RESUMO

The study comprises 26 patients with non-Hodgkin's lymphoma who entered complete remission. Lymphocyte subsets and spontaneous and mitogen (ConA, PWM)-induced DNA synthesis were recorded in purified blood lymphocytes harvested before treatment, in remission and in relapse. The T and B lymphocytopenia noted in untreated patients persisted in complete remission. At diagnosis the spontaneous DNA synthesis was increased and the response to mitogen was decreased as compared to controls. Both functions became normalised during complete remission in most patients under 60 yr. The lymphocyte abnormalities reappeared during relapse. A low response to mitogen (ConA) in complete remission seemed to predict early relapse in patients younger than 60 yr. It is concluded that functional abnormalities of blood lymphocytes in non-Hodgkin's lymphoma are closely related to the presence of active disease.

RESUMO

Antibody-dependent cellular cytotoxicity (ADCC) of human leukaemic blood cells against human RBC treated with IgG isoantibody was studied by the 51Cr-release method. ADCC in this particular system is a property of normal phagocytic cells of the monocytic and myeloid series while lymphocytes are inactive. Well differentiated leukaemic monocytes from patients with acute monocytic leukaemia were highly cytotoxic and engulfed opsonized RBC. Promyelocytic leukaemic cells from two patients with acute promyelocytic leukaemia were cytotoxic and phagocytic. Seven patients with low differentiated acute myeloblastic leukaemia had no cytotoxic or phagocytic blood cells. Leukaemic B cells from patients with chronic lymphocytic leukaemia or prolymphocytic leukaemia lacked cytotoxic and phagocytic properties. It is concluded that ADCC against isoantibody-treated human RBC may be a tool to distinguish between well and poorly differentiated leukaemic cells of the monocytic or myeloid series.

Forty-four adult patients under 60 years of age with acute nonlymphoblastic leukemia were randomized for induction treatment with one of the following three regimens: R 1 = courses of daunorubicin on day 1 + ARA-C on days 1--5; R 2 = courses of daunorubicin on days 1 and 2 + ARA-C on days 4--8; R 3 = courses of daunorubicin-DNA complex on days 1--2 + ARA-C on days 4--8. Out of 14 patients, 9 went into remission on R 1, 6 out of 14 on R 2, and 8 out of 16 on R 3. The preliminary results suggest that daunorubicin-DNA complex has the same efficacy for inducing remission as daunorubicin alone, if the same time intervals and dosages are used.
