AIMS: A high-volume aerosol collector was developed to efficiently capture airborne bacteria in order to assess levels of diversity in the air. METHODS AND RESULTS: Particulate matter was collected on a device designed to filter 1.4 x 10(6) litres of air in a 24 h period on a 1-microm pore size polyester membrane. Methods were optimized for extraction of genomic DNA from the air filter concentrate. Preparation times of 90 s with 0.5-0. 05 mm diameter zirconia/silica beads yielded the highest concentration genomic DNA that was able to support PCR. A 24-h air sample was taken in Salt Lake City, Utah and the microbial composition was determined by the amplification and sequence analysis of 16S ribosomal DNA fragments. CONCLUSIONS: Sequence analysis revealed a large diversity in the type of microbial species present including clones matching the sequence of Clostridium botulinum. The primary components of the aerosol sample included many different spore-forming bacteria as well as more fragile members of the Proteobacteria division. SIGNIFICANCE AND IMPACT OF STUDY: The high-volume air collection and genomic DNA recovery system allows for the rapid detection of both cultivable as well as culture-resistant organisms in the environment.

The contribution of inhalation exposure to the total dose for residents of the Marshall Islands was monitored at occasions of opportunity on several islands in the Bikini and Enewetak Atolls. To determine the long-term potential for inhalation exposure, and to understand the mechanisms of redistribution and personal exposure, additional investigations were undertaken on Bikini Island under modified and controlled conditions. Experiments were conducted to provide key parameters for the assessment of inhalation exposure from plutonium-contaminated dust aerosols: characterization of the contribution of plutonium in soil-borne aerosols as compared to sea spray and organic aerosols, determination of plutonium resuspension rates as measured by the meteorological flux-gradient method during extreme conditions of a bare-soil vs. a stabilized surface, determination of the approximate individual exposures to resuspended plutonium by traffic, and studies of exposures to individuals in different occupational environments simulated by personal air sampling of workers assigned to a variety of tasks. Enhancement factors (defined as ratios of the plutonium-activity of suspended aerosols relative to the plutonium-activity of the soil) were determined to be less than 1 (typically 0.4 to 0.7) in the undisturbed, vegetated areas, but greater than 1 (as high as 3) for the case studies of disturbed bare soil, roadside travel, and for occupational duties in fields and in and around houses.

We have conducted experiments to evaluate the effectiveness of removing contaminated soils from the Nevada Test Site with a large truck-mounted vacuum cleaner. Our results show that this method is effective, relatively easy, and safe for equipment operators. With four passes of the truck-mounted vacuum, 92% of the 241Am (and the accompanying 239 + 240Pu) was removed and resuspension rates were reduced by more than 99%. The ecological impact was, however, serious in terms of soil erosion and destruction of small animal habitats. Compared to standard earth-moving techniques, vacuuming permits a significant reduction in the volume of soil collected to achieve the desired level of decontamination, and the volume reduction could result in cost savings for packaging, shipment, and burial. This cost savings would only be realized for projects involving decontamination of the top 5 cm of soil.

Between 1970 and 1986 the Nevada Applied Ecology Group (NAEG), U.S. Department of Energy, conducted environmental radionuclide studies at weapons-testing sites on or adjacent to the Nevada Test Site. In this paper, NAEG studies conducted at two nuclear (fission) sites (NS201, NS219) and two nonnuclear (nonfission) sites (Area 13 [Project 57] and Clean Slate 2) are reviewed, synthesized and compared regarding (1) soil particle-size distribution and physical-chemical characteristics of 239 + 240Pu-bearing radioactive particles, (2) 239 + 240Pu resuspension rates and (3) transuranic and fission-product radionuclide transfers from soil to native vegetation, kangaroo rats and grazing cattle. The data indicate that transuranic radionuclides were transferred more readily on the average from soil to air, the external surfaces of native vegetation and to tissues of kangaroo rats at Area 13 than at NS201 or NS219. The 239 + 240Pu resuspension factor for undisturbed soil at Area 13 was three to four orders-of-magnitude larger than at NS201 and NS219, the geometric mean (GM) vegetation-over-soil 239 + 240Pu concentration ratio was from ten to 100 times larger than at NS201, and the GM GI-over-soil, carcass-over-soil and pelt-over-soil 239 + 240Pu ratios for kangaroo rats were about ten times larger than at NS201. These results are consistent with the finding that Area 13, compared with NS201 or NS219, has a higher percentage of radioactivity associated with smaller soil particles and a larger percentage of resuspendable and respirable soil. However, the resuspension factor increased by a factor of 27 at NS201 when the surface soil was disturbed, and by a factor of 12 at NS219 following a wildfire. The average (GM) concentration of 239 + 240Pu for the GI (and contents) of Area 13 kangaroo rats and for the rumen contents of beef cattle that grazed Area 13 were very similar (400 vs. 440 Bq kg-1 dry wt, respectively) although the variability between individuals was very large. The GM carcass-over-GI 239 + 240Pu concentration ratio for kangaroo rats at Area 13, Clean Slate 2, and NS201 were similar in value (approximately 2 X 10(-2)), as were the GM GI-over-vegetation concentration ratios (approximately 2 X 10(0)) (no statistical differences).(ABSTRACT TRUNCATED AT 400 WORDS)

Two varieties of ponderosa pine (Pinus ponderosa Dougl. var. scopulorum (Rocky Mountain variety) and P. ponderosa var. ponderosa (Sierran variety)) seedlings were subjected to elevated atmospheric CO(2) for two and a half years. The CO(2) concentrations were ambient, ambient + 75 microl l(-1), ambient + 150 microl l(-1) and ambient + 300 microl l(-1), or approximately 350, 425, 500 and 650 microl l(-1) CO(2). After one and a half years of exposure to elevated CO(2) and until the end of the study, seedlings of both varieties showed symptoms of stress including mottling, mid-needle abscission and early senescence. In both varieties, exposure to CO(2) concentrations greater than ambient + 75 microl l(-1) resulted in lower chlorophyll a, chlorophyll b and carotenoid concentrations. At elevated CO(2) concentrations, the concentrations of pigments in needles of the Sierran variety were lower than those in the Rocky Mountain variety. Also, at elevated CO(2) concentrations, the pigment concentrations in the 1-year-old needles of both P. ponderosa varieties were lower than those in current-season needles.

Estimates are obtained of the fraction of ingested or inhaled 239+240Pu transferred to blood and tissues of a reproducing herd of beef cattle, individuals of which grazed within fenced enclosures for up to 1064 d under natural conditions with no supplemental feeding at an arid site contaminated 16 y previously with Pu oxide. The estimated (geometric mean [GM]) fraction of Pu transferred from the gastrointestinal tract to blood serum was about 5 x 10(-6) (geometric standard error [GSE] = 1.4) with an approximate upper bound of about 2 x 10(-5). These results are in reasonable agreement with the value of 1 x 10(-5) recommended for human radiation protection purposes by the International Commission on Radiological Protection (ICRP) for insoluble Pu oxides that are free of very small particles. Also, results from a laboratory study by Stanley (St75), in which large doses of 238Pu were orally administered daily to dairy cattle for 19 consecutive days, suggest that aged 239+240Pu at this arid grazing site may not be more biologically available to blood serum than fresh 239+240Pu oxide. The estimated fractions of 239+240Pu transferred from blood serum to tissues of adult grazing cattle were: femur (3.2 X 10(-2), 1.8; GM, GSE), vertebra (1.4 X 10(-1), 1.6), liver (2.3 X 10(-1), 2.0), muscle (1.3 X 10(-1), 1.9), female gonads (7.9 X 10(-5), 1.5), and kidney (1.4 X 10(-3), 1.7). The blood-to-tissue fractional transfers for cattle initially exposed in utero were greater than those exposed only as adults by a factor of about 4 for femur (statistically significant) and of about 2 for other tissues (not significant). The estimated (GM) fraction of inhaled Pu initially deposited in the pulmonary lung was 0.34 (GSE = 1.3) for adults and 0.15 (GSE = 1.3) for cattle initially exposed in utero (a statistically significant difference), which may be compared with the expected fraction of 0.11 at the study site using the ICRP lung model for humans.

Seven-year-old ponderosa pine (Pinus ponderosa Dougl. ex P. Laws.) saplings and one- and two-year-old ponderosa pine seedlings of a Sierra Nevada and a Rocky Mountain seed source, respectively, were exposed to CO(2)-enriched atmospheres in an outdoor open-top chamber facility for 2.5 years. Seedling growth (main stem diameter, height, volume) increased with increasing CO(2) concentration, though the two populations exhibited different patterns of response. By the beginning of the last growth season, however, the trees under the highest CO(2) concentrations showed signs of stress that included accelerated needle abscision, chlorosis, and apparent alteration of tolerance to heat. The stress response is at least partly attributable to elevated foliar temperatures resulting from CO(2)-induced stomatal closure, which in turn lowered transpirational cooling of needles.
