RESUMO

Efficiency and diagnostic adequacy of immunoglobulin isolation from rabbit immune serum by reiterated salting out with ammonium sulfate and subsequent separation of DEAE sephadex A-50 was evaluated. Electrophoresis and immunoelectrophoresis in agar gel demonstrated that the resultant immunoglobulin fraction is much more pure than after one salting out. It contains nothing more than antibodies to influenza virus taken for immunization, and after binding to the stain shows high fluorescent activity and induces but a moderate background nonspecific fluorescence in the subsequent preparations. In clinical trials of conjugates based on the new combined method of purification, influenza A or B could be correctly diagnosed in 97-98% cases.

RESUMO

Effects of 0.1-10% ethanol on creatine kinase activity and water-soluble protein amount were studied in extracts of 2 day-old culture of chick embryo myoblasts. Low concentrations of the toxicant stimulated the total creatine kinase activity of the culture whereas middle concentrations exerted no influence, and the highest concentration (10%) oppressed the enzymatic activity. The increase of protein content in extracts was observed when ethanol concentration in the incubation medium was 0.1%. Within the range of 1-5% ethanol the protein content sharply decreased at 10% concentration it did not differs from the control value. The treatment of 5 day-old culture with 1-5% ethanol did not influence either creatine kinase activity or protein content in myoblasts. The obtained data are discussed in the context of chemical hormesis scheme and in terms of unmasked changes involvement in the adaptive reactions of the cell system.

RESUMO

The influence of green-yellow light of copper laser (mean power of impulse-periodic emanation 3 W, working density of power in experiment is 200 mW/cm2) on biochemical indices of chick embryo myoblasts and neuroblasts in primary culture was studied. The content of water extracted protein was found to increase progressively, the total and specific activities of creatine kinase elevated considerably in both the cell cultures under irradiation with the total doses equal to 3 and 15 J. However, with dose of 30 J these parameters corresponded to the level characteristic of nonirradiated cultures. Mechanisms of stimulating action of highly intensive laser emanation on cell functions are discussed.

RESUMO

The effects of exogenous ganglioside GM1 (1 microM) from bovine brain on the morphological state and biochemical parameters (creatine kinase, acetylcholinesterase and adenylate cyclase activities as well as the protein, phospholipid and ganglioside content) have been studied in primary cultures of trypsin-treated dissociated cells of chicken embryonic brain. Ganglioside GM1 accelerated the growth and differentiation of cultured cells, increased the phospho- and glycolipid content and stimulated the activity of the enzymes.

RESUMO

The reaction of chicken myoblasts in competent primary culture during their contact with activated rat neutrophils was evaluated. In the incubation medium dynamics of activities were defined for proteinases and creatine kinase, protein content, as well as for DNA and protein marked precursor increase. An increase in myoblast membrane permeability has been revealed. It is shown that the culture is a sensitive model for investigation of physiological interaction of neutrophils with muscle tissue cells.

RESUMO

Effects of copper laser stimulating doses on biochemical parameters of competent chicken embryo myoblast and neuroblast cultures were studied. It was shown that in both systems biostimulation was characterized by an increase of water-extractable protein level and a considerable rise of total and specific creatine kinase activity. The authors believe that cases of protein drop during myoblast culture exposure to chemical agents described in literature were caused by these agents overdosage and that this drop may be considered as a manifestation of a peculiar biochemical shock induced by overdosage. They suggest to follow up the time course of water-extractable protein levels in the cultures to differentiate between trophic and injurious loading.

RESUMO

A study was made of the influence of creatine phosphate (CP) on creatine kinase activity and the total protein status of the primary chicken myoblast culture. It has been established that the two-hour exposure of the developing culture in the presence of 2 and 4 mM CP is accompanied by the lowering of the content of water extractable protein and a considerable increase of creatine kinase activity. Meanwhile the total activity of the enzyme rises in the non-differentiated and falls in the differentiated one. Electrophoretic analysis has shown CP to produce not only quantitative but also deep qualitative changes in the myoblast protein spectrum. It is assumed that the changes revealed are nonspecific.

RESUMO

Different methods for the preparation of antibodies to rabbit FITC-labeled MM creatine kinase are compared. Highly specific fluorescent antibodies have been employed to detect MM creatine kinase in isolated cells and in tissue culture. Prospects for research and clinical diagnostic application of the direct method for the detection of creatine kinase in biological samples are discussed.

RESUMO

Dynamics of creatine kinase activity was studied in heart muscle, liver tissue, lymphatic glands, intestine and spleen of guinea pigs infected with pseudotuberculosis microbes. The maximal increase in creatine kinase activity was observed in lymphatic glands and in heart muscle within the first day after the pseudotuberculosis infection. The enzymatic activity increase in liver tissue occurred within the fifth day, while in spleen--within twelfth day after pseudotuberculosis infection. The data obtained were considered in correlation with the clinical manifestations of experimental pseudotuberculosis.

The recovery of the acetylcholine esterase (AChE) activity after the irreversible inhibition with an organophosphorus inhibitor B-156 was studied in a developing monolayer culture of chick myoblasts. The culture was obtained from muscles of posterior limbs of the 11 day old chick embryos. The AChE activity was estimated by the modified Ellman method from the moment of inoculation to the stage of spontaneous contractions of muscle fibres. After the B-156 treatment the AChE activity of muscle cells decreased, then started to increase and the maximum recovery of activity, below the initial level, was attained within roughly 2 days after the treatment. The AChE activity in the treated culture somewhat decreased thereafter. The lower the inhibitor concentration, i.e. the lower the value of the initial AChE inhibition, the higher the starting rate and degree of recovery of the AChE activity. The results obtained suggest that, unlike the multilayer culture of muscle tissue at later stages of differentiation no compensatory enhancement of AChE biosynthesis after irreversible inhibition of this enzyme by an organophosphorus inhibitor is observed in the monolayer culture of chick myoblasts at the early stages of myogenesis.

RESUMO

The authors described and analyzed a case of Alzheimer's disease in young age, which eventuated in the death of the patient at 36 years. Major manifestations o the atypical course and syndrome formation are outlined. Data provided by paraclinical studies including computerized tomography are also presented. The clinical diagnosis was confirmed by autopsy findings.

Creatine action on the activity of creatine kinase (ATP: creatine-phosphotransferase; EC 2.7.3.2) and the content of water-soluble proteins in the developing monolayer culture of chick myoblasts are studied. Creatine at concentrations of 1.9-10- minus 3-3.8-10- minus 3 M is shown to increase reliably the creatine kinase activity by 1,1--2,9 times and to reduct considerably the content of water-soluble proteins. Lower concentrations of creatine (3.8-10- minus 5 M) also increased the creatine kinase activity but did not change the contents of water-soluble proteins. The creatine effect was maximal at the period preceding the termination of tissue cells differentiation. In the course of the combined effect of both actinomycin D (50 mcg/plate) and creatine (3.8-10- minus 3 M) the creatine kinase activity was much higher than that in the presence of actinomycin D alone which considerably reduced the enzyme activity as well as the contents of water-soluble proteins.
