RESUMO

Seroprevalence studies on cats are essential for monitoring the occurrence of Toxoplasma gondii infection. The present research investigated anti-T. gondii antibodies, risk factors, clinical signs, hematology and serum biochemistry in cats from different regions of Rio de Janeiro. An overall 18.7% (17/91) of the cats were seroreactive, and age was associated with increased chances of seroprevalence of anti-T. gondii antibodies. Clinical signs, hematology and serum biochemistry parameters did not help achieve an antemortem diagnosis of cat toxoplasmosis. The parasite circulates in cats from three major regions of Rio de Janeiro, and the present data set will contribute to future epidemiological studies in this endemic state of Brazil.

Cryoprotectants are required to reduce damage caused to the cells due to low temperatures during the cryopreservation. Antifreeze proteins (AFP) have a well-known role in cell membrane protection, while resveratrol is a potent antioxidant. This study assessed the effect of the association of resveratrol concentrations and AFP I in a ram semen extender. Pooled semen of four rams was allocated into six treatments in a factorial arrangement: (CONT, only the semen extender); only AFP I (ANT: 0.1 µg/mL of AFP I), only resveratrol, one treatment with two levels (10 µM/mL or 50 µM/mL of resveratrol); and two treatments with the interactions, with one AFP I and one of the two levels of resveratrol (0.1 µg/mL of AFP I with 10 µM/mL resveratrol; 0.1 µg/mL of AFP I with 50 µM/mL resveratrol). No interaction between factors was observed on sperm kinetics, plasma membrane integrity, hypo-osmotic test, and mitochondrial activity parameters. There was a high probability (P = 0.06) of reducing sperm cells with functional membrane percentage in the hypo-osmotic test and increasing the percentage of sperm with high mitochondrial activity (P = 0.07) was observed in AFP presence. An interaction of AFP and resveratrol was observed in non-capacitated sperm (P = 0.009), acrosomal reaction (P = 0.034), and sperm binding (P = 0.04). In conclusion, the association of resveratrol and AFP did not improve the quality of frozen-thawed semen and even promoted deleterious effects compared to their single addition in the semen extender. The supplementation of 50 µM/mL of resveratrol improved the outcomes of frozen-thawed ram sperm, being a potential cryoprotectant.

The hematopoietic system changes during the pregnancy to carry fetal development and maternal needs. This study compared the hematological parameters between ewes with single and twin pregnancies during gestation, delivery, and postpartum. The experiment was conducted on 60 healthy pregnant Dorper ewes that were divided into two experimental groups: Group 1 (G1), with single pregnancies (n=30), and Group 2 (G2), with twin pregnancies (n=30). Blood samples were collected from all ewes at different times: immediately before fixed-time artificial insemination (AI); on day 30, 90, 120, 130, and 140 of pregnancy; immediately after delivery; and at 24h and 48h postpartum. Statistical analysis compared the two groups at different times (P<0.05). Mild, normocytic, and hypochromic anemia was detected in all ewes from AI time and throughout pregnancy from both groups, but did not prove to be of clinical relevance. In the peripartum stage (from the 140th day of pregnancy to 48h postpartum), the ewes with twin pregnancies (G2) exhibited higher erythrogram values and neutrophil: lymphocyte ratio than did ewes with single pregnancies (G1). This indicated a greater hematopoietic adaptation in the body during the development of two fetuses. Except for the eosinophil numbers, all leukogram parameters were influenced by pregnancy in a similar way in both groups, and was characterized mainly by leukocytosis with neutrophilia during peripartum due to the high presence of endogenous cortisol at delivery. Thus, these findings showed that pregnancy was a stressful physiological event that increased the leukocyte count with a slight alteration in the erythrogram of Dorper ewes.


O sistema hematopoiético sofre mudanças durante a gestação para atender o desenvolvimento fetal e as necessidades maternas. Este estudo comparou os parâmetros hematológicos entre ovelhas com gestação simples e gemelar durante a prenhez, parto e pós-parto. O estudo foi realizado em 60 ovelhas Dorper prenhes, saudáveis, divididas em dois grupos experimentais: Grupo 1 (G1) com gestação simples (n = 30); e Grupo 2 (G2) com gestação gemelar (n = 30). Amostras de sangue foram colhidos de todas as ovelhas em diferentes momentos: imediatamente antes da inseminação artificial em tempo fixo (IA); nos dias 30, 90, 120, 130 e 140 de gestação; imediatamente após o parto; e às 24h e 48h pós-parto. A análise estatística comparou os dois grupos nos diferentes momentos (P < 0,05). Anemia leve, normocítica e hipocrômica foi detectada desde a IA e ao longo da gestação, nas ovelhas de ambos os grupos, mas não houve relevância clínica. Na fase periparto (do 140º dia de gestação às 48h pós-parto), as ovelhas com gestação gemelar (G2) mostraram valores do eritrograma e proporção neutrófilos: linfócitos mais elevados do que as ovelhas com gestação simples (G1). Isso indicou maior adaptação hematopoiética da ovelha para o desenvolvimento de dois fetos. Com exceção do número de eosinófilos, todos os parâmetros do leucograma foram influenciados pela gestação de forma semelhante em ambos os grupos, e foi caracterizada principalmente por leucocitose com neutrofilia no periparto devido ao alto nível de cortisol endógeno no parto. Assim, os achados mostraram que a gestação foi um evento fisiológico estressante que aumentou a contagem de leucócitos com leve alteração no eritrograma de ovelhas Dorper.

Background: Wounds that occur with tissue necrosis and that result from the application of medications through the most diverse accesses are described as drug skin medical embolism or Nicholas syndrome in human medicine, with wide description. In veterinary medicine, this subject has not yet been described extensively and specifically in veterinary medicine, especially regarding to wounds that occurred after the application of non-intravenous medications in horses, even though these lesions are recurrent in the clinical routine. This report aims to describe a case of skin necrosis in a horse, due to phenylbutazone infection. Case: A 7 year-old Mangalarga Marchador horse, weighing 400 kg, was admitted to the Veterinary Hospital for Large Animals of the Universiade Federal Rural do Rio de Janeiro (UFRRJ), with a history of phenylbutazone injection to the left side of the neck. The animal had an extensive wound on the neck and face on the left side and was characterized by the presence of cold and devitalized skin, with a hardened and parched appearance and that easily detached. During the anamnesis, a single administration of 10 mL of a non-steroidal anti-inflammatory drug based on phenylbutazone was reported intramuscularly for about 10 days to control the pain resulting from the claudication present for 14 days. The medication was administered in the region of the lateral border of the neck, on the left side. After drug administration, the animal presented an increase in volume at the application site. After 24 h, the lesion spread from the inoculation region, extending to the head and chest of the animal. During debridement, it was found that the lesion did not reach the underlying muscle tissue. In addition to the wound, the animal had upper eyelid palsy, lower lip, and auricular ptosis. Treatment with surgical debridement of devitalized tissue, topical application of ozonated sunflower oil, ketanserin, and a free skin graft was instituted. During hospitalization, the animal had a corneal ulcer in the left eye with an unfavorable prognosis due to paralysis of the upper eyelid, with enucleation of the affected eyeball. The animal was under veterinary care for 180 days and was discharged when his wound was already in an advanced stage of healing. Discussion: The history of the application of phenylbutazone intramuscularly and the location and characteristics of the lesion presented by the patient in the present report suggest that this animal presented aseptic tissue necrosis resulting from the administration of non-steroidal anti-inflammatory drugs, phenylbutazone. Although aseptic tissue necrosis, better known as Nicolau's syndrome or drug embolism cutis, is widely characterized and described in this species, there are studies in the literature that reproduce the syndrome in pigs and rabbits. Phenylbutazone was able to cause arterial damage, mainly in the tunica intima of the artery in which the medication was administered, with perivascular inflammatory infiltrate and subsequent skin necrosis at the site of administration. In addition to the skin lesion, the animal started to show signs compatible with the left facial nerve lesion, evidenced by the immobility of the upper eyelid and labial and ear ptosis. This resulted in corneal ulceration and subsequent enucleation. The animal also developed chewing difficulty in the first months of hospitalization. This dysfunction may be due to a lesion of the mandibular nerve, responsible for innervating the masticatory muscles and the oral mucosa. However, the animal showed improvement in this aspect, no longer showing this condition after 90 days of hospitalization. The treatment used was successful in healing the wound.

This study assessed the effect of l-carnitine (LC) in sheep semen extenders containing or not egg yolk for cryopreservation in sheep. Two extenders (TRIS-egg yolk or the commercial optiXcell™ IMV medium) were used, totaling six groups: IMV - (0, 5 and 10 mM LC) and TRIS - (0, 5 and 10 mM LC). After the freezing-thawing process and throughout incubation at 38 °C for up to 3 h, several parameters were evaluated: sperm kinetics, hypoosmotic, plasma membrane integrity, capacitation status and lipid peroxidation level. The supplementation of either 5 or 10 mM LC randomly affected some parameters and, overall, TRIS was superior (P < 0.05) than IMV extender. In the LC-groups, IMV had greater (P < 0.05) oxidative stress than TRIS. In conclusion, although LC affected isolated parameters, its supplementation in semen extender had no consistently beneficial effect on freezing-thawing ram sperm.

A retrospective study of Cattle Tick Fever was made with animals up to one year old, which occurred from 1986-2007 in Botucatu-SP in the influence area of the Veterinary Hospital of the School of Veterinary Medicine and Animal Science UNESP. There were 232 cases and 57 deaths. The main etiological agent causing the disease on this region is Anaplasma marginale identified as a single agent in 31.5% of the cases. Most of the cases occurred on autumn in mixed-blood animals two to six months old. Predominant clinical signs included apathy, hyporexia or anorexia, dehydration, weight loss and pale mucosa. The high incidence and mortality observed justify prophylactic actions to have the balance between host, agent, vector, and environment, thus avoiding the clinical form of disease and death of livestock herds in the region.


Foi realizado um estudo retrospectivo dos casos de tristeza parasitária bovina (TPB) em bovinos com até um ano de idade ocorridos na região de Botucatu-SP, área de influência do Hospital Veterinário da Faculdade de Medicina Veterinária e Zootecnia de Botucatu UNESP, entre os anos de 1986 e 2007. Ocorreram, nesse período, 232 casos da doença e 57 mortes. O principal agente etiológico causador da doença nesta região foi o Anaplasma marginale, identificado como um único agente em 31,5% dos episódios da doença. A maioria dos casos ocorreu no Outono, em animais mestiços e com dois a seis meses de idade. Os sinais clínicos predominantes foram: apatia, hiporexia ou anorexia, desidratação, perda de peso e palidez das mucosas. A elevada incidência e mortalidade observados justificam ações profiláticas visando o equilíbrio entre hospedeiro, agente, vetor, e meio ambiente, evitando, assim, a forma clínica da doença e morte dos animais em rebanhos da região.

Foram analisados os resultados e complicações decorrentes do emprego da técnica de biopsia pulmonar transtorácica percutânea em 20 ovinos clinicamente sadios. Os animais foram submetidos ao exame clínico seguido da biopsia com agulha semi-automática no 7º espaço intercostal direito, 5cm acima do olécrano. Foram analisados o número de tentativas para a execução do procedimento e o tamanho dos fragmentos. As amostras obtidas foram avaliadas histologicamente. Posteriormente ao abate, foi realizado o exame pos mortem para avaliação de complicações da técnica e das eventuais lesões provocadas. Entre todos os animais submetidos à biopsia apenas dois demonstraram resistência a técnica, sendo obtidos fragmentos pulmonares de 4-7mm de comprimento, com média de 1,8±1 tentativas por fragmento. As principais alterações relacionadas à técnica foram tosse, dispnéia inspiratória, elevação das freqüências cardíaca e respiratória e aumento do ruído broncobronquiolar. Ao exame post mortem observou-se hemorragia dos músculos intercostais e pleura visceral em todos os animais. Das 20 tentativas de execução da técnica, 18 obtiveram sucesso, enquanto que em duas o fígado foi equivocadamente puncionado. As amostras de tecido pulmonar foram consideradas representativas, pois foi possível a visualização de estruturas íntegras, incluindo bronquíolos e alvéolos. Podemos concluir que a biopsia pulmonar é segura e eficaz para obtenção de amostras pulmonares com fins de diagnóstico histológico.(AU)


The results and complications arising from use of the percutaneous transthoracic lung biopsy technique in 20 clinically healthy sheep were analyzed. The animals were subjected to clinical examination followed by lung biopsy with a semi-automatic needle in the 7th right intercostal space, 5cm above the olecranon. The number of attempts to implement the procedure and size of the fragments were evaluated. The samples were evaluated histologically. After the slaughter, a postmortem examination assessed the technical complications and possible injuries. Among all animals which underwent biopsy only two did not tolerate the technique. Lung fragments measuring 4-7mm were obtained by an average of 1.8 attempts per fragment. The main changes related to the procedure were cough, labored breathing, increased heart and breathing rate, and increased bronco-bronchiolar sound. The postmortem examination revealed bleeding of the intercostal muscles and visceral pleura of various extend in all lambs, and a wound up to 14mm length in the lung of two animals that did not tolerate the procedure. From 20 attempts to implement the technique, 18 were successful, while in two of them the liver was mistakenly punctured. Samples of lung tissue were representative. It was possible to visualize intact structures, including bronchioles and alveoli. It could be concluded that lung biopsy is a safe and effective procedure to obtain lung samples for histological diagnosis.(AU)

RESUMO

As espécies reativas do oxigênio (ERO) são moléculas instáveis e extremamente reativas capazes de transformar outras moléculas com as quais colidem. As ERO são geradas em grande quantidade durante o estresse oxidativo, condição em que são afetadas moléculas como proteínas, carboidratos, lipídeos e ácido nucleicos. Neste trabalho, são discutidos os principais conceitos sobre os radicais livres e as ERO: principais tipos, sua formação e a forma como atuam sobre as estruturas celulares, provocando lesão tecidual significativa. Os principais sistemas de defesa antioxidantes e a influência do aumento na produção dessas ERO no trato respiratório de grandes animais também são discutidos, dando ênfase ao envolvimento das ERO em doenças como a pneumonia em ruminantes e na obstrução recorrente das vias aéreas e a hemorragia pulmonar induzida por exercício em equinos.


The reactive oxygen species (ROS) are unstable and highly reactive molecules capable of transforming other molecules with which they collide. The ROS are generated in large numbers during the oxidative stress, a condition in which molecules are affected such as proteins, carbohydrates, lipids and nucleic acid. This research discussed the main concepts of the free radicals and ROS: main types, their training and how they act on the cellular structures causing significant tissue injury. The main antioxidant defense systems and the influence of the increase in the production of ROS in the respiratory tract of large animals are also discussed, with emphasis on the involvement of ROS in diseases such as pneumonia in ruminants and recurrent obstruction of the airways and pulmonary hemorrhage induced by exercise in horses.

Foram analisados os resultados e complicações decorrentes do emprego da técnica de biopsia pulmonar transtorácica percutânea em 20 ovinos clinicamente sadios. Os animais foram submetidos ao exame clínico seguido da biopsia com agulha semi-automática no 7º espaço intercostal direito, 5cm acima do olécrano. Foram analisados o número de tentativas para a execução do procedimento e o tamanho dos fragmentos. As amostras obtidas foram avaliadas histologicamente. Posteriormente ao abate, foi realizado o exame pos mortem para avaliação de complicações da técnica e das eventuais lesões provocadas. Entre todos os animais submetidos à biopsia apenas dois demonstraram resistência a técnica, sendo obtidos fragmentos pulmonares de 4-7mm de comprimento, com média de 1,8±1 tentativas por fragmento. As principais alterações relacionadas à técnica foram tosse, dispnéia inspiratória, elevação das freqüências cardíaca e respiratória e aumento do ruído broncobronquiolar. Ao exame post mortem observou-se hemorragia dos músculos intercostais e pleura visceral em todos os animais. Das 20 tentativas de execução da técnica, 18 obtiveram sucesso, enquanto que em duas o fígado foi equivocadamente puncionado. As amostras de tecido pulmonar foram consideradas representativas, pois foi possível a visualização de estruturas íntegras, incluindo bronquíolos e alvéolos. Podemos concluir que a biopsia pulmonar é segura e eficaz para obtenção de amostras pulmonares com fins de diagnóstico histológico.


The results and complications arising from use of the percutaneous transthoracic lung biopsy technique in 20 clinically healthy sheep were analyzed. The animals were subjected to clinical examination followed by lung biopsy with a semi-automatic needle in the 7th right intercostal space, 5cm above the olecranon. The number of attempts to implement the procedure and size of the fragments were evaluated. The samples were evaluated histologically. After the slaughter, a postmortem examination assessed the technical complications and possible injuries. Among all animals which underwent biopsy only two did not tolerate the technique. Lung fragments measuring 4-7mm were obtained by an average of 1.8 attempts per fragment. The main changes related to the procedure were cough, labored breathing, increased heart and breathing rate, and increased bronco-bronchiolar sound. The postmortem examination revealed bleeding of the intercostal muscles and visceral pleura of various extend in all lambs, and a wound up to 14mm length in the lung of two animals that did not tolerate the procedure. From 20 attempts to implement the technique, 18 were successful, while in two of them the liver was mistakenly punctured. Samples of lung tissue were representative. It was possible to visualize intact structures, including bronchioles and alveoli. It could be concluded that lung biopsy is a safe and effective procedure to obtain lung samples for histological diagnosis.
