There are contradictory data concerning the influence of mesenchymal stromal cells (MSC) on immunoglobulin (Ig) production. Most of them were obtained using MSC from bone marrow. Properties of MSC from other tissues are elusive. In the present work MSC cultures were derived from umbilical cord, adipose tissue, and bone marrow of healthy donors, as well as from bone marrow of patients with autoimmune diseases. MSC from all these sources had similar surface markers phenotype. The influence of co-cultivation with MSC at exponential or stationary phase on IgM and IgE content in Namalva and U266 cells was evaluated. MSC from bone marrow of healthy donors had no effect on IgM and IgE production. Proliferating MSC obtained from patients with Crohn's disease and multiple sclerosis stimulated Ig production. Exponentially growing MSC derived from umbilical cord and adipose tissue also stimulated Ig synthesis. MSC at stationary cultures amplified IgM production in Namalva cells and suppressed IgE synthesis in U266. Thus, MSC with similar phenotype but derived from different sources differ in their capacity to modulate Ig production in B-lymphoid cells. The effect of MSC depends on their growth stage and may differ for lymphoblastoid and myeloma cells.

RESUMO

This paper examines the structural and mechanical properties of composite materials based on chitosan and micro- and nanoparticles of Na-montmorillonite and possibility of application for cultivation and targeted delivery of mesenchymal stem cells and regenerative cells. It's have been shown by addition of Na-montmorillonite biomaterial acquires stability of structural and mechanical properties in the sterilization process the handling of liquid media in cell culture. In vitro studies using dermal fibroblasts and adipose tissue mesenchymal stem cells demonstrated that this material has a set of properties to ensure matrix biocompatibility.

Telomeres are the ends of the chromosomes and represent repeated DNA sequence and protein complex. Telomeres shorten with each cell division, which limits proliferative potential of cells. There is a great progress in clinical application of telomere length now. Additional characteristic of stem cell proliferation activity estimated by telomere length can be an informative indicator of transplant quality. In this work, we analyzed 14 cord blood samples by flow-FISH, ELISA and gematologycal analisator, and also the number of CD34 + CD45dim cells. Average telomere length of leukocytes fraction CB cells was 20.4-4.9 % respectively the control cells 1301 (T-cell lymphoblastic leukemia).

The last years witnessed significant change in therapeutic and infectious pathology of man, its immunoreactivity changed, especially in elderly patients. Thus, older people show the increase of chronic infectious-inflammatory diseases, characterized by flaccid, constantly recurring course, torpid to adequate etiotopic therapy. They are caused by conventionally-pathogenic or opportunistic microbes, often with atypical biological properties, having multiple resistance to antibiotics. About 30 new pathogenic microorganisms were revealed for the last 20-30 years, including HIV-infection agent. The number of patients with allergic and autoimmune diseases increased, thus resulting in high urgency of this problem in gerontology. Decrease of immunologic reactivity, observed practically in all parts of the world, but mostly manifested in Russia, is one of the causes of infectious pathology, including pathology of inner organs, manifestation of allergic and autoimmune diseases. General practitioners, gerontologists understand that to cope with the increased rate of infectious, allergic or autoimmune diseases only by antibiotics or anti-inflammatory medicines is next to impossible. It's clear now that the use of only chemotherapeutic drugs in treatment of elderly and therapeutic patients is a mistake. Immunocorrective medicines, which stimulate reparative processes, allow to accelerate the processes of involution and shorten the general period of treatment. Antibiotic inhibits the agent multiplication, but its complete elimination from the body is a result of immune factors activity. In elderly patients with inner organ pathology and chronic pathology on the background of the suppressed immunoreactivity the effect of antibiotics, as well as antifungal, antiviral and other chemotherapeutic medicines is ineffective or of low effect. Optimal clinical effect in elderly patients can obviously be achieved only if there is synergism in the activity of the body protective forces and antimicrobial drugs. Thus, immunoreactivity dysfunction has a significant part in the pathogenesis of inner organ diseases in elderly patients with the disturbance of immunological status. So in complex of medical means, indicated for this pathology, adequate immunotherapy plays a special role, and the main problem is the choice of particular drugs.

We studied gastric secretion in submaximal and maximal histamine and pentagastrin tests, computer-assisted pH-metry, chromogastroscopy and chromoduodenoscopy in 205 patients with duodenal ulcer (DU) and diabetes mellitus (DM). DU patients with DM type I had low acid gastric secretion in the stimulated phase, an excitable type of gastric secretion, continuous acid-formation of low intensity, subcompensated acid-neutralizing function of the antral portion of the stomach, normoacidic state. DU patients with DM type II had low fasting acid gastric secretion, an inhibitory type of gastric secretion, normal acid-formation, compensated acid-neutralizing function of the antral portion of the stomach, hypoacidic state.

RESUMO

Actovegin, a deproteinized hemoderivative, was used to correct bioenergetic defects of duodenal mucosa, to reduce inflammatory-cell infiltration, to stimulate blood supply of patients with duodenal ulcer associated with ischemic heart disease or diabetes mellitus. As shown by the results obtained in 194 patients (119 males and 75 females), adjuvant actovegin in patients with duodenal ulcer associated with ischemic heart disease and diabetes mellitus stimulates the ulcer healing, prolongs the recurrence-free interval. It can be used both in outpatient departments and specialized hospitals.

RESUMO

Long-term scarring and frequently recurrent ulcers are often found in patients with concomitant diseases such as ischemic heart disease or diabetes mellitus. Laser therapy may be a good alternative in such patients. Gastroduodenal ulcer sites were exposed to laser radiation (He-Ne and AG lasers ATOLL, VITA-01) in 182 patients. The laser treatment was indicated in the failure of a 3-month previous therapy or resistance to antisecretory and antibacterial drugs. Laser treatment is recommended to perform in specialized gastroenterological clinics for better selection of patients, especially those with gastroduodenal ulcer combined with ischemic heart disease and/or diabetes mellitus.

RESUMO

The authors studied the peculiarities of the course taken by experimental gastric ulcer in rabbits under the effect of the norepinephrine precursor 1-dioxyphenylalanine (L-DOPA) injected parenterally in a dose of 10 mg/kg twice a day for 14 days. As compared to the controls (nontreated ulcer), L-DOPA restored significantly more rapidly the bioenergetic processes in the gastric mucosa which were disturbed as the result of ulcerogenesis, provided for stimulation of reparative regeneration, and reduced the terms of ulcer cicatrization.
