RESUMO

Reliable disease management can guarantee healthy plant production and relies on the knowledge of pathogen prevalence. Modeling the dynamic changes in spore concentration is available for realizing this purpose. We present a novel model based on a time-series modeling machine learning method, i.e., a long short-term memory (LSTM) network, to analyze oomycete Plasmopara viticola sporangia concentration dynamics using data from a 4-year field experiment trial in North China. Principal component analysis (PCA)-based high-quality input screening and simulation result calibration were performed to ensure model performance, obtaining a high determination coefficient (0.99), a low root mean square error (0.87), and a low mean bias error (0.55), high sensitivity (91.5%), and high specificity (96.5%). The impact of the variability of relative factors on daily P. viticola sporangia concentrations was analyzed, confirming that a low daily mean air temperature restricts pathogen development even during a long period of high humidity in the field.

RESUMO

OBJECTIVE: In this study, the hypothesis that hBD-3 is upregulated by LPS via epidermal growth factor receptor (EGFR) signaling pathways to enhance metastasis in oral squamous cell carcinoma (OSCC) was tested. STUDY DESIGN: hBD-3 expression in human tissue specimens was evaluated by RT-qPCR and immunohistochemical staining. The presence of hBD-3 peptide in the culture supernatants of each type of treated cells was evaluated by enzyme-linked immunosorbent assay. The chemotaxis response to LPS or hBD-3 protein of SCC-25 cells or siRNA-hBD-3 transfected cells were also measured by chemotaxis assay. Paired, 2-tailed Student t test and analysis of variance was used to assess the statistical significance between 2 groups or many groups. RESULTS: hBD-3 is highly expressed and associated with lymphatic invasion of OSCC. hBD-3 expression and EGFR phosphorylation were markedly upregulated when SCC-25 cells were treated with LPS. When SCC-25 cells were preincubated with EGFR inhibitor or TLR4-neutralizing Ab before LPS stimulation, a decrease in the expression of hBD-3 was observed. hBD-3 markedly enhanced cancer metastasis, and the chemotaxis response to LPS of SCC-25 cells was partly blocked by siRNA target hBD-3. CONCLUSION: These findings indicate that hBD-3 is upregulated by LPS via EGFR signaling pathways to enhance lymphatic invasion of OSCC.

RESUMO

Aim To investigate whether ischemic postconditioning could induce ischemic tolerance to focal cerebral reperfusion injury and comparison with ischemic preconditioning. Methods A reversible middle cerebral artery occlusion (MCAO)was used in this study. Fifty male Sprague-dawley rats were randomized into five groups(n=10): sham-operated group,MCAO group, preconditioning group, postconditioning group and nimodipin group . The neurological function was scored and the infarct volume was measured after operation. The grouping, experimental methods and procedures of another fifty rats were as above except that ipsilateral brain of infarction was dissected and made into homogenate, then the contents of MDA,SOD,GSH and LA were quantitatively measured. Results The postconditioning and preconditioning improved the neurological function, reduced the cerebral infarction volumes and cerebral swelling significantly compared with those of the MCAO group. Contents of LA and MDA were lower, while the activity of SOD in the brain tissues were higher in preconditioning group and postconditioning group than those of the MCAO group. Conclusion Postconditioning could induce brain tolerance, and increase the antioxidant activity, which might be an important mechanism of induction of brain tolerance.
