RESUMO

The growth of Haematococcus pluvialis in two alternative culture media NPK (10:10:10) and ME (macrophyte extract), under mixotrophic conditions using sugarcane molasses as a carbon source were evaluated for 28 days. The molasses was used in two different ways, in a native form (untreated) and a hydrolyzed (pretreated). Cell density of Haematococcus pluvialis in mixotrophic cultivation was higher in pretreated molasses. Growth rate was higher when pretreated molasses were employed in mixotrophic cultivation with NPK culture medium (k=0.5 7th growth day). Biomass, chlorophyll-a, conductivity and total inorganic nitrogen were not significantly different (p>0.05) during the experimental period for two mixotrophic cultivation and culture media. Protein contents of H. pluvialis biomass were higher in NPK culture medium with pretreated molasses (50% dry biomass). Annual biomass production was 520 kg-1 dry biomass with untreated molasses for two culture media, and 650 and 520 kg-1 dry biomass with pretreated molasses for NPK and ME culture media, respectively. The use of NPK and ME culture media in mixotrophic cultivation may be a new protocol for H. pluvialis cultivation due to the low cost and similar annual production.

Abstract Growth and biological conditions of Messastrum gracile were evaluated to compare the effect of photoautotrophic and mixotrophic cultivation on the increase of biomass production and chemical conditions cultured in macrophyte and commercial culture media. The growth rate (k) of M. gracile was different in the culture media, higher in mixotrophic cultivation for Lemna minor culture medium, whilst to Eichhornia crassipes and NPK culture media were higher in photoautotrophic cultivation. Mean lipid contents in photoautotrophic cultivation were 8.2% biomass dry weight, whereas they reached 19% biomass dry weight in mixotrophic cultivation. Protein contents were below 48% biomass dry weight in photoautotrophic cultivation and 30% biomass dry weight in mixotrophic cultivation. Messastrum gracile cultured in macrophyte culture media (E. crassipes and L. minor) and NPK culture medium provided satisfactory results with regard to lipid and protein contents in mixotrophic and photoautotrophic cultivations, respectively. Lipid and protein contents in alternative media were higher or similar to the CHU12 commercial culture medium.


Resumo O crescimento e as condições biológicas da microalga Messastrum gracile foram avaliados para comparar o efeito do cultivo foto-autotrófico e mixotrófico na produção de biomassa e condições químicas em meios de cultura comercial e de macrófitas. A taxa de crescimento (k) de M. gracile foi diferente entre os meios de cultura, sendo maior no cultivo mixotrófico para o meio Lemna minor, enquanto para os meios Eichhornia crassipes e NPK foram maiores no cultivo foto-autotrófico. Os teores de lipídios no cultivo foto-autotrófico foram de 8,2% da biomassa seca, enquanto que no mixotrófico atingiram 19% da biomassa seca. Os teores de proteína em cultivo foto-autotrófico estiveram abaixo de 48% da biomassa seca e 30% de biomassa seca no cultivo mixotrófico. Messastrum gracile cultivada em meios de cultura de macrófitas (E. crassipes e L. minor) e NPK apresentaram resultados satisfatórios em relação aos teores de lipídeos e proteínas nos cultivos mixotróficos e foto-autotróficos, respectivamente. Os teores lipídicos e proteicos em meios alternativos foram maiores ou semelhantes ao meio comercial CHU12.

Resumo Duas espécies de Chlorophyceae, Ankistrodesmus gracilis e Haemotococcus pluvialis foram utilizadas para comparar o efeito do melaço de cana-de-açúcar como fonte de carbono. Nos dois meios de cultura de microalgas, a maior densidade celular foi obtida em meio de cultura comercial (CHU12 e WC). Durante o crescimento exponencial (1º ao 10º dia), detectou-se alta densidade celular em H. pluvialis para o meio de cultura E. crassipes variando entre 0,4 x 105 células mL-1 e 1,7 x 105 células mL-1. Os meios de cultura foram fundamentais para o crescimento em cultivo mixotrófico. O melaço de cana-de-açúcar apresentou resultados diferentes para as duas microalgas em relação ao crescimento, aos teores de proteína e lipídio foram mais elevados para o cultivo de H. pluvialis, exceto lipídio no final do experimento onde A. gracilis apresentou quase o dobro dos níveis de lipídio. O melaço de cana de açúcar pode ser uma fonte alternativa de carbono em condições de laboratório.

Abstract Current study investigated the effectiveness of different macrophytes as culture media for Ankistrodesmus gracilis in laboratory conditions. Significant difference (p < 0.05) was reported in cell density with regard to conventional culture medium (CHU12) and macrophytes culture media. Mean cell density in NPK, Eichhornia crassipes and E. azurea media was higher (p < 0.05) than in conventional culture medium. Chlorophyll-a was higher than 1 g.L-1, except in CHU12 (0.7 ± 0.4 g.L-1) and T. domingensis (0.8 ± 0.3 g.L-1) media. Nitrate decreased sharply as from the 7th-day of the experiment. Ammonium and total phosphorus were highest in culture media and ranged between 0.4 g.L-1 (P. cordata medium) and 1.7 g.L-1 (CHU12 medium) for ammonium, and between 0.8 g.L-1 (CHU12 medium) and 1.9 g.L-1 (T. domingensis medium) for total phosphorus. Results revealed inorganic fertilizer and macrophytes combined with vitamins may be effective as culture media and strongly supports the growth of Ankistrodesmus gracilis, since cell density and biochemical composition are similar to or higher than conventional culture medium (CHU12). Macrophyte is a tool for aquaculture since biological wastes may be used with nutrients to improve the cultivation of microalgae.


Resumo O presente estudo investigou a eficácia na utilização de diferentes macrófitas como meio de cultura para Ankistrodesmus gracilis em condições laboratoriais. Foi observada diferença significativa (p < 0,05) entre a densidade celular em relação aos meios de cultura convencional (CHU12) e de macrófitas. Os meios de cultura com Eichhornia crassipes, E. azurea e NPK apresentaram densidade celular maiores (p < 0,05) que o meio de cultura convencional. Os teores de clorofila-a foram superiores a 1 g.L-1, exceto nos meios de cultura CHU12 (0.7 ± 0.4 g.L-1) e de T. domingensis (0.7 ± 0.3 g.L-1). O nitrato diminuiu acentuadamente a partir do 7º dia de experimento. Dentre os nutrientes, amônia e o fósforo total foram os mais elevados nos meios de cultura, variando entre 0.4 g.L-1 (meio de P. cordata) a 1,7 g.L-1 (meio CHU12) para amônia e, entre 0.8 g.L-1 (meio CHU12) e 1,9 gL-1 (meio de T. domingensis) para o fósforo total. Os resultados do presente estudo mostraram que o uso de fertilizantes inorgânicos e macrófitas, combinados com vitaminas, pode ser eficaz como meio de cultura no crescimento de Ankistrodesmus gracilis, uma vez que a densidade celular e a composição bioquímica foram semelhantes ou superiores ao meio de cultura convencional (CHU12). As macrófitas são ferramentas a serem adotadas na aquicultura, desde que os resíduos biológicos podem ser usados como nutrientes para melhorar o cultivo de microalgas.

Abstract The effects of treated and untreated water inlets with macrophytes on the improvement of water quality and zooplankton community were evaluated in a fish pond with continuous water flow. Water and zooplankton samples were retrieved at four sites during nine months. There were differences (p<0.01) between inlet water from fish pond and inlet water from canal with macrophytes, featuring higher concentrations of nutrient load, mainly TAN and TP in the former. The inlet water from fish pond contained a higher number of abundant species (9 species), whilst the water supply from the canal with macrophytes had a greater richness (31 species) of zooplankton species. Results showed that inlet water without macrophytes directly affected the characteristics of the water column and the dominance of zooplankton species such as Thermocyclops decipiens, and greater abundance of Rotifera species. Since aquatic plants in the inlet water of fish pond analyzed showed lower allochthonous material loads from the previous fish pond, the management adopted with macrophytes may be applied to avoid eutrophication risks, common in farm ponds.


Resumo Os efeitos do abastecimento tratado e não tratado com macrófitas sobre a qualidade da água e comunidade zooplanctônica foram avaliados em um viveiro de piscicultura com fluxo contínuo de água. Houve maior concentração de fósforo total e nitrogênio amoniacal na água não tratada (p<0,01). Na água de entrada sem tratamento de macrófitas foi observado maior número de espécies abundantes (9 espécies), contudo, a entrada de água tratada com macrófitas apresentou maior riqueza de espécies (31 espécies) zooplanctônicas. Os resultados obtidos mostraram que a água de entrada sem macrófitas influenciou as características da coluna de água com predominância de espécies zooplanctônicas, como Thermocyclops decipiens e maior abundância de espécies de Rotifera. A água tratada com macrófita apresentou menor carga de materiais alóctones provenientes do viveiro anterior, indicando que o manejo adotado com macrófitas pode ser aplicado para evitar riscos de eutrofização, comuns em piscicultura.

Abstract The growth rate and percent survival of Betta splendens when submitted to formulated diet and live food treatments are evaluated. The three different diets were used and designated as: formulated diet (basal diet); live food diet (plankton) and mixed diet (formulated diet with plankton). The live food diet contained plankton belonging to an open pond. High mortality was reported with live food (plankton) treatment whereas higher percent survival occurred with formulated diet. Highest specific growth rate, weight gain and final weight were reported in the mixed diet treatment and were significantly different (p<0.01) from those in formulated diet and live food treatments. The gut contents of B. splendens in mixed diet and live food treatments comprised, Rotifera and Bacillariophyceae species in high percentages or rather, over 78% of total organisms. Lecane sp. was the most ingested zooplankton species by B. splendens in both treatments (mixed diet and live food), with the phytoplankton species Asterionella sp. and Melosira sp. respectively in mixed diet and in live food, respectively. Results indicated that the formulated diet influenced the water parameters dissolved oxygen, total suspended solids, total dissolved solids and pH. The live food in the open pond was not enough to improve the growth rate and percent survival of B. splendens. The growth performance of B. splendens; had the best results with mixed diet which was capable of maintaining species’s survival (82%) and development in artificial conditions, benefiting the culture management of ornamental fish.


Resumo Foi avaliado o crescimento e sobrevivência de Betta splendens, quando submetidos à dieta formulada e alimento vivo. Foram utilizadas três dietas experimentais e designadas como: dieta formulada (ração); alimento vivo (plâncton) e dieta mista (ração com plâncton). O plâncton pertence ao tanque externo de cultivo. Alta mortalidade foi observada no tratamento com alimento vivo (plâncton) e a mais alta sobrevivência na dieta formulada. No tratamento com dieta mista foram observadas as maiores taxas de crescimento específico, ganho de peso e peso final, sendo significativamente diferentes (p<0,01) dos outros tratamentos. No conteúdo do trato digestório do B. splendens nos tratamentos dieta mista e alimento vivo, as espécies de Rotifera e Bacillariophyceae foram encontradas acima de 78% do total de organismos observados. Lecane sp. foi a espécie de zooplâncton mais ingerida por B. splendens em ambos os tratamentos (dieta mista e alimento vivo) e de fitoplâncton foi Asterionella sp. (dieta mista) e Melosira sp. (alimento vivo). Os resultados do presente estudo indicaram que a dieta formulada (ração) influenciou alguns parâmetros da água, como oxigênio dissolvido, sólidos totais solúveis, sólidos totais solúveis, sólidos totais suspensos e pH, e o alimento vivo não foi suficiente para incrementar o crescimento e a sobrevivência de B. splendens. A dieta mista promoveu o maior crescimento do B. splendens possibilitando a sobrevivência de 82% e, o desenvolvimento desta espécie em condições artificiais, beneficiando o manejo do cultivo deste peixe ornamental.

The gut contents of B. splendens in mixed diet and live food treatments comprised, Rotifera and Bacillariophyceae species in high percentages or rather, over 78% of total organisms. Lecane sp. was the most ingested zooplankton species by B. splendens in both treatments (mixed diet and live food), with the phytoplankton species Asterionella sp. and Melosira sp. respectively in mixed diet and in live food, respectively. Results indicated that the formulated diet influenced the water parameters dissolved oxygen, total suspended solids, total dissolved solids and pH. The live food in the open pond was not enough to improve the growth rate and percent survival of B. splendens. The growth performance of B. splendens; had the best results with mixed diet which was capable of maintaining species's survival (82%) and development in artificial conditions, benefiting the culture management of ornamental fish.

Current study investigates the effect of two alternative media NPK (20-5-20) fertilizer and NPK plus macrophyte (M+NPK) compared to the commercial medium (WC) under growth rate and physiological parameters in batch culture mode (2-L), and verifies whether the use of fertilizer (NPK) and macrophyte (Eichhornia crassipes) would be a good tool for Haematococcus pluvialis culture in the laboratory. The highest number of cells of H. pluvialis has been reported in NPK medium (5.4 × 105 cells.mL-1) on the 28th day, and in the M+NPK and WC media (4.1 × 105 cells.mL-1 and 2.1 × 105 cells.mL-1) on the 26th day, respectively. Chlorophyll-a contents were significantly higher (p<0.05) in NPK medium (41-102 µg.L-1) and lower in WC and M+NPK media (14-61 µg.L-1). The astaxanthin content was less than 0.04 mg.L-1. Production cost of 10-L of H. pluvialis was low in all media, and NPK and M+NPK media had a cost reduction of 65% and 82%, respectively when compared with commercial medium (WC). The use of a medium based on commercial fertilizer and macrophyte (E. crassipes) produced a new medium formulation that proved to be efficient, at least in batch culture mode, in promoting high density culture of H. pluvialis. NPK and macrophyte (E. crassipes) medium seems to be an adequate alternative to replace the conventional one (WC).

Differences in tilapia growth among ponds were related to stocking density and ecological process affecting tilapia food availability and intraspecific competition. Lettuce wastes addition into the ponds did not produce negative effects, thus this practice may be considered a disposal option and a low-cost input source for tilapia, at least at the amounts applied in this study.

Resumo No sudeste do Brasil, a criação extensiva e semi-intensiva de tilápias é realizada em viveiros escavados com fluxo contínuo, com a disponibilidade e manejo do fluxo de água diferentes nas épocas chuvosa e seca. Na região, os resíduos de alface são um recurso potencialmente de baixo custo para a tilapicultura. Este estudo examinou os processos ecológicos ocorrendo em três viveiros escavados com fluxo contínuo fertilizados com restos de alface para criação extensiva de tilápias durante as épocas chuvosa e seca. Parâmetros de qualidade de água, plâncton e sedimento foram amostrados mensalmente durante um ano. A análise de fator foi utilizada para identificar os processos ecológicos nos viveiros e elaborar um modelo gráfico do funcionamento do ecossistema dos viveiros durante as duas épocas. Os processos relacionados ao ciclo do nitrogênio mostraram diferenças entre as duas épocas, enquanto que os processos relacionados com o ciclo do fósforo não foram influenciados pela época. As diferenças ecológicas entre os viveiros foram principalmente devido aos efeitos do vento, adição de material orgânico e densidade de estocagem. Diferenças no crescimento das tilápias entre os viveiros relacionaram-se com a densidade de estocagem e os processos ecológicos afetando a disponibilidade de alimento e a competição intraespecífica. A adição dos restos de alface nos viveiros não resultou em efeitos negativos, assim esta prática pode ser considerada um destino alternativo e um recurso de baixo custo para a criação extensiva de tilápias, pelo menos nas quantidades investigadas neste estudo.

Resumo O presente estudo investigou o efeito de dois meios alternativos, NPK (20-5-20) e NPK mais macrófita (M+NPK), em relação ao meio comercial (WC) para avaliar a taxa de crescimento e parâmetros fisiológicos em cultivo estático (2-L), a fim de verificar se o fertilizante (NPK) e a macrófita (Eichhornia crassipes) podem ser utilizados no cultivo de Haematococcus pluvialis em laboratório. O maior número de células de H. pluvialis foi observado no meio NPK (5,4 × 105 células.mL–1) no vigésimo oitavo dia, e nos meios M+NPK e WC foi de 4,1 × 105 células.mL–1 e 2,1 × 105.celulas.mL–1 no vigésimo sexto dia, respectivamente. Os teores de clorofila-a foram significativamente maiores (p<0,05) em meio NPK (41-102 g.L–1) quando comparado aos meios WC e M+NPK (14-61 g.L–1). O teor de astaxantina foi menor que 0,04 mg.L–1. O custo de produção de 10-L de H. pluvialis foi baixo em todos os meios sendo que os meios NPK e M+NPK apresentaram uma redução de custos de 65% e 82%, respectivamente, quando comparados ao meio comercial. O meio contendo fertilizante e o de macrófita (E. crassipes) obtiveram resultados eficientes em cultivo estático, com alta densidade celular de H. pluvialis. O meio NPK e o de macrófita (E. crassipes) demonstraram ser uma alternativa adequada para substituir o meio comercial (WC).

Plankton communities and macrofauna associated to aquatic macrophyte stands in a shallow water-supply reservoir (21°14'09″S; 48°18'38″W) on an aquaculture farm were compared to evaluate the relationship between organism densities and some abiotic features of the reservoir. Water and communities associated were sampled at two sites, one in an area with the predominance of Eichhornia azurea (Sw.) Kunth and the other with the predominance of Salvinia auriculata Aublet. Communities associated with macrophytes were sampled with floating quadrants (0.5 m2); the macrophytes were washed and plankton and macrofauna were fixated with 4% formalin and 1% lugol iodine; the specimens were then identified and counted. Plankton and macrofauna communities associated with S. auriculata and E. azurea had a similar diversity of species but different (p<0.05) in the abundance of associated organisms. Eichhornia azurea had the highest contents in dry and wet weight, total phosphorus, total nitrogen and organic matter. Planktonic algae were directly correlated with biomass of E. azurea. The taxa with highest densities were Rotifera and Zygnematophyceae. Results showed that the environmental variables associated with macrophytes presence in the shallow reservoir is a strong predictor of favourable conditions to maintain great diversity plankton community and macrofauna associated with plants. The role of macrophytes is important for not only stabilising the clear-water state and maintaining high diversity of organisms associated, but also it seems to be a good alternative to maintaining desirable water-supply quality for aquaculture farms.

The aim of this work was to investigate the growth performance of Diaphanosoma birgei fed with two Chlorophyceae algae, Ankistrodesmus gracilis and Haematococcus pluvialis using monoalgal diets and simpler mixed diets. D. birgei was daily fed on four treatments: 1) 100% Ankistrodesmus gracilis (Ag); 2) 100% Haematoccocus pluvialis (Hp); 3) 25% A. gracilis + 75% H. pluvialis (Ag-25+Hp-75) and 4) 75% A. gracilis + 25% H. pluvialis (Ag-75+Hp-25). The fecundity curve of D. birgei showed that the mixed feed Ag-25+Hp-75 and temperature 24±2°C triggered fast fecundity at approximately two days. The fecundity was low when based only on H. pluvialis (Hp), albeit with greater longevity (19 days) and a higher number of broods (8). D. birgei fed on Ag and Ag-75+Hp-25 diets in this experiment sustained higher growth rate and higher lipid content in these treatments. The present study showed that A. gracilis diet and mixed microalgae diets tested were able to support the egg production and development of D. birgei.
