RESUMO

Schistosomiasis remains a serious public health problem in tropical regions, affecting more than 250 million people. Sensitive diagnostic methods represent key tools for disease elimination, in particular in areas with low endemicity. Advances in the use of luminol-based chemiluminescent techniques have enabled greater sensitivity and speed in obtaining results in different diagnostic settings. In this study, we developed a luminol-H2O2 chemiluminescence (CL) method to detect Schistosoma mansoni eggs in human fecal sediments processed by the Helmintex (HTX) method. After S. mansoni eggs were incubated with a solution of luminol-H2O2 the light emission was detected and measured by spectrophotometry at 431 nm for 5 min, using detection and counts of eggs by bright field optical microscopy as a reference. CL intensity was found to correlate with different sources and numbers of eggs. Furthermore, our results showed that the CL method can distinguish positive from negative samples with 100% sensitivity and 71% specificity. To our knowledge, this is the first study to report the use of CL for the diagnosis of helminths from fecal samples. The combination of the HTX method with CL represents an important advance in providing a reference method with the highest standards of sensitivity.

BACKGROUND: Stimulation of plant growth by low doses of a toxic compound is defined as a hormetic effect. Exposure of plants to low doses of glyphosate can cause stimulatory effects on growth or other variables. Sugarcane is the major biofuel and sugar-production crop cultivated in Brazil, but its expansion to new areas is limited; therefore, there is a demand for new technologies to improve sugarcane production per unit area. The use of pesticides to stimulate growth through the hormetic effect might be a suitable strategy to increase sugarcane yields. The purpose of this research was to investigate the effect of a low dose of glyphosate on metabolic compound accumulation, leaf phosphorus (P) concentration, and morphological variables across a one-year sugarcane cycle, as well as to determine whether the glyphosate effect was sustained and effective in improving the yield and technological quality of the sugarcane at harvest. RESULTS: The application of a low dose of glyphosate led to higher concentrations of shikimic acid and quinic acid, higher leaf P concentrations, and improved plant growth, yield, and technological quality of the sugarcane, by increasing the Brix% juice, pol% cane, total recoverable sugar, tons of culms per hectare, and tons of pol per hectare, relative to the results for an untreated control. CONCLUSIONS: The increased growth stimuli, observed through several variables, promoted an improvement in sugarcane yield. Therefore, the application of a low dose of glyphosate to sugarcane is a promising practice for crop management. © 2020 Society of Chemical Industry.

The impact of the kinetics of the anti-HLA antibodies after KTx on the occurrence of acute rejection as well as the better time-point to monitor anti-HLA Abs after transplantation is not completely defined. This prospective study followed 150 patients over 12 months after transplantation. Serum IgG anti-HLA Abs were detected by single antigen beads after typing donors and recipients for loci A, B, C, DR, and DQ. Before KTx, 89 patients did not present anti-HLA Abs and 2% developed "de novo" Abs during the 1st year, 39 patients were sensitized without DSAs, and 13% developed DSA after surgery; all of them presented ABMR. Sensitized patients presented higher acute rejection rates (36.4% versus 13.5%, p < 0.001), although 60% of the patients did not present ABMR. Patients, in whom DSA-MFI decreased during the first two weeks after surgery, did not develop ABMR. Those who sustained their levels presented a rate of 22% of ABMR. 85% of patients developed ABMR when MFIs increased early after transplantation (which occurred in 30% of the DSA positive patients). In the ABMR group, we observed an iDSA-MFI sharp drop on the fourth day and then an increase between the 7th and 14th POD, which suggests DSA should be monitored at this moment in sensitized patients for better ABMR prediction.

This paper pretends to demonstrate the effect of the combination of transversus abdominis plane block (TAP block) and Serratus plane block (SP block) techniques in analgesia of 4 dogs undergoing total unilateral mastectomy. Dogs were premedicated with methadone (0.5mg.kg-1) intramuscularly. Anesthesia was induced with propofol (6mg.kg-1) and midazolam (0.3mg.kg-1) and maintained with isoflurane. SP and TAP block were performed unilaterally using ultrasound by the injection of bupivacaine 0.25% (0.3mL kg-1) diluted with NaCl solution 1:1. Heart rate (HR), respiratory rate (f), non-invasive arterial pressure, esophageal temperature (T), oxygen saturation (SpO2) and electrocardiogram were monitored continuously. Animals were monitored for two and four hours after extubation for pain by using the Canine Acute Pain Scale from Colorado State University. Two hours after extubation, tramadol (4mg.kg-1) and dipyrone (25mg.kg-1) was administered to all dogs. It was not observed any alteration on cardiac rhythm. HR, f, T and mean arterial pressure remained below the preincisional values for all dogs. No dog required intraoperative rescue analgesia. Recovery from anesthesia was without any complication. All animals scored 0 (0/5) at pain scale, two and four hours after extubation and none of them expressed concern over the surgical wound. Dogs were able to walk before two hours after extubation. The combination of both techniques is effective in anesthetic blocking the thoracic and abdominal walls and it is suggested both may be included in the multimodal analgesia protocols for this type of surgery.(AU)


Este trabalho pretende demonstrar o efeito analgésico da combinação das técnicas de bloqueio do plano transverso abdominal (TAP block) e bloqueio do plano serrátil (SP block) em 4 cadelas submetidas à mastectomia unilateral total. Os animais foram pré-medicados com metadona (0,5mg.kg-1) por via intramuscular. A anestesia foi induzida com propofol (6mg.kg-1) e midazolam (0,3mg.kg-1) e mantida com isoflurano. Os bloqueios SP e TAP foram realizados unilateralmente, utilizando ultrassonografia, pela injeção de bupivacaína a 0,25% (0,3mL.kg-1), diluída com solução de NaCl a 1:1. A frequência cardíaca (FC), frequência respiratória (f), pressão arterial não invasiva, temperatura esofágica (T), saturação de oxigênio (SpO2) e eletrocardiograma foram monitorados continuamente. Os animais foram monitorizados durante duras e quatro horas após a extubação para a dor usando a Escala de Dor Aguda Canina da Universidade Estadual do Colorado. Duas horas após a extubação, tramadol (4mg.kg-1) e dipirona (25mg.kg-1) foram administrados a todos os cães. Não foi observada qualquer alteração no ritmo cardíaco. HR, f, T e pressão arterial média permaneceram abaixo dos valores basais para todos os cães. Nenhum cão requereu resgate analgésico intra-operatório. Não houve complicações na recuperação anestésica. Todos os animais apresentaram escore 0 (0/5) na escala de dor, duras e 4 quatro horas após a extubação e nenhum expressou desconforto com a ferida cirúrgica. Todos os cães foram capazes de caminhar antes de duas horas após extubação. A combinação de ambas as técnicas é eficaz no bloqueio anestésico das paredes torácica e abdominal e sugere-se que ambos podem ser incluídos nos protocolos de analgesia multimodal para este tipo de cirurgia.(AU)

The photocatalytic degradation of textile effluent was investigated using TiO2, ZnO, and Nb2O5 catalysts under solar irradiation. The procedures were carried out at ambient conditions in April 2014, with pH 3.0 and catalyst concentration of 0.250 g L-1. The photocatalytic activity of the oxides was evaluated by means of kinetic efficiency (rate constant and half-life time), chemical oxygen demand reduction, and absorbance reduction at 228, 254, 284, 310, 350, 500, and 660 nm (λmáx). Mineralization in terms of the formation of inorganic ions and toxicity reduction using bioassays with Artemia salina were performed. TiO2 reduced the absorbance at 660 nm (λmax) after 300 min of solar irradiation around 94 and 93%; and 68 and 60% of COD, respectively. ZnO showed lower photocatalytic activity giving 64 and 42% of absorbance and COD reduction, respectively. The photocatalytic activity of Nb2O5 was very close to TiO2-P25. In this sense, Nb2O5 becomes a promising alternative to replace the commercial TiO2-P25. Bioassays confirmed the efficacy of treatment, increasing the lethal concentration of 27.59 (in natura) to 131.95% in the presence of Nb2O5.

This study reports on the investigation of the photodegradation of biodiesel (B100) in contact with water using TiO2/H2O2. The TiO2 was characterized by X-ray diffraction analysis (XRD), pH point of zero charge (pHpzc) and textural analysis. The results of the experiments were fitted to a quadratic polynomial model developed using response surface methodology (RSM) to optimize the parameters. Using the three factors, three levels, and the Box-Behnken design of experiment technique, 15 sets of experiments were designed considering the effective ranges of the influential parameters. The responses of those parameters were optimized using computational techniques. After 24h of irradiation under an Hg vapor lamp, removal of 22.0% of the oils and greases (OG) and a 33.54% reduction in the total of fatty acid methyl ester (FAME) concentration was observed in the aqueous phase, as determined using gas chromatography coupled with flame ionization detection (GC/FID). The estimate of FAMEs undergo base-catalyzed hydrolysis is at least 3years (1095days) and after photocatalytic treatment using TiO2/H2O2, it was reduced to 33.54% of FAMEs in only 1day.
