RESUMO

We present a lattice determination of the Λ parameter in three-flavor QCD and the strong coupling at the Z pole mass. Computing the nonperturbative running of the coupling in the range from 0.2 to 70 GeV, and using experimental input values for the masses and decay constants of the pion and the kaon, we obtain Λ_{MS[over ¯]}^{(3)}=341(12) MeV. The nonperturbative running up to very high energies guarantees that systematic effects associated with perturbation theory are well under control. Using the four-loop prediction for Λ_{MS[over ¯]}^{(5)}/Λ_{MS[over ¯]}^{(3)} yields α_{MS[over ¯]}^{(5)}(m_{Z})=0.11852(84).

RESUMO

Inflammatory immune activation has been frequently associated with the development of major depression. Microglia might serve as an important interface in this immune system-to-brain communication. Interleukin-4, the major Th2 type cytokine, might be protective against depression due to its ability to counter-regulate inflammation and to inhibit serotonin transporter activity. By using an Interferon-α mouse model, we show that a decreased IL-4 responsiveness of microglia was specifically related to the development of depressive-like behavior. IL-4 deficient mice in a BALB/cJ background showed a considerable increase of depressive-like behavior in the forced swim (FST) and tail suspension test (TST) and reduced avoidance behavior in an active avoidance task. Prior conditioning with unescapable foot shocks further decreased avoidance behavior (learned helplessness) but to a similar level as in the wild type strain. IFN-α treatment was not able to further enhance the already increased level of depressive-like behavior in the FST and TST. Thus, IL-4 seems to be a critical participant in the regulation of depressive-like behavior in an untreated baseline condition. Increase of depressive-like behavior during inflammation in wild-type mice might be mediated to some extent by a reduction of IL-4 signaling.

RESUMO

We discuss the determination of the strong coupling α_{MS[over ¯]}(m_{Z}) or, equivalently, the QCD Λ parameter. Its determination requires the use of perturbation theory in α_{s}(µ) in some scheme s and at some energy scale µ. The higher the scale µ, the more accurate perturbation theory becomes, owing to asymptotic freedom. As one step in our computation of the Λ parameter in three-flavor QCD, we perform lattice computations in a scheme that allows us to nonperturbatively reach very high energies, corresponding to α_{s}=0.1 and below. We find that (continuum) perturbation theory is very accurate there, yielding a 3% error in the Λ parameter, while data around α_{s}≈0.2 are clearly insufficient to quote such a precision. It is important to realize that these findings are expected to be generic, as our scheme has advantageous properties regarding the applicability of perturbation theory.

Maternal infection during pregnancy is a well-established risk factor for schizophrenia in the adult offspring. Consistently, prenatal Poly(I:C) treatment in mice has been validated to model behavioral and neurodevelopmental abnormalities associated with schizophrenia. By using the Poly(I:C) BALB/c mouse model, we investigated the functional profile of microglia by flow cytometry in relation to progressive behavioral changes from adolescence to adulthood. Prenatal Poly(I:C) treatment induced the expected sensory gating deficits (pre-pulse inhibition (PPI) of the acoustic startle response) in 100day-old adult offspring, but only in female not in male descendants. No PPI-deficits were present in 30day-old adolescent mice. Sensory gating deficits in adult females were preceded by a strong M1-type microglia polarization pattern during puberty as determined by flow cytometric analysis of multiple pro- and anti-inflammatory surface markers. Microglia activation in females did not persist until adulthood and was absent in behaviorally unaffected male descendants. Further, the specific activation pattern of microglia was not mirrored by a similar activation of peripheral immune cells. We conclude that prenatal Poly(I:C) treatment induces post pubertal deficits in sensory gating which are specifically preceded by a pro-inflammatory activation pattern of microglia during puberty.

RESUMO

A distinctive feature of the presence of spontaneous chiral symmetry breaking in QCD is the condensation of low modes of the Dirac operator near the origin. The rate of condensation must be equal to the slope of M(π)(2)F(π)(2)/2 with respect to the quark mass m in the chiral limit, where M(π) and F(π) are the mass and the decay constant of the Nambu-Goldstone bosons. We compute the spectral density of the (Hermitian) Dirac operator, the quark mass, the pseudoscalar meson mass, and decay constant by numerical simulations of lattice QCD with two light degenerate Wilson quarks. We use lattices generated by the Coordinated Lattice Simulation (CLS) group at three values of the lattice spacing in the range 0.05-0.08 fm, and for several quark masses corresponding to pseudoscalar mesons masses down to 190 MeV. Thanks to this coverage of parameters space, we can extrapolate all quantities to the chiral and continuum limits with confidence. The results show that the low quark modes do condense in the continuum as expected by the Banks-Casher mechanism, and the rate of condensation agrees with the Gell-Mann-Oakes-Renner relation. For the renormalization-group-invariant ratios we obtain [Σ(RGI)](1/3)/F=2.77(2)(4) and Λ(M̅S)/F=3.6(2), which correspond to [Σ(M̅S)(2 GeV)](1/3)=263(3)(4) MeV and F=85.8(7)(20) MeV if F(K) is used to set the scale by supplementing the theory with a quenched strange quark.

We present a factorization formula for the dependence of light hadron masses and low energy hadronic scales on the mass M of a heavy quark: apart from an overall mass-independent factor Q, ratios such as r_{0}(M)/r_{0}(0) are computable in perturbation theory at large M. The perturbation theory part is stable concerning different loop orders. Our nonperturbative Monte Carlo results obtained in a model calculation, where a doublet of heavy quarks is decoupled, match quantitatively to the perturbative prediction. Upon taking ratios of different hadronic scales at the same mass, the perturbative function drops out and the ratios are given by the decoupled theory up to M^{-2} corrections. We verify-in the continuum limit-that the sea quark effects of quarks with masses around the charm mass are very small in such ratios.

AIMS: To develop and implement an automated virtual slide screening system that distinguishes normal histological findings and several tissue--based crude (texture-based) diagnoses. THEORETICAL CONSIDERATIONS: Virtual slide technology has to handle and transfer images of GB Bytes in size. The performance of tissue based diagnosis can be separated into a) a sampling procedure to allocate the slide area containing the most significant diagnostic information, and b) the evaluation of the diagnosis obtained from the information present in the selected area. Nyquist's theorem that is broadly applied in acoustics, can also serve for quality assurance in image information analysis, especially to preset the accuracy of sampling. Texture-based diagnosis can be performed with recursive formulas that do not require a detailed segmentation procedure. The obtained results will then be transferred into a "self-learning" discrimination system that adjusts itself to changes of image parameters such as brightness, shading, or contrast. METHODS: Non-overlapping compartments of the original virtual slide (image) will be chosen at random and according to Nyquist's theorem (predefined error-rate). The compartments will be standardized by local filter operations, and are subject for texture analysis. The texture analysis is performed on the basis of a recursive formula that computes the median gray value and the local noise distribution. The computations will be performed at different magnifications that are adjusted to the most frequently used objectives (*2, *4.5, *10, *20, *40). The obtained data are statistically analyzed in a hierarchical sequence, and in relation to the clinical significance of the diagnosis. RESULTS: The system has been tested with a total of 896 lung cancer cases that include the diagnoses groups: cohort (1) normal lung--cancer; cancer subdivided: cohort (2) small cell lung cancer--non small cell lung cancer; non small cell lung cancer subdivided: cohort (3) squamous cell carcinoma--adenocarcinoma--large cell carcinoma. The system can classify all diagnoses of the cohorts (1) and (2) correctly in 100%, those of cohort (3) in more than 95%. The percentage of the selected area can be limited to only 10% of the original image without any increased error rate. CONCLUSION: The developed system is a fast and reliable procedure to fulfill all requirements for an automated "pre-screening" of virtual slides in lung pathology.

RESUMO

OBJECTIVE: To develop and implement an Internet-based, automated image measurement system for immunohistochemically stained slides including fluorescence images in online and off-line modes. STUDY DESIGN: An image analyzing system was developed that automatically measures digitized images obtained from immunohistochemically stained slides. It is divided into a common server platform and a specific image quantification system based upon DIAS (University of Jena). After registration, the user fills in an input data form and attaches images to be measured. The server periodically transfers the data to the measurement system. The measurement works on dynamic thresholding and active sampling of objects visualized by fluorescence and conventional chromogens. It includes stereologic algorithms, object quantification, syntactic structure analysis and quality assurance. RESULTS: The system has been tested for diaminobenzidene, alkaline phosphatase and fluorescence images (FITC, etc.). The reproducibility and stability of the system are > 98%. The series of successfully measured images comprises > 1,000 images in total in the online and off-line modes. CONCLUSION: An Internet-based automated image measurement system has been developed that offers worldwide access to the major requests for quantification of immunohistochemically stained slides-tissue array analysis, nuclear stains (MIB, hormones), membrane stains (CerbB2), vascularization and fluorescence in situ hybridization.
