RESUMO

Correlation between clinical structure of motor deficit and indices of stimulation electromyography was studied in 108 patients with hemispheric stroke. Methods of statistic analysis were applied including devising of histograms, Pareto diagrams, determination of the narrowness of correlation ratio. The analysis revealed complex relationship between the signs of motor deficit in clinical picture and the data of stimulation electromyography in patients with cerebral hemispheric stroke.

RESUMO

A new classification of motor disorders in patients after brain hemisphere's stroke as well as with its sequelae was proposed on the basis of clinical electromyographic studies. The classification validity was confirmed by mathematic statistic methods. The classification was composed of motor syndromes and is based on the following criteria: the severity of paresis, the correlation between gravity of paresis of the upper and lower limbs, muscular spasticity, alteration of integral estimation of active movements (the motor algorithm). The most informative signs of classification are the following: the paresis gravity, the degree of muscular spasticity, the alteration of motor algorithm, the parameters of stimulative electromyography. The described classification permits to carry out differentiated actions of neurorehabilitation directed to intensification of effectivity of treatment after hemisphere's stroke.

RESUMO

Inhibition of the soleus H-reflex was studied in hemiparetic postapoplectic patients and in control group of patients without motor disorders. The depression of H-reflex was caused by stimulation of ipsilateral peroneal nerve and presented in two successive waves (D1 and D2). The former of them corresponds to presynaptic inhibition. The size of both waves of inhibition was reduced in postapoplectic patients according to the degree of their muscle spasticity.

RESUMO

The effect of purified preparation of adenylate cyclase isolated from Y. pestis on peritoneal leukocytes of white mice and guinea pigs was investigated. Y. pestis adenylate cyclase was shown to accomplish its pathogenic action via histamine-specific receptors on the surface of eukaryotic cells. The involvement of H1 and H2 histamine receptors on target cells in the adenylate cyclase action leading to development of plague infection is discussed.

RESUMO

In Y. pestis a cyclic AMP-binding protein was detected, isolated to a homogeneous state, and its physico-chemical properties were studied. The protein is a highly molecular compound with a molecular weight of 180 kD, capable of being released into the environment in the process of cell growth and having protein kinase activity, not depending on the presence of cyclic AMP. Y. pestis neuraminidase is one of the substrates appearing due to the action of protein kinase detected in this study. Y. pestis protein kinase may alter the spectrum of protein phosphorylation in the leukocytes of white mice. The direct participation of this protein in the development of infection is supposed.

RESUMO

Stimulation electromyography was used to study inhibition of the soleus muscle H-reflex induced by stimulation of the common peroneal nerve in healthy individuals and in patients who had underwent cerebral apoplexy. Two successive inhibition waves (D1 and D2) were recorded; the first wave demonstrated the development of presynaptic inhibition of group Ia afferent fibers. In post-insult patients the depth of inhibition diminished depending on the degree of the pathologic increase of the tonus of the skeletal musculature. The role of presynaptic inhibition disorders in the pathogenesis of muscle spasticity is discussed.

RESUMO

The data on the influence of the preparation of Y. pestis adhesion pili on peritoneal macrophages in white mice and guinea pigs are presented. Y. pestis adhesion pili have been found to induce the dose-dependent increase of cell chemiluminescence. They have also been found to induce a number of biochemical changes in target cells: the secretion of myeloperoxidase, an increase in the activity of cAMP-dependent protein kinases.

RESUMO

cAMP-binding protein was isolated from the plaque agent and purified to the homogeneous state. Purification process included filtration of the initial preparation through the membrane able to transmit particles with mol. weight to 300,000 Da, chromatography on cellulose "DE-52" and biogel HTP. The protein homogeneity was confirmed by electrophoresis in polyacrylamide gel and precipitation with commercial plague agglutinating serum. The protein with mol. weight of 180,000 Da consisted of two identical subunits (90,000 Da. each) which could dissociate with formation of monomers (mol. weight approximately 18,000 Da), Cu2+, Co2+, Mn2+ ions stimulated activity of cAMP-binding protein of a plague microbe while Fe3+, Ca2+, Zn2+ ions inhibited it by 30-70%. A monospecific rabbit serum to the homogeneous preparation of cAMP-binding protein was obtained. It helped finding the similar protein in the close relative bacterium Yersinia pseudotuberculosis but not in Y. enterocolitica.

RESUMO

The phenomenon of the reversible phosphorylation of proteins has been discovered in Y. pestis cells. Eight proteins with a molecular weight of 30-80 kilodaltons have been found capable of phosphorylation. The intensity of phosphorylation has been found to be influenced by the temperature of cultivation and the composition of the incubation medium. This newly found phenomenon of the chemical modification of proteins is supposed to play a certain role in the organization of rapid responses of the cell to changes in the environment.

RESUMO

Homosynaptic depression of H-reflex was studied in 25 healthy persons and 33 patients with hemiplegia. The mean duration of the homosynaptic depression in healthy persons was 5200 ms. After the initial relative facilitation the time course of the homosynaptic depression corresponded to a logarithmic function of the interval between the conditioning and testing stimuli. The homosynaptic depression in patients with hemiplegia decreased depending on the degree of the muscular spasticity.

Three forms of adenylate cyclase have been detected in Y. pestis: membrane-bound, cytoplasmic and extracellular. Extracellular adenylate cyclase has been purified so as to achieve a homogeneous state, and some of its physicochemical parameters have been investigated. In the process of purification the initial preparation of this enzyme has been subjected to heating at 100 degrees C for 15 minutes, fractionation with ammonium sulfate, and gel filtration on Sephadex G-100. The homogeneity of adenylate cyclase has been confirmed by electrophoresis in 7.5% polyacrylamide gel and precipitation by the plague agglutinating serum. The enzyme has been found to have a molecular weight of 30,000 daltons and to show the optimum activity at pH 7.0-7.2 and at a temperature between 37 and 40 degrees C. Monospecific rabbit serum to the homogeneous preparation of adenylate cyclase has been obtained.
