RESUMO

Coffea canephora plant stem cells can have bioactive compounds with tissue repairing and anti-inflammatory action. This study aimed to develop a liposomal stem cell extract formulation obtained from the leaves of C. canephora (LSCECC) and to investigate its capacity to contribute to the dynamic mechanisms of tissue repair. The liposome cream was developed and characterized through the dynamic light scattering technique, atomic force microscopy, and transmission electron microscopy. The excisional full-thickness skin wound model was used and daily topically treated with the LSCECC formulation or vehicle control. On days 2, 7, 14, and 21 after wounding, five rats from each group were euthanized and the rates of wound closure and re-epithelialization were evaluated using biochemical and histological tests. LSCECC resulted in faster re-epithelialization exhibiting a significant reduction in wound area of 36.4, 42.4, and 87.5% after 7, 10, and 14 days, respectively, when compared to vehicle control. LSCECC treated wounds exhibited an increase in granular tissue and a proper inflammatory response mediated by the reduction of pro-inflammatory cytokines like TNF-α and IL-6 and an increase of IL-10. Furthermore, wounds treated with LSCECC showed an increase in the deposition and organization of collagen fibers at the wound site and improved scar tissue quality due to the increase in transforming growth factor-beta and vascular endothelial growth factor. Our data showed that LSCECC improves wound healing, the formation of extracellular matrix, modulates inflammatory response, and promotes neovascularization being consider a promising bioactive extract to promote and support healthy skin. The graphical presents the action of LSCECC in all four phases of wound healing and tissue repair. The LSCECC can reduce the inflammatory infiltrate in the inflammatory phase by decreasing the pro-inflammatory cytokines like IL-6 and TNF-α, in addition to maintaining this modulation through lesser activation and recruitment of macrophages. The LSCECC can also increase the release of IL-10, an anti-inflammatory cytokine, decreasing local edema. The increase in VEGF provides neovascularization and the supply of nutrients to newly repaired tissue. Finally, signaling via TGF-ß increases the production and organization of collagen fibers in the remodeling phase.

The goal of the present study was to assess the viability of the cryopreserved ovine (Ovis aries) semen, upon supplementation with α-terpineol and rosemary essential oil (Rosmarinus officinalis). The collection of the semen from six rams formed the pool, collected once a week during 7 weeks. The diluted semen was packed into straws (0.25 ml) and frozen in a TK 3000® device. Both α-terpineol and rosemary essential oil were added in the concentrations of 6.25, 12.5 and 25 µg/ml to the TRIS-yolk extender forming six experimental groups; the control group received only the TRIS-yolk extender. The samples were analyzed after thawing regarding motility and vigor, integrity of the plasmatic membrane, thermoresistance test (TT), mitochondrial membrane potential, acrosomal integrity and computer-assisted semen analysis (CASA). The levels of the thiobarbituric acid reactive substances were also analyzed. According to the results obtained with the addition of the concentrations of 6.25, 12.5 and 25 µg/ml of α-terpineol, significantly reduced the parameters assessed through CASA (VSL, LIN and WOB) and TT. Rosemary essential oil did not have deleterious effects on the spermatozoa and did not reduce the oxidative stress in the concentrations studied, although it presented absolute values higher than those of the control in several parameters. Alpha-terpineol in the concentrations studied was not able to reduce the oxidative stress and had toxic effect over the ovine spermatozoa.

There is increasing evidence as to the participation of the ovarian renin-angiotensin system in important reproductive processes. The inhibition of the angiotensin-converting enzyme (ACE) caused an increase in the rate of ovulation and pregnancy in the artificial insemination protocol has fixed time (TFIA). This study aimed to investigate the presence and location of Ang II, Ang- (1-7) and ACE2 in goat ovaries and the possibility of the involvement of these peptides in previous results. Ten ovaries from goats were collected in a slaughterhouse, washed in buffered PBS, perfused with protease inhibitor solution and processed for immunohistochemistry protocol. The search for peptides was performed using the avidin–biotin–peroxidase method. A strong immunoreactivity for Ang II in theca cells of antral follicles and corpus luteum was observed. Antral follicles (theca cells), corpus luteum and oocyte cytoplasm in early antral follicles exhibited strong immunoreactivity for Ang-(1-7). There was strong immunoreactivity for ACE2 in the cytoplasm of luteal cells and theca cells of antral follicles. In this study, for the first time, the presence and location of Ang II, Ang-(1-7) and ACE2 are reported in goat ovary, suggesting that there is participation in follicular development, oocyte maturation and corpus luteum development.


Há evidências crescentes quanto à participação do sistema renina-angiotensina ovariano em processos reprodutivos importantes. A inibição da enzima conversora de angiotensina (ECA) ocasionou aumento na taxa de ovulação e gravidez no protocolo de inseminação artificial por tempo fixo (TFIA). Este estudo teve como objetivo investigar a presença e localização de Ang II, Ang-(1-7) e ECA2 em ovários de cabras e a possibilidade do envolvimento desses peptídeos em resultados anterio-res. Dez ovários de cabras foram coletados em abatedouro, lavados em PBS tamponado, perfundidos com solução inibidora de protease e processados para protocolo de imunohistoquímica. A busca por peptídeos foi realizada usando o método avidina-bio-tina-peroxidase. Foi observada uma forte imunorreatividade para Ang II em células da teca de folículos antrais e corpo lúteo. Os folículos antrais (células da teca), corpo lúteo e citoplasma do oócito nos folículos antrais iniciais exibiram forte imunor-reatividade para Ang-(1-7). Houve forte imunorreatividade para ECA2 no citoplasma das células luteais e células da teca dos folículos antrais. Neste estudo, pela primeira vez, a presença e localização de Ang II, Ang- (1-7) e ECA2 são relatadas em ovário caprino, sugerindo que há participação no desenvolvimento folicular, maturação oocitária e desenvolvimento do corpo lúteo.

The study aimed to investigate the chemical composition and the anti-inflammatory activity of the hydroethanolic rhizomes, stems, and leaf extracts of Renealmia petasites using in vitro and in vivo assays. The chemical composition of the extracts was characterized in a linear iron trap mass spectrometer. Total phenolic, flavonoid, and tannin content were determined by spectrophotometry analyses. In vitro anti-inflammatory activity was investigated in lipopolysaccharide-stimulated macrophages evaluating the influence on the production of superoxide anion (O2-), nitric oxide (NO), and the pro-inflammatory cytokines tumor necrosis factor (TNF-α) and interleukin-6 (IL-6). In vivo effects were determined using the air pouch model in which were inoculated carrageenan and thereafter treated with 50 mg/kg of the hydroethanolic extracts of R. petasites. After 4 and 24 h, the cellular influx, protein exudation, cytokines, and nitric oxide were evaluated. Eight compounds were tentatively identified in the R. petasites extracts, suggesting five diarylheptanoids, one flavonoid, and two fatty alcohols. The in vitro results showed that the extracts were capable of blocking free radicals and/or inhibiting their intracellular actions by inhibiting the production of important mediators of the inflammatory process, such as NO, O2-, TNF-α, and IL-6. In vivo, R. petasites significantly decrease the influx of leukocytes, mainly neutrophils, protein exudation, NO, TNF-α, and IL-6 concentration in the air pouch model. The results evidenced that R. petasites can be considered a promising alternative therapy for the treatment and management of osteoarthritis and other inflammatory diseases.

The objective of this study was to evaluate the behavioral and thermoregulatory characteristics of Dorper sheep. The experiment involved 12 adult females in which the respiratory frequency (RF), heart rate (HR), and rectal temperature (RT) were measured and the following behavioral activities were evaluated: Idling while standing, Idling while lying, Drinking, Ruminating while standing, Ruminating while lying, Defecating, Urinating, Walking, Vocalizing, Biting, Coughing, Playing, Fighting, Rubbing, Self-cleaning, and Sleeping. The physical analysis of the thermal environment in the facility was obtained at the meteorological station of the Technical School of Teresina, where the air temperature (AT) and air relative humidity (ARH) were recorded. The black globe humidity index was obtained using a thermometer inserted in a black globe. A significant difference was detected for AT and ARH, especially the former, which had a higher value in the afternoon period (36.5 ºC). A significant effect was observed for the physiological variables, for which the highest values were found in the afternoon period, as follows: 85.30 mov./min, 93.11 beats/min, and 39.51 ºC (RF, HR, and RT, respectively). Air temperature had positive correlations of 0.572, 0.516, and 0.165 with all thermoregulatory characteristics (RF, HR, and RT, respectively). There was a significant difference for the ‘Idling while standing’, ‘Idling while lying’, ‘Ruminating while lying’, ‘Vocalizing’, ‘Eating’, ‘Playing’, and ‘Sleeping’ behaviors in relation to the evaluated periods of the day. The animals showed a certain level of thermal discomfort, especially in the afternoon period.

The aim of this study was to estimate the variance components and genetic parameters for marbling in the ribeye area (MRA) and body condition score (BCS) using Bayesian inference via mixed linear and threshold animal models. Data were obtained from Santa Ines breed sheep reared in the Brazilian Mid-North region. Analyses considering the Monte Carlo methods were performed with Markov chains from 500000 cycles onward. A 200000-cycle initial burn-in was considered with values taken at every 250 cycles, in a total of 1200 samples. The Monte Carlo Error deviations were low for the means heritability in all chains by both linear and threshold models. Additive variances estimated by threshold model were higher than those estimated by the linear model. Marble meat from the ribeye area and body condition score can be used as selection criteria to obtain genetic progress in Santa Inês sheep. (AU)

Os dados foram obtidos em rebanhos de ovinos da raça Santa Inês criados no Meio-Norte do Brasil. As análises, considerando os métodos de Monte Carlo, foram realizadas com cadeias de Markov a partir de 500000 ciclos. Considerou-se burn-in inicial de 200000 ciclos com valores tomados a cada 250 ciclos, obtendo-se 1200 amostras. Os desvios do erro de Monte Carlo foram baixos para as herdabilidades médias em todas as cadeias, tanto pelo modelo linear quanto de limiar. As variâncias aditivas, estimadas pelo modelo de limiar, foram maiores que aquelas obtidas pelo modelo linear. This study was conducted to evaluate the effect of different concentrations of limonene (R)-(+) (50 µM,100 µM and 150 µM) in supplementing the diluter of bulls freezing semen. Thirty-six ejaculated from fourCurraleiro-Pé-Duro bulls were used for cryopreservation. The epifluorescence microscopy was used todetermine the plasmatic integrity and mitochondrial activity potential. No was observed effect on the integrity ofplasma membrane nor mitochondrial activity potential when cryopreserved by adding limonene R - (+). Theresults obtained in this study allow to conclude that supplementation limonene (R) - (+) on diluter of bullsfreezing semen does not interfere with the integrity of the plasma membrane or the potential of spermmitochondrial activity.

Objective to evaluate the effect of captopril in vitro production of bovine embryos. 472 COCs were usedfrom abattoir ovaries, which were screened, sorted and selected in grades I and II, and submitted to the PIVprocess. The IVM treatments were added in the following concentrations: T1 - control; T2 - 5 mM of captopril;T3 - 10 uM of captopril; and T4 - 15 captopril uM, the COCs were subjected to IVM, IVF CIV. The results showthat there was no statistically significant difference (P > 0.05) between treatments during maturation. Thevariables studied were cleavage and blastocyst rates. The captopril supplementation did not improve (P > 0.05)G1 cleavage rate - 61.84%; G2 - 71.00%; G3 - 68.87%; and G4 - 56.90%. Supplementation of 20μM ofcaptopril influenced the amount of viable embryos.

This study aimed to evaluate the effect of captopril on the in vitro maturation of oocytes. 1627 COCscattle slaughter houses were used, were subsequently screened, classified into grades I and II, according to themorphological quality, and submitted to the IVM process, distributed in four treatments: T1 - control; T2 - 5μMof captopril; T3 - 10μM of captopril; and T4 - 15μM of captopril, incubated in an incubator at 38.5 ° C with anatmosphere of 5% CO2. After 22 hours, the COCs were stripped to evaluate the rate of maturation, which wereas follows: T1 - 56.60% (120/212); T2 - 60.60% (240/396); T3 - 52.88% (174/329) and T4 - 58.30% (207/355).The results show that there was no statistically significant difference ( P> 0.05) between treatments duringmaturation, concluding that captopril supplementation did not affect the maturation of COCs.

This study was conducted to evaluate the effect of different plasminogen activator inhibitor 1concentrations (70 ƞg, 140 and 210 ƞg ƞg) in the kinetic parameters of sperm cryopreserved of Curraleiro FootHard bulls. Sperm kinetics were analyzed by means of a computer aided system (CASA). The variables evaluatedwere: (MT-%) (MOP-um / s), (VCL - um / s), (VSL - um / s), (VAP-um / s), (LIN -%) ( Str-%) (ALH - uM)Wobble (WOB -%) and (BCF-Hz). The cryopreserved sperm in the presence of the inhibitor of plasminogenactivator 1 in a concentration of 210 ƞg decreased velocity parameters in a straight line (VSL - um / s) andlinearity (LIN -%). In conclusion, supplementation of the Inhibitor of plasminogen activator 1 (PAI-1),cryopreservation of bovine semen does not improve the kinetic parameters as compared to the control.

RESUMO

The aim of this research was to evaluate the effects of supplementation of maturation medium withcaptopril in oocyte in vitro maturation (IVM). 470 CCOs were recovered and classified into grades I and II,being divided into four groups: G1 (n = 56), that was control group; G2 (n = 152) 20 mM of captopril; G3 (n =126) 40 uM of captopril; and G4 (n = 136) 80 uM of captopril, then these were subsequently submitted to IVMprocess for 24 hours at 38.5°C in 5% CO2. After 24 hours of maturation, oocytes were denuded and evaluatedfor the first polar body extrusion and were, therefore, considered matured. The addition of the Captopril inmedium of oocytes maturation has not improved the IVM rate.

This study was conducted to evaluate the effect of different concentrations of inhibitor antipain serineprotease (10 g, 50 ug and 100 ug) in supplementing the dilutive of bovine semen freezing. Thirty-six ejaculatedfour Curraleiro Pé-Duro cattle were used for cryopreservation. The epifluorescence microscopy was used todetermine the acrosomal integrity and acrosome reaction rate was calculated after sperm were incubated for sixhours heparin. Spermatozoa cryopreserved in the presence of antipain significantly increased acrosomalintegrity, as they were able to complete the acrosome reaction in vitro. In conclusion, the inhibitor is antipainserine protease is able to preserve the integrity of cryopreserved sperm acrosomal cattle.

This study aims to evaluate the effects of tea of copaíba extract (Copaifera luetzelburgii), in differentconcentrations, added to the Tris-egg yolk extender, the functionality of the plasma membrane sperm ofcryopreserved goats. Were used four goats breeds, adults, male, six of each breed, aged between 1 and 6 years,a pool of ejaculates were formed to analyze. The animals were randomly divided into four experimentaltreatments: TI without adding of tea of copaíba extract (control); TII, added 0,1 mg of tea of copaíba extract;TIII, 0,2 mg; and TIV, 0,5 mg. The samples were frozen and stored, after 30 days were thawed for analysis byhisposmotic test (HOST). Adding tea of copaíba extract showed no significant difference between treatments forHOST, so the use of tea of copaíba extract the seminal dilution was not effective in preserving the functionalityplasma membrane sperm of goats post-thawing.

The semen cryopreservation process causes damages in spermatozoa characteristics. This study aimedto evaluate the effects of tea of Copaíba extract (Copaifera luetzelburgii), in different concentrations, added tothe Tris-egg yolk extender, in the quality of post-cryopreserved spermatozoa from locally adapted breeds ofgoats. The animals were randomly dividedinto four experimental treatments: TI (Control), TII (0,1mg), TIII (0,2mg) and TIV (0,5mg). The samples werefrozen and stored, after 30 days they were thawed for analysis by term-resistance test (TTR). The addition of teaof copaíba extract showed no significant difference between treatments by TTR, therefore the use of tea copaíbaextract in seminal diluting not increment the sperm longevity post-thawing.

This work aimed to evaluate the quantitative parameters of fresh and thawed sperm of Canindé goats in theState of Piauí. Ten sperm samples were collected, using artificial vagina, from four goats at reproductive age. Eachejaculate was evaluated for volume, sperm concentration, mass motility, total and progressive motility, percentage ofmotile spermatozoa and vigor, for further dilution in ACP-101. Then, the sperm was filled in 0.25 mL straws,cryopreserved using the TK3000 machine and stored in nitrogen cylinders at -196 °C, for further analysis at SCA. Thesperm parameters were submitted to the T test for comparison between the fresh and thawed groups. The values forfresh sperm relative to total and progressive motility, vigor and sperm viability were higher (P < 0.05) whencompared to thawed, although both fresh and thawed sperm of these animals could be used in programs of artificial insemination (IA).

This study evaluated the quality of ejaculated from adapted breeds of goats as the morphologic aspects,functional and thermoresistance. An aliquot of the pool of each breed was used for morphological evaluation bywet preparation, quantifying sperm defects and dividing them into classes. The samples were cryopreservedusing TK3000 machine and evaluated by thermoresistance test by checking motility and sperm vigor every hour.For plasma membrane functionality analysis, was used hiposmotic test. There was no difference (P > 0.05)between the breeds in all sperm defects before and after cryopreservation, as well as for the thermoresistancetimes for motility and sperm vigor (P > 0.05). There was a significant reduction in motility and sperm vigor 120minutes after thawing of the samples. There was no difference (P > 0.05) between the percentage of sperm withfunctional membrane between the evaluated breeds. Therefore, the evaluated breeds have morphology,functionality and similar sperm thermoresistance.
