RESUMO

A fluorescence-electrochemiluminescence (FL-ECL) dual-mode sensor for apoE gene detection has been developed, leveraging the unique properties of ruthenium metal organic framework nanosheets (RuMOFNSs). The system utilizes the quenching effect of the Ru(bpy)32+ ECL signal by ferrocene, leading to the synthesis of a multi-electron electrical signal marker, bisferrocene. By immobilizing the P-DNA on RuMOFNSs, bisferrocene quenches both FL and ECL signals. The addition of T-DNA and the consequent formation of double-stranded DNA enable the ExoIII enzyme to excise the bisferrocene fragment, restoring the signals. The sensor demonstrates wide detection linear ranges (1 fM to 1 nM for FL and 0.01 fM to 10 pM for ECL) and remarkable sensitivity (0.048 fM for FL and 0.016 fM for ECL). The dual-mode design offers enhanced reliability through a self-correction feature, reducing false positives. Compared to single-mode sensors, the dual-mode sensor shows significant advantages. Real-world testing confirms the sensor's capacity for robust detection in actual samples, underscoring its promising application in early disease diagnosis. This innovative approach opens up avenues for multi-signal response sensors, offering significant potential for diagnostic technologies.

Seed dormancy and germination are attractive topics in the fields of plant molecular biology as they are key stages during plant growth and development. Seed dormancy is intricately regulated by complex networks of phytohormones and numerous key genes, combined with diverse environmental cues. The transition from dormancy to germination is a very important biological process, and extensive studies have demonstrated that phytohormones abscisic acid (ABA) and gibberellin acid (GA) are major determinants. Consequently, the precise balance between ABA and GA can ensure that the seeds remain dormant under stress conditions and germinate at optimal times. Here we review the role of auxin in seed dormancy and germination. Auxin is one of the classic phytohormones effective during tropism growth and tissue differentiation. Recent studies, however, show that auxin possesses positive effects on seed dormancy, which suggests that auxin is the second phytohormone that induces seed dormancy, besides ABA. We will focus on the synthetic effects in detail between auxin and ABA pathways on seed dormancy and propose future research directions.

RESUMO

Karrikins are a class of signaling molecules discovered in wildfire smoke, which can significantly promote seed germination in some species (such as Arabidopsis and Avena fatua). The structures of Karrikins were first elucidated in 2004. At present, six different types of Karrikins have been documented, and their biological activities vary significantly. So far, studies for Karrikins have become a hot spot in the plant molecular biology field. Recent advances demonstrate that Karrikins regulate plant photomorphogenesis and leaf differentiation effectively, in addition to the effect on seed germination. Furthermore, Karrikins share highly similar molecular structures and signaling transduction pathways with strigolactone. In this review, we summarize the history of discovery, signaling transduction pathways, physiological functions and ecological significance of Karrikins, and further discuss the future research directions.

OBJECTIVE: To explore the associations between urine uric acid excretion (UUAE) and diabetic retinopathy (DR)/lower limb atherosclerotic lesions in hospitalized Chinese patients with type 2 diabetes. METHODS: This cross-sectional study was conducted in 2529 hospitalized Chinese patients with type 2 diabetes. UUAE was determined enzymatically using a single 24-h urine collection. The subjects were stratified into quartile based on UUAE levels. DR was determined by digital fundus photography. Lower limb atherosclerotic lesions were assessed by Doppler ultrasound. Both DR and lower limb atherosclerosis were compared among the UUAE quartile groups, respectively. RESULTS: There was a significant decrease in the prevalence of DR in patients across the UUAE quartiles after adjustment for sex, age and diabetic duration (35.0%, 30.7%, 26.1%, and 21.5%, respectively, p = 0.000001 for trend). A fully adjusted multiple logistic regression analyses revealed that UUAE quartiles were markedly inversely associated with the presence of DR (p = 0.030). The prevalence of lower limb plaque (73.9% vs. 62.6%, p = 0.000044) and stenosis (16.3% vs. 9.7%, p = 0.000015) was markedly higher in the diabetics with DR than in those without DR. However, there was no statistical association between the UUAE and lower limb atherosclerotic lesions in type 2 diabetes. CONCLUSIONS: Decreased UUAE was an independent risk factor for DR but not for lower limb atherosclerosis in hospitalized Chinese patients with type 2 diabetes. In selected populations, such as those with type 2 diabetes, the role of uric acid in atherosclerosis may be result from other concomitantly atherosclerotic risk factors, such as DR.
