Stream restoration projects undertaken as compensatory mitigation pursuant to Sect. 404 of the U.S. Clean Water Act must be evaluated using ecological performance standards that are objective and verifiable and based on the best available science that can be measured or assessed in a practicable manner. While performance standards for physical stream conditions are common, evaluating biological conditions following stream restoration activities has proven more problematic. We developed a macroinvertebrate multimetric index for headwater streams in three Southeastern Plains subecoregions (65 g, 65 h, and 65 l) of Georgia using 76 sites sampled in 2019. An abiotic disturbance gradient based on principal components analysis of instream habitat, physicochemical, and land use variables was employed to assign condition classes (good, fair, poor) among sites within each subecoregion. We identified genus-level macroinvertebrate richness and proportional richness of traits-based metrics (habit and functional feeding groups) that demonstrated high discriminatory power between good and poor abiotic conditions and response to individual stressors. Subecoregion-specific metrics were then standardized and aggregated to develop the final index and biological reference curves. These biological reference curves represent a continuum of relevant regional conditions against which a stream enhancement or restoration project may be assessed relative to other streams throughout the region and allow for the award of mitigation credit, if applicable, to be based directly on the relative improvement of biological conditions. These biological performance standards will supplement other performance standards (hydrologic and geomorphic measures) necessary to evaluate the effectiveness of stream restoration projects in the study area.

BACKGROUND: Although local inhibition of the generation or actions of endothelin-1 has been shown to cause forearm vasodilatation, the systemic effects of endothelin receptor blockade in healthy humans are unknown. We therefore investigated the cardiovascular effects of a potent peptide endothelin ETA/B receptor antagonist, TAK-044, in healthy men. METHODS AND RESULTS: Two randomized, placebo-controlled, crossover studies were performed. In nine subjects, TAK-044 (10 to 1000 mg IV over a 15-minute period) caused sustained dose-dependent peripheral vasodilatation and hypotension. Four hours after infusion of the highest dose (1000 mg), there were decreases in mean arterial pressure of 18 mm Hg and total peripheral resistance of 665 AU and increases in heart rate of 8 bpm and cardiac index of 0.9 L x min(-1) x m(-2) compared with placebo. TAK-044 caused a rapid, dose-dependent increase in plasma immunoreactive endothelin (from 3.3 to 35.7 pg/mL within 30 minutes after 1000 mg). In a second study in eight subjects, intravenous administration of TAK-044 at doses of 30, 250, and 750 mg also caused peripheral vasodilatation, and all three doses abolished local forearm vasoconstriction to brachial artery infusion of endothelin-1. Brachial artery infusion of TAK-044 caused local forearm vasodilation. CONCLUSIONS: The endothelin ETA/B receptor antagonist TAK-044 decreases peripheral vascular resistance and, to a lesser extent, blood pressure; increases circulating endothelin concentrations; and blocks forearm vasoconstriction to exogenous endothelin-1. These results suggest that endogenous generation of endothelin-1 plays a fundamental physiological role in maintenance of peripheral vascular tone and blood pressure. The vasodilator properties of endothelin receptor antagonists may prove valuable therapeutically.

We have cloned a Na,K-ATPase alpha-subunit gene from Caenorhabditis elegans and discovered that it is identical to the gene eat-6, eat-6 mutations cause feeble contractions and slow, delayed relaxations of pharyngeal muscle. The resting membrane potential of eat-6 mutant pharynxes is consistently depolarized compared to wild-type. The action potentials are smaller, and the return to resting potential is slower. To explain these abnormalities, we propose that a reduction of Na,K-ATPase activity in eat-6 mutants leads to a reduction of the ion concentration gradients that power membrane potential changes.

A survey was conducted to establish the prevalence of Actinobacillus seminis in ovine semen and the possible importance of the organism as a cause of ram infertility. A seminis was isolated from three of 16 infertile rams and the infected animals had lesions of the genital tract and produced poor quality semen. A seminis was also isolated from two of 96 fertile rams used as donors of semen for artificial insemination programmes. The strains of A seminis could be identified on the basis of their API ZYM profiles. The results suggest that in the United Kingdom A seminis should be considered as a common cause of infertility in rams.

RESUMO

This experimental evaluation of Cybex II isokinetic measurement was based on 70 normal subjects and 120 patients with chronic low back pain. It considered: reliability and learning effect; discrimination of individual patients versus normal subjects; relationship to clinical measures; assessment of effort; and a prospective 2-year follow-up of normal subjects to predict future low back pain.

The aim of this study was to investigate physical impairment in patients with chronic low back pain, to develop a method of clinical evaluation suitable for routine use, and to consider the relationship between pain, disability, and physical impairment. Twenty-seven physical tests were investigated. Permanent anatomic and structural impairments of spinal deformities, spinal fractures, surgical scarring, and neurologic deficits were excluded as not relevant to the patient with low back pain in the absence of nerve root involvement or previous surgery. Three consecutive 20-patient reproducibility studies were used to develop reliable methods of examination for 23 of the tests. Only four tests were excluded as unreliable: sacral angle, pelvic tilt, and separate lumbar and pelvic extension, none of which are part of routine clinical examination or have any proven relationship to disability. The remaining 23 physical tests were evaluated in 70 asymptomatic subjects and 120 patients with chronic low back pain. Passive knee flexion, passive hip flexion, hip flexion strength, hip abduction strength, pain reproduction on each of these tests, and the prone extension strength test were excluded because they were too closely related to nonorganic and behavioral responses to examination. Eight tests successfully discriminated patients with low back pain from normal subjects and were significantly related to self-report disability in activities of daily living: pelvic flexion, total flexion, total extension, lateral flexion, straight leg raising, spinal tenderness, bilateral active straight leg raising, and sit-up. Factor analysis failed to demonstrate an underlying statistical dimension of physical impairment. However, an empirical combination of total flexion, total extension, average lateral flexion, average straight leg raising, spinal tenderness, bilateral active straight leg raising, and sit-up provided an equally satisfactory alternative. Simple cut-offs from normal subjects made the scale simple and quick to use. This final scale successfully discriminated 78% of patients and normal subjects and explained 25% of the variance of disability, with a specificity of 86% and sensitivity of 76%. This scale provides an objective clinical evaluation that meets the criteria for evaluating physical impairment, yet is simple, reliable, and suitable for routine clinical use. It should, however, be emphasized that all the tests included in the final scale are measures of current functional limitation rather than of permanent anatomic or structural impairment. This raises questions about the physical basis of permanent disability due to chronic low back pain.

The incidence and acquisition of Candida albicans and other yeasts in two wards of a skin hospital is described. Carriage rates on the skin in hospital patients is higher than is generally supposed, and cutaneous sites may act as sources of infection with these organisms.
