There is a gap in the understanding of the relationship between dietary phytate levels and the relative efficacy of phytase to improve amino acid (AA) digestibility in pigs and chickens. Two experiments were conducted to investigate the effect of exogenous phytase on standardized ileal digestibility (SID) of AA and the apparent ileal digestibility (AID) of P in both standard- (SP) and high-phytate (HP) diets for broilers and swine. There were either 40 cages of Cobb 500 male broilers or 10 crossbred barrows (35 kg) fitted with ileal T-cannulas. Both studies were allotted to five dietary treatments (8 replicates). Treatments consisted of four corn-soybean meal-based diets arranged in a 2 × 2 factorial of standard or high phytate and exogenous phytase at 0 or 1 000 phytase units (FYT)/kg; and one N-free diet. Birds were fed a common starter diet from d 0 to 20 and fed experimental diets from d 20 to 25. Birds were euthanized on d 25 via CO2 asphyxiation, and digesta were collected from the terminal ileum. Pigs were fed for a total of four 7-d periods, where digesta were collected on d 6 and 7 of each period. Diet and digesta samples were analyzed for DM, N, Ti, AA, and P to determine AA and P digestibility. The SID of AA was determined by correcting the AID of AA for the basal endogenous losses estimated using the N-free diet. Main effects of the diet type (standard or HP) and phytase (0 or 1 000 FYT/kg), and the interaction of diet type and phytase were evaluated. For both experiments, the HP diets produced lower SID of AA compared to the SP (P < 0.001). For broilers, there was a phytase effect (P < 0.001) for the SID of all AAs evaluated regardless of the diet type. For pigs, phytase improved (P < 0.05) the SID of Met, Lys, Cys, Glu and Ser and tended to improve (P < 0.10) Arg, Leu, Thr, and Tyr. There were no significant interactions for either experiment. For both experiments, AID of P was lower for the HP diets (P < 0.01), and phytase produced greater AID of P for both diet types (P < 0.01). These data indicate that phytase greatly improves the digestibility of P for broilers and pigs and has the ability to significantly increase the digestibility of amino acids for these animals, regardless of the dietary phytate P.

1. Two experiments were conducted to explore the effects of an exogenous sfericase protease on the apparent ileal nutrient digestibility of soybean meal (SBM) and rapeseed meal (RSM) in broiler chickens.2. In each experiment, a total of 256 sixteen-day-old male Cobb 500 broilers were fed one of four semi-purified experimental diets, comprising two different batches (A and B) of samples for either SBM (Exp. 1) or RSM (Exp. 2) without or with an exogenous sfericase (0 or 30,000 NFP/kg). Each experimental diet was fed to eight replicate pens of broiler chickens from 16 to 21 d of age (eight birds per cage), and ileal digesta were collected for measuring the digestibility coefficients.3. In Exp. 1, the amino acid digestibility was greater (P < 0.05) in SBM B compared with SBM A for Arg and Val, and a similar trend (P < 0.1) was observed for Tyr, Leu and Thr. Exogenous sfericase increased (P < 0.10) digestibility of most of amino acids except Gly, His and Trp. There was an interaction between SBM source and sfericase, whereby digestibility of P, N and Asp was increased by sfericase for SBM B but not for SBM A. In Exp. 2, there was no interaction (P > 0.05) between RSM source and sfericase for ileal nutrient digestibility. Digestibility was greater in RSM A compared to RSM B for all non-essential AA and most essential AA (except for Trp), while the reverse was noted for Ca and P (P < 0.05). Exogenous sfericase increased (P < 0.1) digestibility for all amino acids except Cys and Met.4. In conclusion, the current studies showed that both SBM and RSM batches influenced amino acid digestibility. Sfericase protease supplementation increased amino acid digestibility for both SBM and RSM. The digestibility effects were greater in the SBM batch with low digestibility for N and Asp which was in line with an increase in P digestibility.

The objective of these studies was to evaluate the impact of dietary muramidase (MUR) on endogenous amino acids (AA) losses and digestibility of nutrients in wheat and corn-based broiler diets. In experiment 1, the effect of dietary MUR on the flow of endogenous AA (EAA) at the jejunum and terminal ileum of broilers were assessed using either the nitrogen (N) free diet method (NFD) or the highly digestible protein diet method (HDP; 100 g casein/kg diet). Sialic acid and muramic acid concentrations were measured in the jejunal content. In experiment 2, a 2x2x2 factorial arrangement of treatments with 2 base grains (wheat or corn), with low or high metabolizable energy (ME) levels, and without or with MUR supplementation was implemented. All diets contained phytase, xylanase, and cellulase. Apparent ileal digestibility (AID) of dry matter (DM), protein (CP), amino acids (AA), crude fat, and energy, as well as the apparent total tract metabolizability (ATTM) of DM, CP, and gross energy (GE) were determined. The standardized ileal digestibility (SID) of AA was obtained by correcting AID values for basal ileal EAA obtained from chicks fed with NFD or HDP in experiment 1, jejunal EAA flow of all AA was higher (P < 0.001) compared to the ileum, but this effect was method dependent. Jejunal, but not ileal, EAA flow measured with HDP was higher compared to NFD, as well as sialic acid (P < 0.001) and muramic acid (P < 0.004) concentrations. Muramidase inclusion had no effect on basal EAA flow, independently of the segment and the method used. In experiment 2, dietary MUR supplementation increased the AID of CP (P < 0.05), all AA, and tended (P = 0.07) to increase the AID of GE, independently of the cereal type used. However, ATTM of DM and GE, but not CP, increased with MUR inclusion compared with the control treatments, especially in wheat and low ME diets (P < 0.05). In conclusion, MUR supplementation improved AID of CP and AA without affecting EAA losses and increases energy utilization.

An experiment was conducted to determine the standardized ileal digestible (SID) Ca requirement of Arbor Acres Plus male broilers from d 25 to 42 post-hatch. Broilers were obtained at hatch, placed in floor pens, and fed a nutrient adequate diet until d 24 post-hatch. On d 25, twelve hundred birds were weighed and allocated to one of 4 treatments. There were 25 birds per pen and 12 pens per diet. The diets were formulated to contain 0.46, 0.35, 0.24, or 0.13% SID Ca. Available P (avP) was 0.39% in all diets, including 0.16% avP expected from 2,500 FYT/kg of phytase. The SID Ca requirement was estimated using nonlinear models, including quadratic, straight broken-line, and quadratic broken-line. There was no effect of SID Ca on feed intake, body weight gain, feed conversion ratio, or livability. Tibia ash percent was greatest in birds fed 0.35% SID Ca and lowest in birds fed 0.13% SID Ca (quadratic, P = 0.063). Apparent ileal digestibility (AID) of Ca was highest in birds fed the diets containing 0.13% SID Ca and decreased (quadratic, P = 0.014) as dietary SID Ca increased to 0.46%. Apparent digested Ca was highest in birds fed 0.35% SID Ca and lowest in birds fed 0.13% SID Ca (quadratic, P = 0.005). Decreasing the concentration of SID Ca in the diet from 0.46 to 0.13% (P < 0.0001) increased the AID of P and apparent digested P. Litter N or P were lowest in birds fed 0.35% SID Ca and increased (quadratic, P ≤ 0.05) as dietary SID Ca decreased to 0.13%. Non-linear equations, developed using tibia ash percent, digested Ca, or litter P, estimate the SID Ca requirement of Arbor Acres Plus broilers from d 25 to 42 was 0.37, 0.35, or 0.35%, respectively. This corresponds to an SID Ca to available P ratio of 0.95 to 0.90.

An experiment was conducted to compare the use of a total and digestible Ca formulation system in broilers from hatch to d 37 post-hatch. Ross 308 (n = 288) male broilers were obtained and allocated to one of 2 dietary treatments in floor pens. There were 18 birds per pen and 8 replicate pens per diet. One diet was formulated using ingredient and nutrient recommendations for total Ca and the second diet was formulated using ingredient and nutrient recommendations for standardized ileal digestible (SID) Ca. All diets were formulated to be nutrient adequate using a 2-phase feeding program and including 0.19% available P (avP) and 0.209% total Ca or 0.073% digestible Ca from 1,000 FYT/kg of phytase. On d 17 and 37, tibias and ileal contents were obtained. From hatch to d 37, birds fed diets formulated using digestible Ca gained (P < 0.05) more and were more efficient (P < 0.05) compared with birds fed diets formulated using total Ca. There was no impact of formulation system on tibia ash or minerals. Litter pH (P < 0.05) was greater and litter dry matter (P < 0.05) was lower in birds fed the diets formulated using digestible Ca compared with those fed diets formulated using total Ca. Apparent ileal digestibility (AID) of Ca (P < 0.05), AID of P (P < 0.05) and digestible P intake (P < 0.05) were lower in birds fed diets formulated using digestible Ca compared with those formulated using total Ca at d 17 or 37. However, apparent ileal digested Ca and digestible Ca intake were not different between the experimental diets on d 17 or 37. In conclusion, formulating diets using digestible Ca improved weight gain and feed conversion ratio, but reduced the AID of Ca and P compared with birds fed diets formulated using total Ca. These findings might be reflective of the higher total Ca concentration in the diets formulated using digestible Ca compared with those formulated using total Ca.

Male broilers (n = 576) were obtained and allocated to 96 cages with 6 birds per cage. From hatch to d 24, 16 randomly allocated cages were fed a nutrient adequate reference diet. The reference diet was formulated to contain 0.96 and 0.87% total Ca from hatch to d 10 and d 11 to 24, respectively. The remaining 80 cages were fed a nutrient adequate diet, formulated to contain 0.50% standardized ileal digestible (SID) Ca from hatch to d 10. On d 11, five diets containing 0.56, 0.46, 0.36, 0.26, or 0.16% SID Ca were randomly assigned to 16 cages per diet. Means were separated using polynomial contrasts and Dunnett's Multiple Comparison tests. From hatch to d 10, birds fed diets formulated to contain 0.50% SID Ca gained more (P < 0.05) compared with birds fed the reference diet. From d 11 to 24, there was no effect of diet on feed intake or BW gain. Birds fed 0.46 or 0.16% SID Ca were less (P < 0.05) efficient compared with birds fed the reference diet. Birds fed 0.26 or 0.36% SID Ca were more efficient compared with birds fed all other levels of SID Ca (quadratic, P < 0.05). Tibia ash percent was greatest in birds fed 0.56 or 0.46% SID Ca, decreased (quadratic, P < 0.05) as dietary SID Ca decreased and was lower (P < 0.001) in birds fed the 0.26 or 0.16% SID Ca diets compared with birds fed the reference diet. Apparent ileal digestibility (AID) of Ca or retention of P was greater (P < 0.05) in birds fed the diets formulated using SID Ca compared with birds fed the reference diet. Apparent P retention was greatest in birds fed 0.36% SID Ca (quadratic, P < 0.05). Regression equations developed using P retention, AID of Ca, and percent tibia ash estimated the SID Ca requirement of 11 to 24-d-old broilers was 0.380, 0.488, and 0.515%, respectively. This corresponds to a SID Ca to available P ratio of 0.86 to 1.17.

Arbor Acres Plus male broilers (n = 1,152) were obtained at hatch and allocated to 1 of 6 diets from hatch to d 10 post-hatch. There were 16 replicate cages per diet and 12 birds per cage. Five of the diets were formulated to contain graded concentrations of standardized ileal digestible (SID) Ca at 0.60, 0.50, 0.40, 0.30, or 0.20%. A sixth reference diet was formulated using total Ca coefficients for each ingredient and contained 0.96% total Ca. Available P (avP) was maintained at 0.48% in all 6 diets. Data were subjected to an analysis of variance and the model included diet and block. Means were separated using contrasts to determine linear or quadratic effects of SID Ca and using the Dunnett's test to compare the reference diet to all SID Ca diets. There was no effect of graded levels of SID Ca on intake or gain. Birds fed diets containing 0.60, 0.50, 0.30, or 0.20% SID Ca ate (P < 0.05) or gained (P < 0.05) more compared with birds fed the reference diet. Mortality corrected FCR improved (linear, P < 0.05) as the SID Ca concentration in the diet increased from 0.20 to 0.60%. Tibia ash percent was greatest in birds fed 0.50% SID Ca and lowest in birds fed 0.20% SID Ca (quadratic, P < 0.05). Tibia ash percent was lower (P < 0.05) in birds fed diets formulated to contain 0.20% SID Ca compared with birds fed the reference diet. No other differences in tibia ash were reported. Apparent ileal digestibility (AID) or retention of P was greater (P < 0.05) in birds fed diets formulated using SID Ca compared with birds fed the reference diet. The AID of P increased (linear, P < 0.05) as the SID Ca content in the diet decreased from 0.60 to 0.20%. The AID or retention of Ca was similar in birds fed 0.60 or 0.50% SID Ca and increased as SID Ca decreased to 0.20% (quadratic, P < 0.05). Regression equations developed using bone ash and apparent P retention estimate the SID Ca requirement of Arbor Acres Plus broilers from hatch to d 10 post-hatch was 0.53 and 0.49%, respectively. This corresponds to a SID Ca to available P ratio of 1.1 to 1.02.

Starch is the main energy source in broiler diets. However, endogenous amylase secretion in young broilers is suboptimal to completely digest dietary starch, so exogenous α-amylase supplementation may help increase starch digestibility. The objective of this study was to assess the supplementation of increasing doses of an exogenous α-amylase (0, 40, 80, 120, and 160 kilo-novo α-amylase units (KNU)/kg) on corn and on a complete corn-soybean meal diet for 25-day-old broilers. Jejunal and ileal apparent digestibility coefficients of available starch, resistant starch, total starch, and DM, DM total tract retention, as well as dietary AME levels were evaluated. Interactions (P < 0.05) between diets and α-amylase showed that the enzyme had a more evident effect on increasing DM jejunal digestibility and AME on corn compared with the complete diet. Corn DM digestibility increased to a maximum of 67.84% with up to 47 KNU/kg, whereas 89 KNU/kg led to a maximum of 53.92% in the complete diet A maximum increase of 64 kcal AME/kg was obtained with 80 KNU/kg on the complete diet, whereas 109 KNU/kg generated 327 kcal AME/kg on corn (P < 0.05). Increasing the α-amylase dose linearly increased ileal digestibility of resistant starch (P < 0.05), and the effect on DM total tract retention was quadratic (P < 0.05). Corn showed a higher digestibility for DM, resistant and total starch, as well as DM total tract retention and AME, compared with the complete diet (P < 0.05). Treatments had no influence on available starch. The inclusion of exogenous α-amylase improves starch, DM, and energy utilization of corn-based and corn-soybean meal-based diets for broilers.

Dietary starch is the major energy source for broiler chickens; therefore, relevant information on its intestinal utilization is important. The present study was designed to evaluate intestinal starch and energy digestibility of broiler chickens fed diets supplemented with α-amylase. A total of 240 day-0 male broiler chicks were randomly assigned to 3 nutritionally adequate corn-soybean-based experimental diets comprising 3 levels of α-amylase supplementation (0, 80, or 160 KNU/kg diet). Each treatment comprised 8 replicate cages of 10 birds each. At day 21 after hatching, digesta was collected from 4 intestinal sites: the anterior jejunum (AJ), posterior jejunum (PJ), anterior ileum (AI), and posterior ileum. Increasing α-amylase supplementation linearly improved (P < 0.01) overall BW gain and feed efficiency of the birds. There were linear and quadratic (P < 0.01) responses of increasing α-amylase supplementation on starch and energy digestibility at the PJ and AI. The total tract digestibility of starch increased (P < 0.05) with increasing α-amylase supplementation. Starch disappearance and digestible energy (kcal/kg) linearly increased (P < 0.01) with digesta flow from the AJ to PJ as dietary α-amylase supplementation increased. There were linear (P < 0.01) and quadratic (P < 0.05) effects of increasing α-amylase supplementation on the villus height in the jejunum. The viscosity of the jejunal digesta decreased (P < 0.05) with increasing dietary α-amylase supplementation. The results from this study showed the efficacy of exogenous amylase in improving growth performance and starch and energy digestibility in broiler chickens. Furthermore, the digestibility of starch and energy and the impact of the exogenous amylase were higher at the PJ than other intestinal sites.

Growth performance, nutrient digestibility, intestinal health, and endogenous enzyme secretion responses to dietary α-amylase supplementation during 4 growth phases of broiler chickens fed corn-soybean meal-based diets were evaluated in the present study. A total of 1,136 male broiler chicks were assigned at day 0 after hatching to 8 treatments in a 2 × 4 factorial arrangement. There were 2 dietary levels of α-amylase supplementation of 0 or 80 kilo-Novo alpha amylase units per kg diet and 4 posthatching growth phases of day 0 to 11, day 11 to 21, day 21 to 42, or day 42 to 56 in a randomized complete block design. Each treatment comprised 8 replicate pens, with either 25 (day 0-11), 20 (day 11-21), 16 (day 21-42), or 10 (day 42-56) birds per pen. Body weight gain and feed efficiency of birds improved (P < 0.01) with α-amylase supplementation. There were main effects of α-amylase, growth phase, and interaction (P < 0.01) on apparent ileal digestibility (AID) of starch. This ranged from 0.8% during day 11 to 21 to 2.8% during day 0 to 11 after hatching. The total tract retention of starch increased (P < 0.05) with amylase supplementation but was not different across growth phases. Amylase supplementation increased (P < 0.05) AID of gross energy, AME (kcal/kg), and AMEn (kcal/kg). Villus height in the jejunal tissue was increased (P < 0.01) by α-amylase supplementation. During day 11 to 21 after hatching, the viscosity of jejunal digesta and pancreatic amylase activity increased (P < 0.01) with amylase supplementation. In conclusion, dietary amylase supplementation improved growth performance, apparent nutrient digestibility, and digestive enzyme activity of broiler chickens fed a corn-soybean diet. The study indicates that the growth phase of birds may affect response to exogenous amylase.

Trypsin inhibitors (TI) resident in soybeans affects protein utilization. While heat treatment influences residual TI, it simultaneously affects the structure and solubility of the soybean proteins and confounds any response to exogenous proteases. Using purified TI, the effect of exogenous protease to TI can be dissociated from changes in the soybean protein. Thus, the current study was designed to evaluate the growth performance and protein utilization responses of broiler chickens to purified TI and exogenous protease. Soybean meal (SBM) was preanalyzed for basal TI (2,996 TIU/g SBM), formulated into nutritionally adequate experimental diets to contain 1,033 TIU/g diet, and purified TI was added at 9,000 TIU/g diet. A total of 320 Cobb-500 broiler chicks were allocated to 4 diets, each with 8 replicate cages and 10 birds per replicate. The experimental diets were arranged as a 2 × 2 factorial with factors being dietary TI (1,033 or 10,033 TIU/g) and exogenous protease (0 or 15,000 PROT/kg). On day 7, 14, and 21 posthatching, protease supplementation improved the BW gain (P < 0.01) and gain to feed ratio (P < 0.05) of birds. On day 14 and 21 posthatching, the relative weight of pancreas increased (P < 0.05) with added TI but was reduced (P < 0.001) with protease supplementation. Apparent ileal digestibility of all amino acids, except methionine, decreased (P < 0.001) with added TI but increased (P < 0.05) with protease supplementation. Jejunal MUC-2 was downregulated (P < 0.01) and SCL7A-2 was upregulated (P < 0.05) by protease supplementation. Duodenal trypsin and chymotrypsin activities reduced (P < 0.05) with added TI but increased (P < 0.01) with protease supplementation. Exogenous protease produced longer villi (P < 0.05) and deeper crypts (P < 0.01) in the jejunal tissue. In conclusion, dietary addition of purified TI negatively affects nutrient utilization by broiler chickens. Furthermore, the study showed that the efficacy of the exogenous protease might be independent of dietary TI concentration.

A total of 468 male Ross 308 broilers were used in a digestibility study to determine the additivity of apparent or standardized amino acid (AA) digestibility values for corn, soybean meal (SBM), or a mixture of corn and SBM that were supplemented, or not, with either phytase, protease, or a combination of phytase and protease. These treatments generated a total of 12 experimental diets that were arranged in a 3 × 4 design. A nitrogen-free diet was also fed to estimate endogenous AA loss. Apparent and standardized AA digestibility values were assessed on day 28 posthatch. The apparent digestibility of AA in the complete diet was higher (P < 0.05) than expected based on the digestibility of the corn and SBM individually. However, this overestimation was corrected by the adjustment to standardized values. Importantly, addition of protease or the combination of protease and phytase increased (P < 0.05) the digestibility of AA in corn and SBM. Furthermore, these effects were arithmetically coherent with respect to the measured effects of the enzymes in the mixture of corn and SBM, even improving the additivity of AA digestibility values when assessed on an apparent basis. This study demonstrates that the effect of exogenous protease and phytase on AA digestibility in complete diets is predictable based on measurements made in individual ingredients. In addition to improving digestibility values per se, exogenous protease and phytase may enhance precision in least cost formulation systems.

A total of 72 male Ross 308 broilers were reared to day 34 on a standard wheat and soy-based diet and then offered one of the four semisynthetic experimental diets, comprising two different soybean meal sources either without or with exogenous protease (treatments therefore offered in a 2*2 factorial arrangement). Each experimental diet was fed to 18 individually housed birds from 34 to 37 D after which ileal digesta were collected and digestibility coefficients were calculated. The two soybean meal sources were found to be nutritionally divergent (P < 0.01), with one having the apparent ileal amino acid digestibility coefficient of 0.80 and the other 0.71. Exogenous protease increased (P < 0.01) apparent ileal amino acid digestibility coefficients from 0.74 to 0.77. There were no interactions between soybean meal origin and protease effect. On an individual bird level, there were substantial differences in the capacity to digest amino acids with the mean total amino acid digestibility coefficients from 0.54 to 0.80 for one of the soybean meal samples. Exogenous protease addition reduced the coefficient of variation for total amino acids from 11.4 to 9.1% in one soybean meal and from 7.7 to 6.3% in the other. It can be concluded that soybean meal digestibility varies and that some of this variance is associated with heterogeneity in the digestive capacity of broilers.

A total of 144 male Ross 308 broiler chickens were used in a digestibility bioassay to determine the additivity of apparent or standardized amino acid (AA) digestibility values for corn, soybean meal (SBM), or a mixture of corn and SBM. Following the receipt of a standard commercial starter diet from days 1 to 21, broilers were divided into 4 treatments (6 cages per treatment; 6 birds per cage) and received semi-purified diets based on corn, SBM, or a mixture of corn and SBM or a nitrogen-free diet (for the estimation of basal endogenous AA losses). Apparent and standardized ileal AA digestibility values were determined on day 28 and the measured values for the mixture of corn and SBM were statistically compared to calculated values based on the digestibility of the individual raw materials and their concentration in the mixed diet. The use of apparent ileal AA digestibility values for single ingredients resulted in an underestimation (P < 0.05) of the digestibility of the mixed feed for nitrogen, Lys, Arg, Thr, Asp, and Gly. Correction of apparent digestibility values to standardized values entirely corrected the underestimation in the mixed feed and resulted in a predictable linearity from single ingredients to the mixed diet. It can be concluded that standardized ileal AA digestibility values are more additive than apparent values and should be used, where possible, to enhance the accuracy of feed formulation and reduce both the diet cost and the environmental impact of nitrogenous pollution.

An experiment was carried out to evaluate the digestibility and metabolizability of corn and a corn/soy-based diet with the inclusion or not of xylanase and/or phytase in broilers. In the trial, 1,120 broiler chicks were distributed according to a completely randomized experimental design, consisting of 16 treatments, with 10 replicates of 7 birds each. Treatments were evaluated following a factorial arrangement (4 × 2 × 2), with 4 xylanase levels (zero, 50, 100, or 150 fungal ß-xylanase units/kg; FXU), 2 phytase levels (zero or 1,000 phytase units/kg; FTU), and 2 diets (corn/soy or pure corn). The same basal diets were fed from one to 14 d post hatch for all birds, after which the experimental diets were provided until d 25. All birds were euthanized on d 25 for collection of ileal contents. Samples of feed, excreta, and ileal digesta were analyzed for determination of apparent ileal digestibility. The effect of xylanase on the coefficient of apparent dry matter metabolizability and apparent metabolizable energy was increased by the presence of phytase in the complete diet but not in the diet based on pure corn resulting in a diet*phytase*xylanase interaction (P < 0.01; P < 0.001, respectivaly). Equivalent effects were observed for the apparent coefficient of ileal protein digestibility in which xylanase effects were potentiated by the presence of phytase only in the complete diet, resulting in a significant 3-way interaction. In corn there was a limitation in improving digestibility when we added increasing levels of xylanase with phytase. Otherwise in the corn/soy-based diets, the enzymes were potencialized when they were added together. The fact that the effect of xylanase was enhanced by the presence of phytase in complete diets but not in pure corn may be associated with differences in substrate (arabinoxylan and/or phytate) concentration and presentation, diet nutrient balance, or other factors. It can be concluded that the interactive effects of xylanase and phytase can be substantial but may depend on the characteristics of the diet fed.

A study was conducted to evaluate the effects of increased levels of a ß-xylanase on energy and nutrient utilization of broiler chickens fed corn-soy diets. A total of 480 slow feathering Cobb × Cobb 500 male broilers were randomly distributed to 10 treatments having 8 replicates of 6 birds each. Birds were fed a common starter diet to d 14 post hatch (3,050 kcal/kg AMEn, 21.7% CP, 1.05% Ca, and 0.53% nPP). The experimental diets were provided afterwards until 25 d. Two experimental diets, a conventional corn/soy-based basal diet (CS) and the basal diet in which 40% of the diet was displaced by corn (CN), were fed as-is or supplemented with 50, 100, 150, or 200 fungal ß-xylanase units (FXU)/kg. Dietary treatments were distributed factorially as a 2 × 5 arrangement. Samples of feed, excreta, and ileal digesta were analyzed for determination of ileal digestible energy (IDE), metabolizable energy, and total tract retention of protein and lipid. No interactions between diet and xylanase were observed. The CS diets had higher (P < 0.05) energy utilization and nutrient digestibility when compared to the CN diets. AMEn and IDE were improved (P < 0.05) by 192 and 145 kcal/kg, respectively, when diets were supplemented with 100 FXU/kg xylanase. The xylanase added to the CN diet led to quadratic increases (P < 0.05) in IDE (Y = - 0.014x(2) + 2.570x + 3,155; r(2) = 0.60) and in AMEn (Y = - 0.016x(2) + 3.982x + 3,155; r(2) = 0.68). Crude protein digestibility and AMEn were linearly increased (P < 0.05) when xylanase was added to the CN diet. In conclusion, energy utilization and digestibility of crude protein and dry matter increased with xylanase supplementation in corn/soy-based diets. When xylanase was tested in the CS diet, 92 and 124 FXU/kg maximized the energy release effect; however, the maximum energy response in the CN diet or corn was not achieved until 200 FXU/kg.

A study was conducted to evaluate the effects of dietary α-amylase and ß-xylanase supplementation of corn-soy diets, formulated with or without supplemental phytase, on growth performance, energy utilization, and starch digestibility in broiler chickens. A total of 336 slow-feathering, Cobb × Cobb 500 male broilers were randomly distributed to 6 treatments having 8 replicates of 7 birds each. Birds were fed a common starter diet to d 14 post-hatch (3,050 kcal/kg AMEn, 21.7% CP, 1.05% Ca, and 0.53% nPP). The experimental diets were provided afterwards until d 25. A 2 × 3 factorial arrangement of 2 control diets (basal = corn-soy diet without added phytase or PHY = corn-soy diet formulated with 1,000 phytase units/kg) and 3 carbohydrase supplementations (0, 80 kilo-Novo α-amylase units/kg, or 80 kilo-Novo α-amylase units/kg + 100 fungal ß-xylanase units/kg) was used from d 14 to 25. Excreta were collected from 21 to 24 d and all birds were euthanized at 25 d for jejunum and ileum content collection. Samples of feed, excreta, and jejunal and ileal digesta were analyzed for determination of total tract retention and ileal apparent digestibility. No interactions between diet and carbohydrase were observed. Broilers fed diets formulated with phytase or supplemented with amylase + xylanase had higher BW gain (BWG) and lower FCR (P < 0.05) when compared with birds fed diets without carbohydrases. Relative to the basal diet, AMEn was increased (P < 0.01) by 70 kcal/kg and 99 kcal/kg when birds were fed the diet supplemented with amylase and amylase + xylanase, respectively. Starch digestibility in the jejunum and ileum was increased (P < 0.05) by 3.5% and 2.4%, respectively, when birds were fed the diet supplemented with amylase + xylanase. Results from this experiment show that corn-soy diets having phytase and supplemented with amylase and xylanase led to increased growth performance, AMEn, and starch digestibility in broilers. Furthermore, the efficacy of exogenous amylase and xylanase was independent of the presence of microbial phytase.

Since it started as an organized economic activity, poultry production has been undergoing an evolution toward the optimization of its feed formulation features. Notably, advances in the knowledge of birds' nutrient requirements have allowed recommendations that are increasingly closer to the needs of the birds. Over time, availability of nutrients and energy has been incorporated into those recommendations, especially to compensate for the variability in the digestibility of nutrients originating from variable feed ingredient sources. Instead of using the total energy and nutrient content, current tables of nutrient recommendations provide an estimate of the digestible fractions of the nutrients in ingredients. For instance, nonphytate P is preferred instead of total P to account for the unavailable phytate P, and digestible amino acids to account for the differences in digestibility of amino acids in different ingredients, whereas energy is usually expressed as a proportion that has been digested and metabolized (AME). With the increasing interest in the use of exogenous enzymes in poultry feeds, special attention is directed to the feed substrates such that an added enzyme can match it, forming an enzyme-substrate complex that will be followed by a chemical reaction within the gastrointestinal tract. As a consequence of a degradation reaction, nutrients released can be absorbed and metabolized. In general, nutritional data banks used in linear feed formulation software have limited data on the proportions of fractions of ingredients that are indigestible. Therefore, estimations of the presence of many substrates in the feeds, and therefore the benefits of adding exogenous enzymes, are frequently limited because of the scarcity of adequate information. The objective of this review paper is to provide insights into the use of expanded nutrient databanks to include all the molecules considered potentially indigestible for poultry such that the inclusion of exogenous enzymes allows the estimation of the values of the product originated by their hydrolysis.

The use of exogenous enzymes in feeds for poultry has increased dramatically between 1990 and 2013. Today, the use of enzymes is broad, going beyond phytases and ß-glucanases and xylanases to include other carbohydrases and proteases as well as lipases. The number of scientific articles and publications related to enzymes in feed clearly shows that this has been an area of intense and broad interest for scientists and nutritionists. However, knowledge of the different substrates available in the feed and how these substrates change depending on feed ingredient selection has not received the same level of attention. Understanding substrates is key to better developing and implementing exogenous enzymes. Of importance today is to potentiate endogenous digestive capabilities and use exogenous enzymes to optimize nutrient digestion and use. Our aim with this symposium was to call attention to the importance of having a more in-depth knowledge about substrates and to fill the large gaps in our current understanding of the digestive processes in poultry.

A case is made for the application of true ileal amino acid digestibility and true ileal reactive lysine digestibility (lysine availability) in poultry nutrition. Technical aspects of the true ileal digestibility assays are reviewed, as are factors influencing amino acid digestibility in the broiler. There is considerable variation in amino acid digestibility and lysine availability both within and among diverse feedstuffs. Differences in mean amino acid digestibility among feedstuffs, and the variability in the digestibility of an amino acid within a feedstuff should both be taken into account during least-cost dietary formulation.
