RESUMO

OBJECTIVE: To explore the functional role of the drug-dependent mesenchymal-epithelial transition (Met)-axiation "π" structural module of neurogenesis after processing by three components of Qingkailing injection in neurogenesis and angiogenesis in cerebral ischemia. METHODS: We used a Glutathione S-transferase (GST)-pull down assay, isothermal titration calorimetry assay, and other related methods to identify the relationships among Met, inositol polyphosphate phosphatase like 1 (Inppl1), and death associated protein kinase 3 (Dapk3) in this allosteric module. The biological effects of the modules of neurons generation composed of Met, Inppl1, and Dapk3 were measured through Western blot, apoptosis analysis, and double immunofluorescence labeling. RESULTS: The GST-pull down assay revealed that proline-serine-threonine rich domain of Met binds to the Src homology domain of Inppl1 to form a protein-protein complex; Dapk3 with a C-terminal domain interacts weakly with the protein kinase C domain of Met in the intracellular region. Thus, we obtained a "π" structuring module considered a neural regeneration module. The biological effects of angiogenesis and neurogenesis modules composed of Met, Inppl1, and Dapk3 were also verified. CONCLUSION: The study suggested that understanding the functional modules that contribute to pharmaceutics might provide novel signatures that can be used as endpoints to define disease processes under stroke or cerebral ischemia conditions.

RESUMO

AIM: To study protective effects of the extract from Toona sinensis seeds on gastric mucosal injury in experimental mice. Objective To study the similarity of chemical fingerprints of different proportion of Gualoupi-Gualouzi and Fruc?tus Trichosanthis,so as to provide the theoretical basis for the reasonable proportion of Gualoupi-Gualouzi. Methods The HPLC fin?gerprints and the common patterns of 6 the proportion of the 6 kinds of compatibility(30:70,35:65,40:60,45:55,50:50 and 55:45)of Gualoupi-Gualouzi and Fructus Trichosanthis were established,and the similarity of chemical fingerprints of different compati?bility proportion of Gualoupi-Gualouzi and Fructus Trichosanthis between their common patterns were compared. Results Compared with the common pattern of Fructus Trichosanthis,the similarities of the common patterns of chemical fingerprints of the above 6 kinds of compatibility proportion of Gualoupi-Gualouzi were 0.8808 ± 0.0407,0.8930 ± 0.0236,0.8995 ± 0.0195,0.9033 ± 0.0190,0.9363 ± 0.0082 and 0.8904 ± 0.0144,respectively. Among them,Gualoupi-Gualouzi ratio of 50:50 had the highest similarity,and compared with other groups,the differences were statistically significant(P<0.05 or 0.01). Conclusion The optimum proportion of Gualoupi-Gualouzi can be determined by the fingerprint technology,which provides the theoretical basis for the reasonable proportion of Gualou?pi-Gualouzi.

RESUMO

Aim To investigate the protective effects of total triterpenoid from Prunella vulgaris L. ( TTP) on CCl4-induced hepatic fibrosis in rats and its mecha-nism. Methods Rat liver fibrosis was induced by 50% CCl4 twice a week for 12 weeks. From the 5th week, all the therapeutic groups were treated with the TTP(25, 50, 100 mg·kg-1 ) and the colchicine (0. 1 mg· kg-1 ) respectively once a day for 8 weeks. At the end of the twelfth week, the levels of ALT, AST, HA, PCⅢ, CⅣ, MDA, SOD, GSH-Px, Hyp were measured . HE and Masson staining were used to evalu-late the degree of hepatic fibrosis. The mRNA expres-sion ofα-SMA, procollagen I, Smad2, Smad3, Smad7 in liver was detected by RT-PCR, and the p-ERK pro-tein expression was evaluated by Western blot. Results Compared with the model group, TTP(25, 50, 100 mg·kg-1 ) not only reduced serum content of ALT, AST, HA, PCⅢ, CⅣand Hyp, MDA in liver tissue, improved the morphologic changes of hepatic fibrosis, but also increased SOD and GSH-Px activity. Moreo-ver, it decreased the α-SMA, procollagen I, Smad2, Smad3 mRNA expression and increased Smad7 mRNA expression in liver tissues obviously. Furthermore, TTP reduced the protein expression of p-ERK. Conclusions TTP can protect rats from CCl4-induced liver fibro-sis. The mechanism of this process may involve inhibi-ting the expression of p-ERK and interference with TGF-β1/Smad signal transduction pathway.

RESUMO

Objective: To study the effect of Baihuzhuifeng Pills (BHZFP) on interleukin 1?(IL 1?) secreted by synoviocytes in adjuvant arthritis (AA) rats. Methods: Synovium of knee joints from AA rats were digested by collagenase and tripsin to prepare DSSC(digested synovial single cells). The content of the supernatant fluid was assayed by the radioimmunoassay. Results: BHZFP down regulated IL 1? productions of DSSC.Conclusion: It is suggested that BHZFP be probably inhibit the over proliferation of synoviocyte in AA rats by diminishing IL 1? secretion.
