RESUMO

The fermentation and recovery of the EcoRl restriction endonuclease with a genetically modified Escherichia coli strain is investigated. Vast amounts of product could be obtained after cultivation in a 20-L computer-coupled pilot fermentor and purification of the recovered wet cells. It was found that in the end the product is at least inhibitory and probably lethal to the cells (the lethality has been proven with genetic experiments) so that optimum yield requires an optimized choice for the time instant of induction. Growth after induction and product formation require substantial amounts of oxygen, which must be supplied if a high population level is to be achieved. pH control may alleviate the burden of high oxygen supply. Quantitative assessment after the different purification stages indicate that approximately 15% active enzyme can be obtained from the total amount produced.

The role of computers in the monitoring and control of fermentation processes has increased steadfastly. The ultimate utility of the machines will not depend on the availability of online sensors but also on the availability of techniques that combine direct measurements, leading towards estimates of variable closely related to the microbial process or its control. In this article, a methodology for on-line and noninterfering evaluation of the volumetric mass-transfer coefficient K(l)a is developed. A detailed presentation of the procedure, called "the static method," is given. Its feasibility is proved through implementation of the method on an antibiotic fermentation process. These experiments indicate that operator actions meant to modify the oxygen-transfer conditions can be checked on-line. The quantitative value of the static method is ascertained by comparing the experimental results with K(l)a estimates obtained with the "gassing-out" method. A sensitivity analysis was carried out, revealing the need for temperature and pressure corrections and showing that the precision of the oxygen analyzer determines the precision of the static method.

RESUMO

Sister chromatids of human metaphase chromsomes from cells which have replicated twice in medium containing 5-bromodeoxyuridine exhibit unequal fluorescence when stained with the dye 33258 Hoechst. Sister chromatid exchanges occurring in these chromosomes are apparent as interchanges of brightly and dully fluorescing chromatids. A technique for detecting such exchanges by computer analysis of chromsome images has been developed and found to campare favorably with manual methods. The exchanges have been localized in the context of quinacrine banding patterns.
