RESUMO

The efficiency and specificity of RNA-protein cross-linking in the 30S subunit of Escherichia coli ribosomes, induced by low-intensity (10(15) photons cm-2 s-1, 254 nm) and high-intensity [(1.6-6.8) X 10(24) photons cm-2 s-1, 266 nm, pulse duration 10(-8) s] ultraviolet radiation, are studied. Under the former conditions proteins S4, S7 and S9/S11, and under the latter conditions these proteins together with S3, S18 and S20, are cross-linked to 16S RNA. Biphotonic processes operate in the latter case. In the presence of 2-mercaptoethanol cross-linking occurs either directly, via a higher excited state or via activated intermediates with life-times less than 25 ns. Cross-links thus formed are specific, i.e. they are formed between regions of macromolecules which are in contact in the native (non-disturbed) complex prior to excitation. The efficiency of cross-linking (per photon absorbed) is 20-100 times higher upon two-step excitation as compared with single-step excitation and an analysable number of cross-links are produced in a single pulse. Only base U-1239 of 16S RNA is cross-linked to protein S7 by low-intensity radiation, whereas the adjacent base, G-1240 is also involved in laser-induced cross-linking. A transition from the former to the latter conditions allows one to reduce the duration of irradiation from several minutes to several nanoseconds.

RESUMO

The action of high-intensity ultraviolet pulse laser radiation on a 161 bp fragment of pBR 322 DNA (EcoRI-MspI fragment) was studied. At doses up to 5 X 10(18) photons/cm2 the N-glycosidic bond splitting is negligible. The action of hot piperidine on irradiated DNA leads to chain splitting at the residues, modified via biphotonic processes. The modification and, hence, splitting efficiencies depend on the type of base (G greater than T greater than A greater than C) and on its position in the sequence. Preferentially modified bases in the opposite strands of double-stranded DNA belong, mainly, to the same or adjacent base pairs. Residues in the Pribnow box are modified considerably less, than in the sequences, immediately upstream and downstream. This approach seems to be useful in footprinting of DNA secondary structure peculiarities and alterations, conjugated with the functional role and state of the respective fragment.

RESUMO

Some main products of laser photolysis and gamma-radiolysis of thymine were shown to undergo subsequent transformations during DNA hydrolysis by formic acid to the bases. The method has been developed for the quantitative determination of the character and the degree of thymine transformations under the effect of laser- and gamma-radiations with a reference to the end-products of the irradiated thymine hydrolysis.

RESUMO

UV-irradiation (lambda = 254 nm) of liquid aqueous solutions of deoxyguanosine in the presence of oxygen at pH less than 7 causes an intensive degradation of nucleoside. The quantum yield estimated from A254 decrease for the reaction mixture was found to be 1.5X10(-4) at doses to 150 E/mole. The rate of A254 decrease was found to grow with increasing doses. The structures of the products isolated from the reaction mixture after irradiation suggest that one of the ways of deoxyguanosine degradation is a breakdown of a purine cycle without splitting of N-glycoside bond. Simultaneously another type of photoinduced modification of guanine nucleus takes place, which is followed by appearance of free 2-deoxyribose. Deoxyguanosine degradation in both directions is kinetically one-step process proceeding with comparable quantum yields of approximately 1X10(-4).

RESUMO

UV (lambda = 254 nm) irradiation of bacteriophage MS2 or its treatment with bisulfite induce covalent crosslinkage of the RNA to the coat protein. epilsonN-(2-oxopyrimidyl-4)-lysine was found in the phage hydrolysates after either type of treatment. An equimolar mixture of 0-methylhydroxylamine and bisulfite causes complete disappearance of the cross-links. This led to the conclusion that one of the factors responsible for the UV-induced polynucleotide-protein crosslinkage and the main factor in treatment with bisulfite is substitution of the exocyclic amino group of the activated cytosine nucleus by the lysine residue epilson-amino group of the protein.

RESUMO

The principal UV-induced (lambda = 2546nm) reaction of N4-hydroxy- and N4methoxycytidines and N6-methoxyadenosine in neutral aqueous solutions is cleavage of the exocyclic N-O bond with the respective formation of cytidine and adenosine. Quantum yields are 2.8x10(-3) and 2.2x10(-3) M/E for the first two compounds and 9.1x10(-3) M/E for N6-methoxyadenosine.

It has been shown that the quantum yield of the photochemical conversion of adenine and the corresponding nucleosides and nucleoside 5'-phosphates in liquid (pH 5.6 and 2.0) and frozen aqueous solutions do not exceed 10(-4). The quantum yield of the photoconversion of guanine-containing nucleosides and nucleoside 5'-phosphates in liquid aqueous solution (pH 5.6) after removal of oxygen by passing through nitrogen and in the frozen state do not exceed 0.3 x 10(-4). The quantum yield in oxygen-containing liquid aqueous solutions increase to 0.3 x 10(-3), i.e. to values commensurate with the quantum yield of pyrimidine photolysis.
