RESUMO

This study investigated the effects of sericin on the testicular growth hormone (GH)/insulin-like growth factor-1 (IGF-1) axis in rats with type 2 diabetes mellitus. Forty rats were randomly assigned to normal control, type 2 diabetes mellitus, sericin and metformin treated groups. Type 2 diabetes was established by repeated intraperitoneal injection of streptozotocin, and identified by blood glucose ≥16.7 mmol/L at 1 week. The diabetic rats were given no other treatment, these rats in the sericin group were intragastrically perfused with 2.4 g/kg sericin and the metformin treated rats were intragastrically perfused with 55.33 mg/kg Metformin daily for 35 consecutive days. Enzyme-linked immunosorbent assays were used to determine serum testosterone, growth hormone and IGF-1 levels. Immunohistochemical staining, western blotting and reverse transcription-PCR were used to determine testicular growth hormone, growth hormone receptor and IGF-1 expression. The sericin significantly reduced serum growth hormone levels, downregulated growth hormone expression, increased serum testosterone and IGF-1 levels, and upregulated testicular growth hormone receptor and IGF-1 expression. Moreover, there were no significant differences in any of the parameters between the sericin and metformin treated groups. These findings indicated that sericin improved spermatogenic function through regulating the growth hormone/IGF-1 axis, thereby protecting reproductive function against diabetes-induced damage.

The aim of the present study was to investigate the effects of urantide on the expression status of C-reactive protein (CRP) and the inflammatory cytokines monocyte chemotactic protein (MCP)-1 and transforming growth factor (TGF)-ß in the aortas of rats with atherosclerosis (AS), and to identify its underlying mechanisms. The effects of urantide in a rat model of AS and in cultured rat vascular smooth muscle cells (VSMCs) were analyzed via hematoxylin and eosin staining, immunohistochemical staining and ELISA. The results in vivo demonstrated that urantide downregulated the expression of inflammatory mediators CRP and MCP-1 and upregulated the expression of TGF-ß. The results in vitro indicated that urantide inhibited the proliferation of VSMCs. In addition, urantide reduced the expression of CRP and downregulated the secretion of TGF-ß in the culture supernatant. In conclusion, urantide ameliorated the arterial inflammatory damage that was observed in the AS rat model at the cell and tissue levels by controlling the expression of CRP and the inflammatory cytokines MCP-1 and TGF-ß. Therefore, urantide may be a potential agent for the complementary treatment of AS.

OBJECTIVE: To observe the effects of color silk cocoon extraction-sericine on transforming growth factor-beta1 (TGF-beta1) and Smad3 protein expression in kidney of diabetic nephropathy (DN) rats. METHODS: 60 male SD rats were randomly divided into 5 groups (n = 12): normal control group, DN model group, sericine treatment group, metformin group and sericine prevention group. The rats in model group, sericine treatment group, metformin group and sericine prevention group were all established DN rats model by intraperitoneally injected streptozotocin (STZ). Blood glucose > or = 16.7 mmol/L was taken as standard to judge if the rats model were successfully established. After the rats model were successfully established, the rats in sericine treatment group were lavaged with sericine (2.4 g/(kg x d), 35 d). The rats in metformin group were lavaged with metformin (55.33 mg/(kg x d), 35 d). The rats in sericine prevention group were lavaged with the same dose sericine for 35 d before injecting STZ. The blood glucose and kidney weight/body weight of rats in each group were respectively detected. Immunohistochemical staining was used to observe the expression of TGF-beta1 and Western blot to detect the expression of Smad3 in kidney. RESULTS: Compared with normal control rats: the blood glucose, kidney weight/body weight, TGF-beta1 and Smad3 expression in kidney of rats in model group increased obviously (P < 0.01). The blood glucose, TGF-beta1 and Smad3 expression in kidney of rats in sericin treatment group, sericin prevention group and metformin group were significantly lower than that of model group (P < 0.01). Moreover, there were no obvious differences between sericin treatment group, sericin prevention group and metformin group (P > 0.05). The kidney weight/body weight of rats in sericin treatment group, sericin prevention group and metformin group were significantly lower than that of model group (P < 0.01). Moreover, the kidney weight/body weight of rats in sericin treatment group and sericin prevention group were obviously lower than that of metformin group (P < 0.05). CONCLUSION: Sericin can inhibit activation of TGF-beta1/Smad3 signal pathway in kidney of DN rats, lighten glomerulosclerosis and renal interstitial fibrosis, so has protective and preventive effects on kidney injury of DN rats. Moreover, the therapeutical and preventive effects of sericin on DN are similar with metformin.

To study the influence of resources thresholds on plant growth is a major theme in restoration ecology. Based on the simulation of the natural thresholds of soil moisture, nitrogen (N), and phosphorus (P) under drought condition in the arid valley of Mingjiang River, a full factorial experiment was designed to study the dynamics of Bauhinia faberi seedlings survival rate, growth, biomass production, and resources use efficiency across one growth season. High soil moisture (40% field water capacity), high soil P (24 mg P x kg(-1)), and low N (100 mg N x kg(-1)) increased the seedlings survival rate, and promoted the seedlings growth, biomass production, and water use efficiency. There was a significant coupling effect between soil N and P, but the interactions between soil moisture and soil N and P were not obvious. High N (240 mg N x kg(-1)) restrained the seedlings growth markedly, while high P mitigated the negative effects of high N via increasing root area, root length, and root mass to promote the seedlings N and P uptake. The N and P use efficiency across one growth season kept steady, and had significant positive correlation with root/shoot mass ratio. The combination of high soil moisture, low N, and high P promoted the seedlings growth effectively, while that of low soil moisture, low P, and high N inhibited the seedlings growth markedly.

RESUMO

In this text the interface of hardware and software for laboratory analyzer is analyzed. Adopting VC++ and multi-thread technique, the real-time communication without errors between LIS and the laboratory analyzer is realized. Practice proves that the system based on the technique is stable in running and all data are received accurately and timely.

Sediment gyration sampling was conducted in June 2006 to analyze the profile distribution of sediment particle sizes at a check-dam built in 1954 in a typical small watershed of hilly and gully Loess plateau. There were at least 28 layers of sediment gyration at the check-dam, with the layer depth varied from 2 cm to 60 cm. In the sediment profile, silt (0.05-0.002 mm) occupied > 60%, its particle size being mainly within the range of 0.01-0.05 mm, while coarse sand (> 0.05 mm) accounted for 23.09%. The variation degree of sediment particle sizes increased with increasing depth, and an abrupt change was observed at the depth 2 m. Both longitudinal gradient and hydrodynamic condition caused the variation of sediment particle sizes. There was an accumulated plough layer of 72 cm thickness in the profile, which had indicative role for the sediment erosion environment of check dam.

OBJECTIVE: To study scutellaria thermosensitive gel in situ. METHOD: The phase transition temperature of gel with various concentration polymer solutions was studied by using stirring method and the viscosity of gel was monitored under different temperatures. Eye irritation experiments were performed with rabbit. The duration of residence time in rabbit eyes of scutellaria thermosensitive gel was observed with 2% fluorescein. RESULT: 20% poloxmar407 and 10% poloxmar 188 were suitable to scutellaria thermosen-sitive gel in situ. The results suggested that the scutellaria thermosensitive gel in situ based on poloxmar were nonirritant and retention time on the surface of eye was (150 +/- 8) min. CONCLUSION: The scutellaria thermosensitive gel in situ forms gel in eye and has longer release time than that of eye drops.
