RESUMO

OBJECTIVE: To develop a highly sensitive method for the determination of platelet-rich plasma serotonin concentration for every-day clinical practice. MATERIAL AND METHODS: The method was developed for determination of serotonin in plateled-rich plasma. Serotonin was determined using high performance liquid chromatography with fluorescent detection and hematologic analyzer. RESULTS AND CONCLUSION: In healthy people, concentrations of platelet serotonin were similar to those reported in literature: from 2.19 to 4.87 nmol/109 platelets. In patients with depression (n=30), platelet serotonin concentrations were lower (from 0.11 to 2.89 nmol/109 platelets) and correlated with the severity of depression. The method may be used in clinical purposes.

RESUMO

Authors studied the cytotoxic activity (CA) of natural killer cells (NK) in 16 schizophrenic patients, aged from 18 to 25 years, with the first episode of psychosis. A significant decrease in CA NK was found in the total group of patients compared to healthy controls that suggested the dysregulation of natural immunity at this stage of disease. The addition of serotonin to the cell culture in vitro in concentrations 10-9 M stimulated the NK activity while the elimination of monocytes resulted in the significant reduction of CA NK. The parallel use of two methods for assessment of CA NK (a radioactive method and an easy and safe method using multifunctional particle size analyzer Multisizer MS-4) revealed similar results.

RESUMO

The essential amino acid tryptophan is metabolized on the methoxyindole pathway to serotonin, melatonin and 5-hydroxyindoleacetic acid and on the kynurenine pathway to kynurenine and related neuroactive metabolites, including 3-hydroxykynurenine, kynurenic acid, quinolinic acid and xanthurenic acid. Kynurenine and related metabolites play a significant role in the pathogenesis of major depressive disorder and schizophrenia. This paper is the review of literature data on the most modern state of this problem.
