In recent years, some microorganisms have shown resistance to conventional treatments. Considering this increase in resistant pathogens, treatment alternatives are needed to promote greater treatment efficiency. In this sense, antimicrobial photodynamic therapy (aPDT) has been an alternative treatment. This technique uses a photosensitizer that is activated by light with a specific wavelength producing reactive species, leading to the death of pathogenic microorganisms. In this study, bacteriochlorophyll derivatives such as bacteriochlorin metoxi (Bchl-M) and bacteriochlorin trizma (Bchl-T) obtained from purple bacterium (Rhodopseudomonas faecalis), were evaluated as photosensitizers in the aPDT. Photodynamic inactivation (PDI) of the microorganisms Staphylococcus aureus, Micrococcus luteus, Candida albicans and Pseudomonas aeruginosa was investigated with both bacteriochlorins (Bchl-M and Bchl-T) at different concentrations (1, 15 and 30 µM for S. aureus; 1, 15, 30, 45, 60 and 75 µM for M. luteus; 30, 60, 90, 105, 120 and 150 µM for C. albicans; and 200 µM for P. aeruginosa) and different doses of light (20 and 30 J/cm2 for S. aureus and M. luteus; 30 and 45 J/cm2 for C. albicans; and 45 J/cm2 for P. aeruginosa) to inactivate them. Both photosensitizers showed good activation against S. aureus and for M. luteus, we observed the inactivation of these microorganisms at approximately 3 log, showing to be a good photosensitizers for these microorganisms.

This article describes the impregnation of copper nanoparticles (CuNP) in a polyester fibre filter that can be used in solid-gas filtration to retain the spread of pathogen microorganisms in indoor environments. The impregnation of the CuNP was achieved by spraying the suspension on the surface of filter media. An acid pretreatment was also evaluated to increase the adhesion between fibre and nanoparticle. The synthesis of the CuNP was done by chemical reduction. The bacterial effect was measured through the contact method for Escherichia coli and Staphylococcus aureus, and we demonstrate that the presence of CuNP to filter media reduced up to 99.99% of gram-negative and 99.98% of gram-positive bacteria. The pretreatment with HCl was a good alternative to filter modification due to the higher adhesion between CuNP and the fibre while the high efficiency against pathogen microorganisms was kept. The modification of filters with CuNP can improve the air quality of indoor environments, vanishing the pathogen microorganisms circulating in the air.

Inhalation of bioaerosols has been linked to many health problems. Filter media impregnated with antimicrobial material can provide effective removal and inactivation of bioaerosols. In this study, fibres were spun on a substrate by centrifugal spinning, obtaining filter media denoted 5THY and THY. Thyme essential oil was used as an antimicrobial agent. For 5THY, the thyme essential oil was added to the polyacrylonitrile (PAN) solution, while for THY, it was sprayed onto the medium after the fibres had been produced. The THY medium presented a higher collection efficiency, compared to the substrate or 5THY, with efficiencies of 99% for microparticles and 58% for nanoparticles. Using the plaque assay method, THY provided the highest reductions of the bacteria Escherichia coli and Staphylococcus aureus, with efficiency of 99.999%. The findings demonstrated that filter media covered with spun fibres and containing thyme essential oil provided excellent antimicrobial action and filtration performance.

Photodynamic therapy presents a therapeutic choice that can be utilized to treat diverse neoplasms. In this technique, the critical element is a photosensitive molecule that absorbs light energy and transfers it to molecular oxygen or biological molecules to form reactive oxygen species, thus inducing irreversible damage to target cells and ultimately leading to cell death. Bacteriochlorin derivatives are employed as photosensitizers (PSs), possessing light-absorbing capacity in the near-infrared region. The objective of this study was to prepare a semi-synthetic bacteriochlorin from Rhodopseudomonas faecalis and adding Trizma® to improve solubility. Cell viability tests, flow cytometry (apoptotic and necrotic cells were identified by Annexin V and propidium iodide), and confocal microscopy were used to evaluate the photoactivity of bacteriochlorin-Trizma (Bchl-T) in fibroblast (HFF-1-control cells) and breast cancer (MCF-7 cells-target cells) cells. At concentrations above 0.5 µM, Bchl-T demonstrated 80 % cell death, presenting the highest PS interaction (via fluorescence microscopy) with lysosomes, mitochondria, and the endoplasmic reticulum; the cell death type was revealed as apoptosis (via cytometry). Our findings indicated the suitability of Bchl-T for future application in photodynamic therapy against cancer cells by inducing apoptosis.

Antimicrobial treatment failure has been increasing at alarming rates. In this context, the bactericidal properties of biocompatible antimicrobial agents have been widely studied. F18 is a recently developed bioactive glass that presents a much wider working range when compared to other bioactive glasses, a feature that allows it to be used for coating metallic implants, sintering scaffolds or manufacturing fibers for wound healing applications. The aim of this study was to investigate the in vitro bactericidal and anti-biofilm activity of F18 glass as a powder and as a coating on steel samples, and to explore the effects of its dissolution products at concentrations from 3 mg/mL to 50 mg/mL against the Staphylococcus aureus and methicillin-resistant Staphylococcus aureus (MRSA) biofilms. Furthermore, we intend to verify whether changes in the medium pH could influence the bactericidal activity of F18. The results indicated that F18 presented bactericidal activity in preformed S. aureus and MRSA biofilms, reducing more than 6 logs of the viable cells that remained in contact with 50 mg/mL for 24 h. Moreover, an anti-biofilm activity was observed after 12 h of direct contact, with a drop of more than 6 logs of the viable bacterial population. Neutralization of the F18 solution pH decreased its bactericidal efficacy. These results indicate that the F18 glass could be considered as an alternative material for controlling and treating infections by S. aureus.

Crop-livestock system implies in the balance of appropriate animal foraging, minimizing effects on grain production on the same crop season. This study aims to assess canopy heights (20 and 30 cm) and cuts number (no cut, 1, 2 and 3 cuts) effects on wheat productivity and bromatological composition of forage, and grain yield of BRS Umbu and BRS Tarumã cultivars. Regardless cuts number and cultivars, 30 cm provided higher forage productivity (1032 and 1348 kg DM ha-1) than 20 cm (554 and 909 kg DM ha-1), for BRS Umbu and Tarumã, respectively, in average crop seasons. For both cultivars and crop seasons, mean values of 26% crude protein, 55% neutral detergent fiber and 26% acid detergent fiber were obtained. At 20 cm no difference in grain yield was observed as a function of cuts number with 3780 kg ha-1 for BRS Umbu, in average crop seasons and 4306 kg ha-1 in 2014, for BRS Tarumã. However, at 30 cm, the grain yield was penalized at least in 30% as a result only third cut. These results indicate that wheat as dual-purpose can be employed in southern Brazil in grazing systems in forage shortage times and then produce grains without yield penalties as a function to the management used.


A integração lavoura-pecuária implica no equilíbrio do forrageamento adequado dos animais, minimizando os impactos nas áreas agrícolas graníferas. Os objetivos deste trabalho foram verificar, em trigo, os efeitos das alturas pré-corte (20 e 30 cm) e do número de cortes (sem corte, 1, 2 e 3 cortes) sobre a produtividade e composição bromatológica da forragem, e a produtividade de grãos dos cultivares BRS Umbu e BRS Tarumã. Independente do número de cortes, a altura de 30 cm proporcionou maior produtividade de forragem (1032 e 1348 kg MS ha-1) do que 20 cm (554 e 909 kg MS ha-1), para os cultivares BRS Umbu e BRS Tarumã, respectivamente, na média das safras. Para ambos cultivares e safras, obteve-se valores médios de 26% de proteína bruta, 55% de fibra em detergente neutro e 26% de fibra em detergente ácido. A 20 cm não foram encontradas diferenças na produtividade de grãos em função do número de cortes, com 3780 kg ha-1 para BRS Umbu na média das safras e 4306 kg ha-1 em 2014 para BRS Tarumã. Porém, para ambos os cultivares a 30 cm, o rendimento de grãos foi prejudicado em 30% no terceiro corte. Estes resultados indicam que o trigo duplo-propósito pode ser empregado no sul do Brasil em sistemas de pastejo em épocas de escassez de forragem e ainda produzir grãos sem penalidades, de acordo com o manejo empregado.

Mixotrophy is a metabolic strategy in which an organism is autotrophic and heterotrophic simultaneously. Considering that the aquatic environment provides several organic sources of carbon, it is probably common for microalgae to perform mixotrophy and not only photoautotrophy, but little is known about microalgae mixotrophy. The present work aimed at investigating the growth, photosynthetic activity, morphology, and biochemical composition of the microalga Chlorella sorokiniana in mixotrophic and photo-mixotrophic conditions, comparing it with photoautotrophy. The results showed pH changes after glucose addition, reaching pH 11.62 in mixotrophic and 10.47 in sequential photo-mixotrophic cultures, which limited the microalgal growth. Highest biomass was obtained in the mixotrophic culture in comparison with the sequential photo-mixotrophic one. Rapid light saturation curves showed that α (photosynthetic efficiency, 1.69) and relative electron transport rate (rETR; 565.61) were higher in the mixotrophic cultures, whereas the highest Ik (irradiance saturation, 386.68) was obtained in the photoautotrophic ones. In the sequential photo-mixotrophic cultures, photosynthetic activity varied during glucose consumption, decreasing the maximum quantum yield Fv/Fm after glucose addition, indicating change in metabolism, from photoautotrophy to mixotrophy by the microalga. The results showed that the mixotrophic cultures had higher production of chlorophyll a (6.26 mg mL-1), cell density (6.62 × 107 cell mL-1), and lipids (0.06 pg µm-3). Sequential photo-mixotrophic cultures showed the highest biovolume (360.5 µm3 cell-1) and total carbohydrates (0.026 pg µm-3). The protein concentration was 3.2 and 2.4 times higher in photoautotrophy and photo-mixotrophic growth, respectively, than in mixotrophy, but lipids were three times higher under mixotrophy. The biochemical changes we observed indicate that the microalga's plasticity in face of new environmental characteristics, such as the presence of organic carbon, can change the flow of energy through natural ecosystems.

ABSTRACT: Root rot (RR), which can occur in different cropping systems, affects soybean seed yield. The aim of this study was to determine the incidence of RR in soybean crop systems and its relationship with soybean yield. The study was carried out for two growing seasons in a no-till system consisting of four soybean crop systems: soybean-oat+soybean-oat, soybean-maize+soybean-maize, soybean-wheat+soybean-wheat and soybean-pasture+soybean-pasture. Data were submitted to Pearson's correlation. Incidence of RR, mainly charcoal rot and sudden death, was more than 50% in soybean roots for all the soybean crop systems and consequently decreased yield by around 22kg ha-1 of seed for every 1% increase in RR. Root rot led to a decline in seed yield from 20 to 102kg ha-1 in the first season and 9 to 32kg ha-1 in the second season, considering all the cropping systems. Soybean + pasture was the lowest productive system, with a negative RR impact of 74.9 and 32.9kg ha-1 in the first and second season, respectively.


RESUMO: As podridões radiculares podem ocorrer em diferentes sistemas de cultivos e afetar a produtividade. Objetivou-se neste trabalho quantificar a incidência de podridões radiculares em distintos sistemas de sucessão de cultivos e sua relação com a produtividade de sementes soja. A pesquisa foi realizada por duas safras agrícolas, na Região de Ponte Serrada, Santa Catarina, em lavouras de sementes de soja cultivadas em plantio direto que foi subdividida em quatro sistemas de sucessão de cultivos: aveia-soja+aveia-soja, milho-soja+milho-soja, trigo-soja+trigo-soja e pasto-soja+pasto-soja. Os dados foram submetidos à correlação de Pearson e ao teste de médias de Duncan ou F (P<0,05). Constatou-se que a incidência de podridões radiculares, principalmente podridão cinzenta e morte súbita, na média dos sistemas, está acima de 50% e como consequência gerou redução de produtividade na ordem de 22kg ha-1 de semente de soja para cada 1% de aumento na incidência destas doenças. As podridões radiculares reduzem a produtividade de 20 a 102kg ha-1 na primeira safra e, de 9 a 32kg ha-1 na segunda safra, considerando todos os sistemas de cultivo. O sistema soja+pasto, é o menos produtivo com impacto negativo das PR de 74,9 e 32,9kg ha-1 na primeira e segunda safra, respectivamente.

Banana is the most consumed fruit in the world as it is a delicious and aromatic fruit. However, its production is limited mainly due post-harvest problems such as diseases, especially anthracnose, which accelerates senescence and causes early fall. The aim of this work was to evaluate the agronomic traits and the resistance of banana genotypes (Fhia 18, Fhia 01, Thap Mae, Caipira, Prata (control) and Maçã (control)) to the fungus Colletotrichum musae. The weight of bunch (Wb), fruit length (Fl), fruit diameter (Fd), the number of fruits per bunch (Nfc), number of hands per bunch (Nhb), yield (Yd), pH, total sugars (Brix) and finger detachment were evaluated. The AUDPC (area under the disease progress curve) showed that the genotypes Fhia 01, Maçã and Prata (15.77, 15.55 and 11.60 respectively) are more susceptible to the fungus C. musae than Caipira, Fhia 18 and Thap maeo (6.66, 7.29 and 8.76 respectively). The experiment with Fhias 18 and Thap Maeo genotypes showed lower finger detachment of 18 and 15%, and Thap maeo, Fhia18 and Caipira were resistant to C musae and presented the best agronomic characteristics.


A banana é fruta mais consumida no mundo, tradicionalmente exploradas em várias regiões do mundo, pois é uma fruta de muito saborosa e aromática. Contudo sua produção é limitada principalmente por problemas pós-colheita que mais acometem o fruto, dentre elas aparecimento de doenças principalmente a antracnose que acelera a senescência e o despencamento precoce. O objetivo desde trabalho foram avaliadas as características agronômicas e a resistência dos genótipos de banana Fhia 18, Fhia 01, Thap Mae, Caipira, Prata (controle) e Maçã (Controle), ao fungo Colletotrichum musae. A avaliação das características agronômicas como; peso do cacho (PC), comprimento dos frutos (CF), diâmetro do fruto (DF), número de frutos por cacho (NFC), número de pencas por cacho (NPC), Produtividade (Pd), pH, açúcares totais (Brix) e despencamento. A AACPD Área abaixo da curva da progressão da doença nos genótipos Fhia 01, Maçã e Prata (15,77, 15,55 e 11,60 respectivamente) mostraram se mais suscetível ao fungo C. musae que os demais genótipos Caipira, Fhia 18 e Thap maeo (6,66, 7,29 e 8,76 respectivamente), enquanto que para o experimento de despencamento genótipos Fhias 18 e Thap Maeo foram os que tiveram menor despencamento 18 e 15%, e a Os genótipos Thap maeo Fhia18 e Caipira foram resistentes a C musae e tiveram as melhores características agronômicas.

OBJECTIVE: The aim of this study was to investigate the effects of the antimicrobial photodynamic therapy (aPDT) using the association of curcumin with the surfactant sodium dodecyl sulfate (SDS) for oral decontamination in orthodontic patients. BACKGROUND DATA: The installation of the orthodontic appliances promotes an increase in the retentive area that is available for microbial aggregation and makes difficult the oral health promotion. However, aPDT is one possible approach that is used for the reduction of oral microbial load. MATERIALS AND METHODS: Twenty-four patients (n = 24) were randomly distributed into four groups: Light group: which was treated only with the blue light, no drug; PDT group, which was treated with curcumin and blue light; PDT + S group, which was treated with curcumin plus surfactant and irradiated with blue light; and Chlorhex group, which was treated with chlorhexidine. The photosensitizer agent was prepared by adding 0.1% of SDS to a curcumin solution of 1 g/L. Two distinct LED devices emitting blue light (450 ± 10 nm) were used as follows: extra-oral irradiation (200 mW, 80 mW/cm(2), 36 J and 14 J/cm(2)) and intra-oral irradiation (1200 mW, 472 mW/cm(2), 216 J and 85 J/cm(2)).The collection of nonstimulated saliva (n = 3; 3 mL/collection) was performed at the following steps: (1) immediately before swishing (curcumin, chlorhexidine, or water); (2) after swishing; and (3) after performing aPDT treatments. The colony-forming units (CFU) were counted visually, and the values were adjusted to CFU/mL. RESULTS: There was significant Log reduction for PDT (from 6.33 ± 0.92 to 5.78 ± 0.96, p < 0.05), PDT + S (from 5.44 ± 0.94 to 3.83 ± 0.71, p < 0.01), and Chlorhex (from 5.89 ± 0.97 to 2.55 ± 1.80, p < 0.01) groups. The survival rate was significantly reduced in both PDT + S and Chlorhex groups compared with all situations (p < 0.05). However, there was no significant difference between PDT + S and Chlorhex groups (p ≥ 0.05). CONCLUSIONS: These results indicate that when associated with the surfactant SDS, the aPDT can be used as an adjutant and a convenient agent to promote the oral decontamination in clinical practice.

RESUMO: A utilização de bioestimulantes pode auxiliar a mitigar os prejuízos ao milho, ocasionados pela má qualidade de estande. Este trabalho foi conduzido objetivando avaliar o efeito do tratamento de sementes com bioestimulante sobre o desempenho agronômico do milho, submetido a diferentes níveis de variabilidade na distribuição espacial das plantas na linha de semeadura. O experimento foi implantado em Lages, SC, durante o ano agrícola de 2013/2014. O delineamento experimental foi de blocos ao acaso, dispostos em parcelas subdivididas. Nas parcelas principais, testaram-se três níveis de desuniformidade espacial na linha: 0, 50 e 100% do Coeficiente de Variação (CV). No nível 0 % de CV, todas as sementes foram uniformemente distribuídas no sulco de semeadura, a uma distância de 17cm entre si. Nos demais níveis de CV, as distâncias entre sementes variaram conforme o tratamento. Nas subparcelas, testou-se a presença ou ausência do tratamento de sementes com bioestimulante. O aumento da variabilidade na distribuição espacial das plantas reduziu a área foliar e o teor de clorofila da folha índice no espigamento, bem como o diâmetro do colmo na colheita, independentemente do tratamento de sementes com bioestimulante. Isso contribuiu para reduzir o número de grãos por espiga e o rendimento de grãos dos tratamentos com 50 e 100% de CV na variabilidade espacial, tanto na presença quanto na ausência do bioestimulante. Portanto, o tratamento de sementes com bioestimulante não mitigou os prejuízos ao rendimento de grãos do milho ocasionados pela distribuição espacial irregular das plantas na linha de semeadura.


ABSTRACT: The use of biostimulants may help to mitigate damages to maize caused by poor stand quality. This research was carried out aiming to evaluate the effect of seed treatment with biostimulant on the agronomic performance of maize submitted to different levels of plant spatial distribution at the sowing row. The experiment was set in Lages, SC, during the 2013/2014 growing season. A randomized block design disposed in split plots was used. Three levels of plant spatial unevenness at the sowing row were tested in the main plots: 0, 50 and 100% of the variation coefficient (CV). At the level 0% of CV all seeds were evenly distributed with a distance of 17cm from each other. At the other levels of CV, the distance between seeds varied according to the treatment. The presence or absence of seed treatment with biostimulant was assessed in the split plots. The increase in plant spatial distribution variability at the sowing row decreased leaf area and leaf chlorophyll content at silking, as well as stem diameter at harvest, regardless of seed treatment with biostimulant. Such behavior contributed to decrease the number of kernels per ear and grain yield of treatments with 50 and 100% C.V. of spatial variability, with and without biostimulant seed treatment. Therefore, seed treatment with biostimulant did not mitigate damages to maize grain yield caused by irregular spatial plant distribution at sowing row.

The effective treatment of antimicrobial modalities continues to be a serious challenge, mainly due to the increasing number of multidrug resistance pathogenic microorganisms. Microbial bioinhibition is an alternative method that has shown to be effective. This study investigated and described the effect of the visible light on five different microorganisms. The studied groups were composed by the species Acanthamoeba polyphaga, Candida albicans, Mycobacterium massiliense, Pseudomonas aeruginosa, and Staphylococcus aureus. These microorganisms were analyzed after six light doses exposition with three different wavelengths: 450, 520, and 630 nm. The present study indicates two different behaviors: bioinhibition and/or biostimulation. The bioinhibition effect was calculated using different percentages of the microorganism population, compared to the control group, in which the maximum value corresponds to 94% growth inhibition. The biostimulation effect was evaluated by the microorganism population increment for specific light doses. Our results showed a 132% population growth as the maximum value. These results were assessed by variance analysis. The Tukey's test was used for differentiating or comparing, depending on the circumstances. The obtained results suggested a visible light phototherapeutic effect that could be used as a microorganism inactivation method for the studied microorganisms. In some approaches, the biostimulation effect might also be a very interesting effect to be considered. This study supports the relevance of understanding the important role that phototherapy plays as a useful method for microbiological control studies and applications.

Nitrogen and phosphorus present in sewage can be used for microalgae growth, possibiliting cost reduction in the production of microalgae at the same time that it decreases the eutrophication potential of the effluent. This research aimed at monitoring the native community of microalgae and coliform bacteria in a secondary effluent from anaerobic municipal sewage treatment. Two treatments (aerated and non-aerated) were performed to grow microalgae under semi-controlled conditions in semi-closed photobioreactors in a greenhouse. The results showed no significant pH and coliforms (total and Escherichia coli ) variation between treatments. Nutrient concentrations were reduced supporting microalgae growth up to 10 (7) cells.mL (-1) independent of aeration. Exponential growth was obtained from the first day for the non-aerated, but a 5 day lag phase of growth was obtained for the aerated. Chlorella vulgaris was the dominant microalgae (99.9%) in both treatments. In the aerated, 5 algae classes were detected (Chlorophyceae, Cyanophyceae, Chrysophyceae, Bacillariophyceae and Euglenophyceae), with 12 taxa, whereas in the non-aerated, 2 classes were identified (Chlorophyceae and Cyanophyceae), with 5 taxa. We concluded that effluent is viable for microalgae growth, especially Chlorella vulgaris, at the same time that the eutrophication potential and coliforms are decreased, contributing for better quality of the final effluent.

Objective: To describe the microorganisms present on surface areas of surgical rooms, in a medium-sized hospital in Sao Paulo state (Brazil). Materials and method: Sixty samples were collected with the aid of sterile swabs soaked in peptone water and rubbed into quadrants of 20 cm 2 . The surfaces investigated were: medication tables, surgical tables, marble countertops and air conditioning grilles. Results: Staphylococcus aureus , coagulase negative, was the microorganism most frequently found on the surgical tables and on the medication tables (50.7% of the samples). This microorganism is also the most frequent cause of post-surgical infection at the same hospital. Conclusions: Prophylactic measures should include proper hand washing, the use of personal protective equipment, appropriate uniforms, and cleaning and sterilization of surface and medical and hospital equipment.


Objetivo: Describir los microorganismos presentes en las superficies del área de quirófanos de un hospital de tamaño medio en el estado de Sao Paulo (Brasil). Materiales y métodos: Se recolectaron y cultivaron 60 muestras con la ayuda de hisopos estériles en agua peptonada y aplicadas sobre cuadrantes de 20 cm 2 . Las superficies investigadas fueron: tabla de medicamentos, mesa operatoria, terminaciones de mármol de la sala y grillas del aire acondicionado. Resultados: El organismo aislado de manera más frecuente fue Staphylococcus aureus coagulasa negativa y se encontró sobre la mesa operatoria y en la mesa de drogas (50,7% de las muestras). Este es el microorganismo reportado como la causa más frecuente de infecciones post-quirúrgicas en el mismo hospital. Conclusiones: Las medidas profilácticas deben incluir un apropiado lavado de manos, uso de equipo personal protector y limpieza y esterilización del equipo médico y hospitalario y de las superficies de trabajo.

The objectives of this study were to evaluate physical properties and antibacterial activity of a light-activated composite modified with silver nanoparticles. Discs were produced with unmodified resin (control group - CG) and modified resin with silver nanoparticles at two concentrations, 0.3% wt (MR03) and 0.6% wt (MR06). Streptococcus mutans and Lactobacillus acidophilus biofilms were induced in vitro by incubation of discs in a 20% sucrose medium, followed by sonication and counting of viable cells after 1, 4 and 7 days (n=9). The arithmetic roughness of all three groups was evaluated by atomic force microscopy (n=9). Compression assay was conducted in all groups to measure the compressive strength at failure and elasticity modulus (n=5). Data were subjected to ANOVA and Tukey's tests (α=0.05%). At all three time points the number of viable cells was statistically lower for MR03 and MR06 compared with CG, for both specimens. MR03 and MR06 showed no significant differences. Microscopic analysis demonstrated no significant differences for roughness among the three groups (p>0.05). The MR03 was stronger to compression than CG, and MR06 was statistically lower than CG and MR03. It was concluded that the MR03 were less conducive to biofilm growth, without compromising the strength in compression and surface roughness.

Os objetivos do estudo foram avaliar as propriedades físicas e a atividade antibacteriana de uma resina composta fotopolimerizável modificada com nanopartículas de prata. Discos foram produzidos com resina não modificada (grupo controle - GC) e resina modificada com nanopartículas de prata em duas concentrações 0,3 e 0,6% em massa (MR03 e MR06, respectivamente). Biofilmes de Streptococcus mutans e Lactobacillus acidophilus foram induzidos in vitro pela incubação dos discos em meio líquido com 20% de sacarose, seguida pela sonicação e contagem de células viáveis após 1, 4 e 7 dias. A rugosidade aritmética de todos os grupos foi avaliada por microscopia de força atômica (MFA). Os dados foram submetidos à ANOVA e ao Teste de Tukey (α=0,05%). Nos três períodos de tempo, o número de células viáveis foi estatisticamente mais baixo para MR03 e MR06, em comparação com GC, para ambas as espécies. MR03 e MR06 não apresentaram diferenças significantes. A análise por meio de MFA demonstrou que não houve diferença significante para a rugosidade entre os três grupos. MR03 apresentouse mais resistente em compressão do que GC, e o desempenho em compressão em MR06 foi estatisticamente pior do que em GC e MR03. Foi possível concluír que as resinas modificadas de MR03 inibiram o crescimento dos biofilmes, sem comprometimento da resistência à compressão e da rugosidade superficial.

Biofilms are less sensible to antibiotics and therefore are more difficult to eradicate. Linezolid and N-acetylcysteine (NAC), have demonstrated to be active against gram-positive microorganisms. Therefore and since linezolid and NAC have different modes of action, the main objective of this work was to investigate the single and synergistic effect of linezolid and NAC against S. epidermidis biofilms. Methods: This work reports the in vitro effect of linezolid and NAC against S. epidermidis biofilms, treated with MIC (4 mgml−1) and 10×MIC of NAC, and MIC (1 μgml−1) and peak serum concentration (PS = 18μgml−1) of linezolid alone and in combination. After exposure of S. epidermidis biofilms to linezolid and/or NAC for 24 h, several biofilm parameters were evaluated, namely the number of cultivable cells [colony forming unit (CFU) enumeration], total biofilm biomass and cellular activity. Results: When tested alone, NAC at 10 × MIC was the most effective agent against S. epidermidis biofilms. However, the combination linezolid (MIC) + NAC (10×MIC) showed a synergistic effect and was the most biocidal treatment tested, promoting a 5 log reduction in the number of biofilm viable cells. Conclusion: This combination seems to be a potential candidate to combat infections caused by S. epidermidis biofilms, namely as a catheter lock solution therapy


Introducción: Staphylococcus epidermidis es un organismo comúnmente asociado con infecciones causadas por biofilms. Los biofilms son menos sensibles a los antibióticos y, por lo tanto, más difíciles de erradicar. El linezolid y la N-acetilcisteína (NAC) han demostrado ser activos contra los microorganismos grampositivos. Por lo tanto, y puesto que el linezolid y la NAC tienen diferentes modos de acción, el objetivo principal de este trabajo fue investigar el efecto individual y sinérgico del linezolid y la NAC frente a biofilms de S. epidermidis. Métodos: Este trabajo reporta el efecto in vitro del linezolid y la NAC solos y en combinación contra biofilms de S. epidermidis, tratados con NAC en las concentraciones de MIC (4mg·ml–1) y 10×MIC, y linezolid en las concentraciones MIC (1mg·ml–1) y concentración sérica máxima (PS = 18μg ml–1). Después de la exposición de los biofilms de S. epidermidis al linezolid y/o la NAC durante 24h, fueron evaluados varios parámetros del biofilm, a saber, el número de células cultivables (recuento de unidades formadoras de colonias [UFC]), la biomasa total del biofilm y la actividad celular. Resultados: Durante el ensayo solo, la NAC en la concentración de 10×MIC fue el agente más eficaz contra biofilms de S. epidermidis. Sin embargo, la combinación linezolid (MIC)+NAC (10×MIC) mostró un efecto sinérgico y fue el tratamiento con mayor efecto biocida, promoviendo una reducción logarítmica de 5 en el número de células viables del biofilm. Conclusión: Esta combinación parece ser un potencial candidato en el combate de las infecciones causadas por biofilms de S. epidermidis, es decir, como una terapia de solución de bloqueo del catéter

INTRODUCTION: The ability of staphylococci to produce biofilm is an important virulence mechanism that allows bacteria both to adhere and to live on artificial surfaces and to resist to the host immune factors and antibiotics. Staphylococcal infections have become increasingly difficult to treat due their antibiotic resistance. Therefore, there is a continuous need for new and effective treatment alternatives against staphylococcal infections. The main goal of this study was to test N-acetylcysteine (NAC) and vancomycin alone and in combination against S. epidermidis and S. aureus biofilms. METHODS: Biofilms were treated with NAC at minimum inhibitory concentration (MIC) and 10 × MIC concentrations and vancomycin at MIC and peak serum concentrations. RESULTS: The use of NAC 10 × MIC alone showed a significant antibactericidal effect, promoting a 4-5 log10 CFU/ mL reduction in biofilm cells. The combination of NAC 10 × MIC with vancomycin (independently of the concentration used) reduced significantly the number of biofilm cells for all strains evaluated (5-6 log10). CONCLUSION: N-acetylcysteine associated to vancomycin can be a potential therapeutic strategy in the treatment of infections associated to biofilms of S. epidermidis or S. aureus.
