RESUMO

Activities of ectophosphatases (alkaline phosphatase, 5'-nucleotidase and ecto-ATPase) from different subpopulation of lymphoma NKLy and ovary tumor of rats were investigated. A strong positive correlation has been shown between these activities in different subpopulations. A negative correlation between proliferative activity and activities of ectophosphatases was determined. The role of ectophosphatases in cell proliferation is discussed.

RESUMO

A study was made of the antioxidative activity (AOA), of the activity of glytathionreductase and catalase, and of SH-group concentration in suspensions of intact cells from eight fractions of the rat ascitic ovary tumor. A correlative bond was detected between AOA, the activity of antioxidative enzymes, and SH-group concentration. It was shown that antioxidative properties of the fractions containing resting (G0) and polyploid cells were higher than those of proliferating diploid cells.

RESUMO

Studies have been made on the activity of erythrocyte acetylcholinesterase in 18 species of wild and laboratory rodents. Significant differences in the enzymic activity unrelated to taxonomic position of rodents were revealed. Negative correlation was found between the enzymic activity and body mass in rodents. The relationship between these parameters for wild rodents is approximated by an allometric equation Y = 1.1 . X-0.73, for laboratory species--Y = 1.5 . X-0.41. It is suggested that the revealed relationship indicates the existence of a connection between the activity of erythrocyte acetylcholinesterase and the intensity of metabolism in small rodents.

RESUMO

The NKLy ascitic tumor cells in the stationary phase of growth were fractionated by velocity sedimentation method. Cells from the obtained fractions were characterized by measurements of DNA contents and 3H-thymidine incorporation. The surface properties of the cells from five fractions, differing in proliferative capacity, stage of the cell cycle and ploidy were considered using cell electrophoresis, two polymer aqueous phase system and Alcian blue sorption. A correlation between electrophoretic mobility and cell partition constant for these fractions has been obtained. No correlation was found between these parameters and dye absorption. The surface charge of the cells from G0/G1 and S fractions was higher than that of other cells. The polyploid NKLy cells demonstrated a lower surface charge. The surface properties of the tumor cells differing in proliferative capacity, stage of the cell cycle and ploidy are discussed.

RESUMO

A study was made of sensitivity of NKLy tumor fractions, differing in the proliferative activity, position in the cell cycle, and ploidy, to membrane-toxic action of natural killer cells from mouse spleen. The membrane toxicity index for cells of three out of five fractions under study was shown to decrease after exposure to 4 Gy radiation. It is concluded that there is no correlation between NKLy cell sensitivity to membrane-toxic effect of natural killer cells and their radiosensitivity as well as their ability to repair potentially lethal damages.

RESUMO

A study of membrane toxicity of allogenic splenocytes with regard to irradiated mouse NKLy lymphosarcoma cells (tested by the release of 3H-uridine label from target cells in the presence of exogenous RNAses) has displayed an insignificant increase, as compared with the control, in the membrane toxicity with radiation doses of 1-2 Gy and its decrease at higher doses.

RESUMO

The peripheral blood leukocytes in chronic myeloid leukaemia (CML) patients and healthy donors are separated on ficoll-verografin one-step gradients with 1.077 g/ml density. It is shown that 95-98% of donor granulocytes had the density above 1.077 g/ml. Granulocytes of CML patients consisted of two populations having the density above and below 1.077 g/ml (high density granulocytes-HDG, low density granulocytes-LDG). The electrophoretic mobility (EPM) of LDG is 8-34% higher than that of HDG. As a result of EPM definition of granulocytes affected by hyaluronidase, neuraminidase and RNase it is shown that the high EPM of LDG is due to an increase in the density of sialic acids on their surface.

RESUMO

The sedimentation fractionation of ascitic form of Plyss lymphosarcoma was performed in the linear 1-3% sucrose gradient with 1 g. Fractions were different in cell sizes, protein content and also in intensity of the 3H-thymidine incorporation. Fractions which contained cells in the G1, G1 + S and S + G2 phases were transplanted in vivo in a solid form. Cells with the low sedimentation rate had the maximum growth capacity. The growth rate of G1 + S cells was similar to that of the initial tumour. Cells of high-rate sedimentation were characterized by the low growth activity. The values of D0 for these 3 fractions were 1.64, 2.05 and 2.11 Gy, and those of n 1.9, 2.9 and 1.6, respectively. These differences were not a result of selective injury of cells in fractions or their contamination by nontumour cells.

RESUMO

The relation was studied between the density of the electrical charge of the surface (DECS) and the karyological structure of the rat ovary tumour (OT) cells. For this purpose the sedimentation rate and electrical mobility of cells from two OT clones were measured. The OT1 and OT2 clones differed in their modal chromosome numbers (44 and 80-90, respectively). An increase in DECS by 4-5% on the average occurs in cells with the chromosome set close to the tetraploid one. The further increase in the chromosome number to the octaploid one does not change DECS. The results obtained show stability of DECS with significant changes in the structure of the OT cells karyotype.

RESUMO

Irradiation of rats with a dose of 7 Gy increases the density of electrical change of bone marrow cells by (8-15) X 10(3) elementary charge X micron-2 which makes up 12-26% of original density. The maximum effect was registered 30 min after irradiation. After 3 days the density of electrical charge decreased to reach the original values. In the peripheral blood erythrocytes, marked changes in the charge were only detected in 3 days when the charge decreased as compared to the control.

RESUMO

The surface changes of the rat bone marrow cells was measured. For identification of cells, differences in sedimentational characteristics were used. With this purpose the velosity of sedimentation and electrophoretic mobility of cells were measured simultaneously. The decrease in the surface charge of myeloid cells was shown in the media with lowered ionic force.

RESUMO

The electrophoretic mobility of the ascitic tumor cells with different rates of sedimentation was measured by photographic registration of summarizing traces in the electrophoretic chamber. Changes in electrophoretic mobility of these cells during tumour growth are described. A conclusion is made that these changes are not connected with the proliferative activity of the tumour. It was shown that cells with low densities appeared in the late period of the tumor growth.

RESUMO

The described method of analysis of cell populations' heterogeneity involves two parameters simultaneously: the sedimentation velocity and the electrophoretic mobility of cells. The method is based on the photographic registrarion of resulting traces of cell velocity in the fields of gravition and electricity with consequence of the elements of these traces on the photographs. The experimental data on the distribution of these parameters in the ascitic tumor cell population are given.

RESUMO

Changes of specific resistance and electrophoretic mobility of the cells from regenerative liver of rats are investigated. It is shown that specific resistance decreases and electrophoretic mobility increases during 30 h after hepathectomy. Exogenous ribonuclease decreases electrophoretic mobility by 10% and does not reduce significantly specific resistance. Neuraminidase treatment caused a marked increase (approximately 40%) of specific resistance without reduction in the mobility of cells from both intact and regenerative liver. It is concluded that there is no difference in the sensitivity of the cell surface of normal and regenerative liver to ribonuclease and neuraminidase.

Ascite cells were treated with naphthenic acid preparation, containing a mixture of naphthenic acids with average molecular weight equal to 240, and with a fraction of this preparation differing in the boiling temperature. A 5-10(-7)M and higher concentrations of the preparation and its fraction brought about an increased permeability in ascite cells for K-ions. The effectiveness of naphthenic acids is associated with their hydrophobic properties, which was characterized in the present communication by the value of hydrophilic-lipophilic balance calculated according to the Davies formula.
