RESUMO

The method for the extraction of native surface protein antigen with a mol. wt. of 92 kD from vegetative cells of B.anthracis STI-1 and its purification was developed. The antigen was extracted with 3% sodium lauryl sarcosylate at 4 degrees C from bacterial mass previously treated with 0.1 M tris-HCl buffer solution, pH 8.0 Purification was carried out by adsorption chromatography on hydroxylapatite. The isolated protein antigen with a mol. wt. of 92 kD was electrophoretically and immunochemically homogeneous. The study of the protective properties of this protein in combination with Freund's adjuvant, made on white mice, revealed that it had good prospects for use in the creation of chemical vaccines against the causative agent of anthrax.

RESUMO

In guinea pigs and noninbred white mice, infected subcutaneously with anthrax which resulted in their death, characteristic generalized infection with the hematogenic contamination of their organs and the signs of intoxication and shock could be observed. In inbred white rats (Fisher 344) the invasion and dissemination of B. anthracis are relatively slightly pronounced, the phenomena of intoxication and shock being clearly prevalent.

B. anthracis virulent and vaccine strains differ from the strains of species closely related to B. anthracis, such as B. cereus and B. thuringiensis, in their plasmid spectrum. The use of their plasmid spectrum. The use of the plasmid analysis of the strain is recommended for laboratory practice as the main differential diagnostic test.

RESUMO

The pathomorphological picture of experimental B. anthracis infection in white rats (strain Fisher-344) essentially corresponds to experimental anthracic intoxication with very moderately pronounced morphological manifestation of B. anthracis invasion. This indicates that specific anthracic intoxication is an essential component of the pathological process in B. anthracis infection.

In the process of batch cultivation in a synthetic medium B. anthracis cells actively consume free amino acids. The maximum amino acid consumption per unit of biomass occurs at the exponential phase of growth, but the consumption of serine is maintained at a high level also during the stationary phase. At the same time the consumption of proline by both vaccine and virulent strains is insignificant at the stationary phase of growth. In contrast to B. anthracis virulent strains, vaccine strains have been shown to consume no lysine, histidine, arginine and tryptophan.

A synthetic culture medium is proposed. This medium satisfies the nutritional requirements of most cultivated strains under study by a single amino acid: valine. The possibility of detecting strains with other nutritional requirements has been shown. The determination of LD50 in white mice has revealed no differences between the virulence of valine-dependent strains and that of strains with other nutritional requirements. During the batch cultivation of strain STI-1 in a liquid medium glucose shows the effect of substrate limitation and inhibition. The optimum concentration of glucose can be different, depending on the composition of the medium. The influence of different amino acids, the sources of carbon and energy, and nitrogen on the growth of the strain is evaluated.
