alters the clinical presentation of COVID-19.

RESUMO

Animal feeding research has revealed a close relationship between the chemical composition and nutritional value of cow rations, the number of rumen bacterial communities and animal productivity. Our present research aimed to investigate the outcome of inclusion of different levels of protein concentrate in rations of Ayrshire dairy cows in relation to the rumen microbiome, reproductive traits and economic value. Forty-five Ayrshire cows were divided into three groups (15 in each). The first control group 0 AM was fed the basal ration, while the second 1 AM and third 2 AM groups were fed the basic ration with the sunflower cake replaced by different levels of protein concentrate Agro-Matic (1 and 1.5 kg/head/day, respectively). Ruminal fluid samples, reproductive parameters and economic value were studied. During the early lactation period, 120 days in milk (DIM), the number of pathogenic microorganisms decreased in both the 1 AM and 2 AM groups when compared with the control group 0 AM; moreover, a significant decrease in Peptococcus bacteria was recorded in the 1 AM group, while Fusobacterium decreased in the 2 AM group. At the end of lactation, the total number of cellulolytic bacteria increased with the use of protein concentrate in animals of the 1 AM group when compared with the control group. Regarding undesirable bacteria, the 2 AM group recorded the highest value for Lactobacilli and Actinobacteria when compared with the 0 AM group (0.18 and 8.90 vs. 0.04 and 4.24), and the differences were significant (p < 0.05). The insemination index and the duration of the days open period decreased in the 2 AM group, while the differences were p > 0.05. The profitability of milk production increased by 2.76% and 6.28% in both supplemented groups, and the differences compared to the 0 AM group were significant. We conclude that the supplementation of Agro-Matic caused no deviations from the normal standards of cellulolytic, amylolytic, transit and pathogenic bacteria, no impact on reproductive functions and significantly improved the profitability of the milk production process of Ayrshire dairy cows.

RESUMO

Lacrimal gland ductulitis is a rare infection of the lacrimal gland ductules. Individuals affected report a history of chronic mucopurulent conjunctivitis with "stringy" discharge. All patients are clinically noted to have an inflamed lacrimal gland ductule. Prior reports have suggested Actinomyces species as the "probable" etiology. We report the findings of a 42-year-old male found to have lacrimal gland ductulitis with a lacrimal gland abscess, confirmed by radiologic studies. Surgical drainage was necessitated and performed. Histopathologic analysis confirmed colonization by Actinomyces species, as well as the presence of sulfur granules. To our knowledge, this is the first report of Actinomyces lacrimal gland ductulitis resulting in a secondary lacrimal gland abscess.

RESUMO

Routinely used anti-inflammatory drugs are associated with off-target effects such as cyclooxygenase (COX)-1 inhibition and gastric ulcers. The aim of this study is to examine the anti-inflammatory potential and gastroprotective effects of synthetic amino acid derivatives of 2-mercaptobenzimidazole (MBAA1, MBAA2, MBAA3, MBAA4 and MBAA5). The results showed that compound MBAA5 possess a potential anti-inflammatory action by inhibition of 15-LOX and COX-2. MBAA5 also attenuated the pro-inflammatory cytokines and mediators (TNF-α, IL-1ß and COX-2) in rat hind paw in carrageenan-induced inflammatory model of rat. 2-mercaptobenzimidazole derivative, MBAA5 also inhibited gastric H+/K+ ATPase and demonstrated a better selectivity index for COX-2 (SI 27.17) in comparison to celecoxib (SI 41.43). Molecular docking studies predicted the binding interactions of the synthesized compounds with retrieved target proteins of H+/K+ ATPase, COX-1, COX-2, and 15-LOX. The results of in silico and molecular docking analysis of amino acid derivatives of 2-mercaptobenzimidazoles further explained their pharmacological activities. Moreover, these compounds presented better antimicrobial activity against three clinical isolates of Helicobacter pylori. Together, our findings suggested that these synthetic 2-mercaptobenzimidazole derivatives are safer therapeutic candidates for inflammation.

RESUMO

Over the past decades, construction delays have been perceived as a global challenge that hinders the time delivery, budget, and quality of construction projects. It is the most common, expensive, and risky problem associated with both private and public construction projects. Within this context, the aim of this study is to investigate the significant factors that influence delays in construction projects in Hargeisa, using the road and building projects as a baseline. Through a questionnaire survey with 51 critical delay factors that are categorized into seven major groups, data were collected from 51 construction stakeholders selected based on simple random sampling from the different construction companies. Feedback from the respondents was analyzed using Relative Importance Index (RII) for ranking purposes. Results showed that delay in honoring payment progressively, underestimation or overestimation of the project cost, and delay in the approval of major changes in the work scope were ranked as the three major causes of delays in construction projects in Hargeisa. The implications of these results are vital to future projects as they clearly demonstrate how less attention is given to the application of project management tools such as robust cost, scope and risk management in construction projects. As such, construction stakeholders are recommended to make efforts to use the appropriate project management practices needed to manage the 16 identified critical delay factors, when executing future construction projects. Although, the current study focused on Somaliland construction projects, it expands and improves the understanding of delay factors in the global context, and as such can be applied to other countries and future studies.

RESUMO

Prolactin (PRL) and its receptor (PRLR) were considered as potential genetic markers for milk production and quality traits in cattle. However, little information is available regarding PRLR genetic diversity and association studies with milk traits in Egyptian water buffaloes. Therefore, the present study was conducted to search for mutations in PRLR and determine their associations with milk performance in these animals. Exon3 (E3) and E10 of PRLR were screened for polymorphisms using single strand conformation polymorphism (SSCP) and sequencing in 400 buffaloes. The associations between haplotypes and milk production (fat%, protein%, lactose%, and solid%) traits as well as mRNA and protein levels of PRL and PRLR were studied. Two single nucleotide polymorphisms (SNPs) in E10 were detected: g.11685G>A (p.Ala494Thr) and g.11773T>C (p.Val523Aal). The G and T alleles were wild (ancestral) alleles, while the A and C alleles were mutant alleles. These SNPs resulted in four haplotypes; AC, AT, GC, and GT. Buffaloes with wild GT haplotypes showed significantly higher milk yield, fat% and protein%, mRNA and protein levels of PRL and PRLR in milk somatic cells than other animals. Animals carrying mutant AC haplotype had inferior milk traits and lowest levels of associated mRNAs and proteins. With these results, we could conclude that the selection of buffaloes with wild GT haplotypes for g.11685G>A and g.11773T>C SNPs of the PRLR gene might improve the milk production traits of Egyptian water buffaloes.

RESUMO

We describe a 75-year-old woman with rapid onset orbital inflammatory syndrome as her initial manifestation of relapsing polychondritis. Patient presented after the development of right eyelid swelling, erythema, and proptosis over a 48-hour period. Visual acuity was 20/30 in both eyes. Intraocular pressure was elevated in her right eye along with severe restriction of extraocular motility. Magnetic resonance imaging of the orbits revealed thickened right medial and inferior recti muscles. Serologic laboratory data was unrevealing. Patient demonstrated marked improvement within 12 hours of administration of intravenous corticosteroids. She was symptom-free after 1 week. A diagnosis of relapsing polychondritis was confirmed 3 weeks later after new onset complaints of right ear pain and a rash.

RESUMO

Purpose: Postoperative hydrophilic intraocular lens opacification can lead to decreased vision and may require intraocular lens exchange. This study aims to identify the incidence of scleral-fixated Akreos AO60's (Bausch + Lomb) lens opacification and risk factors for this phenomenon. Methods: This is a retrospective case series of all patients who underwent scleral-fixated Akreos AO60 lens at our institution between January 1, 2015 and December 31, 2019. The following data were recorded: age, sex, medical history, indication for Akreos AO60 implantation, laterality, ocular history, previous ocular surgical procedures, subsequent intraocular surgical procedures after the Akreos implantation, lens opacification, visual significance of opacification, and Akreos explantation. Intraoperative and postoperative complications were recorded. Main outcome measures were the overall incidence of Akreos lens opacification as well as the incidence of these eyes undergoing subsequent intraocular surgery. Results: A total of 262 eyes of 257 patients underwent Akreos lens implantation. Overall, 2% (5 of 262) developed lens opacification. Two patients had Descemet stripping automated endothelial keratoplasty (DSAEK) concurrently with Akreos implantation. One patient underwent subsequent Baerveldt glaucoma implantation and DSAEK. The fourth patient had vitrectomy with sulfur hexafluoride gas followed by DSAEK. This represents a 25% (4 of 16) opacification rate among all patients who underwent DSAEK (P ≤ .01, Fisher exact test). One patient developed opacification after undergoing 2 vitrectomies for retinal detachment in the absence of DSAEK. Conclusions: Akreos lens opacification can be visually significant and may occur after a retinal or corneal procedure that involves the use of intraocular gas or air.

RESUMO

The synthesis of a molecularly diverse library of tetrasubstituted alkenes containing a barbiturate motif is described. Base-induced condensation of N1-substituted pyrimidine-2,4,6(1H,3H,5H)-triones with 5-(bis(methylthio)methylene)-2,2-dimethyl-1,3-dioxane-4,6-dione gave 3-substituted 5-(methylthio)-2H-pyrano[2,3-d]pyrimidine-2,4,7(1H,3H)-triones ('pyranopyrimidinones'), regioselectively. A sequence of reactions involving ring-opening of the pyran moiety, displacement of the methylthio group with an amine, re-formation of the pyran ring, and after its final cleavage with an amine, gave tetrasubstituted alkenes (3-amino-3-(2,4,6-trioxotetrahydropyrimidin-5(2H)-ylidene)propanamides) with a diversity of substituents. Cleavage of the pyranopyrimidinones with an aniline was facilitated in 2,2,2-trifluoroethanol under microwave irradiation. Compounds were tested against Escherichia coli, Staphylococcus aureus, the yeast Schizosaccharomyces pombe, and the pathogenic fungus Candida albicans. No compounds exhibited activity against E. coli, whilst one compound was weakly active against S. aureus. Three compounds were strongly active against S. pombe, but none was active against C. albicans.

RESUMO

In plant tissues, sugar levels are determined by the balance between sugar import, export, and sugar synthesis. So far, water soluble carbohydrate (WSC) dynamics have not been investigated in a diurnal context in wheat stems as compared to the dynamics in flag leaves during the terminal phases of grain filling. Here, we filled this research gap and tested the hypothesis that WSC dynamics interlink with gene expression of TaSUT1. The main stems and flag leaves of two genotypes, Westonia and Kauz, were sampled at four hourly intervals over a 24 h period at six developmental stages from heading to 28 DAA (days after anthesis). The total levels of WSC and WSC components were measured, and TaSUT1 gene expression was quantified at 21 DAA. On average, the total WSC and fructan levels in the stems were double those in the flag leaves. In both cultivars, diurnal patterns in the total WSC and sucrose were detected in leaves across all developmental stages, but not for the fructans 6-kestose and bifurcose. However, in stems, diurnal patterns of the total WSC and fructan were only found at anthesis in Kauz. The different levels of WSC and WSC components between Westonia and Kauz are likely associated with leaf chlorophyll levels and fructan degradation, especially 6-kestose degradation. High correlation between levels of TaSUT1 expression and sucrose in leaves indicated that TaSUT1 expression is likely to be influenced by the level of sucrose in leaves, and the combination of high levels of TaSUT1 expression and sucrose in Kauz may contribute to its high grain yield under well-watered conditions.

RESUMO

Camels belong to a group of animals, where the structure of placenta does not allow intrauterine transfer of maternal immunoglobulins to the fetus and maternal immunity is exclusively transferred by colostrum to the newborn calf. There are few studies on the passive transfer of maternal immunity in the dromedary camel. This study determined total immunoglobulin G concentration, heavy chain antibody (HCAbs) levels, and neutrophils to lymphocytes ratio (NLR) in female camels and their newborn calves. For this, samples were collected from nine she-camels (blood and colostrum) and their calves (blood). IgG concentration and HCAb level were determined in mother serum and colostrum as well as in calf serum using ELISA. The NLR was calculated after the estimation of relative fractions of neutrophils and lymphocytes in collected blood samples using a blood cell analyzer. Both IgG and HCAbs were higher concentrated in camel colostrum than in mother serum. At parturition and before the first colostrum intake, calf serum did not contain any measurable concentration of IgG and only low levels of HCAbs. After colostrum consumption, a rise in IgG and HCAb levels was observed in calf serum. For total IgG, a minimum was reached on day 30 postnatum. While a significant increase in IgG concentration was seen on day 60 postnatum, no significant rise was measured in HCAbs at that age. Only post-colostrum IgG levels in calf serum correlated positively with IgG levels in mother colostrum. Directly after birth, newborn calves showed significantly higher NLR than their mothers. This indicates a pro-inflammatory nature of the calf immune response. The decrease of the NLR on day 60 postnatum may argue for the maturation of the calf immune response at this age.

RESUMO

Efforts aimed at minimizing the spread of COVID-19 and "flattening the curve" may be affecting clinical care delivery for non-COVID-19 cases that include otolaryngologic and orbital conditions. We are witnessing changes in the manner that patients present, as well as modifications in clinical management strategies. An improved understanding of these phenomena and the contributing factors is essential for otolaryngologists to provide sound clinical care during this unprecedented pandemic.
