RESUMO

Two new Sm3+ complexes with pyrazolic ß-diketones bearing a CF3 group acting as main ligands and with 2,2'-bipyridine or 1,10-phenanthroline being the ancillary ligand were studied, and their energy level structure was established. Stark splitting observed in the photoluminescence spectra of the complexes points to their non-cubic symmetry, confirmed by the calculated Judd-Ofelt intensity parameters. Internal quantum yields obtained for the compounds by the Judd-Ofelt calculations were of the order of 5.5%, whereas the measured external quantum yields were 0.75% and 1.5% for Sm3+ complexes involving 2,2'-bipyridine and 1,10-phenanthroline ancillary ligands, respectively, with the corresponding sensitization efficiencies calculated as 0.16 and 0.26. It was demonstrated that replacing the 1,10-phenanthroline ancillary ligand with 2,2'-bipyridine provides an increase in the intensity of 650 nm emission of the Sm3+ complexes, with the branching ratio reaching 55%. Intensive emission of the studied complexes at 650 nm offers hope for their use as spectrally pure red emitters.

DNA aptamers (oligonucleotides) interacting with thrombin exosite I contain G-quadruplex, two T-T, and one T-G-T loops in their structure. They prevent exosite I binding with fibrinogen and thrombin receptors on platelet surface, thereby suppressing thrombin-stimulated formation of fibrin from fibrinogen and platelet aggregation. Earlier, we synthesized original antithrombin aptamer RE31 (5'-GTGACGTAGGTTGGTGTGGTTGGGGCGTCAC-3') that contained (in addition to G-quadruplex) a hinge region connected to six pairs of complementary bases (duplex region). In this study, we compared properties of RE31 aptamer and its analogues containing varying number of bases in the duplex region and nucleotide insertions in the hinge region. Reduction in the number of nucleotides in the duplex region by 1 to 4 pairs (in comparison with RE31 aptamer) resulted in the decrease of the structural stability of aptamers (manifested as lower melting temperatures) and their ability to inhibit thrombin-stimulated fibrin formation in human blood plasma in tests of thrombin, prothrombin, and activated partial thromboplastin times. However, an increase in the number of bases by 1 to 2 pairs did not cause significant changes in the stability and antithrombin activity of the aptamers. Insertions into the hinge region of RE31 aptamer decreased its antithrombin activity. Investigation of RE31 antithrombotic properties demonstrated that RE31 (i) slowed down thrombin formation in human blood plasma (thrombin generation test), (ii) accelerated lysis of fibrin clot by tissue plasminogen activator in in vitro model, and (iii) suppressed arterial thrombosis in in vivo model. Based on the obtained data, RE31 aptamer can be considered as a potentially effective antithrombotic compound.

AIM: To present the results of fast track rehabilitation after anatomical lung resection. MATERIAL AND METHODS: Single-center prospective non-randomized trial has included patients for the period December 2014 - December 2016. Conventional protocol was applied in 124 patients, 58 patients after atypical lung resections or pneumonectomy were excluded from the study. Thus, there were 66 patients aged 61 (51; 67) years. Men/women ratio was 37:29. Lobectomy (n=55) and segmentectomy (n=11) were performed for lung cancer, metastatic injury and various inflammatory diseases in 53 (80.3%), 8 (12.1%) and 5 (7.6 %) cases, respectively. ASA risk score was II (16), III (46), IV (4). Video-assisted/open procedures ratio was 42 (63.6%) / 24 (36.4%). RESULTS: 30-day postoperative morbidity was 7.6% (5 out of 66 patients, 95% CI 3.3- 16.5). Pleural drainage tube was removed within the 1st postoperative day in 49 (74.2%) out of 66 patients. Prolonged insufficient aerostasis was observed in 3 patients followed by effective conservative treatment. Overall mortality was 3% (n=2, 95% CI 0.8- 10.4) due to pulmonary embolism and sudden cardiac death. Median of postoperative hospital-stay was 7 (6; 9) days without significant differences between groups of lobectomy and segmentectomy (p>0.05). CONCLUSION: Fast track rehabilitation protocol in thoracic surgery is safe and effective. Further studies are needed to justify early rehabilitation in high risk patients.

AIM: To optimize fluid therapy in transhiatal eshophagectomy by using of goal-oriented infusion therapy based on stroke volume variation. MATERIAL AND METHODS: Our trial enrolled 30 patients who underwent transhiatal esophagectomy followed by repair for the period 2011-2014. Patients were divided into 2 groups. The first group (LT) included 16 patients with liberal fluid therapy. The second group (GDT) consisted of 14 patients in whom goal-oriented fluid therapy was performed. Goal-oriented fluid therapy was implemented via stroke volume variation (SVV). RESULTS: Infusion rate was 6.7 ml/kg/h and 11.5 ml/kg/h in the main and control groups, respectively. Morbidity rate was 28.6% (n=4) and 62.5% (n=10) in the main and control groups respectively. Clavien-Dindo IV complications were lung atelectasis (n=2, 14%), pneumonia (n=1, 7%). Hydrothorax required puncture was noted in 1 (7%) case. Acute respiratory failure as complication IVa was in 1 (9%) patient. In the control group complications were registered in 10 (62.5%) patients. Complications I-II degree included lung atelectasis (n=4, 25%), cervical anastomosis failure (n=1, 6%); complications IVa were observed in 8 cases (50%). It was significant respiratory failure with reduced PO2/FiO2<300. Patients of the main group required less time for postoperative mechanical ventilation (120 [90-300] vs. 315 [215-810] min (p=0.02) and ICU-stay (0.83 [0.7-0.8] vs. 1.75 [1.25-2.75] (p=0.0022).

RESUMO

The capability of memantine, a noncompetitive antagonist of the NMDA receptors, to prevent impairments of cognitive functions in rats was investigated in the lithium-pilocarpine model of epilepsy. After status epilepticus, rats exhibited impaired exploratory behavior and spatial memory, and a decline of extinction of orienting behavior. Memantine administration prevented these disturbances. Thus, the blockade of the NMDA receptors immediately after status epilepticus allowed prevention of the development of the possible cognitive impairments.

This communication presents synthesis and spectral characterization of metalloporphyrin [Zn(X)TMPyP4] (TMPyP4 is 5,10,15,20-tetrakis (N-methylpyridinium-4-yl)porphyrin), and studies its binding onto anionic surface sites of synthetic double stranded polynucleotide Poly(A)-Poly(U). [Zn(X)TMPyP4] binding with Poly(A)-Poly(U) was monitored by UV-Vis absorbance spectroscopy, two fluorescence spectroscopies and 1H NMR in a working aqueous medium of 0.15M ionic strength, pH7.0 and at 25°C. The evidence provided by spectroscopic measurements and multivariate data analysis suggests the use of this metalloporphyrin as a probe for investigation of the polynucleotide surface. In contrast to TMPyP4 intercalation, an outside adsorption of [Zn(X)TMPyP4] induces an attenuation of luminescence intensity and has little influence on the shape of luminescence band. Special attention was paid to the quantitative description of the interaction between neighboring ligands on the Poly(A)-Poly(U) surface. The intrinsic binding constant to an isolated binding site lgKin 5.8±0.1, the cooperativity parameter ω 1.8±0.2, and number of monomers occupied by a ligand n=2 (25°C; pH7.0) were calculated based upon the recently proposed non-linear least-squares fitting procedure. The discovered cooperativity of binding of [Zn(X)TMPyP4] metalloporphyrin to Poly(A)-Poly(U) is significantly lower as compared to free porphyrin TMPyP4, reflecting minimal mutual influence between the nearest neighboring ligands bound with functional PO4(-) groups of the polynucleotide surface.

BACKGROUND: Rejection from prolonged mechanical ventilation with conversion to support ventilation modes during the surgery and subsequent immediate extubation at the end of the surgery could be the method ofprophylaxis ofpostoperative respiratory complications. THE AIM: To improve the results of surgical treatment of patients with infrarenal aorta injury due to the development and implementation to the anesthetic management complex the modes of support ventilation. MATERIALS AND METHODS: 2-staged clinical trial on patients undergoing surgery on infrarenal aorta was conducted. At the 1st stage patients were assessed for opportunity of immediate or early extubation and support ventilation initiation. At the 2nd stage support modes during the intraoperative ventilation were introducted into clinical praxis. RESULTS: Based on received data we concluded that not everyone patient needed intra- and postoperative mechanical ventilation in spite ofprolonged duration of the surgery and large surgical trauma. Inclusion in the protocol of anesthesia support ventilation strategy decreases requirement ofparalytic agents and their side effects. CONCLUSION: Applying the support ventilation modes during the anesthesia gives a chance of immediate and early extubation after the surgery and decreases the number of critical events and respiratory complications.

Cognitive impairment in six-week -old rats has been studied in the lithium-pilocarpine model of adolescent temporal lobe epilepsy in humans. The pilocarpine-treated rats (n =21) exhibited (a) a decreased exploratory activity in comparison with control rats (n = 20) in the open field (OP) test and (b) a slower extinction of exploratory behavior in repeated OP tests. The Morris Water Maze (MWM) test showed that the effect of training was less pronounced in the pilocarpine-treated rats, which demonstrated disruption of predominantly short-term memory. Therefore, our study has shown that lithium-pilocarpine seizures induce substantial changes in exploratory behavior and spatial memory in adolescent rats. OP and MWM tests can be used in the search of drugs reducing cognitive impairments associated with temporal lobe epilepsy.

The aim of this study was to discover the effects of chronic intraperitoneal administration of proinflammatory cytokine interleukin-1ß (IL-1ß) on rat investigative behavior and spatial memory. Rats were injected with a moderate pyrogenic dose of IL-1ß (0.5 mkg/kg) daily during 14 days (7 days before tests and 7 testing days). The behavior was examined in 23.5 hours after the previous injection of cytokine. The test battery included: "The open field" (within 3 consecutive days), "The exploration of novel objects", and "The Morris water maze". The animals treated with IL-1ß differed from the control animals in an essential decrease of locomotor activity, slight increase of anxiety and suppression of exploratory behavior. The impairment of spatial memory was not revealed.

Features of the existential organization of a bioelectric activity of a brain at posterity from rats with a lateral injury of the brain were investigated. The EEG analysis revealed in one month animals significantly increased the proportion of advanced and lagging decrease in the activity of the right frontal derivation towards the right occipital region. Change between structural ties in terms of the rats were correlated with the severity of behavioral disorders in the test "open field" (OF). The most significant change in the existential organization of a bioelectrical activity (the reduction of synchronous communications between the frontal derivations) recorded in rats with severe conduct disorder compared with animals of the same groups, in which the behavior was not significantly different from the norm. The violation of the behavior in the test OF was accompanied by a decrease in synchronous activity between the left frontal and right occipital region or the decline in the anticipatory activity in the right frontal region in relation to the right occipital region. By maintaining the integrity of the behavior of these parameters were increased in comparison with an intact control.

RESUMO

The effectiveness of the use of a stabilized composition of alpha 2-macroglobulin concentrate obtained from fraction III of plasma proteins according to Cohn in burn disease in the experiment on guinea pigs was studied. Under the influence of intraabdominal administration for 4 days of alpha 2-macroglobulin concentrate with a stabilizer, healing of the burn wounds, normalization of a state of the animals occurred 2 times more rapidly than in control ones, activity of acid proteinases restored by day 5 (in control--by day 14) due to increase in activity of proteolytic enzyme inhibitors.

RESUMO

The technique of dermotension, peculiarities and expediency of its use in postburn and mechanical skin deformation in 50 children ranging in age from 2 to 14 years are described. It is expedient to perform operative elimination of the postburn alopecia, severe contracture of the neck, cicatricial bandage of the chest and abdomen by means of the dermotensive flaps. This permits to reduce a number of operative steps, create a sufficient reserve of healthy tissue identical to the lost one both in texture and in colour. Dermotension has the advantages over the pediculate plasty and can be widely used in children with the burn sequelae.

RESUMO

The experience with performing operations in 17 patients with contracture of the neck, face, trunk with the use of composite cutaneous arterialized flaps from the lateral surface of the thoracic wall and inclusion of the latissimus dorsi has been analysed. It is established that such a plasty is attended by the lesser risk of circulatory impairement in the transferred tissues as compared with the available methods of non-free plasty.

In patients with pathological scars, especially in children, the activity of proteolytic enzymes (prekallikrein, acid and neutral proteinases) is increased. Summary inhibitory activity and content of the alpha 2-macroglobulin in the serum and scar tissue is also increased. This is indicative of continued inflammation.

RESUMO

Biochemical and electron microscopy study of the collagen molecule from 14 biopsy specimens of the postburn scar (in 9 patients before and in 5 after cryoeffects) revealed that the postburn scar contains I, II, III and VIII collagen types changed according to the amino acid composition. It was found that after cryogenic effects on the scar there was a tendency to normalization of the collagen structure.
