RESUMO

Although the world has entered the post-pandemic period, the mental health and life satisfaction of college students still need to be addressed. However, previous literature has primarily focused on negative variables and has paid little attention to positive variables, such as self-compassion and the capacity to be alone. Therefore, this longitudinal study aims to investigate the relationships between the capacity to be alone, self-compassion, life satisfaction, depression, and anxiety among college students. This study analyzed data from 1460 Chinese college students who completed an online survey at two time-points one year apart. We employed cross-lagged analysis and constructed longitudinal mediation models to explore the relationships between five variables (i.e., capacity to be alone, self-compassion, life satisfaction, depression, and anxiety). Our findings indicate that depression and life satisfaction could negatively predict each other over time. Self-compassion in wave 1 could negatively predict depression and anxiety in wave 2. Higher life satisfaction in wave 1 was associated with a lower capacity to be alone in wave 2. We also found reciprocal positive predictive relationships between depression and anxiety, and life satisfaction and self-compassion. Life satisfaction mediated the relationship between self-compassion and psychopathological variables (i.e., depression and anxiety). Additionally, self-compassion mediated the association between life satisfaction and psychopathological variables and the association between capacity to be alone and psychopathological variables. Our study highlights the significance of early identification and intervention in depression and anxiety. We also discovered the possible self-soothing function of self-compassion as well as the importance of fostering positive personal characteristics.

RESUMO

BACKGROUND: Umbilical cord blood-derived therapeutics, such as serum (UCS) and platelet-rich plasma (UCPRP), are popular treatment options in clinical trials and can potentially be utilized to address a clinically unmet need caused by preservatives, specifically benzalkonium chloride (BAK), present in ophthalmic formulations. As current clinical interventions for secondary injuries caused by BAK are suboptimal, this study will explore the feasibility of utilizing UCS and UCPRP for cornea treatment and investigate the underlying mechanisms associated with this approach. METHODS: Mice's corneas were administered BAK to induce damage. UCS and UCPRP were then utilized to attempt to treat the injuries. Ocular tests were performed on the animals to evaluate recovery, while immunostaining, RNA-seq, and subsequent bioinformatics analysis were conducted to investigate the treatment mechanism. RESULTS: BAK administration led to widespread inflammatory responses in the cornea. Subsequent treatment with UCS and UCPRP led to the downregulation of immune-related 'interactions between cytokine receptors' and 'IL-17 signaling' pathways. Although axonal enhancers such as Ngf, Rac2, Robo2, Srgap1, and Rock2 were found to be present in the injured group, robust axonal regeneration was observed only in the UCS and UCPRP treatment groups. Further analysis revealed that, as compared to normal corneas, inflammation was not restored to pre-injury levels post-treatment. Importantly, Neuropeptide Y (Npy) was also involved in regulating immune responses, indicating neuroimmune axis interactions. CONCLUSIONS: Cord blood-derived therapeutics are feasible options for overcoming the sustained injuries induced by BAK in the cornea. They also have potential applications in areas where axonal regeneration is required.

RESUMO

BACKGROUND: Umbilical cord blood has been widely used in clinical transplantation. Blood gas analysis of umbilical cord blood is routinely used to evaluate neonatal asphyxia. This study aimed to evaluate an improved umbilical cord blood collection method that does not affect the results of umbilical cord blood gas analysis and hematopoietic stem cell transplantation-related indices. METHODS: Three hundred pregnant women were recruited between December 2019 and August 2022. In total, 270 umbilical cord blood samples were included and randomly divided into 3 groups. Group A was defined as the group in which both umbilical cord blood samples for hematopoietic stem cell transplantation and blood gas analysis were collected. Group B was defined as the group from which umbilical cord blood was collected for hematopoietic stem cell transplantation. Group C was defined as that wherein umbilical cord blood was collected only for blood gas analysis. Hematopoietic stem cell transplantation-related indices were detected in groups A and B, and blood gas analysis was performed in groups A and C. RESULTS: Hematopoietic stem cell transplantation-related indices were not significantly different between groups A and B. The pH, base excess, and lactic acid values were not significantly different between groups A and C. CONCLUSION: The cord blood double collection method would not affect the results of umbilical cord blood gas analysis and hematopoietic stem cell transplantation-related indices. It is suitable for cord blood collection when preparing for hematopoietic stem cell transplantation and blood gas analysis.

RESUMO

Object: Repeated quarantine policies over the past 3 years have led to poor psychological consequences for the public. Previous studies have proved that the quarantine policy leaves individuals vulnerable to anxiety, depression, and insomnia, especially among college students. This study aims to explore whether psychological problems during isolation continue with the release of isolation. Methods: Overall, 2,787 college students both answered a web-based survey during and after the closure management was lifted. The Patient Health Questionnaire, Generalized Anxiety Disorder Scale, and Youth Self-rating Insomnia Scale were measured. The cross-lagged path model was used to explore the influence of psychological impact during isolation on the individual after the release. Results: We found that anxiety and sleep disturbance levels alleviated significantly after quarantine, except for depression. As expected, a bidirectional relationship exists between anxiety, depression, and sleep disturbance. Moreover, depression and sleep disturbance can predict post quarantine depression, sleep disturbance, and anxiety, yet anxiety cannot predict sleep disturbance afterward. Conclusion: Timely and effective intervention for anxiety, depression, and insomnia during isolation is essential for individuals to repair themselves quickly after the release.

RESUMO

Perceived organizational support (POS) in the relationship between neuroticism and job burnout among firefighters received little attention in China. A sampling of 716 firefighters in China, we drew on perceived organization support theory and the notion of support as a buffer in job burnout, examining moderating effects of POS on the relationship between neuroticism and three components of burnout (emotional exhaustion, depersonalization and reduced personal accomplishment). Also, this study explored the mediating effect of burnout on the relationship between neuroticism and mental health (i.e., anxiety and depression). We found that two components (depersonalization and emotional exhaustion) of burnout have significantly mediated the relationship between neuroticism and anxiety and depression. At the same time, POS reinforced the relationship between neuroticism and depersonalization and emotional exhaustion. Therefore, organizations can take our analysis into account when taking actions to improve firefighters' mental health. The implications of these findings were discussed.

RESUMO

BACKGROUND: The efficacy of erector spinae plane block (ESPB) for pain control in other surgeries remains an interesting topic of discussion. This study aimed to evaluate the safety and efficacy and quality of recovery of ultrasound-guided bilateral ESPB in laparoscopic surgery for colon cancer. MATERIAL AND METHODS: In this study 50 patients were included and randomly divided into the intervention group (E group, n = 25) and the control group (C group, n = 25). Patients in the E group received general anesthesia with preoperative bilateral ultrasound-guided ESPB, whereas patients in the C group received general anesthesia with saline injection in the erector spinae plane preoperatively. Data on intraoperative and postoperative anesthetic effects and the effect on enhanced recovery after surgery were recorded and analyzed. RESULTS: Rocuronium consumption in the intervention group was 82.80 ± 21.70 mg, which was lower than that in the control group (P < 0.05). Visual analog scale scores at 2, 6, and 24 h after surgery in the intervention group were lower than those in the control group (Fbetween = 34.034, P = 0.000). The time to ambulation, consumption of ketorolac tromethamine, time to oral intake and hospital stay after operation in the intervention group were significantly lower than those in the control group (P < 0.05). The block area at the different baselines was significant (Fbetween = 3.211, P = 0.009). The association between baseline and time was significant (Fbaseline * time = 3.268, P = 0.001). CONCLUSION: This study confirmed that ultrasound-guided ESPB technology is safe and beneficial for patients with colon cancer undergoing laparoscopic colon surgery.

RESUMO

Angiogenesis in the field of tissue engineering has attracted significant attention. Graphene oxide has become a promising nanomaterial in tissue engineering for its unique biochemical properties. Therefore, herein, a series of chitosan (CS)/graphene oxide (GO) hydrogel scaffolds were synthesized by crosslinking CS and GO at different concentrations (0.1, 0.5, and 1.0 wt.%) using genipin. Compared with the CS hydrogel scaffolds, the CS/GO hydrogel scaffolds have a better network structure and mechanical strength. Then, we used endothelial progenitor cells (EPCs) extracted from human umbilical cord blood and cocultured these EPCs with the as-prepared scaffolds. The scaffolds with 0.1 and 0.5 wt.%GO showed no considerable cytotoxicity, could promote the proliferation of EPCs and tube formation, and upregulated the expressions of CD34, VEGF, MMP9, and SDF-1 in EPCs compared to the case of the scaffold with 1.0 wt.%GO. This study shows that the addition of graphene oxide improves the structure of chitosan hydrogel and enhances the proliferation activity and angiogenic capacity of EPCs.

RESUMO

BACKGROUND: Diabetic foot is caused by ischemic disease of lower extremities of diabetic patients, and the effective therapy is very limited. Mesenchymal stem cells (MSCs) based cell therapy had been developed into a new treatment strategy for diabetic foot clinically. However, the underlying molecular mechanism remains to be fully addressed. Exosomes (extracellular vesicles) secreted by MSCs may play crucial role in the processes of MSCs mediated inhibition of inflammatory microenvironment as well as pro-angiogenesis of ischemic tissue of diabetic foot. METHODS: Exosomes were isolated from MSCs using ultracentrifugation, and further characterized by the nanoparticle tracking analyzer and flow cytometry. Moreover, RNA sequencing, Western Blot, in vitro cell proliferation, in vivo pro-angiogenesis, as well as ischemic repairment of diabetic foot through rat model were performed to evaluate exosome physiological functions. RESULTS: We found that inflammatory cytokines (tumor necrosis factor α and interleukin-6) and vascularcelladhesion molecule-1 induced MSCs to secrete exosomes heterogeneously, including exosome size and quantity. Through RNA sequencing, we defined a new proangiogenic miRNA, miRNA-21-5p. Further knockdown and overexpression of miRNA-21-5p by manipulating MSCs validated the biological activity of exosome miRNA-21-5p, including in vitro cell proliferation, in vivo pro-angiogenesis in Chick Chorioallantoic Membrane (CAM) assay, and in vivo pro-angiogenesis experiments (tissue injury and repair) in diabetic rat models. Furthermore, we discovered that exosomemiRNA-21-5p promoted angiogenesis through upregulations of vascular endothelial growth factor receptor (VEGFR) as well as activations of serine/threonine kinase (AKT) and mitogen-activated protein kinase (MAPK). Together, our work suggested miRNA-21-5p could be a novel mechanism by which exosomes promote ischemic tissue repair and angiogenesis. Meanwhile, miRNA-21-5p could be potentially developed into a new biomarker for exosomes of MSCs to treat diabetic foot. CONCLUSIONS: miRNA-21-5p is a new biomarker and a novel mechanism by which exosomes promote ischemic tissue repair and angiogenesis of diabetic foot. Our work could not only provide new scientific evidences for revealing pro-angiogenesis mechanism of MSCs, but also eventually benefit MSCs-based clinical therapy for diabetic foot of diabetes patients.

RESUMO

Background: Disabilities have a negative effect on mental health and individuals with such conditions are especially susceptible to mental disorders. Previous research has established that in normal population, social support is significantly correlated with subjective well-being (SWB) through mediating variables; however the internal mechanism underlying this in people with physical disability remains unclear. This study aims to examine whether self-esteem and perceived discrimination play a mediating role in the relationship between social support and SWB in a sample of people with physical disability in China. Methods: A total of 210 people with physical disability of Chinese ethnicity were recruited to complete a series of questionnaires. This included the Chinese Social Support Rating Scale (CSSRS), Rosenberg Self-esteem Scale (RSES), Disability Discrimination Perception Questionnaire (DDPQ), and the Chinese Happiness Inventory (CHI). Path analysis was implemented on the data. Results: The model showed excellent fit to data: χ 2 = 2.314, p > 0.05; root-mean-square error of approximation (RMSEA) = 0.079; standardized root-mean residual (SRMR) = 0.035; comparative fit index (CFI) = 0.989; and Tucker-Lewis index (TLI) = 0.936. The results showed that self-esteem significantly mediated the relationship between social support and SWB, and perceived discrimination in people with physical disability. However, there is no mediating effect of perceived discrimination between social support and SWB. Conclusion: These findings demonstrate that self-esteem may be a critical resource in mediating the relationships between social support, SWB, and perceived discrimination in people with physical disability.

RESUMO

A novel fluorescence quenching immunochromatographic sensor (ICS) was developed for detecting chromium (Cr(3+)) within 15 min utilizing the fluorescence quenching function of gold nanoparticles (Au-NPs). The sensor performed with a positive readout. When the low concentrations of Cr(3+) samples were applied, detection signals of the test line (T line) were quenched, whereas when higher concentration Cr(3+) samples (1.56 ng/mL) were applied, the detection signal of the T line appeared. The detection signal intensity of the T line increased with increasing concentrations of Cr(3+). The low detection limit of developed fluorescence quenching ICS was 1.56 ng/mL. The fluorescence quenching ICS has a linear range of detection of Cr(3+) comprising between 6.25 ng/mL to 800 ng/mL. The recoveries of the fluorescence quenching ICS to detect Cr(3+) in tap water ranged from 94.7% to 101.7%. This result indicated that the developed sensor gave higher sensitivity and reliable reproducibility. It could provide a general detection method for small analyte in water samples.
